The asialoglycoprotein (ASGP) receptor is a wellcharacterized hepatic receptor that is recycled via the common cellular process of receptor-mediated endocytosis (RME). The RME process plays an integral part in the proper traff icking and routing of receptors and ligands in the healthy cell. Thus, the missorting or altered transport of proteins during RME is thought to play a role in several diseases associated with hepatocyte and liver dysfunction. Previously, we examined in detail alterations that occur in hepatocellular RME and associated receptor functions as a result of one particular liver injury, alcoholic liver disease (AlD). The studies revealed profound ethanolmediated impairments to the ASGP receptor and the RME process, indicating the importance of this receptor and the maintenance of proper endocytic events in normal tissue. To further clarify these observations, studies were performed utilizing knockout mice (lacking a functional ASGP receptor) to which were administered several liver toxicants. In addition to alcohol, we examined the effects following administration of antiFas (CD95) antibody, carbon tetrachloride (CCl4) and lipopolysaccharide (lPS)/galactosamine. The results of these studies demonstrated that the knockout mice sustained enhanced liver injury in response to all of the treatments, as shown by increased indices of liver damage, such as enhancement of serum enzyme levels, histopathological scores, as well as hepatocellular death. Overall, the work completed to date suggests a possible link between hepatic receptors and liver injury. In particular, adequate function and content of the ASGP receptor may provide protection against various toxinmediated liver diseases.

The feed additive ractopamine hydrochloride was fortified at four concentrations into batch vials containing soils that differed in both biological activity and organic matter（OM）.Sampling of the liquid layer for 14 days demonstrated that ractopamine rapidly dissipated from the liquid layer. less than 20% of the fortified dose remained in the liquid layer after 4 hr,and recoveries of dosed ractopamine ranged from 8 to 18% in the liquid layer at 336 hr. Sorption to soil was the major fate for ractopamine in soil：water systems, i.e., 42%–51% of the dose at14 days. The major portion of the sorbed fraction was comprised of non-extractables; a smaller fraction of the sorbed dose was extracted into water and acetone, portions which would be potentially mobile in the environment. Partitioning coefficients for all soils suggested strong sorption of ractopamine to soil which is governed by hydrophobic interactions and cation exchange complexes within the soil OM. Ractopamine degradation was observed, but to mostly non-polar compounds which had a higher potential than ractopamine to sorb to soil. The formation of volatiles was also suggested. Therefore, despite rapid and extensive soil sorption,these studies indicated a portion of ractopamine, present in manures used to fertilize soils,may be mobile in the environment via water-borne events.

AIM:To investigate the association of variations in the cyclooxygenase-2(COX2) and uridine diphosphate glucuronosyltransferase 1A6(UGT1A6) genes and non-steroidal anti-inflammatory drugs(NSAIDs) use with risk of colon ***:NSAIDs,which are known to reduce the risk of colon cancer,act directly on COX2 and reduce its *** studies have associated variations in the COX2 gene with colon cancer risk,but others were unable to replicate this ***,enzymes in the UGT1A6 gene have been demonstrated to modify the therapeutic effect of NSAIDs on colon *** in the UGT1A6 gene have been statistically shown to interact with NSAID intake to influence risk of developing colon adenomas,but not colon *** we examined the association of tagging single nucleotide polymorphisms(SNPs) in the COX2 and UGT1A6 genes,and their interaction with NSAID consumption,on risk of colon cancer in a population of 422 colon cancer cases and 481 population ***:No SNP in either gene was individually statistically significantly associated with colon cancer,nor did they statistically significantly change the protective effect of NSAID consumption in our *** others,we were unable to replicate the association of variants in the COX2 gene with colon cancer risk(P > 0.05),and we did not observe that these variants modify the protective effect of NSAIDs(P > 0.05).We were able to confirm the lack of association of variants in UGT1A6 with colon cancer risk,although further studies will have to be conducted to confirm the association of these variants with colon ***:Our study does not support a role of COX2 and UGT1A6 genetic variations in the development of colon cancer.

Background: The use of feed grade amino acids can reduce the cost of lactation feed. With changing genetics,increasing feed costs, and higher number of pigs weaned with heavier wean weights further evaluation of higher inclusion levels of feed-grade amino acid in lactation diets than previously published is warranted. Two experiments(Exp.) were conducted to determine the optimal inclusion level of l-lysine HCl to be included in swine lactation diets while digestible lysine levels remain constant across dietary treatments and allowing feed grade amino acids to be added to the diet to maintain dietary ratios relative to lysine to maximize litter growth rate and sow reproductive performance. Furthermore, the studies were to evaluate minimal amino acid ratios relative to lysine that allows for optimal litter growth rate and sow reproductive ***: Exp. 1: Increasing l-lysine HCl resulted in similar gilt feed intake, litter, and reproductive *** litter gain from birth to weaning was 2.51, 2.49, 2.59, 2.43, and 2.65 kg/d when gilts were fed 0.00, 0.075,0.150, 0.225, and 0.30% l-lysine HCl, respectively. Exp. 2: The average litter gain from birth to weaning was 2.68,2.73, 2.67, 2.70, and 2.64 kg/d(P l-lysine HCl plus valine,respectively. No other differences among dietary treatments were ***: Collectively, these studies demonstrate corn-soybean meal based lactation diets formulated with a constant SID lysine content for all parities containing up to 0.40% l-lysine HCl with only supplemental feed grade threonine and a methionine source have no detrimental effect on litter growth rate and subsequent total born.
