The use of feed-grade amino acids in lactating sow diets

Background: The use of feed grade amino acids can reduce the cost of lactation feed. With changing genetics,increasing feed costs, and higher number of pigs weaned with heavier wean weights further evaluation of higher inclusion levels of feed-grade amino acid in lactation diets than previously published is warranted. Two experiments(Exp.) were conducted to determine the optimal inclusion level of L-lysine HCl to be included in swine lactation diets while digestible lysine levels remain constant across dietary treatments and allowing feed grade amino acids to be added to the diet to maintain dietary ratios relative to lysine to maximize litter growth rate and sow reproductive performance. Furthermore, the studies were to evaluate minimal amino acid ratios relative to lysine that allows for optimal litter growth rate and sow reproductive ***: Exp. 1: Increasing L-lysine HCl resulted in similar gilt feed intake, litter, and reproductive *** litter gain from birth to weaning was 2.51, 2.49, 2.59, 2.43, and 2.65 kg/d when gilts were fed 0.00, 0.075,0.150, 0.225, and 0.30% L-lysine HCl, respectively. Exp. 2: The average litter gain from birth to weaning was 2.68,2.73, 2.67, 2.70, and 2.64 kg/d(P < 0.70) when sows were fed 0.1, 0.2, 0.3, 0.4, and 0.4% L-lysine HCl plus valine,respectively. No other differences among dietary treatments were ***: Collectively, these studies demonstrate corn-soybean meal based lactation diets formulated with a constant SID lysine content for all parities containing up to 0.40% L-lysine HCl with only supplemental feed grade threonine and a methionine source have no detrimental effect on litter growth rate and subsequent total born.

^(99m)TC-Methylene diphosphonate uptake at injury site correlates with osteoblast differentiation and mineralization during bone healing in mice

99mTc-Methylene diphosphonate （99mTc-MDP） is widely used in clinical settings to detect bone abnormalities. However, the mechanism of 99mTc-MDP uptake in bone is not well elucidated. In this study, we utilized a mouse tibia injury model, single-photon emission computed tomography （gamma scintigraphy or SPECT）, ex vivo micro-computed tomography, and histology to monitor 99mTc-MDP uptake in injury sites during skeletal healing. In an ex vivo culture system, calvarial cells were differentiated into osteoblasts with osteogenic medium, pulsed with 99mTc-MDP at different time points, and quantitated for 99mTc-MDP uptake with a gamma counter. We demonstrated that 99mTc-MDP uptake in the injury sites corresponded to osteoblast generation in those sites throughout the healing process. The 99mTc-MDP uptake within the injury sites peaked on day 7 post-injury, while the injury sites were occupied by mature osteoblasts also starting from day 7. ~mTc-MDP uptake started to decrease 14 days post-surgery, when we observed the highest level of bony tissue in the injury sites. We also found that 99mTc-MDP uptake was associated with osteoblast maturation and mineralization in vitro. This study provides direct and biological evidence for 99mTc-MDP uptake in osteoblasts during bone healing in vivo and in vitro.

Variation in resource competition traits among Microcystis strains is affected by their microbiomes

Freshwater harmful algal blooms are often dominated by Microcystis,a phylogenetically cohesive group of cyanobacteria marked by extensive genetic and physiological *** have previously shown that this genetic diversity and the presence of a microbiome of heterotrophic bacteria influences competitive interactions with eukaryotic *** this study,we sought to explain these observations by characterizing Monod equation parameters for resource usage(maximum growth rateμmax,half-saturation value for growth Ks,and quota)as a function of N and P levels for four strains(NIES-843,PCC 9701,PCC 7806[WT],and PCC 7806ΔmcyB)in presence and absence of a microbiome derived from Microcystis isolated from Lake *** indicated limited differences in maximum growth rates but more pronounced differences in half-saturation values among Microcystis *** largest impact of the microbiome was reducing the minimal nitrogen concentration sustaining growth and reducing half saturation values,with variable results depending on the Microcystis *** strains also differed from each other in their N and P quotas and the extent to which microbiome presence affected *** data highlight the importance of the microbiome in altering Microcystis-intrinsic traits,strain competitive hierarchies,and thus bloom *** quota,μmax,and Ks are commonly used in models for harmful algal blooms,our data suggest that model improvement may be possible by incorporating genotype dependencies of resource-use parameters.

Magnesium-ion-conducting solid polymer electrolytes have been studied for rechargeable Mg metal batteries,one of the beyond-Li-ion *** this paper,magnesium polymer electrolytes with magnesium bis(trifluoromethane)sulfonimide(Mg(TFSI)2)salt in poly(ε-caprolactone-co-trimethylene carbonate)(PCL-PTMC)were investigated and compared with the poly(ethylene oxide)(PEO)*** thermal properties and vibrational spectroscopy indicated that the total ion conduction in the PEO electrolytes was dominated by the anion conduction due to strong polymer coordination with fully dissociated Mg2+.On the other hand,in PCL-PTMC electrolytes,there is relatively weaker polymer–cation coordination and increased anion–cation *** Mg-and F-rich particles were observed on the Cu electrodes after polarization tests in Cu|Mg cells with PCL-PTMC electrolyte,suggesting that Mg was conducted in the ion complex form(MgxTFSIy)to the copper working electrode to be reduced which resulted in anion ***,the Mg metal deposition/stripping was not favorable with either Mg(TFSI)2 in PCL-PTMC or Mg(TFSI)2 in PEO,which inhibited quantitative analysis of magnesium conduction.A remaining challenge is thus to accurately assess transport numbers in these systems.

Climate Change Adaptation and Vulnerability: A Case of Rain Dependent Small-Holder Farmers in Selected Districts in Zambia

Food crop production by small-holder farmers in Africa is particularly vulnerable to climate change, given high dependence on rainfall coupled with limited adaptive capacity. In Zambia, smallholder farmers contribute about 79% of national stable food requirements particularly maize. This paper attempted to establish levels of food security in each of the three agro-ecological zones of Zambia, and evaluated the current adaptive measures of rain dependent small-holder farmers against climate change risks. The challenges farmers are facing in adapting to the change risks were identified and livelihood vulnerability assessed. The findings indicate that rain dependent small-holder farmers in Zambia are highly vulnerable to weather related shocks which impact greatly on their food production;and that the levels of vulnerability vary across gender and per agro-ecological zone. After the evaluation of scenarios including staple food crop yields (maize), the authors conclude that most rain-fed small-holder farmers in Zambia (about 70%) are facing considerable hardships in adapting to the changing climate, which in turn, undermines their contribution to food security. While efforts by government have been made to assist farmers towards climate change adaptation, there still remains many challenges to achieve the desired outcomes. Most farmers (66%) are unable to afford certain alternatives, such as those of agro-forestry or conservation. Difficulties in accessing markets, poor road infrastructure, fluctuating market prices, high costs and late deliveries of farming in-puts were found to be among the major challenges that farmers are facing in Zambia. There are also no systematic early warning systems in place against natural hazards and disasters. This makes farming a difficult undertaking in Zambia.

Alcohol-Induced cardiomyopathy Presenting with Left Ventricular Apical Thrombus

Background: Chronic excessive alcohol consumption has been strongly associated with alcohol-induced cardiomyopathy (AC) in patients with no evidence of coronary artery disease (CAD). AC may cause cardiovascular complications and significant impact on the quality of life. We discuss an interesting case of dilated cardiomyopathy, associated complication, diagnostic work-up and management. Case Report: A young male presented to our service with worsening dyspnea, orthopnea, and scrotal and lower extremity edema. On average, he consumed a pack of 12 beers every day and had a 30-pack-years smoking history. He was found to be in acute heart failure with evidence of pulmonary edema and cardiomegaly on chest imaging. He had biventricular dilatation and severely reduced left ventricular ejection fraction (LVEF) 15% in addition to a thrombus in the LV apex. The cardiac catheterization was unremarkable for CAD. He was diuresed appropriately resulting in significant weight loss and resolution of symptoms. LV thrombus was treated with unfractionated heparin infusion that was transitioned to warfarin. He was maintained on guidelines-directed medical therapy for heart failure. Extensive counseling was provided regarding alcohol and tobacco cessation. On follow-up echocardiogram, his LVEF improved and there was no evidence of LV thrombus. We think, the readership will benefit from our experience of treating a case of AC, and the importance of clinical history. Conclusion: Chronic excessive alcohol use is detrimental to cardiac function leading to alcohol-induced cardiomyopathy. A careful approach to clinical history of alcohol consumption and prompt diagnostic workup negative for ischemic causes may confirm the diagnosis. cardiac function improves with guidelines-directed medical therapy for heart failure and abstinence from alcohol.

An exploratory study of food safety and food handling: Examining ready-to-eat foods in independent delicatessen operations

The US Center for Disease Control and Prevention (CDC, 2009) reports that each year 76 million cases of foodborne illness occur with over 300,000 people hospitalized and 5000 deaths. This study, phase I of a continuing study, identifies key areas for food safety improvement. This study is designed as an exploratory evaluation of independently owned and operated delicatessen operations, using Escherichia coli and Staphylococcus aureus as indicators to assess food handling and the public’s risk for pathogenic contamination from commonly served ready-to-eat/take-away foods. The analysis consisted of a comprehensive strategy of laboratory testing of samples for pathogenic contamination, informal field observation of food handling procedures, and the examination of the most recent health inspection reports for each of the 18 operations visited. The deli turkey, cream cheese and lettuce were tested using bacteria indicator plates. The results showed widespread levels of contamination. Of the 54 samples tested for Escherichia coli, 26 showed positive results which are ~45% for E. coli contamination. Of the 54 samples tested for Staphy-lococcus aureus, 31 showed positive results which are ~57% for S. aureus contamination. One issue discovered while conducting the study was the lack of consistent uniform international standards of contamination tolerance levels. The informal field observations and health report analyses revealed widespread temperature violations and numerous instances of poor food handling. The study offers independent practitioners a strategy designed to improve their health inspections scores, food handling, and mitigation of operator liability. Independent operators traditionally do not enjoy the resources of centralized supervision and expert on-staff training;yet in aggregate they account for significant volume in both dollars and the quantity of product served. This study adds needed scrutiny to this important food niche.
