Arbuscular mycorrhizal fungi（AMF） are important during revegetation of mining sites, but few studies compared AMF community in revegetated sites with pristine adjacent ecosystems. The aim of this study was to assess AMF species richness in a revegetated iron-mining site and adjacent ecosystems and to relate AMF occurrence to soil chemical parameters. Soil samples were collected in dry and rainy seasons in a revegetated iron-mining site（RA） and compared with pristine ecosystems of forest（FL）, canga（NG）,and Cerrado（CE）. AMF species were identified by spore morphology from field and trap cultures and by LSU r DNA sequencing using Illumina. A total of 62 AMF species were recovered, pertaining to 18 genera and nine families of Glomeromycota. The largest number of species and families were detected in RA, and Acaulospora mellea and Glomus sp1 were the most frequent species. Species belonging to Glomeraceae and Acaulosporaceae accounted for 42%–48% of total species richness. Total number of spores and mycorrhizal inoculum potential tended to be higher in the dry than in the rainy season, except in RA. Sequences of uncultured Glomerales were dominant in all sites and seasons and five species were detected exclusively by DNA-based identification. Redundancy analysis evidenced soil p H,organic matter, aluminum, and iron as main factors influencing AMF presence. In conclusion, revegetation of the iron-mining site seems to be effective in maintaining a diverse AMF community and different approaches are complementary to reveal AMF species, despite the larger number of species being identified by traditional identification of field spores.

Polyhydroxyalkanoates are natural,biodegradable,thermoplastic and sustainable polymers with a huge potential in fabrication of bioresorbable implantable devices for tissue *** describe a comparative evaluation of three medium chain length polyhydroxyalkanoates(mcl-PHAs),namely poly(3-hydroxyoctanoate),poly(3-hydroxyoctanoate-co-3-hydoxydecanoate)and poly(3-hydroxyoctanoate-co-3-hydroxydecanoate-co-3-hydroxydodecanoate),one short chain length polyhydroxyalkanoate,poly(3-hydroxybutyrate),P(3HB)and synthetic aliphatic polyesters(polycaprolactone and polylactide)with a specific focus on nerve regeneration,due to mechanical properties of mcl-PHAs closely matching nerve *** vitro biological studies with NG108-15 neuronal cell and primary Schwann cells did not show a cytotoxic effect of the materials on both cell *** mcl-PHAs supported cell adhesion and *** the three mcl-PHAs,P(3HO-co-3HD)exhibited superior properties with regards to numbers of cells adhered and viable cells for both cell types,number of neurite extensions from NG108-15 cells,average length of neurite extensions and Schwann ***,similar characteristics were observed for flat P(3HB)surfaces,high rigidity of this biomaterial,and FDA-approved polymers such as PLLA,limits their applications in peripheral nerve ***,we have designed,synthesized and evaluated these materials for nerve tissue engineering and regenerative medicine,the interaction of mcl-PHAs with neuronal and Schwann cells,identifying mcl-PHAs as excellent materials to enhance nerve regeneration and potentially their clinical application in peripheral nerve repair.

Claudin-11 and occludin are major contributors to Sertoli cell tight junction function, in vitro

The Sertoli cell tight junction （T J） is the key component of the blood-testis barrier, where it sequesters developing germ cells undergoing spermatogenesis within the seminiferous tubules. Hormonally regulated claudin-11 is a critical transmembrane protein involved in barrier function and its murine knockout results in infertility. We aimed to assess quantitatively the significance of the contribution of claudin-11 to TJ function, in vitro, using siRNA-mediated gene silencing. We also conducted an analysis of the contribution of occludin, another intrinsic transmembrane protein of the TJ. Silencing of claudin-11 and/or occludin was conducted using siRNA in an immature rat Sertoli cell culture model. Transepithelial electrical resistance was used to assess quantitatively TJ function throughout the culture. Two days after siRNA treatment, cells were fixed for immunocytochemical localization of junction proteins or lyzed for RT-PCR assessment of mRNA expression. Silencing of claudin-11, occludin, or both resulted in significant decreases in TJ function of 55% （P 〈 0.01）, 51% （P 〈 0.01）, and 62% （P 〈 0.01）, respectively. Data were concomitant with significant decreases in mRNA expression and marked reductions in the localization of targeted proteins to the Sertoli cell TJ. We provide quantitative evidence that claudin-11 contributes significantly （P 〈 0.01） to Sertoli cell TJ function in vitro. Interestingly, occludin, which is hormonally regulated but not implicated in infertility until late adulthood, is also a significant （P 〈 0.01） contributor to barrier function. Our data are consistent with in vivo studies that clearly demonstrate a role for these proteins in maintaining normal TJ barrier structure and function.

Fertilizer-intensive agriculture is a leading source of reactive nitrogen(Nr)emissions that damage climate,air quality,and human *** has long been studied as a soil amendment,but its influence on Nr emissions remains insufficiently *** recently,the pyrolysis of light hydrocarbons has been suggested as a source of hydrogen fuel,resulting in a solid zero-valent carbon(ZVC)byproduct whose impact on soil emissions has yet to be *** incorporate carbon amendment algorithms into an agroecosystem model to simulate emission changes in the year following the application of biochar or ZVC to the *** *** simulations predicted that the impacts of biochar amendments on Nr emissions would vary widely(−17%to+27%under 5 ton ha^(−1) applications,−38%to+18%under 20 ton ha^(−1) applications)and depend mostly on how nitrification is ***-dose biochar application(5 ton ha^(−1))stimulated emissions of all three nitrogen species in 75%of simulated agricultural areas,while high-dose applications(20 ton ha^(−1))mitigated emissions in 76%of simulated *** zero-valent carbon at 20 ton ha^(−1) exhibited similar effects on nitrogen emissions as biochar applications at 5 ton ha^(−1).Biochar amendments are most likely to mitigate emissions if applied at high rates in acidic soils(pH<5.84)with low organic carbon(<55.9 kg C ha^(−1))and inorganic nitrogen(<101.5 kg N ha^(−1))*** simulations could inform where the application of carbon amendments would most likely mitigate Nr emissions and their associated adverse impacts.

The use of statins for bone regeneration is a promising and growing area of research. Statins, originally developed to treat high cholesterol, are inhibitors of the enzyme 3-hydroxy-3-methylglutaryl, the rate-limiting enzyme of the mevalonate pathway. Because the mevalonate pathway is responsible for the synthesis of a wide variety of important biochemical molecules, including cholesterol and other isoprenoids,the efects of statins are pleiotropic. In particular, statins can greatly afect the process of bone turnover and regeneration via efects on important cell types, including mesenchymal stem cells, osteoblasts, endothelial cells, and osteoclasts. Statins have also been shown to have anti-inlammatory and antimicrobial properties that may be useful since infection can derail normal bone healing. his review will explore the pleiotropic efects of statins, discuss the current use of statins for bone regeneration, particularly with regard to biomaterials-based controlled delivery, and ofer perspectives on the challenges and future directions of this emerging area of bone tissue engineering.

Enriched Opportunistic Pathogens Revealed by Metagenomic Sequencing Hint Potential Linkages between Pharyngeal Microbiota and COVID-19

One of the most important tools used to evaluate kidney function in the context of chronic kidney disease or other renal function related pathologies is the exploration of glomerular filtration rate (GFR). Iohexol is up to this moment a good candidate molecule for the GFR assessment since it exhibits minimum protein binding rates and minimum extra-renal clearance, being neither secreted nor reabsorbed at the tubular level. This study proposes and evaluates a new LC-MS/MS method for the iohexol determination from capillary blood, prelevated using volumetric absorbative microsampling (VAMS) systems. As an alternative to VAMS, a brand new HemaPEN■ device for micro-prelevation was also tested. A new high throughput sample preparation protocol adapted for iohexol quantification from whole blood VAMS samples was developed. The medium term stability study of iohexol in dried whole blood VAMS samples that was conducted showed a good stability of this molecule for up to 12 days. By collecting only 10 mL of blood, iohexol can be analyzed from dried whole blood VAMS samples for concentration ranges between 1 and 250 mg/mL. Due to the analyte stability in VAMS for up to 12 days, this approach might be successfully applied for GFR assessment for clinical cases allowing minimum invasiveness and even delayed analysis.
