The thymine DNA glycosylase （TDG） is a multifunctional enzyme,which is essential for embryonic *** mediates the base excision repair （BER） of G：T and G：U DNA mismatches arising from the deamination of 5-methyl cytosine （5-meC） and cytosine,*** studies have pointed at a role of TDG during the active demethylation of 5-meC within CpG *** interacts with the histone acetylase CREB-binding protein （CBP） to activate CBP-dependent *** addition,TDG also interacts with the retinoic acid receptor α （RARα）,resulting in the activation of RARα target *** we provide evidence for the existence of a functional ternary complex containing TDG,CBP and activated RARα.Using global transcriptome profiling,we uncover a coupling of de novo methylation-sensitive and RA-dependent transcription,which coincides with a significant subset of CBP target *** introduction of a point mutation in TDG,which neither affects overall protein structure nor BER activity,leads to a significant loss in ternary complex stability,resulting in the deregulation of RA targets involved in cellular networks associated with DNA replication,recombination and *** thus demonstrate for the first time a direct coupling of TDG＇s epigenomic and transcription regulatory function through ternary complexes with CBP and RARα.

In tropical montane forests,compositional and structural changes are commonly driven by broad-scale altitudinal ***,given the lack of knowledge on small-scale vegetation changes and temporal dynamics,we address the effects of small-scale variations in soil and altitude on tree community structure,temporal dynamics and phylogenetic diversity in a semi-deciduous tropical forest of the Atlantic Forest Domain,southeastern *** 2010 and 2015 we sampled thirty plots of 400 m^(2),set up along an altitudinal gradient between 1000 and 1500 m a.s.I..In each plot,we collected soil samples for chemical and textural *** fitted linear models to test the effects of altitude and soil on community dynamics and phylogenetic *** and soil explained the spatial variation in number of individuals and phylogenetic diversity *** lower to higher altitudes,we found decreasing fertility,increasing tree density and decreasing phylogenetic *** significantly influenced the increases in total biomass(from 240.9 to 255.4 t ha^(-1))and individual biomass(from 0.15 to 0.17 t)recorded in the *** while community temporal dynamics had rates of 1.96%for mortality,1.02%for recruitment,1.61%for biomass loss and 2.81%for biomass gain,none of them were explained by altitude or *** species substitution averaged0.1 in the ***,these results suggest that the small-scale variations in altitude and soil likely determine the conditions and resources that drive community assembly and structure,which are expressed by spatial variations along the altitudinal *** the same time,temporal patterns were not influenced by altitude-related environmental variation,resulting in a similar dynamic behaviour across the gradient,suggesting that broad-scale factors may play a more important role than local ones.

From a hermeneutic and comparative perspective,the purpose of this paper is to discuss investigative paths of ethnological and ethnographic aspects in Nietzsche’s writings that became relevant for different elaborations of the concept of synthesis in specific thought cycles of his *** a first step,these aspects are verified in his school,philological and philosophical texts,described in relation to his studies on ancient Greek culture and compared under the parameter of the different conceptions of synthesis developed by him in each identified ***,the concept of synthesis is discussed as physiology(considering body,consciousness,and language),explaining human life as a creative expression of nature *** this explanation,it becomes possible to understand the processes related to the concept of synthesis as the task of those he terms as good Europeans,and the great politics in his philosophy of the *** identification between the feeling of resistance(Widerstandsgefühl)and the imminence point of the preliminary stage of dominion structures(Herschafts-Gebilde)in the last analyzed conception of synthesis is the logical and ethical path towards the demonstration intended *** article concludes by demonstrating the creative and dynamic character of the metaphilosophical scope of the task proposed by him in the mid-1880s and the possibilities opened up from then on for future discussions of culture,aesthetics,knowledge,logic,ethics,and politics.

Adenoid Cystic Carcinoma of the Cervix: Case Report

Adenoid cystic carcinoma (ACC) is usually an epithelial neoplasm of the salivary glands but can also occur in the lacrimal glands and the mucous glands of the respiratory tract, breasts and skin. Very rarely, it may affect the uterine cervix. We report here a case of a woman with postmenopausal bleeding for 7 months, who on physical examination presented with a vegetative and friable tumor lesion in the uterine cervix, measuring about 5 cm in diameter. The abdominal and pelvic computed tomography scans showed a hypodense and heterogeneous mass in the cervix and uterine isthmus. Histopathological examination and immunohistochemical assays confirmed the diagnosis of ACC. The different pathological aspects, therapeutic options and prognostic factors of ACC are discussed.

Shortly after President John F. Kennedy's assassination, with a nation deep in mourning and the world looking on in stunned disbelief, Jacqueline Kennedy found the strength to set aside her own personal grief for the sake of posterity and begin the task of documenting and preserving her husband's legacy. In January of 1964, she and Robert F. Kennedy approved a planned oral-history project that would capture their first-hand accounts of the late President as well as the recollections of those closest to him throughout his extraordinary political career. For the rest of her life, the famously private Jacqueline Kennedy steadfastly refused to discuss her memories of those years, but beginning that march, she fulfilled her obligation to future generations of Americans by sitting down with historian Arthur Schlesinger, Jr., and recording an astonishingly detailed and unvarnished account of her experiences and impressions as the wife and confidante of John F. Kennedy. The tapes of those sessions were then sealed and later deposited in the John F. Kennedy Presidential Library and museum upon its completion, in accordance with mrs. Kennedy's wishes.

Parental imprinting is an epigenetic process leading to monoallelic expression of certain genes depending on their parental *** diseases are characterized by growth and metabolic issues starting from birth to *** are mainly due to methylation defects in imprinting control region that drive the abnormal expression of imprinted *** currently lack relevant animal or cellular models to unravel the pathophysiology of growth failure in these *** aimed to characterize the methylation of imprinting regions in dental pulp stem cells and during their differentiation in osteogenic cells(involved in growth regulation)to assess the interest of this cells in modeling imprinting *** collected dental pulp stem cells from five controls and four patients(three with Silver-Russell syndrome and one with Beckwith-Wiedemann syndrome).methylation analysis of imprinting control regions involved in these syndromes showed a normal profile in controls and the imprinting defect in *** results were maintained in dental pulp stem cells cultured under osteogenic ***,we confirmed the same pattern in six other loci involved in imprinting diseases in *** also confirmed monoallelic expression of H19(an imprinted gene)in controls and its biallelic expression in one *** imprinting control regions methylation analysis shows the strong potential of dental pulp stem cells in modeling imprinting diseases,in which imprinting regions are preserved in culture and during osteogenic *** will allow to perform in vitro functional and therapeutic tests in cells derived from dental pulp stem cells and generate other cell-types.

Type 1 neurofibromatosis is an inherited multisystem neurocutaneous disease predisposing to tumors development. Serious skin and ophthalmologic complications, although rare, can occur throughout life. Furthermore in children, unawareness of early symptoms may delay diagnosis. We report the case of A.T. 8 years old, admitted for exophthalmosis and facial deformity dating back to the age of 2 years. The diagnosis of neurofibromatosis was suspected in the presence of light brown skin spots scattered all over the body and subcutaneous nodules. Ophthalmologic examination revealed bilateral exophthalmosis, eyelids neurofibromas, blepharoptosis, Lisch nodules, corneal edema, and optic atrophy. Head CT scan clarified the nature and the extent of ophthalmologic lesions. Treatment was symptomatic. Neurofibromatosis is rarely reported in children in our setting;it is probably under diagnosed. Clinicians should think of this diagnosis in presence of certain specific symptoms and make a clinical assessment.

Background: Although many authors advise to postpone surgical intervention until skeletal maturity is reached, the Langenskiöld procedure—entailing physiolysis and the release of Vickers’ ligament—is a known procedure in case of madelung’s deformity in children. Although advised in textbooks and recent reviews, the results and late sequelae of this procedure have been poorly documented. The aim of this study is to portray the late effects of this procedure for three patients and emphasize a very careful patient selection. methods: A retrospective study was performed of the medical records of three patients who underwent a Langenskiöld procedure with release of Vickers’ ligament for madelung’s deformity of the wrist. They were recalled for function evaluation and their results are presented in the current study. Results: We report the long term results of three patients (five wrists) who underwent a Langenskiöld procedure with release of Vickers’ ligament in detail. mean follow-up was twelve years (range: 11 - 14 years). In all of the operated wrists severe pain recurred due to destruction of the distal radio-ulnar joint. Further salvaging surgeries were necessary, some with highly dissatisfying results. Conclusions: Based on our experience we advise not to perform this procedure until further studies have proven its effectiveness and clear indications. Other techniques are available for later correction of madelung’s deformity, with similar results on pain relief, wrist function and cosmetics. However, we will advocate conservative treatment until skeletal maturity is reached.

Dear Editors,Eculizumab is a therapeutic complement C5 inhibitor approved for the treatment of atypical hemolytic uremic syndrome(aHUS),paroxysmal nocturnal proteinuria(PNH),generalized myasthenia gravis and neuromyelitis optica spectrum *** dosing of eculizumab is important to ensure complete complement *** activity is often monitored by measuring either formation of the C5 activation product C5b-9 in vitro using an ELISA,independent of red blood cells,or by hemolytic assays measuring hemoglobin release from *** approaches allow analysis of complement activation via the classical(CP)and alternative(AP)pathways.
