Optogenetic inhibition of medial prefrontal cortex projections to the nucleus accumbens core and methyl supplementation via L-Methionine attenuates cocaine-primed reinstatement

DNA methylation has been identified as a powerful and activity-dependent regulator of changes in the brain that may underlie neuroadaptations in response to various types of stimuli,including exposure to drugs of ***,the medial prefrontal cortex(mPFC)projections to the nucleus accumbens(NAc)are critically important for reinstated cocaine-seeking in a rodent model of cocaine *** circuitry undergoes several epigenetic modifications following cocaine exposure,including changes in DNA methylation that are associated with drug-seeking *** have previously shown that methyl supplementation via L-Methionine(MeT)administration attenuates cocaine-seeking behavior and reverses expression and methylation patterns of the immediate early gene c-fos,suggesting that MeT may act by altering the excitability of this circuitry during cocaine *** the current study,male rats were microinjected with an adeno-associated virus overexpressing halorhodopsin in the mPFC,optical fibers were surgically implanted into the NAc,and the rats were given injections of MeT *** underwent acquisition of cocaine self-administration(0.75 mg/kg/infusion,2-h sessions)followed by extinction training in the absence of drug-paired *** reinstatement tests were conducted:cue-induced reinstatement without optogenetic manipulations and cocaine-primed reinstatement with optogenetic inhibition of mPFC-to-NAc *** were no group differences before the cocaine-primed reinstatement session,and all groups showed robust cue-induced *** rats treated with MeT and rats that received mPFC-to-NAc inhibition showed an abolishment of cocaine-primed reinstatement,suggesting that systemic methyl supplementation may act through this critical circuity.

endometrial mesenchymal stem cells as a cell based therapy for pelvic organ prolapse

Pelvic organ prolapse(POP) occurs when the pelvic organs(bladder, bowel or uterus) herniate into the vagina, causing incontinence, voiding, bowel and sexual dysfunction, negatively impacting upon a woman's quality of life. POP affects 25% of all women and results from childbirth injury. For 19% of all women, surgical reconstructive surgery is required for treatment, often augmented with surgical mesh. The surgical treatment fails in up to 30% of cases or results in adverse effects, such as pain and mesh erosion into the bladder, bowel or vagina. Due to these complications the Food and Drug Administration cautioned against the use of vaginal mesh and several major brands have been recently been withdrawn from market. In this review we will discuss new cell-based approaches being developed for the treatment of POP. Several cell types have been investigated in animal models, including a new source of mesenchymal stem/stromal cells(MSC) derived from human endometrium. The unique characteristics of endometrial MSC, methods for their isolation and purification and steps towards their development for good manufacturing practice production will be described. Animal models that could be used to examine the potential for this approach will also be discussed as will a rodent model showing promise in developing an endometrial MSC-based therapy for POP. The development of a preclinical large animal model for assessing tissue engineering constructs for treating POP will also be mentioned.

Histone Acetyltransferase AtGCN5/HAG1 Is a Versatile Regulator of Developmental and Inducible Gene expression in Arabidopsis

Histone acetylation/deacetylation is a dynamic process and plays an important role in gene regulation. Histone acetylation homeostasis is regulated by antagonist actions of histone acetyltransferases （HAT） and deacetylases （HDAC）. Plant genome encodes multiple HATs and HDACs. The Arabidopsis HAT gene AtGCNS/HAGlplays an essential role in many plant development processes, such as meristem function, cell differentiation, leaf and floral organogenesis, and responses to environmental conditions such as light and cold, indicating an important role of this HAT in the regulation of both long-term developmental switches and short-term inducible gene expression. AtGCN5 targets to a large number of promoters and is required for acetylation of several histone H3 lysine residues. Recruitment of AtGCN5 to target promoters is likely to be mediated by direct or indirect interaction with DNA-binding transcription factors and/or by interaction with acetylated histone lysine residues on the targets. Interplay between AtGCN5 and other HATand HDAC is demonstrated to control specific regulatory pathways. Analysis of the role of AtGCN5 in light-inducible gene expression suggests a function of AtGCN5 in preparing chromatin commitment for priming inducible gene activation in plants.

Individual and local scale interactions and adaptations to wind energy development:A case study of Oklahoma,USA

Wind energy development receives broad support but is often opposed at the local level due to nuisance concerns and uncertainties about how it affects the landowners living due to the turbines and the broader ***-cal opposition to wind energy development can be a powerful force slowing or even ending its implementation in a given ***,USA is currently ranked as 4^(th)in the United States in current wind energy production and has seen significant pushback from some local communities as a renewable energy *** studies have examined wind energy development’s impact on rural education income,and property values of different communities in ***,funding information on how wind energy development affects the individu-als living alongside the turbines are *** fifteen interviews with landowners,site-managers,community representatives,and pro-wind non-profit organization representatives,this study finds that individuals who live in proximity to wind energy development,particularly those involved in the agricultural industry,have created novel and unique uses for wind farm *** also finds that local perceptions of wind energy production are mostly positive and provides increased knowledge of how wind energy development affects the individuals and communities that are hosting the turbines and related infrastructure.

Agroecosystem modeling of reactive nitrogen emissions from *** soils with carbon amendments

Fertilizer-intensive agriculture is a leading source of reactive nitrogen(Nr)emissions that damage climate,air quality,and human *** has long been studied as a soil amendment,but its influence on Nr emissions remains insufficiently *** recently,the pyrolysis of light hydrocarbons has been suggested as a source of hydrogen fuel,resulting in a solid zero-valent carbon(ZVC)byproduct whose impact on soil emissions has yet to be *** incorporate carbon amendment algorithms into an agroecosystem model to simulate emission changes in the year following the application of biochar or ZVC to the *** *** simulations predicted that the impacts of biochar amendments on Nr emissions would vary widely(−17%to+27%under 5 ton ha^(−1) applications,−38%to+18%under 20 ton ha^(−1) applications)and depend mostly on how nitrification is ***-dose biochar application(5 ton ha^(−1))stimulated emissions of all three nitrogen species in 75%of simulated agricultural areas,while high-dose applications(20 ton ha^(−1))mitigated emissions in 76%of simulated *** zero-valent carbon at 20 ton ha^(−1) exhibited similar effects on nitrogen emissions as biochar applications at 5 ton ha^(−1).Biochar amendments are most likely to mitigate emissions if applied at high rates in acidic soils(pHen(ere the application of carbon amendments would most likely mitigate Nr emissions and their associated adverse impacts.

Claudin-11 and occludin are major contributors to Sertoli cell tight junction function, in vitro

The Sertoli cell tight junction （T J） is the key component of the blood-testis barrier, where it sequesters developing germ cells undergoing spermatogenesis within the seminiferous tubules. Hormonally regulated claudin-11 is a critical transmembrane protein involved in barrier function and its murine knockout results in infertility. We aimed to assess quantitatively the significance of the contribution of claudin-11 to TJ function, in vitro, using siRNA-mediated gene silencing. We also conducted an analysis of the contribution of occludin, another intrinsic transmembrane protein of the TJ. Silencing of claudin-11 and/or occludin was conducted using siRNA in an immature rat Sertoli cell culture model. Transepithelial electrical resistance was used to assess quantitatively TJ function throughout the culture. Two days after siRNA treatment, cells were fixed for immunocytochemical localization of junction proteins or lyzed for RT-PCR assessment of mRNA expression. Silencing of claudin-11, occludin, or both resulted in significant decreases in TJ function of 55% （P 〈 0.01）, 51% （P 〈 0.01）, and 62% （P 〈 0.01）, respectively. Data were concomitant with significant decreases in mRNA expression and marked reductions in the localization of targeted proteins to the Sertoli cell TJ. We provide quantitative evidence that claudin-11 contributes significantly （P 〈 0.01） to Sertoli cell TJ function in vitro. Interestingly, occludin, which is hormonally regulated but not implicated in infertility until late adulthood, is also a significant （P 〈 0.01） contributor to barrier function. Our data are consistent with in vivo studies that clearly demonstrate a role for these proteins in maintaining normal TJ barrier structure and function.

Previous studies have shown cost effectiveness and quality-of-life benefit of pneumatic compression therapy(PCT)for lymphedema(Le).Insurers,such as the Centers for Medicare/Medicaid(CMS),however,desire visual proof that PCT moves *** infrared fuorescence lymphatic imaging(NIRFLI)was used to visualize lymphatic anatomy and function in four subjects with primary and cancer treatment-related Le of the lower extremities before,during,and after *** transparent and windowed PCT garments allowed visualization of lymph movement during single,1h PCT treatment *** revealed significant extravascular and lymphatic vascular movement of intradermally injected dye in all *** one subject with sufficient patent lymphatic vessels to allow quantification of lymph pumping velocities and frequencies,these values were significantly increased during and after PCT as compared to pre-treatment *** contractile activity in patent lymphatic vessels occurred in concert with the sequential cycling of *** visualization revealed increased lymphatic function,during and after PCT therapy,in all Le-affected *** studies are warranted to assess the effects of PCT pressure and sequences on lymph uptake and movement.

Decidual natural killer （dNK） cells express an array of activation receptors to regulate placental immunity and development during early pregnancy. We investigated the functional character of human dNK cells during the first and second trimester of gestation and the interaction between dNK and trophoblast cells. Although the frequency of CD56＋CD16-dNK among the total CD45＋ leukocytes did not change over this period, the expression of the activating receptors, NKp80 and NKG2D, was greatly upregulated. We observed a significantly higher number of extravillous trophoblast cells in proximity to the dNK cells in the first trimester in comparison with the second trimester decidua. NKG2D expression by first trimester dNK cells was decreased when co-cultured with the HTR-8 trophoblast cell line. In the second trimester, functional markers of dNK activation, i.e., angiogenic factor production （e.g., vascular endothelial growth factor, interleukin-8, interferon-gamma）, remained stable despite an increase in NKp80 or NKG2D surface expression. Furthermore, the degranulation capacity of dNK cells, as assessed by CD107a, was decreased in the second trimester. We suggest that in the first trimester, trophoblast-dNK interactions generate a population of dNK cells with a suppressed activating phenotype. In the second trimester, the loss of trophoblast-dNK interactions led to the inhibition of dNK cell function, although their activating receptor expression was increased. We speculate that during pregnancy, two mechanisms operate to modulate the dNK cell activation：suppression of activating receptor levels in the first trimester by trophoblasts and disengagement of receptor-ligand coupling in the second trimester.
