Optogenetic inhibition of medial prefrontal cortex projections to the nucleus accumbens core and methyl supplementation via l-methionine attenuates cocaine-primed reinstatement

Decidual natural killer （dNK） cells express an array of activation receptors to regulate placental immunity and development during early pregnancy. We investigated the functional character of human dNK cells during the first and second trimester of gestation and the interaction between dNK and trophoblast cells. Although the frequency of CD56＋CD16-dNK among the total CD45＋ leukocytes did not change over this period, the expression of the activating receptors, NKp80 and NKG2D, was greatly upregulated. We observed a significantly higher number of extravillous trophoblast cells in proximity to the dNK cells in the first trimester in comparison with the second trimester decidua. NKG2D expression by first trimester dNK cells was decreased when co-cultured with the HTR-8 trophoblast cell line. In the second trimester, functional markers of dNK activation, i.e., angiogenic factor production （e.g., vascular endothelial growth factor, interleukin-8, interferon-gamma）, remained stable despite an increase in NKp80 or NKG2D surface expression. Furthermore, the degranulation capacity of dNK cells, as assessed by CD107a, was decreased in the second trimester. We suggest that in the first trimester, trophoblast-dNK interactions generate a population of dNK cells with a suppressed activating phenotype. In the second trimester, the loss of trophoblast-dNK interactions led to the inhibition of dNK cell function, although their activating receptor expression was increased. We speculate that during pregnancy, two mechanisms operate to modulate the dNK cell activation：suppression of activating receptor levels in the first trimester by trophoblasts and disengagement of receptor-ligand coupling in the second trimester.

AIm To investigate the impact of medication beliefs, illness perceptions and quality of life on medication adherence in people with decompensated *** One hundred adults with decompensated cirrhosis completed a structured questionnaire when they attended for routine outpatient hepatology review. measures of self-reported medication adherence(morisky medication Adherence Scale), beliefs surrounding medications(Beliefs about medicines Questionnaire), perceptions of illness and medicines(Brief Illness Perception Questionnaire), and quality of life(Chronic liver Disease Questionnaire) were examined. Statistical significance was set at alpha = *** medication adherence was " High " in 42 % o f participants, "medium" in 37%, and "low" in 21%. Compared to patients with "High" adherence, those with "medium" or "low" adherence were more likely to report difficulty affording their medications(P lower perception of treatment helpfulness(P = 0.003) and stronger medication concerns relative to medication necessity beliefs(P = 0.003). AIm： To examine the relationships between pre-diag- nostic biomarkers and colorectal cancer risk and assess their relevance in predictive ***： A nested case-control study was designed to include all first primary incident colorectal cancer cases diagnosed between inclusion in the SUpplemen- tation en VItamines et min^raux AntioXydants cohort in 1994 and the end of follow-up in 2007. Cases （n = 50） were matched with two randomly selected con- trols （n = 100）. Conditional logistic regression models were used to investigate the associations between pre- diagnostic levels of hs-CRP, adiponectin, leptin, soluble vascular cell adhesion molecule-1 （sVCAm-1）, soluble intercellular adhesion molecule-I, e-selectin, monocyte chemoattractant protein-1 and colorectal cancer risk. Area under the receiver operating curves （AUC） and relative integrated discrimination improvement （RIDI） statistics were used to assess the discriminatory poten- tial of the models. ReSUlTS： Plasma adiponectin level was associated with decreased colorectal cancer risk （P for linear trend -- 0.03）. Quartiles of sVCAm-1 were associated with increased colorectal cancer risk （P for linear trend = 0.02）. No association was observed with any of the other biomarkers. Compared to standard models with known risk factors, those including both adiponectin and sVCAm-1 had substantially improved performance for colorectal cancer risk prediction （P for AUC improve- ment = 0.01, RIDI = 26.5%）. CONClUSION： These results suggest that pre-diag- nostic plasma adiponectin and sVCAm-1 levels are as- sociated with decreased and increased colorectal cancer risk, respectively. These relationships must be confirmed in large validation studies.

AIm:To determine if the addition of hip-strengthening exercises decreases pain and improves function in patients with patellofemoral pain ***:The authors completed a systematic reviewsearching eight databases(i.e.,Pub med,Cochrane,CINHAl,meDlINe,Sports Discus,emBASe,APTA Hooked on evidence,and PeDro).Two independent reviewers screened and excluded studies if they did not meet the following inclusion criteria:subjects had a primary diagnosis of patellofemoral pain syndrome(PFPS),intervention group included hip-strengthening exercises,control group included a traditional physical therapy intervention,study included outcome measures of pain and/or function,study used a randomized controlled trial design,PeDro score was ≥ 7,and study was published in a peer-reviewed *** outcome measures were subjective scales of pain and *** measures were converted to standardized mean difference [effect size(eS)],and a random-effects model was used to calculate the overall ***:Two hundred eighty-three studies were screened for inclusion in our *** studies were deemed suitable for data extraction and analysis.A total of 426 subjects were used in the nine ***,there was a significant positive effect of hip-strengthening exercises on measures of pain and function in subjects with PFPS(eS = 0.94,P = 0.00004).None of the individual studies had a negative eS,with study eS ranging from 0.35 to *** of the high degree of between-study variance(I2 = 76%;Q = 34.0,P meta-analyses and meta-regressions were *** of the potential moderator variables that were investigated(e.g.,outcome type,hip region targeted,duration of treatment) could explain a significant amount of the between-study variance in eS(P ≥ 0.23).CONClUSION:Overall,the addition of hip-strengthening exercises to traditional physical therapy produced greater improvements in measures of pain and function.
