AIM To determine the variability/conservation of the domain of hepatitis B virus(HBV) pre S1 region that interacts with sodium-taurocholate cotransporting polypeptide(hereafter, NTCP-interacting domain) and the prevalence of the rs2296651 polymorphism(S267 f, NTCP variant) in a Spanish population. METHODS Serum samples from 246 individuals were included and divided into 3 groups: patients with chronic HBV infection(CHB)(n = 41, 73% Caucasians), patients with resolved HBV infection(n = 100, 100% Caucasians) and an HBV-uninfected control group(n = 105, 100% Caucasians). Variability/conservation of the amino acid(aa) sequences of the NTCPinteracting domain,(aa 2-48 in viral genotype D) and a highly conserved pre S1 domain associated with virion morphogenesis(aa 92-103 in viral genotype D) were analyzed by next-generation sequencing and compared in 18 CHB patients with viremia > 4 log IU/mL. The rs2296651 polymorphism was determined in all individuals in all 3 groups using an in-house real-time PCR melting curve *** The HBV pre S1 NTCP-interacting domain showed a high degree of conservation among the examined viral genomes especially between aa 9 and 21(in the genotype D consensus sequence). As compared with the virion morphogenesis domain, the NTCPinteracting domain had a smaller proportion of HBV genotype-unrelated changes comprising > 1% of the quasispecies(25.5% vs 31.8%), but a larger proportion of genotype-associated viral polymorphisms(34% vs 27.3%), according to consensus sequences from Gen Bank patterns of HBV genotypes A to H. Variation/conservation in both domains depended on viral genotype, with genotype C being the most highly conserved and genotype E the most variable(limited finding, only 2 genotype E included). Of note, proline residues were highly conserved in both domains, and serine residues showed changes only to threonine or tyrosine in the virion morphogenesis domain. Transcriptome analysis of the Tityus serrulatusscorpion venom gland

The Tityus serrulatus scorpion is considered the most dangerous scorpion in Brazil and is responsible for several cases of human envenomation annually. In this study, we performed transcriptome profiling of the T. serrulatus venom gland. In addition to transcripts with housekeeping functions, such as those related to protein synthesis, energy supply and structural processes, transcripts from thirty-five families of venom peptides or proteins were identified. These transcripts included three new complete sequences of toxins and more than a dozen putative venom gland proteins/peptides. The venom gland transcriptome profile was verified by comparison with the previously determined proteomic profile. In conclusion, this transcriptome data provides novel insights into the putative mechanisms underlying the venomous character of T. serrulatus. The collected data of scorpion transcripts and proteins/peptides described herein may be an important resource for identifying candidate targets of molecular therapies and preventative measures.

Anti-M3Muscarinic Acetylcholine Receptor Antibodies in Systemic Lupus Erythematosus

Background: Evidences have shown that anti-M3 muscarinic acetylcholine receptor IgG (anti-M3 mAChR IgG) are clinically useful autoantibody that exert a cholinergic pharmacologic effect binding and interacting with M3 mAChR at the level of exocrine gland (salivary and ocular). Aims: The aim of this study was to determine the associations between serum level of anti-M3 mAChR IgG in patients with systemic lupus erythematosus (SLE) and other autoantibodies, serum prostaglandin E2 (PGE2), and clinical manifestations. Methods: Serum autoantibodies against M3 mAChR synthetic peptide were measured by enzyme-linked immuno absorbent assay (ELISA) using, as an antigen, a 25-mer peptide K-R-T-V-P-D-N-Q-C-f-I-Q-f-L-S-N-P-A-V-T-f-G-T-A-I corresponding to the amino acid sequence of the second extracellular loop of the human M3 mAChR. Serum levels of antinuclear antibodies (ANA), anti-Smith (Sm) antibodies, anti-phospholipid (APL) antibodies, and PGE2 were determined by ELISA in patients with SLE. Results: We found significantly enhanced titers of anti-M3 mAChR IgG in sera from SLE patients compared with healthy individuals (control). In addition, serum levels of PGE2 were significantly higher in SLE patients than in control patients and were significantly higher in active than in non-active SLE. No correlation was found with other autoantibodies present in SLE. By contrast, a positive correlation was found between anti-M3 mAChR IgG and PGE2 serum levels in SLE. Conclusions: As anti-M3 mAChR antibodies present in the sera of SLE patients may be another factor in the pathogenesis of this disease, and the increment of PGE2 in the sera of SLE has a modulatory action on the inflammatory process, suggesting that the presence of these autoantibodies against M3 mAChR may contribute to sustained immune deregulation and the strong inflammatory component observed in SLE.

Geothermal Diatoms: Seasonal Variability in the El Tatio Geothermal field (Altiplano, Chile)

Diatom floras were examined in a high-altitude geothermal field, 4200 - 4500 m (29°19'S 68°W'), located in the Central Andean dry Puna ecoregion or southern Altiplano. These locations include hostile environments subjecting living organisms to extreme conditions. The aim of the present study was to investigate the spatial patterns and describe the response of diatom assemblages to differences in physical and chemical variables. Different shallow (Achnanthidium exiguum (Grunow) Czarnecki, Cocconeis placentula var. lineata (Ehrenberg) Van Heurck, Eolimna minima (Grunow) Lange-Bertalot, Staurosirella pinnata (Ehrenberg) Williams and Round, Navicula gregaria Donkin, Nitzschia inconspicua Grunow, Nitzschia palea (Kützing) Smith, Nitzschia perminuta (Grunow) Peragallo, and Planothidium lanceolatum (Brébisson ex Kützing) Lange-Bertalot. As expected, the 20 to 200 μm-size fraction contained the highest numbers of diatom taxa (53 species), although an unexpectedly high number (47 species) were also found in the smaller 5 to 20 μm-size fraction, more associated to fumaroles and saline systems. The 180 to 2000 μm size fraction contained only two species, including rosette-forming diatom Ulnaria ulna (Nitzsch) Compère, and the unicellular species Surirella chilensis Janisch, both species exclusively reported in freshwater systems. Canonical correspondence analysis (CCA) and Monte Carlo permutation tests showed clear correlations between species, conductivity, TP (total phosphorous), NO3- , HO3- , Mg2+, temperature and dissolved oxygen (DO). The gradient of ionic composition values explaining most variation in diatom assemblages ranged from waters dominated by Ca2+ and SO4- to waters with higher proportions of Na+, K+, f-, Li+, Mg2+ and Cl-. Influence of Early Life Stress on Alcohol and Crack Dependents

Background: Early life stress is a significant public health problem associated with increased rates of psychiatric disorders, especially those related to drug abuse. Objective: To identify the prevalence of early life stress in drug users, to compare the intensity of trauma in alcohol and crack users, and to relate the power of injury to the severity of drug dependence. Method: Cross-sectional analytical study with a sample of 105 alcohol and crack users treated by an outpatient service. The instruments for data collection were sociodemographic data questionnaire, the Mini-International Neuropsychiatric Interview, and the Severity of Dependence Scale. The categorical variables association was analyzed using the Chi-squared test, considering p Results: High prevalence of early stress and severity of dependence were identified, with higher rates among crack users. The early stress revealed in the Alcohol group high rates of emotional (88.7%) and physical (94.3%) neglect and in the Crack group significant frequency of physical (61.5%) emotional abuse (51.9%), sexual (46.2%), and emotional (78.8%) and physical (90.4%) neglect. Crack users are 2.6 times more likely to have been emotionally abused, and 2.1 times more likely to have been sexually abused during childhood when compared to the alcohol group. Conclusion: Early stress was prevalent with significant intensity in drug users, and evaluation of this problem is essential for a better understanding of these disorders.

Streptococcus pneumoniaeUpper Respiratory Carriage in Costa Rican Children with Otitis Media before the Introduction of the Heptavalent Conjugated Vaccine in the National Immunization Program

Objective: The aim of this study was to analyze the NP/OP S. pneumoniae serotype distribution and potential vaccine coverage in Costa Rican children with Otitis Media (OM) before the introduction of PCV-7 in the National Immunization Program (NIP). Methods: Between 2002 and 2006, NP and OP samples were obtained from 641 children from 6 to 79 months of age, at the time of OM diagnosis. S. pneumoniae serotyping and antimicrobial susceptibility were performed. Results: 386 S. pneumoniae isolates were recovered. The most common S. pneumoniae serotypes (ST) were: ST 6B, ST 14, ST 19f. Penicillin non-susceptibility was observed among 57% of the isolates obtained from children f age. 15% strains were multidrug resistant. Potential vaccine coverage was: PCV-7: 60%;PCV-10: 62%;and PCV-13: 76% and against penicillin non-susceptible and multidrug resistant isolates was: PCV-7;59% and 83%, respectively;PCV-10: 60% and 85%, respectively and PCV-13: 74% and 96%, respectively. Conclusions: S. pneumoniae was isolated from the NP and/or OP in the majority (59%) of studied children with OM. At a statistical significant level, only serotype 3 was more frequently isolated among children >24 months of age. Antibiotic non-susceptibility and MDR were significantly higher in children f age. This study demonstrates that PCV-13 offers the highest potential vaccine coverage and serves to assess the impact of introduction of one of the conjugated vaccines in the NIP in Costa Rica.

Unsteady magnetohydrodynamic stagnation point flow — closed-form analytical solutions

This paper investigates the unsteady stagnation point flow and heat transfer of magnetohydrodynamic(MHD) fluids over a moving permeable flat surface. The unsteady Navier-Stokes(NS) equations are transformed into a similarity nonlinear ordinary differential equation, and a closed form solution is obtained for the unsteadiness parameter of 2. The boundary layer energy equation is transformed into a similarity equation,and is solved for a constant wall temperature and a time-dependent uniform wall heat flux case. The solution domain, velocity, and temperature profiles are calculated for different combinations of parameters including the Prandtl number, mass transfer parameter, wall moving parameter, and magnetic parameter. Two solution branches are obtained for certain combinations of the controlling parameters, and a stability analysis demonstrates that the lower solution branch is not stable. The present solutions provide an exact solution to the entire unsteady MHD NS equations, which can be used for validating the numerical code of computational fluid dynamics.
