Cortex cinnamomi extract prevents streptozotocin- and cytokine-induced β-cell damage by inhibiting NF-κB

AIM： To clarify the mechanism underlying the antidiabetic activities of cortex cinnamomi extract （CCE）. METHOdS： To induce in vivo diabetes, mice were injected with streptozotocin （STZ） via a tail vein （100 mg STZ/kg body weight）. To determine the effects of CCE, mice were administered CCE twice daily for 7 d by oral gavage starting 1 wk before the STZ injection. Blood glucose and plasma insulin concentration were measured as an index of diabetes. Also, to induce cytotoxicity of RINm5F cells, we treated with cytokines （IL-1β （2.0 ng/mL） and IFN-γ （100 U/mL））. Cell viability and nitric oxide production were measured colorimetrically. Inducible nitric oxide synthase （iNOS） mRNA and protein expression were determined by RT-PCR and Western blotting, respectively. The activation of NF-κβ was assayed by using gel mobility shift assays of nuclear extracts. RESULTS： Treatment of mice with STZ resulted in hyperglycemia and hypoinsulinemia, which was further evidenced by immunohistochemical staining of islets. However, the diabetogenic effects of STZ were completely prevented when mice were pretreated with CCE. The inhibitory effect of CCE on STZ-induced hyperglycemia was mediated through the suppression of iNOS expression. In rat insulinoma RINm5F cells,CCE completely protected against interleukin-1β and interferon-y-mediated cytotoxicity. Moreover, RINm5F cells incubated with CCE showed significant reductions in interleukin-1β and interferon-y-induced nitric oxide production and in iNOS mRNA and protein expression, and these findings correlated well with in vivo observations. CONCLUSION： The molecular mechanism by which CCE inhibits iNOS gene expression appears to involve the inhibition of NF-κβ activation, These results reveal the possible therapeutic value of CCE for the prevention of diabetes mellitus progression,

We demonstrate multi-gigahertz continuous-wave mode-locking of a Yb:KLuW waveguide laser. A femtosecondlaser-inscribed Yb:KLuW channel waveguide in an extended laser cavity delivers a fundamentally mode-locked laser near 1030 nm. A tunable few-centimeter-long cavity containing a single-walled carbon nanotube saturable absorber as mode-locker generates self-starting femtosecond pulses with average output powers exceeding210 mW at repetition rates of 2.27, 2.69, and 3.55 GHz. The laser cavity, which includes a wedged waveguide,is extended by using a lens pair that controls the laser fluence on the saturable absorber for reliable mode-locked operation without instability. The presented laser performance, mode-locked up to 3.55 GHz, highly suggests the potential of crystalline Yb:KLuW waveguides for realizing high-power ultrafast lasers with higher GHz repetition rates in a quasi-monolithic cavity.

AIM:To describe and evaluate the efficacy of Ahmed glaucoma valve implantation(AGV)combined with pars plana vitrectomy(PPV)in a single surgical act for the treatment of advanced neovascular glaucoma(NVG).METHOdS:Retrospective observational case series included 51 eyes from 50 patients with severe NVG treated with PPV,AGV,and panretinal photocoagulation and/or cryotherapy in a single surgical act during a 13-year period(2005-2018).Preoperative,intraoperative and postoperative data at day 1 and months 1,3,6,21,and 24 were systematically *** of surgical success was stablished at IOP between 6 and 21 mm Hg with or without topical ***:Main indications for surgery were NVG secondary to proliferative diabetic retinopathy(39.2%)and central retinal vein occlusion(37.3%).Mean(±Sd)preoperative IOP was 42.0±11.2 mm Hg decreasing to 15.5±7.1 mm Hg at 12 mo and 15.8±9.1 mm Hg at 24 mo of follow *** incidence of success of IOP control was 76.0%at first postoperative month,reaching 88.3%at 6 *** of successful IOP control at long term was 74.4%at 12 mo and 71.4%at 24 *** evisceration for unsuccessful NVG management was required in 1 case(2.0%).CONCLUSION:Combination of AGV implantation and PPV in a single act may be a suitable option for severe forms of NVG in a case-by-case basis for effective IOP control and a complete panretinal photocoagulation.

We report the preparation of poly(3,4-ethylene dioxythiophene)(PEdOT)-modified polyvinylidene fluoride electrospun fibers and their use as a novel adsorbent material for the removal of the anionic dye Methyl Orange(MO)from aqueous *** novel adsorbent material can be used to selectively remove MO on a wide p H range(3.0-10.0),with a maximum capacity of 143.8 mg/g at p H *** used in a recirculating filtration system,the maximum absorption capacity was reached in a shorter time(20 min)than that observed for batch mode experiments(360 min).Based on the analyses of the kinetics and adsorption isotherm data,one can conclude that the predominant mechanism of interaction between the membrane and the dissolved dye molecules is ***,considering the estimated values for the Gibbs energy,and entropy and enthalpy changes,it was established that the adsorption process is spontaneous and occurs in an endothermic *** good mechanical and environmental stability of these membranes allowed their use in at least 20 consecutive adsorption/desorption cycles,without significant loss of their *** suggest that the physical-chemical characteristics of PEdOT make these hybrid mats a promising adsorbent material for use in water remediation protocols and effluent treatment systems.

An Approach for Micropropagation of Blueberry (<i>Vaccinium corymbosum</i>L.) Plants Mediated by Temporary Immersion Bioreactors (TIBs)

A new procedure for blueberry (Vaccinium corymbosum L.) micropropagation in programmed Temporary Immersion Bioreactors (TIBs based on two separate bottles) was developed for the commercial genotypes Biloxi, Sharp Blue and Brillita. Plant cultures were developed in a controlled environment with 0.4 MPa CO2 enrichment, sucrose-reduced medium, and light intensity of 60 mM m-2·s-1. Principal component analysis showed that component 1 (C1) grouped 64.08% of the total variability, while the first two components accounted for 86.97%. Representation of the principal components demonstrated three clusters corresponding with the blueberry genotypes, and within each cluster plants micropropagated in agar-base medium grouped separately from those plants multiplied in TIBs. Both plant number and total internodes traits (related to the productive efficiency) were demonstrated superior in blueberries propagated in TIBs. Additionally, when transferred to greenhouse conditions, blueberries propagated in TIBs showed higher adaptability and growing rates than those cultured by the conventional approach, altogether evidencing the occurrence of a photomixotrophic stage in the vitroplantlets cultured in TIBs.

We fit various color–magnitude diagrams(CMds) of the high-latitude Galactic globular clusters NGC 5024(M53),NGC 5053,NGC 5272(M3),NGC 5466,and NGC 7099(M30) by isochrones from the dartmouth Stellar Evolution database and Bag of Stellar Tracks and Isochrones for α–enrichment [α/Fe] = +*** the CMds,we use data sets from Hubble Space Telescope,Gaia,and other sources utilizing,at least,25 photometric filters for each *** obtain the following characteristics with their statistical uncertainties for NGC 5024,NGC 5053,NGC 5272,NGC 5466,and NGC 7099,respectively:metallicities [Fe/H] =-1.93 ± 0.02,-2.08 ± 0.03,-1.60 ± 0.02,-1.95 ± 0.02,and-2.07 ± 0.04 dex with their systematic uncertainty 0.1 dex;ages 13.00 ± 0.11,12.70 ± 0.11,11.63 ± 0.07,12.15 ± 0.11,and 12.80 ± 0.17 Gyr with their systematic uncertainty 0.8 Gyr;distances(systematic uncertainty added) 18.22 ± 0.06 ± 0.60,16.99 ± 0.06 ± 0.56,10.08 ± 0.04 ± 0.33,15.59 ±0.03 ± 0.51,and 8.29 ± 0.03 ± 0.27 kpc;reddenings E(B-V) = 0.023 ± 0.004,0.017 ± 0.004,0.023 ± 0.004,0.023 ± 0.003,and 0.045 ± 0.002 mag with their systematic uncertainty 0.01 mag;extinctions AV= 0.08 ± 0.01,0.06 ± 0.01,0.08 ± 0.01,0.08 ± 0.01,and 0.16 ± 0.01 mag with their systematic uncertainty 0.03 mag,which suggest the total Galactic extinction AV= 0.08 across the whole Galactic dust to extragalactic objects at the North Galactic *** horizontal branch morphology difference of these clusters is explained by their different metallicity,age,mass-loss efficiency,and loss of low-mass members in the evolution of the core-collapse cluster NGC 7099 and loose clusters NGC 5053 and NGC 5466.

Surface channel waveguides(WGs) were fabricated in a monoclinic Tm^(3+):KLu(WO_4)_2 crystal by femtosecond direct laser writing(fs-dLW). The WGs consisted of a half-ring cladding with diameters of 50 and 60 μm located just beneath the crystal surface. They were characterized by confocal laser microscopy and μ-Raman spectroscopy,indicating a reduced crystallinity and stress-induced birefringence of the WG cladding. In continuous-wave(CW)mode, under Ti:sapphire laser pumping at 802 nm, the maximum output power reached 171.1 mW at 1847.4 nm,corresponding to a slope efficiency η of 37.8% for the 60 μm diameter WG. The WG propagation loss was0.7 0.3 d B∕cm. The top surface of the WGs was spin-coated by a polymethyl methacrylate film containing randomly oriented(spaghetti-like) arc-discharge single-walled carbon nanotubes serving as a saturable absorber based on evanescent field coupling. Stable passively Q-switched(PQS) operation was achieved. The PQS60 μm diameter WG laser generated a record output power of 150 m W at 1846.8 nm with η = 34.6%. The conversion efficiency with respect to the CW mode was 87.6%. The best pulse characteristics(energy/duration)were 105.6 nJ/98 ns at a repetition rate of 1.42 MHz.

Chronic exposure to excess iron promotes EMT and cancer via p53 loss in pancreatic cancer

Based on the evidence that hemochromatosis, an iron-overload disease, drives hepatocellular carcinoma, we hypothesized that chronic exposure to excess iron, either due to genetic or environmental causes, predisposes an individual to cancer. Using pancreatic cancer as our primary focus, we employed cell culture studies to interrogate the connection between excess iron and cancer, and combined in vitro and in vivo studies to explore the connection further. Ferric ammonium citrate was used as an exogenous iron source. Chronic exposure to excess iron induced epithelial-mesenchymal transition(EMT) in normal and cancer cell lines, loss of p53, and suppression of p53 transcriptional activity evidenced from decreased expression of p53 target genes(p21, cyclin d1, Bax, SLC7A11). To further extrapolate our cell culture data, we generated EL-KrasG12d( EL-Kras) mouse(pancreatic neoplastic mouse model) expressing Hfe+/+ and Hfe-/- genetic background. p53 target gene expression decreased in EL-Kras/Hfe-/- mouse pancreas compared to EL-Kras/Hfe+/+ mouse pancreas. Interestingly, the incidence of acinar-to-ductal metaplasia and cystic pancreatic neoplasms(CPN) decreased in EL-Kras/Hfe-/- mice, but the CPNs that did develop were larger in these mice than in EL-Kras/Hfe+/+ mice. In conclusion, these in vitro and in vivo studies support a potential role for chronic exposure to excess iron as a promoter of more aggressive disease via p53 loss and SLC7A11 upregulation within pancreatic epithelial cells.

目的分析社会隔离量表(Social Isolation Scale,SIS)在葡萄牙社区老年人中应用的信度和效度。方法选取250名社区老年人(≥65岁)进行横向描述性调查研究。调查工具包括社会人口学问卷、SIS、老年抑郁量表-4 (Geriatric depression Scale-4,GdS-4)、6项卢本社会网络量表(6-item Lubben Social Network Scale,LSNS-6)、孤独自我评估问题和生活满意度量表(Satisfaction with Life Scale,SWLS)。分析结构效度(验证性因素分析)和收敛效度,评估ISI内部信度(综合信度)、外部信度(重测、组内相关系数)和评分者间信度(Cohen's kappa系数)。结果验证性因素分析显示双因素模型具有良好的拟合指数。SIS得分与LSNS-6(r=0.47)、SWLS(r=0.26)、孤独自我评估(r=0.35)和GdS-4(r=-0.16)得分相关。SIS组合信度良好(0.708)。组内相关系数0.84～0.98。Cohen's kappa系数为0.936～1。结论 SIS可作为评估葡萄牙社区老年人社会隔离的有效和可靠工具。医疗保健专业人员,特别是在社区工作的护士,可使用SIS评估老年人的社会隔离状况,以设计、实施和评估干预措施。
