Designing Cyclopentapeptide Inhibitor of Neuraminidase H5N1 Virus Through Molecular and Pharmacology Simulations

Highly Pathogenic Avian Influenza（HPAI） H5N1 has attracted much attention as a potential pandemic virus in humans, which makes death inevitable in humans. Neuraminidase（NA） has an important role in viral replication. Thus, it is an attractive target when designing anti-influenza virus drug. However, evolving viruses cause some anti-viral drugs to be ineffective, as they show resistance to them. Selection of peptides as drug candidates is important for the peptide-receptor activity and good selectivity. Cyclic bonds in the peptide ligand design aim to improve the stability of the system and remove the obstacles in drug metabolism. The design is based on the polarity of the ligand and amino acid residues in the active site of NA. The results are 4200 cyclic pentapeptides as potential lead compounds. Docking simulations were conducted using MOE 2008.10 and were screened based on the value of the binding energy（？Gbinding）. ADME-Tox prediction assay was conducted on the selected ***- and inter-molecular interactions, as well as changes in the form of bonds, were tested by molecular dynamics simulations at temperatures of 310 K and 312 K. The results of the docking simulations and toxicity prediction assay show that there are two ligands that have a residual interaction with the target protein： CLDRC and CIWRC. These two ligands have ？Gbindingvalues of –40.5854 and –39.9721 kcal/mol（1 kcal/mol = 4.18 k J/mol）. These ligands are prone to be mutagenic and carcinogenic, and they have a good oral bioavailability. The results show that the molecular dynamics of both ligand CLDRC and CIWRC are more feasible at the temperature of 312 K. At the end,both CIWRC and CLDRC ligands can be used as the drug candidates against H5N1 virus.

Engineering magnetic nanostructures with inverse hysteresis loops

Top-down lithography techniques allow the fabrication of nanostructured elements with novel spin configurations, which provide a new route to engineer and manipulate the magnetic response of sensors and electronic devices and understand the role of fundamental interactions in materials science. In this study, shallow nanostructure-patterned thin films were designed to present inverse magnetization curves, i.e., an anomalous magnetic mechanism characterized by a negative coercivity and negative remanence. This procedure involved a method for manipulating the spin configuration that yielded a negative coercivity after the patterning of a single material layer. Patterned NiFe thin films with trench depths between 15%-25% of the total film thickness exhibited inverse hysteresis loops for a wide angular range of the applied field and the trench axis. A model based on two exchange-coupled subsystems accounts for the experimental results and thus predicts the conditions for the appearance of this magnetic behavior. The findings of the study not only advance our understanding of patterning effects and confined magnetic systems but also enable the local design and control of the magnetic response of thin materials with potential use in sensor engineering.

Coronavirus disease 2019(COVID-19)is a viral infection caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2;formerly designated as 2019-nCoV),a novel betacoronavirus firstly identified during a burst of respiratory illness cases in Wuhan City,Hubei Province,China.1 Unfortunately,within a few weeks,the SARS-COV2 virus started to spread globally,producing a pandemic of an extremely spreadable and potentially fatal disease,becoming a cause of great concern for global public health.1 Despite the current estimates of COVID-19 case fatality rate suggest that this coronavirus is less deadly than other pathogens driving other large-scale outbreaks,such as SARS,Middle East respiratory syndrome,or Ebola,the main concern is that this infection is able to spread more easily than other diseases,including seasonal influenza.

Early detection of tomato bacterial canker by reflectance indices

Early detection of infected asymptomatic plants could prevent the dissemination of tomatobacterial canker throughout the crop and, thus, reduce associated economic losses. Reflectance spectroscopy has been proposed for the detection of diseases in plants, mainlycaused by foliar fungal pathogens. In contrast, bacterial canker is a vascular disease thatdoes not produce visible foliar symptoms until it is too late to avoid contagion to otherplants. The effects of the disease on the conductivity of the xylem have an impact onthe water content of the leaves;initially, a subtle effect that cannot be assessed with thenaked eye but can be evaluated by reflectance spectroscopy. The objective of this workwas to identify indices, based on the reflectance spectral signature of the plants, for thedetection of tomato plants affected by bacterial canker prior to the appearance of symptoms. To verify that the proposed methodology is applicable to a broad range of tomatogenotypes, two experiments were carried out with different tomato cultivars grown in potsand another on the ground in a greenhouse. Plants were inoculated with the pathogenicbacteria and the reflectance spectrum of the apical leaflet of two leaves per plant wereobtained before symptoms expression;several spectral indices were calculated from thereflectance spectra. Three indices, of the shortwave-infrared zone, allowed the detectionof bacterial canker inoculated plants in a fast and non-destructive way, up to one weekbefore symptoms were visible: Normalized Difference Water Index (NDWI), Simple Ratioof Water Index (SRWI) and Water Index 1 180 (WI1180). Our research demonstrates, forthe first time, the usefulness of spectral indices sensitive to water content for the identifi-cation of diseased plants. These indices arise as a promising tool for the early detection ofvascular plant diseases, that could be integrated into sustainable management plans.

Myostatin(MSTN) is one of the key factors regulating myogenesis. Because of its role as a negative regulator of muscle mass deposition, much interest has been given to its protein and, in recent years, several studies have analysed MSTN gene regulation. This review discusses the MSTN gene promoter, focusing on its structure in several animal species, both vertebrate and invertebrate. We report the important binding sites considering their degree of phylogenetic conservation and roles they play in the promoter activity. Finally, we discuss recent studies focusing on MSTN gene regulation via promoter manipulation and the potential applications they have both in medicine and agriculture.

Workers'task specialization and division of labor are critical features of social insects'ecological *** has been proposed that the division of labor relies on response threshold models:individuals varying their sensitivity(and responsiveness)to biologically relevant stimuli and performing a specific task when a stimulus exceeds an internal *** this work,we study carbohydrate and protein responsiveness and their relation to worker task specialization in Vespula germanica,an invasive social *** sucrose and peptone responsiveness of two different subcastes,preforagers and foragers,was determined by stimulating the antenna of the wasps with increasing concentrations of the solution and quantifying whether each concentration elicited a licking *** studied responsiveness in five different ways:(1)response threshold,(2)concentration 50(concentration to which at least 50%of wasps responded),(3)maximum response,(4)mean scores and(5)median *** results suggest that V germanica foragers are more sensitive to sucrose(lower thresholds)than preforager ***,we found no differences for peptone thresholds(i.e.,a protein resource).Nonetheless,this is the first study to investigate response thresholds for protein *** intercaste variation in sucrose responsiveness shown in our work contributes to the existing knowledge about response threshold theory as a mechanism for task specialization observed in V germanica.

Sensitivity analysis is a powerful method for discovering the significant factors that contribute to understanding the interaction between variables in multivariate datasets. A number of sensitivity analysis methods fall into the class of local analysis, in which the sensitivity is defined as the partial derivatives of a target variable with respect to a group of independent variables.
