Photooxidative stress,when combined with elevated temperatures,triggers various defense mechanisms leading to physiological,biochemical,and morphological changes in fruit tissue.Furthermore,during sun damage,apple fruit undergo textural changes characterized by high flesh firmness compared to unexposed fruit.Fuji and Royal Gala apples were suddenly exposed to sunlight on the tree and then sampled for up to 29 days.Cell wall components and lignin biosynthetic pathway analyses were carried out on the fruit tissue.At harvest,Fuji apples with different sun exposure levels,such as exposed to direct sunlight(Exp),shaded(Non-Exp),and with severe sun damage(Sev),were also characterized.In fruit suddenly exposed to sunlight,the expression levels of phenylpropanoid-related genes,phenylalanine ammonia lyase(MdPAL),chalcone synthase(MdCHS),and flavanone-3-hydroxylase(MdF3H),were upregulated in the skin and flesh of Exp and Sev.Exposure had little effect on the lignin-related genes caffeic acid Omethyltransferase 1(MdCOMT1)and cinnamyl alcohol dehydrogenase(MdCAD)in the skin;however,the expression of these genes was highly induced in the flesh of Exp and Sev in both cultivars.Lignin deposition increased significantly in skin with sun injury(Sev);in flesh,this increase occurred late during the stress treatment.Additionally,the ethylene biosynthesis genes 1-aminocyclopropane-1-carboxylate synthase(MdACS)and 1-aminocyclopropane-1-carboxylate oxidase(MdACO)were highly expressed in the skin and flesh tissues but were more upregulated in Sev than in Exp during the time-course experiment,which paralleled the induction of the phenylpropanoid pathway and lignin accumulation.At harvest,flesh from Sev fruit exhibited higher firmness than that from Non-Exp and Exp fruit,although no differences were observed in the alcohol-insoluble residues(AIR)among groups.The fractionation of cell wall polymers revealed an increase in the uronic acid contents of the water-soluble pectin fraction(WSF)in Exp and Sev tissues compared to Non-Exp tissues,wh

We present a fast Galerkin spectral method to solve logarithmic singular equations on segments. The proposed method uses weighted first-kind Chebyshev polynomials. Conver- gence rates of several orders are obtained for fractional Sobolev spaces H^-1/2 （or H00^-l/2）. Main tools are the approximation properties of the discretization basis, the construction of a suitable Hilbert scale for weighted L2-spaces and local regularity estimates. Background:In the context of the search for cost-efficient treatments,Ulmo(Eurcyphia cordifolia)honey is an excellent alternative for treating burn wounds and could have a profound medical,social,and economic impact.Ascorbic acid is an enzymatic co-factor necessary for the synthesis of collagen and the proliferation of fibroblasts and has been proposed as a coadjuvant to strengthen the healing effects of honey.The aim of this work was to evaluate by morphometric studies the healing wounds caused by burns treated with Ulmo honey alone and supplemented with ascorbic acid in guinea pig(Cavia porcellus).Methods:Fifteen guinea pigs were used and divided randomly into three groups:positive control(C+),experimental with unsupplemented honey(H),and experimental with supplemented honey(SH).A uniform deep burn covering 1 cm2of the back skin was performed.The following indices were calculated for the morphometric study:superficial contraction index of the wound,deep contraction index of the wound,wound severity index,global healing index,global contraction index,and dermal proliferation area.Results:The superficial contraction index of the wound,global healing index,global contraction,and dermal proliferation area values of the experimental with supplemented honey group were higher than the other groups(P<0.05).Conclusions:According to these results,the combination of honey with an antioxidant(ascorbic acid)promotes an appropriate action to support the healing effect.This study showed that by supplementing the Ulmo honey with ascorbic acid,the healing and contraction effects can be strengthened in burn wounds compared to unsupplemented honey.These results were proof of the synergy between honey and ascorbic acid in healing burn wounds.

Variable retention harvesting: conceptual analysis according to different environmental ethics and forest valuation

Background:Conceptual clarity is important to attain precise communication of scientific knowledge and to implement appropriate technological and policy actions.Many concepts referring to forest management are widely used by decision-makers,regardless of their complexity.Although the scientific and methodological issues of forestry practices are frequently discussed in the literature,their normative dimensions are rarely treated.Thus,linguistic uncertainty increases when different environmentally ethical perspectives and ways of valuing forests are considered.The objective was to compare different conceptualizations on the silvicultural systems suggested for forest management and the implications they have for conservation.We have conceptually contrasted highintensity forestry practices with variable retention harvesting,considering different environmentally ethical perspectives and forest valuation alternatives.Results:Clear boundaries between clear-cutting,selective logging,and variable retention harvesting can be evidenced when different ethical points of view and alternatives in the human-nature relationships are considered.We have found a variety of definitions of variable retention harvesting that can be analyzed under different ethical positions.Sharply contrasting views on variable retention harvesting can be evidenced if nature is considered to be purely at human’s service or if it is conceptualized as humans co-inhabiting with nature.The latter position implies that the maintenance of ecological,evolutionary,and historical processes supported by unmanaged forest stands is a crucial step for forest management proposals based on variable retention harvesting.Conclusions:Forestry practices that are focused on forest yields and that misinterpret functional uncertainty of forest functioning would be risky.Moreover,forestry with variable retention harvesting could imply good yields with reasonable conservation management in some contexts,while it could be unacceptable in other socioecological contexts.The imp

Prevalence of Acute Myocardial and Brain Toxicity in Emergency Department Patients Exposed to Carbon Monoxide

Introduction: Due to its severe toxicity, carbon monoxide poisoning is an emergency that leads to cardiac and brain involvement, and emergency physicians should aim to master this diagnosis. This study is intended to describe the expected prevalence of these severe toxicities in the emergency department. Materials and Methods: A retrospective cohort of consecutive patients over 16 years of age presenting with carbon monoxide poisoning to the emergency department of the Hospital Universitario Austral, Argentina, during the period from January 2018 to June 2022. The prevalence of myocardial and brain toxicity was assessed regarding percentage, continuous variables with mean and standard deviation, categorical variables with percentage and absolute frequency. Positive findings of acute neurological toxicity included seizures, syncope and coma, while cardiovascular toxicity encompassed acute myocardial infarction (MI) diagnosed by electrocardiogram or elevated troponin levels, arrhythmias, and the development of pulmonary edema/ congestive heart failure (CHF) confirmed by chest X-ray with suggestive signs or clinically compatible symptoms. Patients were followed-up for 90 days to estimate hospitalization and mortality. Results: A total of 67 patients were evaluated;44.77% of them were males with a mean age of 38.5 ± 14.97 years. The prevalence of acute myocardial toxicity was 7.46% (n: 5). Among these patients, 3 had overweight as a risk factor, and 3 showed ECG abnormalities with negative T-waves. The prevalence of acute brain toxicity was 8.9% (n: 6). In total, 37% (n: 25) of patients met the criteria for hyperbaric oxygen therapy, of which 32% (n: 8) underwent more than one session. A total of 7.46% of patients (n: 5) required hospitalization. Mortality at 90 days was 0%. Conclusion: Among the patients who presented to the emergency department, the prevalence of acute myocardial toxicity was 7.46% (n: 5), which is lower than the prevalence reported in other studies to date. A Brief Review on Hydroxyapatite Nanoparticles Interactions with Biological Constituents

With the pursuit of new cancer therapies and more effective treatment to diseases in the last decades, nanotechnology has been an important ally for healthcare professionals and patients in critical clinical conditions. Nanomaterials offer an alternative way to deliver toxic chemotherapeutic drugs to specific biological tissues, specific cells or specific microbial beings, resulting in avoidance of strong side effects or resilience to effective drugs. Among these materials, stands out the hydroxyapatite nanoparticles, a ceramic class of calcium phosphates that present chemical and structural similarities with the mineral phase of the human skeleton’s bone matrix, resulting in important biological features, such as biocompatibility, osteoconductive, osteoinduction and osteoaffinity, which led to a lot of scientific researches to apply these nanoparticles for bone diseases diagnosis and therapeutics. Due to the hydroxyapatite biological activities and due to the possibility to promote chemical and physical modifications in these nanoparticles, they can interact with biological cells or microorganisms in different ways, resulting in multiple potentialities to be explored such as apoptosis induction to cancerous cells, osteogenesis promotion, cellular proliferation, angiogenesis and tissue recovery, in addition to promote cell adhesion and cell uptake. Furthermore, chemical and physical modifications, such as surface functionalization, dopant inclusions and radiolabeling process, allow scientists to track the particle activities in biological environments. In the last decades of scientific productions, the literature brings together important data on how hydroxyapatite nanoparticles interact with biological tissues and such data are crucial for the development of more effective therapeutic and diagnostic agents. Pancreatic cancer(PC)continues to pose a major clinical challenge.There has been little improvement in patient survival over the past few decades,and it is projected to become the second leading cause of cancer mortality by 2030.The dismal 5-year survival rate of less than 10%after the diagnosis is attributable to the lack of early symptoms,the absence of specific biomarkers for an early diagnosis,and the inadequacy of available chemotherapies.Most patients are diagnosed when the disease has already metastasized and cannot be treated.Cancer interception is vital,actively intervening in the malignization process before the development of a full-blown advanced tumor.An early diagnosis of PC has a dramatic impact on the survival of patients,and improved techniques are urgently needed to detect and evaluate this disease at an early stage.It is difficult to obtain tissue biopsies from the pancreas due to its anatomical position;however,liquid biopsies are readily available and can provide useful information for the diagnosis,prognosis,stratification,and follow-up of patients with PC and for the design of individually tailored treatments.The aim of this review was to provide an update of the latest advances in knowledge on the application of carbohydrates,proteins,cell-free nucleic acids,circulating tumor cells,metabo-lome compounds,exosomes,and platelets in blood as potential biomarkers for PC,focusing on their clinical relevance and potential for improving patient outcomes.

Practice change in the management of metastatic urothelial carcinoma after ASCO 2020

Metastatic urothelial carcinoma(mUC)is an incurable and aggressive disease.In the past decades there have been few effective treatment options that have impacted the prognosis of mUC patients.However,in the last few years,several drugs have emerged as new treatment choices that are changing the therapeutic landscape of mUC.Immune checkpoint inhibitors(ICIs)and targeted agents are useful treatment strategies that have been incorporated into our clinical practice.Nevertheless,cisplatin-based chemotherapy is still the standard of care in the first-line of metastatic disease.The results of the JAVELIN Bladder 100 phase 3 trial were presented at ASCO 2020,this trial evaluated the role of avelumab,an ICI,as maintenance therapy in patients who had not progressed after first-line platinum-based chemotherapy.The trial met its primary endpoint demonstrating an overall survival benefit with avelumab maintenance.In addition,new drugs and combinations are being evaluated to improve the outcomes of second and subsequent lines.Fibroblast growth factor receptor(FGFR)inhibitors and immunotherapy combinations were some of the strategies presented at ASCO 2020 that have shown promising results.Finally,the development of predictive biomarkers that help us in the decision-making process will be one of the most important challenges in the next years.

Non-invasive diagnosis of gastric mucosal atrophy in an asymptomatic population with high prevalence of gastric cancer

AIM: To validate a non-invasive method to detect gastric mucosal atrophy in a Chilean population with high prevalence of gastric cancer and a poor survival rate. METHODS: We first determined the optimal cut-off level of serum pepsinogen (PG)-1, PG-1/PG-2 ratio and 17-gastrin in 31 voluntary symptomatic patients (mean age: 66.1 years), of them 61% had histologically confirmed gastric atrophy. Then, in a population-based sample of 536 healthy individuals (209 residents in counties with higher relative risk and 327 residents in counties with lower relative risk for gastric cancer), we measured serum anti-H pylori antibodies, PG and 17-gastrin and estimated their risk of gastric cancer. RESULTS: We found that serum PG-1 13.3 pmol/L had a high specificity (91%-100%) and a fair sensitivity (56%-78%) to detect corpus-predominant atrophy. Based on low serum PG-1 and PG-1/PG-2 ratio together as diagnostic criteria, 12.5% of the asymptomatic subjects had corpus-predominant atrophy (0% of those under 25 years and 20.2% over 65 years old). The frequency of gastric atrophy was similar (12% vs 13%) but H pylori infection rate was slightly higher (77% vs 71%) in the high-risk compared to the low-risk counties. Based on their estimated gastric cancer risk, individuals were classified as: low-risk group (no H pylori infection and no atrophy; n = 115; 21.4%); moderate-risk group(H pylori infection but no atrophy; n = 354, 66.0%); and high-risk group (gastric atrophy, with or without H pylori infection; n = 67, 12.5%). The high-risk group was significantly older (mean age: 61.9 ± 13.3 years), more frequently men and less educated as compared with the low-risk group. CONCLUSION: We propose to concentrate on an upper gastrointestinal endoscopy for detection of early gastric cancer in the high-risk group. This intervention model could improve the poor prognosis of gastric cancer in Chile.

Chronic exposure to excess iron promotes EMT and cancer via p53 loss in pancreatic cancer

Based on the evidence that hemochromatosis, an iron-overload disease, drives hepatocellular carcinoma, we hypothesized that chronic exposure to excess iron, either due to genetic or environmental causes, predisposes an individual to cancer. Using pancreatic cancer as our primary focus, we employed cell culture studies to interrogate the connection between excess iron and cancer, and combined in vitro and in vivo studies to explore the connection further. Ferric ammonium citrate was used as an exogenous iron source. Chronic exposure to excess iron induced epithelial-mesenchymal transition(EMT) in normal and cancer cell lines, loss of p53, and suppression of p53 transcriptional activity evidenced from decreased expression of p53 target genes(p21, cyclin D1, Bax, SLC7A11). To further extrapolate our cell culture data, we generated EL-KrasG12D( EL-Kras) mouse(pancreatic neoplastic mouse model) expressing Hfe+/+ and Hfe-/- genetic background. p53 target gene expression decreased in EL-Kras/Hfe-/- mouse pancreas compared to EL-Kras/Hfe+/+ mouse pancreas. Interestingly, the incidence of acinar-to-ductal metaplasia and cystic pancreatic neoplasms(CPN) decreased in EL-Kras/Hfe-/- mice, but the CPNs that did develop were larger in these mice than in EL-Kras/Hfe+/+ mice. In conclusion, these in vitro and in vivo studies support a potential role for chronic exposure to excess iron as a promoter of more aggressive disease via p53 loss and SLC7A11 upregulation within pancreatic epithelial cells.
