Estimation of the carbon pool in soil and above-ground biomass within mangrove forests in southeast Mexico using allometric equations

We report the results of carbon stored in soil and aboveground biomass from the most important area of mangroves in Mexico, with dominant vegetation of Red mangrove （Rhizophora mangle L.）, Black mangrove （Avicennia germinans L.）, white mangrove （Laguncularia racemosa Gaertn.） and button mangrove （Conocarpus erectus L.）. We sampled soils with high fertility during the dry season in 2009 and 2010 at three sites on Atasta Peninsula, Campeche. We used allometric equations to estimate above ground biomass （AGB） of trees. AGB was higher in C. erectus （253.18±32.17 t？ha-1）, lower in A. germinans （161.93±12.63 t？ha-1）, and intermediate in R. mangle （181.70±16.58 t？ha-1） and L. racemosa （206.07±19.12 t？ha-1）. Of the three studied sites, the highest absolute value for AGB was 279.72 t？ha-1 in button mangrove forest at any single site. Carbon stored in soil at the three sites ranged from 36.80±10.27 to 235.77±66.11 t？ha-1. The Tukey test （p 〈0.05） made for AGB was higher for black mangrove showed significant differences in soil carbon content between black mangrove and button mangrove. C. erectus had higher AGB compared with the other species. A. germinans trees had lower AGB because they grew in hypersaline environments, which reduced their development. C. erectus grew on higher ground where soils were richer in nutrients. AGB tended to be low in areas near the sea and increased with distance from the coast. A. germinans usually grew on recently deposited sediments. We assumed that all sites have the same potential to store carbon in soil, and then we found that there were no significant differences in carbon content between the three samples sites： all sites had potential to store carbon for long periods. Carbon storage at the three sampling sites in the state of Campeche, Mexico, was higher than that reported for other locations.

Carbamoyl phosphate synthetase i （CPs1） deficiency （CPs1D） is an inborn error of the urea cycle having autosomal （2q34） recessive inheritance that can cause hyperammonemia and neonatal death or mental retardation. We analyzed the effects on CPs1 activity, kinetic parameters and enzyme stability of missense mutations reported in patients with CPs1 deficiency that map in the 20-kDa C-terminal domain of the enzyme. This domain turns on or off the enzyme depending on whether the essential allosteric activator of CPs 1, N-acetyl- L-glutamate （NAG）, is bound or is not bound to it. To carry out the present studies, we exploited a novel system that allows the expression in vitro and the purification of human CPs1, thus permitting site-directed mutagenesis. These studies have clarified disease causation by individual mutations, identifying functionally important residues, and revealing that a number of mutations decrease the affinity of the enzyme for NAG. Patients with NAG affinity-decreasing mutations might benefit from NAG site saturation therapy with N-carbamyl-L- glutamate （a registered drug, the analog of NAG）. Our results, together with additional present and prior site-directed mutagenesis data for other residues mapping in this domain, suggest an NAG-triggered conformational change in the 134-~4 loop of the C-terminal domain of this enzyme. This change might be an early event in the NAG activation process. Molecular dynamics simulations that were restrained according to the observed effects of the mutations are consistent with this hypothesis, providing further backing for this structurally plausible signaling mechanism by which NAG could trigger urea cycle activation via CPs1.

BACKGROUND Bowel preparation in children can be *** To describe the efficacy, safety, and tolerability of sodium picosulfate, magnesium oxide, and citric acid(sPMC) bowel preparation in *** Phase 3, randomized, assessor-blinded, multicenter study of low-volume, divided dose sPMC enrolled children 9-16 years undergoing elective colonoscopy. Participants 9-12 years were randomized 1:1:1 to sPMC ? dose × 2, sPMC 1 dose × 2, or polyethylene glycol(PEG). Participants 13-16 years were randomized 1:1 to sPMC 1 dose × 2 or PEG. PEG-based bowel preparations were administered per local protocol. Primary efficacy endpoint for quality of bowel preparation was responders(rating of ‘excellent' or ‘good') by modified Aronchick scale. secondary efficacy endpoint was participant's tolerability and satisfaction from a 7-item questionnaire. safety assessments included adverse events(AEs) and laboratory *** 78 participants were randomized, 48 were 9-12 years, 30 were 13-16 years. For the primary efficacy endpoint in 9-12 years, 50.0%, 87.5%, and 81.3% were responders for sPMC ? dose × 2, sPMC 1 dose × 2, and PEG groups, respectively. Responder rates for 13-16 years were 81.3% for sPMC 1 dose × 2 and 85.7% for PEG. Overall, 43.8% of participants receiving sPMC 1 dose × 2 reported it was ‘very easy' or ‘easy' to drink, compared with 20.0% receiving PEG. Treatment-emergent AEs were reported by 45.5% of participants receiving sPMC 1 dose × 2 and 63.0% receiving *** sPMC was an efficacious and safe for bowel preparation in children 9-16 years, with comparable efficacy to PEG. Tolerability for sPMC was higher compared to PEG.

Prophylaxis with ketotifen in rats with portal hypertension:involvement of mast cell and eicosanoids

BACKGROUND:since we have previously shown an increase of mast cells in the small bowel and in the mesenteric lymph nodes in the rats with prehepatic portal hypertension,it can be hypothesized that this essential inflammatory cell would be involved in the pathogeny of the splanchnic changes related to portal hypertension. METHODs:To verify this hypothesis,we first studied mast cell infiltration in the ileum and in the mesenteric lymph nodes in sham-operated male Wistar rats(n=12) and in short-term prehepatic portal hypertensive rats (n=12),and the serum levels of rat mast cell protease Ⅱ(RMCP-Ⅱ)by *** a second set of experiments ketotifen,a mast cell stabilizer drug,was administered to sham-operated(n=10)and portal hypertensive(n=12) rats 24 hours before the intervention and prostanoids (PGE2,PGI2,TXB2)and leukotrienes(LTC4,LTB4)were assayed by RIA,mast cell infiltration in the ileum and in the mesenteric lymph nodes and the serum levels of RMCP-Ⅱwere also studied,to show its effectiveness to prevent the mesenteric alterations produced by the inflammatory mediators released by the mast cell. REsULTs:Forty-eight hours after the intervention RMCP-Ⅱ (Pserum levels decreased and mast cell number and RMCP-Ⅱlevels increased in mesenteric lymph nodes in portal hypertensive *** administration of ketotifen reduced portal pressure(Pserum levels of PGE2(Psenteric lymph nodes. CONCLUsIONs:In acute portal hypertension in the rat,the mast cell translocation from intestinal mucosa to mesenteric lymph nodes,where they are activated and degranulates,would represent a defence mechanism to avoid the activation of an acute and massive inflammatory response in this *** administration of ketotifen is able to reduce the splanchnic inflammatory changes related to acute portal hypertension in the rat.

A comparison of two methods to assess audience-induced changes in male mate choice

Multidirectional communicative interactions in social networks can have a profound effect on mate choice behavior. Male Atlantic molly Poecilia mexicana exhibit weaker mating preferences when an audience male is presented. This could be a male strategy to reduce sperm competition risk： interacting more equally with different females may be advantageous because ri- vals might copy mate choice decisions. In line with this hypothesis, a previous study found males to show a strong audience effect when being observed while exercising mate choice, but not when the rival was presented only before the choice tests. Audience effects on mate choice decisions have been quantified in poeciliid fishes using association preference designs, but it remains un- known if patterns found from measuring association times translate into actual mating behavior. Thus, we created five audience treatments simulating different forms of perceived sperm competition risk and determined focal males＇ mating preferences by scoring pre-mating （nipping） and mating behavior （gonopodial thrusting）. Nipping did not reflect the pattern that was found when association preferences were measured, while a very similar pattern was uncovered in thrusting behavior. The strongest response was observed when the audience could eavesdrop on the focal male＇s behavior. A reduction in the strength of focal males＇ preferences was also seen after the rival male had an opportunity to mate with the focal male＇s preferred mate. In comparison, the reduction of mating preferences in response to an audience was greater when measuring association times than actual mating behavior. While measuring direct sexual interactions between the focal male and both slimulus females not only the male＇s motivational state is reflected but also females＇ behavior such as avoidance of male sexual harassment [Current Zoology 58 （1）： 84-94, 2012].

Chronic model of tympanic perforation in rats with mitomycin C and dexamethasone

A rat model of chronic tympanic membrane perforation was developed to be used in the search of new materials for the sealing of these perforations. A longitudinal study was carried out in rats subjected to incisional myr-ingotomy followed by the application of mitomycin C alone or with dexamethasone. Rats were checked at days 3, 7,1 0,1 4 and weekly thereafter until perforation closure, for up to 6 months. The addition of dexamethasone is a key component in order to obtain a chronic opening. Myringotomies treated w ith saline had a mean healing time of 8.5 days. At 8 weeks, between 62.5% and 77.7% of tympanic membranes treated w ith mitomycin C and dexamethasone remained perforated and at 6 months this number fell to 21.4%. This technique is able to maintain most tympanic membrane perforations patent for at least 8 weeks. This rat model is adequate for its use in preclinical or translational research.
