Unveiling plant defense arsenal: metabolic strategies in Brassica oleracea during black rot disease

Alterations in plant metabolism play a key role in the complex plant-pathogen ***,there is still a lack of knowledge about the connection between changes in primary and specialized metabolism and the plant defense against diseases that impact ***,we aim to study the metabolic reprograming in Brassica oleracea plants upon infection by Xanthomonas campestris ***(Xcc).To accomplish this,we utilized a combination of untargeted and targeted metabolomics,through UPLC-Q-TOF-MS/MS and 1H-NMR,in two crop lines differing in resistance thatwere evaluated at two-and four-week intervals following inoculation(T1 and T2,respectively).Besides,to depict the physiological status of the plant during infection,enzymatic activities related to the carbohydrate pathway and oxidative stress were *** results revealed different temporal dynamics in the responses of the susceptible *** crops *** *** line suppresses carbohydrate metabolism contributing to limit nutrient supplies to the bacterium and prioritizes the induction of defensive compounds such as indolic glucosinolates,salicylic acid,phenylpropanoids and phytoalexins precursors at early infection *** contrast,the susceptible line invests in carbohydratemetabolism,including enzymatic activities related to the hexoses turnover,and activates defense signaling related to reactive oxygen ***,each line triggers a different metabolic strategy that will affect how the plant overcomes the disease in terms of resistance and *** work provides first insights of a fine-tuned metabolic regulation during Xcc infection in *** that will contribute to develop new strategies for plant disease management.

The present work reports the discovery and the complete characterisation of an ancient cultivated rose variety found growing in a private garden in the southwest of the Principality of Asturias(northern Spain).The variety is here given the name *** majority of roses currently cultivated belong to the so-called group of‘Modern Roses’,all of which were obtained after 1867 via artificial crosses and improvement *** are destined for ornamental *** the 19th century,the great majority of the many ancient cultivated roses in Europe were used in perfumery and cosmetics,or had medicinal *** damascena and Rosa centifollia are still grown and used by the French and Bulgarian perfume *** Asturian Massif of the Cantabrian Mountain Range provides a natural habitat for some 75%of the wild members of the genus Rosa,but until now there was no evidence that this area was home to ancient cultivated roses.A complete botanical description is here provided for a discovered ancient *** is also characterised according to a series of sequence tagged microsatellite sites,and its agronomic features are *** addition,a histological description(optical and scanning electronic microscope studies)of the petals is offered,along with an analysis of the volatile compounds present in these organs as determined by solid phase microextraction and gas chromatography-mass *** results reveal the uniqueness of this ancient type of rose and suggest it may be of interest to the perfume industry.

Lichens are symbiotic organisms that comprise a fungus and a photosynthetic partner wich are recognized as a good indicator of climate ***,our understanding of how aridity affects the diversity of saxicolous lichens in drylands is still *** evaluate the relationship between saxicolous lichen diversity and aridity in a central México dryland,a geographical transect was established of 100 km to build an aridity gradient in the semiarid zone of the State of Querétaro,Mexico,comprising ten sampling sites with a 10 km *** richness,abundance and diversity of soil lichen species were recorded using two sampling methods:the quadrat-intercept and the line-intercept method,to compare their performance in assessing soil lichen diversity in *** number of species and Shannon diversity of saxicolous lichens were higher at intermediate values of the aridity index(AI=0.10-0.34).Quadrat intercept and point intercept methods gave quite similar results,which means that the selected method does not influence the results in a significant *** study confirms the role of saxicolous lichens as climate change indicators and reveals the importance of the sampling method selection in the evaluation of different parameters of soil lichen diversity in drylands.

The Mediterranean Sea is the largest and deepest enclosed sea on Earth covering 0.7% of the world's ocean *** is one of the major reservoirs of marine and coastal biodiversity,with 17,000 species corresponding to the 7.5% of the world's marine fauna and 18% of its marine flora and 28% of endemic *** basin is also subjected to a strong human pressure due to a high degree of urbanisation and tourism in the coastal areas and several activities such as shipping,gas and oil extraction,fishing,and *** this contest ARs represent a tool for protecting sensitive habitats from fishing activities;restoring depleted habitats;enhancing biodiversity improving populations of aquatic organisms by providing shelter for juvenile and mature individuals as well as for adults during delicate life stages(e.g.,moulting season for crustaceans);enhancing professional fisheries;providing a mean to manage coastal activities and reduce conflicts and,more recently,developing recreational activities(e.g.,diving and recreational fishing).Establishment of ARs in the Mediterranean Sea is regulated by international,European,regional and national laws and a few guidelines have been produced at international and national *** hundred and fifty-three ARs have been deployed from 1960 s to present,most of them being realized in 1990 s and 2000 *** highest concentration of ARs is located along the Spanish coast,followed by Italy,Turkey and *** protection and fisheries enhancement have been the most common goals,however the interest towards ARs for recreational activities(i.e.,diving and recreational fishing) has been increasing over the years with the aim of promoting sustainable coastal tourism and reducing the tourists' pressure on the natural *** and modules also evolved over time with the aim of developing multi-functional infrastructures that protect the environment while allowing for a variety of economic activities by local stakeholders.

Detection of KRAS G12D in colorectal cancer stool by droplet digital PCR

AIMTo assess KRAS G12D mutation detection by droplet digital PCR (ddPCR) in stool-derived DNA from colorectal cancer (CRC) *** this study, tumor tissue and stool samples were collected from 70 patients with stage I-IV CRC diagnosed by preoperative biopsy. KRAS mutational status was determined by pyrosequencing analysis of DNA obtained from formalin-fixed paraffin-embedded (FFPE) tumor tissues. The KRAS G12D mutation was then analyzed by ddPCR in FFPE tumors and stool-derived DNA from patients with this point mutation. Wild-type (WT) tumors, as determined by pyrosequencing, were included as controls; analysis of FFPE tissue and stool-derived DNA by ddPCR was performed for these patients as *** the total 70 patients included, KRAS mutations were detected by pyrosequencing in 32 (45.71%), whereas 38 (54.29%) had WT tumors. The frequency of KRAS mutations was higher in left-sided tumors (11 located in the right colon, 15 in the left, and 6 in the rectum). The predominant point mutation was KRAS G12D (14.29%, n = 10), which was more frequent in early-stage tumors (I-IIA, n = 7). In agreement with pyrosequencing results, the KRAS G12D mutation was detected by ddPCR in FFPE tumor-derived DNA, and only a residual number of mutated copies was found in WT controls. The KRAS G12D mutation was also detected in stool-derived DNA in 80% of all fecal samples from CRC patients with this point *** is a reliable and sensitive method to analyze KRAS G12D mutation in stool-derived DNA from CRC patients, especially at early stages. This non-invasive approach is potentially applicable to other relevant biomarkers for CRC management.

Design and Experimentation of a 1 MW Horizontal Axis Wind Turbine

In this work was carried out the aerodynamics design of a 1 MW horizontal axis wind turbine by using blade element momentum theory (BEM). The generated design was scaled and built for testing purposes in the discharge of an axial flow fan of 80 cm in diameter. Strip theory was used for the aerodynamic performance evaluation. In the numerical calculations was conducted a comparative analysis of the performance curves adding increasingly correction factors to the original equation of ideal flow to reduce the error regarding real operating values got by the experimental tests. Correction factors introduced in the ideal flow equation were the tip loss factor and drag coefficient. BEM results showed good approximation using experimental data for the tip speed ratio less than design. The best approximation of the power coefficient calculation was for tip speed ratio less than 6. BEM method is a tool for practical calculation and can be used for the design and evaluation of wind turbines when the flow rate is not too turbulent and radial velocity components are negligible.

Assessment of drug-induced hepatotoxicity in clinical practice: A challenge for gastroenterologists

Currently, pharmaceutical preparations are serious contributors to liver disease; hepatotoxicity ranking as the most frequent cause for acute liver failure and post-commercialization regulatory decisions. The diagnosis of hepatotoxicity remains a difficult task because of the lack of reliable markers for use in general clinical practice. To incriminate any given drug in an episode of liver dysfunction is a step-by-step process that requires a high degree of suspicion, compatible chronology, awareness of the drug’s hepatotoxic potential, the exclusion of alternative causes of liver damage and the ability to detect the presence of subtle data that favors a toxic etiology. This process is time-consuming and the final result is frequently inaccurate. Diagnostic algorithms may add consistency to the diagnostic process by translating the suspicion into a quantitative score. Such scales are useful since they provide a framework that emphasizes the features that merit attention in cases of suspected hepatic adverse reaction as well. Current efforts in collecting bona fide cases of drug-induced hepatotoxicity will make refinements of existing scales feasible. It is now relatively easy to accommodate relevant data within the scoring system and to delete low-impact items. Efforts should also be directed toward the development of an abridged instrument for use in evaluating suspected drug-induced hepatotoxicity at the very beginning of the diagnosis and treatment process when clinical decisions need to be made. The instrument chosen would enable a confident diagnosis to be made on admission of the patient and treatment to be fine-tuned as further information is collected.

Phenolic compounds are related to high biological activity,avoiding oxidation in food and human *** are by-products derived from the pecan nut processing that contain important amounts of phenols which biological activity must be *** research aimed to evaluate the antioxidant(DPPH,ABTS,FRAP and hemolysis)and anti-inflammatory activities of shell extracts from pecan nuts harvested during the crop production cycle 2018 and 2019,as well as the in vitro digestibility of their phenolic compounds,including *** showed that extracts from the crop production cycle 2018 obtained the highest yield,while those from 2019 contained the highest concentration of phenolic compounds,flavonoids and antioxidant capacity determined by DPPH(22.96 mmol ET),ABTS(91.55 mmol ET)and inhibition of hemolysis(92.12%).The anti-inflammatory activity exhibited an inhibition of the elastase enzyme up to 50 min and the bioaccessibility of phenolic compounds reached up to 32%.These results showed that pecan nutshell extracts are an important source of biologically active compounds,thus,they are suitable to be used as commodities in different fields such as agricultural,food and pharmaceutical *** studies must be carried out in order to elucidate the activity of nutshell extracts within in vivo systems.
