Manipulation of bicarbonate concentration in sperm capacitation media improves in vitro fertilisation output in porcine species

Background: The in vivo concentration of bicarbonate(HCO3-), one of the essential sperm capacitating effectors,varies greatly in the different environments sperm go through from cauda epididymis to the fertilisation site. On the contrary, porcine in vitro sperm capacitation and fertilisation media usually contains a standard concentration of25 mmol/L, and one of the main problems presented is the unacceptable high incidence of polyspermy. This work hypothesised that by modifying the HCO3-concentration of the medium, the output of in vitro sperm capacitation and fertilisation could be ***: Once exposed to the capacitation medium, the intracellular pH(pHi) of spermatozoa increased immediately even at low concentrations of HCO3-, but only extracellular concentrations of and above 15 mmol/L increased the substrates protein kinase A phosphorylation(pPKAs). Although with a significant delay, 15 mmol/L of HCO3-stimulated sperm linear motility and increased other late events in capacitation such as tyrosine phosphorylation(Tyr-P) to levels similar to those obtained with 25 mmol/L. This information allowed the establishment of a new in vitro fertilisation(IVF)system based on the optimization of HCO3-concentration to 15 mmol/L, which led to a 25.3% increment of the viable zygotes(8.6% in the standard system vs. 33.9%).Conclusions: Optimising HCO3-concentrations allows for establishing an IVF method that significantly reduced porcine polyspermy and increased the production of viable zygotes. A concentration of 15 mmol/L of HCO3-in the medium is sufficient to trigger the in vitro sperm capacitation and increase the fertilisation efficiency in porcine.

Preventive medicine and food industry have shown an increased interest in the development of natural antioxidants,since those most commonly used synthetic antioxidants may have restricted use in *** could explain why there is currently much research on the antioxidant properties from natural products such as *** mushrooms have been reported to possess antioxidant properties,which enable them to neutralize free *** oxygen molecule is a free radical,which lead to the generation of the reactive oxygen species and can damage the *** damage caused by free radicals appears to be a major contributor to aging and degenerative *** antioxidant components are found in fruit bodies,mycelium and culture both,which include polysaccharides,tocopherols,phenolics,carotenoids,ergosterol and ascorbic acid among *** bodies or mycelium can be manipulated to produce active compounds in a relatively short period of time,which represent a significant advantage in antioxidant compounds extraction from *** compounds may be extracted to be used as functional additives or mushrooms can be incorporated into our food regime,representing an alternative source of food to prevent damage caused by oxidation in the human body.

This paper describes a method based on an energy minimizing deformable model applied to the 3D biomechanical modeling of a set of organs considered as regions of interest (ROI) for radiotherapy. The initial model consists of a quadratic surface that is deformed to the exact contour of the ROI by means of the physical properties of a mass-spring system. The exact contour of each ROI is first obtained using a geodesic active contour model. The ROI is then parameterized by the vibration modes resulting from the deformation process. Once each structure has been defined, the method provides a 3D global model including the whole set of ROIs. This model allows one to describe statistically the most significant variations among its structures. statistical ROI variations among a set of patients or through time can be analyzed. Experimental results are presented using the pelvic zone to simulate anatomical variations among structures and its application in radiotherapy treatment planning.

A new polynomial formulation of variable step size linear multistep methods is pre- sented, where each k-step method is characterized by a fixed set of k - 1 or k parameters. This construction includes all methods of maximal order （p = k for stiff, and p = k ＋ 1 for nonstiff problems）. supporting time step adaptivity by construction, the new formulation is not based on extending classical fixed step size methods; instead classical methods are obtained as fixed step size restrictions within a unified framework. The methods are imple- mented in matLAB, with local error estimation and a wide range of step size controllers. This provides a platform for investigating and comparing different multistep method in realistic operational conditions. Computational experiments show that the new multi- step method construction and implementation compares favorably to existing software, although variable order has not yet been included.

Vaccines and recommendations for their use in inflammatory bowel disease

The patient with inflammatory bowel disease will be predisposed to numerous infections due their immune status. It is therefore important to understand the immune and serologic status at diagnosis and to put the patient into an adapted vaccination program. This program would be applied differently according to two patient groups: the immunocompromised and the non-immunocom-promised. In general, the first group would avoid the use of live-virus vaccines, and in all cases, inflammatory bowel disease treatment would take precedence over vaccine risk. It is important to individualize vaccination schedules according to the type of patient, the treatment used and the disease *** addition, patient with inflammatory bowel disease should be considered for the following vaccines: varicella vaccine, human papilloma virus, influenza, pneumococcal polysaccharide vaccine and hepatitis B vaccine.

Crosslinked hyaluronic acid with liposomes and crocin for management symptoms of dry eye disease caused by moderate meibomian gland dysfunction

One potential drawback of compost-based passive bioreactors,which is a promising biotechnology for acid mine drainage（AMD） treatment,is the transport of dissolved organic matter（DOM）-metal complexes in surface *** address this problem,the objective of this study was to assess the maximum capacity of organic substrates to release soluble DOM-metal complexes in treated water. The reactivities of DOM in maple wood chips and sawdust,composted poultry manure,and leaf compost were quantified toward Cd2＋,Ni2＋,Fe2＋,and Cu2＋ using fluorescence *** DOM showed the highest reactivity toward Fe,but a limited number of available sites for sorption,whereas DOM-Cd complexes exhibited the lowest fluorescence ***,the DOM from a mixture of wastes formed higher concentrations of DOM-metal complexes relative to sole *** DOM-metal complexes, the concentrations of DOM-Ni complexes were the *** reaching steady-state,low concentrations of DOM-metal complexes were released in treated water,which is in agreement with theoretical predictions based on geochemical ***,in addition to physicochemical characterization,fluorescence quenching technique is recommended for the substrate selection of bioreactors.
