Identification and evaluation of antioxidant activity of bioactive casein-derived peptides during in vitro digestion and transepithelial transport in Caco-2 cells

Dairy products are important sources of bioactive(antibacterial,antithrombotic,antihypertensive,immuno-modulatory,and antioxidative)peptides,that need to be released from the target *** the present study,bovine milk casein protein was hydrolyzed by pancreatin and trypsin(4 and 24 h)to evaluate the hydrolyses efficiency(ninhydrin reaction);to elucidate the sequence of casein-derived peptides(SDS-PAGE,LC-ESI-TOF-MS);and to assess the peptides antioxidant activity(DPPH and ORAC),bioactivity in silico,bioaccessibility after in vitro digestion,and bioavailablility by transepithelial transport in Caco-2 *** hydrolysis degree increased with the incubation time,with evident degradation of casein-derived proteins into low molecular weight peptides(<5 kDa)after 4 and 24 h *** showed higher hydrolysis degree(91%)than trypsin(21%).An increase in antioxidant activity was observed with increasing incubation time,with higher antioxidant effect observed for trypsin derived peptides(47%reduction of free radicals;32112μM Trolox equivalents)than pancreatin derived peptides(15%reduction;2862μM Trolox equivalents).A total of 69 ca-seins derived peptides were identified after trypsin(33 peptides)and pancreatin(36 peptides)hydrolysis for 24 h;with 26 peptides released fromβ-casein,21 fromα-s1-casein,15 fromα-s2-casein,and 7 from k-casein *** of these peptides were described as antioxidant,anti-inflammatory,and *** in vitro digestion 123 peptides were found in gastric(56 peptides),duodenal(43 peptides)and colonic(24 peptides)fraction;while five peptides were bioavailable by crossing the intestinal barrier,with increased peptides’abundance after trypsin hydrolysis treatment.

Cold spells and severe weather may have detrimental effects in wild birds,but owing to the stochastic nature of these events,it is not always possible to measure their *** January 2021,a strong snowstorm known as‘Filomena’affected Spain,mainly the region of Madrid(Central Spain),which was fully covered with snow and suffered freezing temperatures for a *** bird surveys conducted in the winter-spring of 2020 and 2021(before and after Filomena)at same locations,we addressed the possible impact of the cold spell in three widespread gamebirds:Wood Pigeon(Columba palumbus),Eurasian Magpie(Pica pica)and Red-legged Partridge(Alectoris rufa).No significant abundance differences were found between years for none of the species and hunting pressure was only significantly reduced for Partridges,hence this species may have been negatively affected by *** suggest that in response to Filomena,Pigeons conducted partial migration in search of better conditions,while Magpies aggregated in certain areas of Madrid and Partridges endured the harsh conditions within their home *** results highlight the importance to alleviate negative impacts of extreme weather events in wild birds through management actions,especially for sedentary species.

Complex perianal fistulas have a negative impact on the quality of life of sufferers and should be *** diagnosis,characterization and classification of the fistulas are essential to optimize ***, in the case of patients whose fistulas are associated with Crohn's disease,complete closure is particularly difficult to *** medical treatments(antibiotics, thiopurines and other immunomodulatory agents,and, more recently,anti-tumor necrosis factor-αagents such as infliximab)have been tried with varying degrees of *** medical(including infliximab)and less aggressive surgical therapy(drainage and seton placement)offer the best outcomes in complex Crohn's fistulas while more aggressive surgical procedures such as fistulotomy or fistulectomy may increase the risk of *** review will focus on emerging novel treatments for perianal disease in Crohn's patients. These include locally applied infliximab or tacrolimus, fistula plugs,instillation of fibrin glue and the use of adult expanded adipose-derived stem cell *** welldesigned controlled studies are required to confirm the effectiveness of these emerging treatments.

The industry is continuously searching for new methods to reduce the use of chemical preservatives while still produce safer foods for the *** whey is a waste from the cheese industry which is primarily treated as a *** is rich in different nutrients and has the potential to be used as a matrix for lactic acid bacteria *** work aims to find a revalorization of whey for the production of a natural origin antifungal agent by a fermentation that can be used as food *** different whey culture mediums were developed and *** of different antifungal molecules in whey culture mediums in comparation to a fermentation of De Man,Rogosa and Sharpe(MRS)culture medium evidenced that the production of this antifungal metabolites was *** in vitro qualitative and quantitative assays evidenced that the whey-based mediums increased the antifungal activity of the CFS in comparation to the *** fermented whey produced bigger inhibition halos(above 1 cm)in the agar diffusion test in comparation to the fermented MRS(bellow 0.6 cm).Alike,in the minimal fungicidal concentration test the lowest antifungal concentrations were detected on the fermented whey mediums,averaging antifungal concentrations lower than 130 g/***,a multivariable data analysis determined that more antifungal metabolites yet to be detected were affecting in the antifungal activity from the whey *** fermented whey has the potential to be a tool in reducing the use of chemical preservatives in foods.

The Iberian hare(Lepus granatensis)is an endemic species distributed in Spain and *** outbreaks affecting this species were detected in 2018 in Central and Southern Spain,spreading *** to evaluate factors affecting the status of hare population after the arrival of myxomatosis,we conducted 108 nocturnal hare counts in Central Spain during two study periods(winter/spring and summer/autumn)in 54 different hunting grounds,covering 1071 km and observing 884 *** mean density in winter/spring was 7.66 hares/100 ha,(range 6.14-9.54/100 ha),while in summer/autumn,it was 3.4 hares/100 ha(range 2.6-4.4/100 ha).Densities of hares were not affected by the dominant habitat and the presence/absence of myxomatosis *** were more abundant at hunting grounds at a higher altitude and in those conducting targeted management,while detection of myxomatosis was related to lower altitude and higher levels of game management.A MaxEnt model used to generate a risk map for myxomatosis occurrence showed that the temperature annual range was the most important predictor,which suggests that environmental factors affecting myxomatosis vectors(mosquitoes,fleas,and ticks)could play a key role in disease *** myxomatosis in hares is becoming endemic,hare densities may be improved by game management and the monitoring and surveillance of this emerging *** surveillance programs could be the basis of effective collaborations between hunters,researchers,and environmental managers.

It has been found in recent years that using setpoint temperatures based on adaptive thermal comfort models is a successful method of energy *** studies using adaptive setpoint temperatures incorporate international models from ASHRAE Standard 55 and *** study,however,has instead considered a regional Brazilian adaptive comfort *** study investigates the energy demand arising from the use of a local Brazilian comfort model in order to assess the energy implications from the use of the worldwide ASHRAE Standard 55 adaptive model and various fixed setpoint *** of Brazil’s climate zones,full air-conditioning,mixed-mode building operating modes,present-day climate change scenarios,and future scenarios—specifically Representative Concentration Pathways(RCP)2.6,4.5,and 8.5 for the years 2050 and 2100—have all been taken into account in building energy *** use of adaptive setpoint temperatures based on the Brazilian local model considering mixed-mode has been found to significantly reduce energy consumption when compared to static setpoint temperatures(average energy-saving values ranging from 52%to 58%)and the ASHRAE 55 adaptive model(average values ranging from 15%to 21%).Considering climate change and the mixed-mode Brazilian model,the overall energy demand for the three groups of climatic zones(annual average outdoor temperatures≤21℃,>21 and≤25℃and>25℃)ranged between 2%decrease and 5%increase,4%and 27%increase,and 13%and 45%increase,*** is concluded as a consequence that setting setpoint temperatures based on the Brazilian local adaptive comfort model is a very efficient energy-saving method.
