Occurrence of Nanobacteria-Like Particles in Renal Stones of a Southern Brazilian Population

Purpose: Identifying the source of stone formation in recurrent stone formers has always been a big problem. Material and Methods: In this study kidney stones from 52 patients were submitted to direct examination by scanning electronic microscopy (SEM) after manual fracture and 27 calculi were cultured in Eagle’s Minimum Essential Medium (E-MEM) and Brain-Heart Infusion (BHI) for eight weeks at 37°C in 5% CO2 atmosphere. Twenty-seven powdered and demineralized stones were suspended in sterile PBS, filtered through 0.22 m-pore-size sterile filters Millex (Millipore, Massachusetts, USA) and submitted to DNA extraction (Quiagen-Brazil). Platinum PCR SuperMIX (GIBCO-BRL), primers (Invitrogen), and Ultra Pure Water (Advanced Biotechnologies, Columbia, MD) were used for PCR (Polymerase Chain Reaction), which was generally conducted for 30 or 35 cycles with annealing of primers for 40 sec at 55°C and extension for 1 min at 72°C. Results: In 36 out 52 (69%) kidney stones it was detected the presence of biofilm coating the mineral surface of the stone when examined by SEM, consisting of coccoid particles, isolated or clustered, with diameter of 500 nm or less. Eleven out 27 (41%) kidney stone cultures produced white-colored sediment on the bottom of the tubes after eight-week incubation, revealing tiny structures similar to those observed directly by SEM. These structures were similar in size and morphology to spherical apatite particles previously observed in human kidney stones and named as nanobacteria (NB). No PCR products were observed in the samples. Conclusion: We found a strong correlation between renal stones and calcifying nanoparticles (CNP) in this study and these results open a new insight on this area to explore the etiology of stone formation. Whether NB/CNP are truly microorganisms or self-propagating mineral compounds is still controversial and its contribution, if any, in apatite nucleation and crystal growth remains uncertain.

Global epidemiology of sexually transmitted diseases

Sexually transmitted diseases （STDs） are among the first ten causes of unpleasant diseases in young adult males in developing countries and the second major cause of unpleasant diseases in young adult women. Adolescents and young adults （15-24 years old） make up only 25% of the sexually active population, but represent almost 50% of all new acquired STDs. In general, STDs are epidemics and present an enormous health and economic consequences. An adequate screening for STDs should be done on a routine basis in every part of the world. Many STDs are asymptomatic and therefore can difficult to control. The purpose of reporting of STDs is to ensure that persons who are infected will be quickly diagnosed and appropriately treated to control the spread of infection and also so that partners are notified, tested and appropriately treated. It is estimated that reported cases of STDs represent only 50%-80% of reportable STD infections in the United States, reflecting limited screening and low disease reporting. High-risk sexual behavior is a highly contributive factor of this process as it often leads to teenage pregnancies and HIV/AIDS. One possible explanation for this behavior is that people do not have enough information about the transmission of STDs or ignore the precautions required for safe sex. Approximately 60% of new HIV infections worldwide occur in young people. The frequency of high-risk behaviors among youths may also be influenced by opportunity to engage in them, particularly the amount of time that they are unsupervised by adults. However, in diagnosing and treating these patients, we can effectively prevent the spread of HIV/AIDS. Individuals infected with STDs are 5-10 times more likely than uninfected individuals to acquire or transmit HIV through sexual contact. The breaking of the genital tract lining or skin creates a portal of entry for HIV and, hence, HIV-infected individuals with other STDs are more likely to shed HIV in their genital secretions. T

Aim: To identify the association between access to obstetric and neonatal hospital service and neonatal death rates. Method: Quantitative and retrospective research retrieved from Declaration of Live Newly-born Children;Declaration of Death;Investigation Chart on Municipal Child Mortality, between 2000 and 2009, at the Nucleus of Information on Mortality Rates. Effect of genistein supplementation of thawing medium on characteristics of frozen human spermatozoa

In this study, we evaluated the effects of genistein supplementation of the thawing extender on frozen-thawed human semen parameters. We analyzed the effect of supplementation on sperm motility, capacitation （membrane lipid disorder）, reactive oxygen species （ros） generation, chromatin condensation and DNA damage. Using this preliminary information, it maybe possible to improve the cryopreservation process and reduce the cellular damage. We have confirmed that the isoflavone genistein （10 μmol L-1） has antioxidant properties on the frozen-thawed spermatozoa. This results in a decreased ros production that shows a slight improvement in the sperm motility, and decreases the membrane lipid disorder and DNA damage caused by cryopreservation. These results suggest an effect of genistein on sperm functionality that could be of interest for assisted reproduction treatments using frozen- thawed human spermatozoa, but further studies will be necessary to confirm our findings and to evaluate the possible clinical applications.

Climatological Hydric Balance and the Trends Analysis Climatic in the Region of Machado in Minas Gerais State,Brazil

Lately the planet’s climate has been constantly changing, caused mainly by global warming which has exposed a great deal of concern to the population over the years. In order to understand the possible impacts that such changes may have on the environment and society in general, the importance of the analysis of climate and hydrological events trends and their performance in a region is justified. The objective of the present work was to perform the climatic classification and to evaluate the behavior of the Climatological Hydric Balance—CHB, from the region of Machado state of Minas Gerais—MG, taking into account a historical series of 55 years of climatic season data of the National Institute of Meteorology—INMET;to verify the occurrence of climatic changes by the temporal trends of precipitation and the average temperature, using the Mann-Kendall and Pettitt method;and the influence of these possible climate changes on CHB behavior and on the region’s climate classification. Based on the results found it verified the increase in the water deficit between the months of June to September and a reduction in the water surplus from November to February. By means of the trend analysis, there was a positive trend of increase in the average temperature of 1.6°C until the year 2100. The continuity and occurrence of these trends may have impacts on the economy, agriculture, the hydrological cycle, and consequently on the fauna, the flora and the population.

In vivo testing of mucus-permeating nanoparticles for oral insulin delivery using Caenorhabditis elegans as a model under hyperglycemic conditions

The aim was to evaluate the potential of mucus-permeating nanoparticles for the oral administration of *** nanocarriers,based on the coating of zein nanoparticles with a polymer conjugate containing PEG,displayed a size of 260 nm with a negative surface charge and an insulin payload of 77 mg/*** intestinal pig mucus,the diffusivity of these nanoparticles(PPA-NPs)was found to be 20-fold higher than bare nanoparticles(NPs).These results were in line with the biodistribution study in rats,in which NPs remained trapped in the mucus,whereas PPA-NPs were able to cross this layer and reach the epithelium *** therapeutic effcacy was evaluated in Caenorhabditis elegans grown under high glucose *** this model,worms treated with insulin-loaded in PPA-NPs displayed a longer lifespan than those treated with insulin free or nanoencapsulated in *** finding was associated with a signifcant reduction in the formation of reactive oxygen species(ros)as well as an important decrease in the glucose and fat content in *** effects would be related with the mucus-permeating ability of PPA-NPs that would facilitate the passage through the intestinal peritrophic-like dense layer of worms(similar to mucus)and,thus,the absorption of insulin.

Climate Characterization of the Machado-Mg Region through Geotechnology Techniques and Tools

The characteristics of a region and its environmental changes are directly associated with the characteristics of the climate and the changes that occur in it. The importance of the study of the climate of a region is widely known in agriculture, by the size of the impact that the climate has on the productivity and final quality of the products. From the studies of these spatial and temporal changes in climatic and environmental information, excellent results have been obtained in understanding the variability of atmospheric phenomena that occur throughout. The objective of the present work was to perform the climatic characterization of the Machado-MG region, analyzing data collected on the elements of the climate, applying methodologies that allow its spatialization, from geotechnology tools, more precisely, the techniques of interpolation and manipulation of spatial data in the Geographic Information Systems (GIS) environment. For this purpose, it has been used the climatic data obtained from meteorological stations of the National Institute of Meteorology-INMET, located in the state of Minas Gerais, referring to the monthly and annual averages of a historical series of 55 years (1961-2015). The results obtained allowed to characterize the climate of the municipality, which presented an average temperature around 15°C to 25°C and with average rainfall between 100 - 200 mm in the rainiest periods. The relative humidity of the air presents throughout the year values in the average of 60% to 80%. From the spatialization maps of temperature, precipitation and relative humidity of the air, it becomes possible to analyze the spatial behavior of the climatic elements and thus to have the perception of the climate dynamics in the region.
