Pramipexole, a dopamine D3/D2 receptor-preferring agonist, attenuates reserpine-induced fibromyalgia-like model in mice

Fibromyalgia(FM) is a complex pathology described as persistent hyperalgesia including somatic and mood dysfunctions, depression and anxiety. Although the etiology of FM is still unknown, a significant decrease in biogenic amines is a common characteristic in its pathogenesis. Here, our main objective was to investigate the role of dopamine D3/D2 receptor during the reserpine-induced pain in mice. Our results showed that pramipexole(PPX) – a dopaminergic D3/D2 receptor agonist – inhibited mechanical allodynia and thermal sensitivity induced by reserpine. Relevantly, PPX treatment decreased immobility time and increased the number of grooming in the forced swimming test and splash test, respectively. Animals that received PPX remained longer in the open arms than the reserpine group using elevated plusmaze apparatus. The repeated PPX administration, given daily for 4 days, significantly blocked the mechanical and thermal allodynia during FM model, similarly to pregabalin, although it failed to affect the reserpine-induced thermal nociception. Reserpine administration induced significant downregulation of dopamine concentration in the central nervous system, and repeated treatment with PPX restored dopamine levels in the frontal cortex and spinal cord tissues. Moreover, PPX treatment inhibited oxidants production such as DCFH(2′,7′-dichlorodihydrofluorescein) and nitrite, also decreased oxidative damage(carbonyl), and upregulated the activity of superoxide dismutase in the spinal cord. Together, our findings demonstrated the ability of dopamine D3/D2 receptor-preferring agonist in reducing pain and mood dysfunction allied to FM in mice. All experimental protocols were approved by the Universidade Federal de Santa Catarina(UFSC) Ethics Committee(approval No. 2572210218) on May 10, 2018.

Rudimentary food fermentation can be defined as a spontaneous process of conversion of food components through enzy-matic action.A great variety of fermented foods are produced using spontaneous approaches;however,cocoa and coffee represent the most important agricultural commodities on international markets.As a manner to increase the efficiency of these processes,starter cultures have been developed and applied under field conditions.The selection process begins with the recovery of microbial strains from spontaneous fermentation through phenotypic and metabolic traits.Next,mutation-based breeding is used to develop and improve well-adapted starter cultures.With advances in synthetic biology,especially in the last decade,the development of robust cellular fabrications with high fermentative capacity has become easier-largely due to the development of genomic approaches,such as next-generation sequencing techniques,CRISPR-Cas system and bioinformatics tools.This review brings prospects on the use of synthetic biology to design new robust strains for use in cocoa and coffee fermentations,but which can be extended to other rudimentary foods.In addition,metabolic traits and target genes(e.g.,UvrA,RecA,GPD1,and GPP2)are proposed as a starting point for the improvement of cocoa and coffee starters.Finally,the regulatory and safety requirements for these food crops are addressed.This review aims to stimulate research on the process of fermentation and the associated synthetic biology tools to produce fermented food efficiently and sustainably.

High-throughput phenotyping of two plant-size traits of Eucalyptus species using neural networks

In forest modeling to estimate the volume of wood,artificial intelligence has been shown to be quite effi-cient,especially using artificial neural networks(ANNs).Here we tested whether diameter at breast height(DBH)and the total plant height(Ht)of eucalyptus can be pre-dicted at the stand level using spectral bands measured by an unmanned aerial vehicle(UAV)multispectral sensor and vegetation indices.To do so,using the data obtained by the UAV as input variables,we tested different configurations(number of hidden layers and number of neurons in each layer)of ANNs for predicting DBH and Ht at stand level for different Eucalyptus species.The experimental design was randomized blocks with four replicates,with 20 trees in each experimental plot.The treatments comprised five Eucalyptus species(E.camaldulensis,E.uroplylla,E.saligna,E.gran-dis,and E.urograndis)and Corymbria citriodora.DBH and Ht for each plot at the stand level were measured seven times in separate overflights by the UAV,so that the multispectral sensor could obtain spectral bands to calculate vegetation indices(VIs).ANNs were then constructed using spectral bands and VIs as input layers,in addition to the categorical variable (species), to predict DBH and Ht at the stand level simultaneously. This report represents one of the first appli-cations of high-throughput phenotyping for plant size traits in Eucalyptus species. In general, ANNs containing three hidden layers gave better statistical performance (higher esti-mated r, lower estimated root mean squared error-RMSE) due to their greater capacity for self-learning. Among these ANNs, the best contained eight neurons in the first layer, seven in the second, and five in the third (8 − 7 − 5). The results reported here reveal the potential of using the gener-ated models to perform accurate forest inventories based on spectral bands and VIs obtained with a UAV multispectral sensor and ANNs, reducing labor and time.

Spatiotemporal dynamics in butterfly hybrid zones

Evaluating whether hybrid zones are stable or mobile can provide novelinsights for evolution and conservation biology. Butterflies exhibit high sensitivity to envi-ronmental changes and represent an important model system for the study of hybrid zoneorigins and maintenance. Here, we review the literature exploring butterfly hybrid zones,with a special focus on their spatiotemporal dynamics and the potential mechanisms thatcould lead to their movement or stability. We then compare different lines of evidenceused to investigate hybrid zone dynamics and discuss the strengths and weaknesses ofeach approach. Our goal with this review is to reveal general conditions associated withthe stability or mobility of butterfly hybrid zones by synthesizing evidence obtained us-ing different types of data sampled across multiple regions and spatial scales. Finally, wediscuss spatiotemporal dynamics in the context of a speciation/divergence continuum, therelevance of hybrid zones for conservation biology, and recommend key topics for futureinvestigation.

Rock slope hazard assessment is an important part of risk analysis for open pit mines.The main parameters that can lead to rock slope failures are the parameters traditionally used in geomechanical classifications,the slope geometrical parameters and external factors like rainfall and blasting.This paper presents a methodology for a hazard assessment system for open pit mine slopes based on 88 cases collated around the world using principal components analysis,discriminant analysis and confidence ellipses.The historical cases used in this study included copper,gold,iron,diamond,lead and zinc,platinum and claystone mines.The variables used in the assessment methodology are uniaxial compressive strength of intact rock;spacing,persistence,opening,roughness,infilling and orientation of the main discontinuity set;weathering of the rock mass;groundwater;blasting method;and height and inclination of the pit.While principal component analysis was used to quantify the data,the discriminant analysis was used to establish a rule to classify new slopes about its stability condition.To provide a practical hazard assessment system,confidence ellipses were used to propose a hazard graph and generate the HAS-Q.The discriminant rule developed in this research has a high discrimination capacity with an error rate of 11.36%.

Native and exotic seed dispersal by the stone marten(Martes foina):implications for the regeneration of a relict climactic forest in central Portugal

The stone marten(Martes foina)is a carnivorous mammal that often consumes fleshy fruits,thus potentially promoting seed dispersion.The present study was developed in Bussaco National Forest,central Portugal,and aimed to assess the potential role of the stone marten in dispersing native and exotic plants in different forest landscape types.Seeds from stone marten scats and fleshy fruits were collected monthly and were thereafter identified and sowed in a nursery,following a randomized experimental setup.Plant emergence was monitored fortnightly.Generalized linear models were used to test for differences in time and success of emergence between seeds from scats and fruits of 3 native species(Rubus ulmifolius,Arbutus unedo and Celtis australis)and 1 exotic plant species with invasive behavior(Prunus laurocerasus).Fruit consumption by the stone martens significantly increased and accelerated the germination of the native R.ulmifolius but had no effect on the other 2 native species or on the invasive species.This suggested that stone martens contribute to gene flow and forest regeneration by dispersing native plant seeds.However,although the germination was not enhanced in the invasive species,the preference of stone martens for these fruits may potentially contribute to the proliferation of P.laurocerasus.Our study represents a contribution to better understanding the fauna and flora interactions,enabling for a more conscious and effective decision-making in forest management.

In vitro antileishmanial effects of Physalis angulata root extract on Leishmania infantum

Objective： In the present study, we evaluated the effects of the aqueous extract of Physalis angulata root （AEPa） on Leishmania infanturn proliferation, morphology, and the driving mechanism in leishmanicidal activity and modulatory action on macrophages. Methods： I., infantum promastigotes were treated with 50 and 100 μg/mL AEPa for 72 h and then antipro- mastigote assay was performed by counts in a Newbauer chamber, morphological changes were analyzed by transmission electron microscopy and the mechanism of the leishmanicidal activity was detected. In addition, macrophages were infected with L. infantum and were used to evaluate anti-amastigote activity of AEPa and effects of AEPa on cytokine secretion after 72-hour treatment. Results： Treatment with AEPa reduced the numbers ofL infantum promastigotes （50% inhibitory concen- tration （ICso） = 65.9μg/mL; selectivity index （SI）= 22.1） and amastigotes （ICso = 37.9 μg/mL; SI = 38.5） compared with the untreated control. Amphotericin B reduced 100% of the promastigote numbers after 72 h of treatment （IC50 = 0.2μg/mL）. AEPa induced several morphological changes and increased the production of reactive oxygen species and apoptotic death in promastigotes after treating for 72 h. AEPa （100 μg/mL） promoted tumor necrosis factor-α secretion in macrophages infected with L. infantum after 72 h of treatment, but did not induce an increase in this cytokine in noninfected macrophages. In addition, AEPa showed no cytotoxic effect on J774-A1 cells （50% cytotoxic concentration 〉1000μg/mL）. Conclusion： AEPa presented antileishmanial activity against the promastigotes and amastigotes of I.. infantum without macrophage cytotoxicity; these results show that natural products such as P. angulata have leishmanicidal potential and in the future may be an alternative treatment for leishmaniasis.

Fluidization of binary mixtures of sisal residue and sand:A new model for deriving the final fluidization velocity

The influence of different factors on the fiuidization of a binary mixture of sisal residue and sand was investigated.The particle sizes of the sand and sisal residue were varied from 0.2to 0.8mm and the biomass mass fractions from 2% to 9%.Some segregation was noted,and a significant relationship was found among the final fluidization velocity (Uff),the biomass and sand sizes,and the biomass mass fraction.A novel model was developed for predicting Uff,leading to an average discrepancy of 12.69% between the measured and predicted Uff compared with the best match of 15.32% when using a model from a previous paper.The new model was applied to data from studies using other biomass and a broad range of particle characteristics.The average divergences from measured values when using the new model were 7.9% for corn cob and walnut shell,and 20.5% for sweet sorghum bagasse,tobacco residue, and soy hulls.These were superior to the values derived using other models.Our results confirm the accuracy of the model developed in this work and show that it represents a viable alternative way to calculate Uff for a binary mixture of sand and biomass.
