Fungal contamination of stored automobile-fuels in a tropical environment

Because of the lack of reports, the base levels of microbial contamination on stored fuels are unknown in tropical regions and it is unclear whether these levels have some influence on fuel quality parameters. Therefore, fungal quality in automobile fuels stored across Costa Rican territory was evaluated during two years according to the standard ASTM D6974-04. For a total of 96 samples, counts and identification of molds and yeasts were performed on regular gas, premium gas and diesel taken from the bottom and superior part of the container tanks. The highest contamination was found on the bottom of the tanks, where an aqueous phase was usually identified, showing populations over the ones present in the hydrocarbon itself （up to 108 CFU/L）. Diesel was the most contaminated fuel （up to 107 CFO/L）; however, an alteration on the physicochemical parameters was not observed in any kind of fuel. Seventy-five mold strains were isolated, Penicillium sp. being the most common genus （45.8% of the samples）, and ten yeast strains, from the genera Candida sp. and Rhodotorula sp. Four of the yeasts were able to grow on diesel as the sole carbon source, at concentrations ranging from 0.5% to 25%. Increasing the frequency of tank cleaning, adding antimicrobial agents and monitoring microbial populations are recommended strategies to improve microbial quality of stored fuels.

Removal of carbofuran is not affected by co-application of chlorpyrifos in a coconut fiber/compost based biomixture after aging or pre-exposure

Biomixtures constitute the biologically active part of biopurification systems（BPS）, which are used to treat pesticide-containing wastewater. The aim of this work was to determine whether co-application of chlorpyrifos（CLP） affects the removal of carbofuran（CFN）（both insecticide/nematicides） in a coconut fiber–compost–soil biomixture（FCS biomixture）, after aging or previous exposure to CFN. Removal of CFN and two of its transformation products（3-hydroxycarbofuran and 3-ketocarbofuran） was enhanced in pre-exposed biomixtures in comparison to aged biomixtures. The co-application of CLP did not affect CFN removal, which suggests that CLP does not inhibit microbial populations in charge of CFN transformation.Contrary to the removal behavior, mineralization of radiolabeled14C-pesticides showed higher mineralization rates of CFN in aged biomixtures（with respect to freshly prepared or pre-exposed biomixtures）. In the case of CLP, mineralization was favored in freshly prepared biomixtures, which could be ascribed to high sorption during aging and microbial inhibition by CFN in pre-exposure. Regardless of removal and mineralization results, toxicological assays revealed a steep decrease in the acute toxicity of the matrix on the microcrustacean Daphnia magna（over 97%） after 8 days of treatment of individual pesticides or the mixture CFN/CLP.Results suggest that FCS biomixtures are suitable to be used in BPS for the treatment of wastewater in fields where both pesticides are employed.

In the tropics,frequent nitrogen(N)fertilization of grazing areas can potentially increase nitrous oxide(N_(2)O)emissions.The application of nitrification inhibitors has been reported as an effective management practice for potentially reducing N loss from the soil-plant system and improving N use efficiency(NUe).The aim of this study was to determine the effect of the co-application of nitrapyrin(a nitrification inhibitor,NI)and urea in a tropical Andosol on the behavior of N and the emissions of N_(2)O from autotrophic and heterotrophic nitrification.A greenhouse experiment was performed using a soil(pH 5.9,organic matter content 78 g kg^(-1),and N 5.6 g kg^(-1))sown with Cynodon nlemfuensis at 60%water-filled pore space to quantify total N_(2)O emissions,N_(2)O derived from fertilizer,soil ammonium(NH_(4)^(+))and nitrate(NO_(3)^(-)),and NUe.The study included treatments that received deionized water only(control,NI).No significant differences were observed in soil NH_(4)^(+)content between the UR and UR+NI treatments,probably because of soil mineralization and NO_(3)^(-)produced by heterotrophic nitrification,which is not effectively inhibited by nitrapyrin.After 56 d,N_(2)O emissions in UR(0.51±0.12 mg N_(2)O-N concluded that the soil organic N mineralization and heterotrophic nitrification are the main processes of NH_(4)^(+)and NO_(3)^(-)production.Additionally,it was found that N_(2)O emissions were partially a consequence of the direct oxidation of the soil's organic N via heterotrophic nitrification coupled to denitrification.Finally,the results suggest that nitrapyrin would likely exert significant mitigation on N_(2)O emissions only if a substantial N surplus exists in soils with high organic matter content.

Biomaterial engineering strategies for modeling the Bruch's membrane in age-related macular degeneration

Age-related macular degeneration,a multifactorial inflammatory degenerative retinal disease,ranks as the leading cause of blindness in the elderly.Strikingly,there is a scarcity of curative therapies,especially for the atrophic advanced form of age-related macular degeneration,likely due to the lack of models able to fully recapitulate the native structure of the outer blood retinal barrier,the prime to rget tissue of age-related macular degeneration.Standard in vitro systems rely on 2D monocultures unable to adequately reproduce the structure and function of the outer blood retinal barrier,integrated by the dynamic interaction of the retinal pigment epithelium,the Bruch's membrane,and the underlying choriocapillaris.The Bruch's membrane provides structu ral and mechanical support and regulates the molecular trafficking in the outer blood retinal barrier,and therefo re adequate Bruch's membrane-mimics are key for the development of physiologically relevant models of the outer blood retinal barrie r.In the last years,advances in the field of biomaterial engineering have provided novel approaches to mimic the Bruch's membrane from a variety of materials.This review provides a discussion of the integrated properties and function of outer blood retinal barrier components in healt hy and age-related macular degeneration status to understand the requirements to adequately fabricate Bruch's membrane biomimetic systems.Then,we discuss novel materials and techniques to fabricate Bruch's membrane-like scaffolds for age-related macular degeneration in vitro modeling,discussing their advantages and challenges with a special focus on the potential of Bruch's membrane-like mimics based on decellularized tissue.

Role of climatic variability in shaping intraspecific variation of thermal tolerance in Mediterranean water beetles

The climatic variability hypothesis(CVH)predicts that organisms in more thermally variable environments have wider thermal breadths and higher thermal plasticity than those from more stable environments.However,due to evolutionary trade-offs,taxa with greater absolute thermal limits may have little plasticity of such limits(trade-off hypothesis).The CVH assumes that climatic variability is the ultimate driver of thermal tolerance variation across latitudinal and altitudinal gradients,but average temperature also varies along such gradients.We explored intraspecific variation of thermal tolerance in three typical Mediterranean saline water beetles(families Hydrophilidae and Dytiscidae).For each species,we compared two populations where the species coexist,with similar annual mean temperature but contrasting thermal variability(continental vs.coastal population).We estimated thermal limits of adults from each population,previously acclimated at 17,20,or 25℃.We found species-specific patterns but overall,our results agree with the CVH regarding thermal ranges,which were wider in the continental(more variable)population.In the two hydrophilid species,this came at the cost of losing plasticity of the upper thermal limit in this population,supporting the trade-off hypothesis,but not in the dytiscid one.Our results support the role of local adaptation to thermal variability and trade-offs between basal tolerance and physiological plasticity in shaping thermal tolerance in aquatic ectotherms,but also suggest that intraspecific variation of thermal tolerance does not fit a general pattern among aquatic insects.Overlooking such intraspecific variation could lead to inaccurate predictions of the vulnerability of aquatic insects to global warming.

Alzheimer’s disease is a neurodegenerative disorder characterized by the amyloid accumulation in the brains of patients with Alzheimer’s disease.The pathogenesis of Alzheimer’s disease is mainly mediated by the phosphorylation and aggregation of tau protein.Among the multiple causes of tau hyperphosphorylation,brain insulin resistance has generated much attention,and inositols as insulin sensitizers,are currently considered candidates for drug development.The present narrative review revises the interactions between these three elements:Alzheimer’s disease-tau-inositols,which can eventually identify targets for new disease modifiers capable of bringing hope to the millions of people affected by this devastating disease.

Digital Social Connectedness as a Lifeline for Older People: Use and Non-use of VinclesBCN During the Pandemic

This article presents the results of a study that evaluated VinclesBCN during COVID-19. This digital-based public social service aims to prevent loneliness and isolation in +65-year-old adults living in Barcelona. Through service user(N = 12) and professional(N = 6) interviews and a questionnaire with service users(N = 255), we demonstrate the pivotal role of digital connectedness in transforming VinclesBCN into a lifeline during the pandemic. The analysis revealed the importance of sociability, social support, and, especially, entertainment in coping with pandemic fatigue and facilitating social connectivity and support among users. Users engaged in activities such as sharing images, songs, memes, and daily greetings to provide proximity, sociability, and care among users, whether they belonged to preexisting groups or were newly introduced to the platform. It also facilitated the identification of individuals who needed companionship, comfort, or more specialized support. The findings emphasize the significance of entertainment as a resilience-building strategy during times of uncertainty. Despite the positive impact, not all users equally used the platform. Non-use was strongly associated with being a woman, having a low educational level, having preexisting social relationships, less time of enrolment in the platform, as well as a high perception of loneliness, poor self-reported health, and low mood. The article underscores the need for further research into older adults' digital engagement during crises, its role in building resilience, and advocating for inclusive digital interventions that take into account diverse older adults' needs and experiences in crisis contexts.

We calculate numerically the optical chiral forces in rectangular cross-section dielectric waveguides for potential enantiomer separation.Our study considers force strength and time needed for separating chiral nanoparticles,mainly via quasi-Te guided modes at short wavelengths(405 nm)and the 90°-phase-shifted combination of quasi-Te and quasi-TM modes at longer wavelengths(1310 nm).Particle tracking simulations show successful enantiomer separation within two seconds.These results suggest the feasibility of enantiomeric separation of nanoparticles displaying sufficient chirality using simple silicon photonic integrated circuits,with wavelength selection based on the nanoparticle size.

Colorectal cancer(CRC)remains one of the most commonly diagnosed and deadliest types of cancer worldwide.CRC displays a desmoplastic reaction(DR)that has been inversely associated with poor prognosis;less DR is associated with a better prognosis.This reaction generates excessive connective tissue,in which cancer-associated fibroblasts(CAFs)are critical cells that form a part of the tumor microenvironment.CAFs are directly involved in tumorigenesis through different mechanisms.However,their role in immunosuppression in CRC is not well understood,and the precise role of signal transducers and activators of transcription(STATs)in mediating CAF activity in CRC remains unclear.Among the myriad chemical and biological factors that affect CAFs,different cytokines mediate their function by activating STAT signaling pathways.Thus,the harmful effects of CAFs in favoring tumor growth and invasion may be modulated using STAT inhibitors.Here,we analyze the impact of different STATs on CAF activity and their immunoregulatory role.

Influenza pandemics are unpredictable recurrent events with global health,economic,and social consequences.The objective of this review is to provide an update on the latest developments in early diagnosis and specific treatment of the disease and its complications,particularly with regard to respiratory organ failure.Despite advances in treatment,the rate of mortality in the intensive care unit remains approximately 30%.Therefore,early identification of potentially severe viral pneumonia is extremely important to optimize treatment in these patients.The pathogenesis of influenza virus infection depends on viral virulence and host response.Thus,in some patients,it is associated with an excessive systemic response mediated by an authentic cytokine storm.This process leads to severe primary pneumonia and acute respiratory distress syndrome.Initial prognostication in the emergency department based on comorbidities,vital signs,and biomarkers(e.g.,procalcitonin,ferritin,human leukocyte antigen-DR,mid-regional proadrenomedullin,and lactate)is important.Identification of these biomarkers on admission may facilitate clinical decision-making to determine early admission to the hospital or the intensive care unit.These decisions are reached considering pathophysiological circumstances that are associated with a poor prognosis(e.g.,bacterial co-infection,hyperinflammation,immune paralysis,severe endothelial damage,organ dysfunction,and septic shock).Moreover,early implementation is important to increase treatment efficacy.Based on a limited level of evidence,all current guidelines recommend using oseltamivir in this setting.The possibility of drug resistance should also be considered.Alternative options include other antiviral drugs and combination therapies with monoclonal antibodies.Importantly,it is not recommended to use corticosteroids in the initial treatment of these patients.Furthermore,the implementation of supportive measures for respiratory failure is essential.Current recommendations are limited,heterogeneous,an
