Low-cost,simple,and scalable self-assembly of DNA origami nanostructures

Despite demonstrating exciting potential for applications such as drug delivery and biosensing,the development of nanodevices for practical applications and broader use in research and education are still hindered by the time,effort,and cost associated with DNA origami *** and robust methods to perform and scale the DNA origami self-assembly process are critical to facilitate broader use and translation to industrial or clinical *** report a simple approach to fold DNA origami nanostructures that is fast,robust,and *** demonstrate fabrication at scales approximately 100-1,500-fold higher than typical *** further demonstrate an approach we termed low-cost efficient annealing (LEAN) self-assembly involving initial heating at 65 ℃ for 10 min,then annealing at 51 ℃ for 2 h,followed by brief quenching at 4 ℃ that leads to effective assembly of a range of DNA origami structures *** contrast to other methods for scaling DNA origami assembly,this approach can be carried out using cheap and widely available equipment (e.g.,hot plates,water baths,and laboratory burners) and uses standard recipes and materials so is readily applied to any existing or new DNA origami *** envision these methods can facilitate device development for commercial applications and facilitate broader use of DNA origami in research and education.

Generalized Uncertainty Relations, Curved Phase-Spaces and Quantum Gravity

Modifications of the Weyl-Heisenberg algebra are proposed where the classical limit corresponds to a metric in (curved) momentum spaces. In the simplest scenario, the 2D de Sitter metric of constant curvature in momentum space furnishes a hierarchy of modified uncertainty relations leading to a minimum value for the position uncertainty . The first uncertainty relation of this hierarchy has the same functional form as the stringy modified uncertainty relation with a Planck scale minimum value for at . We proceed with a discussion of the most general curved phase space scenario (cotangent bundle of spacetime) and provide the noncommuting phase space coordinates algebra in terms of the symmetric and nonsymmetric metric components of a Hermitian complex metric , such . Yang’s noncommuting phase-space coordinates algebra, combined with the Schrodinger-Robertson inequalities involving angular momentum eigenstates, reveals how a quantized area operator in units of emerges like it occurs in Loop Quantum Gravity (LQG). Some final comments are made about Fedosov deformation quantization, Noncommutative and Nonassociative gravity.

A Novel Derivation of Black Hole Entropy in all Dimensions from Truly Point Mass Sources

It is explicitly shown how the Schwarzschild Black Hole Entropy (in all dimensions) emerges from truly point mass sources at r=0due to a non-vanishing scalar curvature involving the Dirac delta distribution. In order to achieve this, one is required to extend the domain of r to negative values −∞≤r≤+∞. It is the density and anisotropic pressure components associated with the point mass delta function source at the origin r=0which furnish the Schwarzschild black hole entropy in all dimensions D≥4after evaluating the Euclidean Einstein-Hilbert action. Two of the most salient results are i) that the observed spacetime dimension D=4is precisely singled out from all the other dimensions when the strong and weak energy conditions are met, and ii) the point mass source described in this work is not the result of a spherically symmetric gravitational collapse of a star as described by the Oppenheimer-Snyder model because we are not neglecting the pressure. As usual, it is required to take the inverse Hawking temperature βHas the length of the circle Sβ1obtained from a compactification of the Euclidean time in thermal field theory which results after a Wick rotation, it=τ, to imaginary time. This approach can be generalized to the Reissner-Nordstrom and Kerr-Newman metrics. The physical implications of this finding warrant further investigation since it suggests a profound connection between the notion of gravitational entropy and spacetime singularities.

On Discrete Hopf Fibrations, Grand Unification Groups, the Barnes-Wall, Leech Lattices, and Quasicrystals

A discrete Hopf fibration of S15 over S8 with S7 (unit octonions) as fibers leads to a 16D Polytope P16 with 4320 vertices obtained from the convex hull of the 16D Barnes-Wall lattice Λ16. It is argued (conjectured) how a subsequent 2-1 mapping (projection) of P16 onto a 8D-hyperplane might furnish the 2160 vertices of the uniform 241 polytope in 8-dimensions, and such that one can capture the chain sequence of polytopes 241,231,221,211in D=8,7,6,5dimensions, leading, respectively, to the sequence of Coxeter groups E8,E7,E6,SO(10)which are putative GUT group candidates. An embedding of the E8⊕E8and E8⊕E8⊕E8lattice into the Barnes-Wall Λ16 and Leech Λ24 lattices, respectively, is explicitly shown. From the 16D lattice E8⊕E8one can generate two separate families of Elser-Sloane 4D quasicrystals (QC’s) with H4 (icosahedral) symmetry via the “cut-and-project” method from 8D to 4D in each separate E8 lattice. Therefore, one obtains in this fashion the Cartesian product of two Elser-Sloane QC’s Q×Qspanning an 8D space. Similarly, from the 24D lattice E8⊕E8⊕E8one can generate the Cartesian product of three Elser-Sloane 4D quasicrystals (QC’s) Q×Q×Qwith H4 symmetry and spanning a 12D space.

DNA Barcodes in Fig Cultivars (Ficus carica L.) Using ITS Regions of Ribosomal DNA, the psbA-trnH Spacer and the matK Coding Sequence

Molecular markers provide a useful method for genotype characterization and allow a high precision determination of the genetic relationship between cultivars and varieties. A system based on DNA sequences—which is known as DNA barcoding—will choose one or several standard loci which can be sequenced and compared to differentiate between species. In this research, the ITS, matK, and trnH-psbA sequences were evaluated for the molecular identification of seven F. carica genotypes, generating complete sequences for the first two loci, but unable to produce bidirectional sequences by using the trnH-psbA sequence. The ITS sequence presented the highest variation rates, while the phylogeny constructed with the matK sequence obtained the highest percentage of solved monophyletic groups. Through Pearson’s correlation analysis, it was possible to determine the existence of a significant correlation between the ITS region and psbA-trnH, and the matK and psbA-trnH sequences, but not between ITS and matK. The phylogenies constructed with the ITS + matK barcodes and ITS + matK + psbA-trnH presented the highest percentage for resolution. However, considering the cost efficiency and the facilitated recovery by using PCR, the matK + ITS combination is recommended.

Influence of Seven Growth Regulators on Fruit Set, Pollen Germination and Pollen Tube Growth of Almonds

To improve fruit set in ＂Carmel＂ almond （Prunus dulcis） trees grown in the 6th region of Chile, seven different growth regulators （GRs） were sprayed onto flowering branches： gibberellic acid （GA3）, Promalin, naphthaleneacetic acid （NAA）, Thidiazuron, Kinetin, Point-tomatomone and Brassinolide. The percentage of fruit set was determined by comparing the initial number of flowers present on each treated branch with the total number of almonds on the branches 50 days after anthesis. In comparison to the controls, Brassinolide, Point-tomatomone and Promalin achieved increases in fruit set of 19.4%, 14.5% and 12.9%, respectively. A parallel experiment was done in vitro, on the effects of these GRs on pollen germination and pollen tube growth. The greatest pollen germination （4 hours at 20 ~C） occurred with Brassinolide and GA3, which were significantly higher than the control, while NAA and Promalin significantly reduced germination. For pollen tube growth （24 hours at 20 ~C）, the greatest lengths were observed with Point-tomatomone and Brassinolide, which were significantly longer than the control, while NAA, Thidiazuron and Promalin were significantly shorter than the control, and the least growth was observed with Kinetin.

Surface Plasmon Resonance of a Few Particles Linear Arrays

We present a study of the enhancement of the electric field due to localized surface plasmons in a short chain of metallic nanoparticles with different shapes: spheres, cylinders and spheroids. We calculate numerically the external radiation effect on these chains and analyze besides the shape, also the influence on size, interparticle distances and number of nanoparticles, corroborating that each one plays a definitive role for the enhancement of the electric field. Particularly, we focus on the main features of the electric field in the inter-particle regions, where an enormous increasing is expected due to the longitudinal localized plasmons. The electric field distribution along the chain shows a maximum in the middle of the chain. This fact could be related to a hybridization effect as the gap between particles decreases below 2 nm, we also observe a strong enhancement with the number of nanoparticles. Also regarding the shape we find agreement with reported results on spheroids, moreover we show that lateral coupled cylinders are more flexible to tune the enhancement factor than all other.

To compare liver proteolysis and proteasome activation in steatotic liver grafts conserved in University of Wisconsin (UW) and Institut Georges Lopez-1 (IGL-1) *** liver grafts from male obese Zücker rats were conserved in UW and IGL-1 solutions for 24 h at 4 °Cand subjected to “ex vivo” normo-thermic perfusion (2 h; 37 °C). Liver proteolysis in tissue specimens and perfusate was measured by reverse-phase high performance liquid chromatography. Total free amino acid release was correlated with the activation of the ubiquitin proteasome system (UPS: measured as chymotryptic-like activity and 20S and 19S proteasome), the prevention of liver injury (transaminases), mitochondrial injury (confocal microscopy) and inflammation markers (TNF 1 alpha, high mobility group box-1 (HGMB-1) and PPAR gamma), and liver apoptosis (TUNEL assay, cytochrome c and caspase 3).RESULTSProfiles of free AA (alanine, proline, leucine, isoleucine, methionine, lysine, ornithine, and threonine, among others) were similar for tissue and reperfusion effluent. In all cases, the IGL-1 solution showed a significantly higher prevention of proteolysis than UW (P < 0.05) after cold ischemia reperfusion. Livers conserved in IGL-1 presented more effective prevention of ATP-breakdown and more inhibition of UPS activity (measured as chymotryptic-like activity). In addition, the prevention of liver proteolysis and UPS activation correlated with the prevention of liver injury (AST/ALT) and mitochondrial damage (revealed by confocal microscopy findings) as well as with the prevention of inflammatory markers (TNF1alpha and HMGB) after reperfusion. Liver ischemia-reperfusion injury(IRI) is an inherent feature of liver surgery and liver transplantation in which damage to a hypoxic organ(ischemia) is exacerbated following the return of oxygen delivery(reperfusion). IRI is a major cause of primary nonfunction after transplantation and may lead to graft rejection, regardless of immunological considerations. The immediate response involves the disruption of cellular mitochondrial oxidative phosphorylation and the accumulation of metabolic intermediates during the ischemic period, and oxidative stress during blood flow restoration. Moreover, a complex cascade of inflammatory mediators is generated during reperfusion, contributing to the extension of the damage and finally to organ failure. A variety of pharmacological interventions(antioxidants, anticytokines, etc.) have been proposed to alleviate graft injury but their usefulness is limited by the local and specific action of the drugs and by their potential undesirable toxic effects. Polyethylene glycols(PEGs), which are non-toxic water-soluble compounds approved by the FDA, have been widely used as a vehicle or a base in food, cosmetics and pharmaceuticals, and also as adjuvants for ameliorating drug pharmacokinetics. Some PEGs are also currently used as additives in organ preservation solutions prior to transplantation in order to limit the damage associated with cold ischemia reperfusion. More recently, the administration of PEGs of different molecular weights by intravenous injection has emerged as a new therapeutic tool to protect liver grafts from IRI. In this review, we summarize the current knowledge concerning the use of PEGs as a useful target for limiting liver IRI.
