colon cancer and the immune system:The role of tumor invading T cells

colon cancer is still one of the leading causes of cancer death worldwide. Although the host immune system has been shown to react against tumor cells, mainly through tumor infi ltrating lymphocytes and NK cells, tumor cells may utilize different ways to escape anti-tumor immune response. Tumor infi ltration of cD8+ and cD4+ (T-bet+) effector T cells has been attributed to a beneficial outcome, and the enhancement of T cell activation through T cell receptor stimulation and co-stimulatory signals provides promising strategies for immunotherapy of colon cancer. Growing evidence supports a role for the Fas/FasL system in tumor immunology, although the mechanisms and consequences of FasL activation in colon cancer are not completely understood. In animal models, depletion of regulatory T cells (cD4+ cD25+ T cells) can enhance the anti-tumor immune response under certain conditions. Taken together, recent insights in the immune reaction against colon carcinoma have provided new approaches to immunotherapy, although much remains to be learned about the exact mechanisms.

chemokine receptor cXcR4-prognostic factor for gastrointestinal tumors

To review the implication of cXcR4 for gastrointestinal cancer, a ＂Pubmed＂ analysis was performed in order to evaluate the relevance of cXcR4 and its ligands for gastrointestinal cancers. Search terms applied were ＂cancer, malignoma, esophageal, gastric, colon, colorectal, hepatic, pancreatic, cXcR4, SDF- 1α, and SDF-1β＂. cXcR4 expression correlated with dissemination of diverse gastrointestinal malignomas. The cXcR4 ligand SDF-1α might act as ＂chemorepellent＂ while SDF-1β might act as ＂chemorepellent＂ for cTLs, inducing tumor rejection. The paracrine expression of SDF-1α was furthermore closely associated with neoangiogenesis. cXcR4 and its ligands influence the dissemination, immune rejection, and neoangiogenesis of human gastrointestinal cancers. Inhibition of cXcR4 might be an interesting therapeutic option.

Biochemical, anatomical, and physiological characteristics of the brain make it especially vulnerable to insult. Specifically, some of these characteristics such as myelin and a high energy requirement provide for the introduction of free radical-induced insult. Recently, the biochemistry of free radicals has received considerable attention. It also has become increasingly suggestive that many drug and chemical-induced toxicities may be evoked via free radicals and oxidative stress. Major points addressed in this work are the regulation of neural-free radical generation by antioxidants and protective enzymes, xenobiotic-induced disruption of cerebral redox status, and specific examples of neurotoxic agent-induced alterations in free radicals as measured by the fluorescent probe dichlorofluorescein. This article considers that free radical mechanisms may contribute significantly to the properties of several diverse neurotoxic agents and proposes that free radicals may be common phenomena of neurotoxicity.

Implications of cMS analysis of photon-photon interactions for photon PDFs

As part of a recent analysis of exclusive two-photon production of W＋W- pairs at the LHc, the cMS experiment used di-lepton data to obtain an ＂effective＂ photon-photon luminosity. We show how the cMS analysis on their 8 TeV data, along with some assumptions about the likelihood for events in which the proton breaks up to pass the selection criteria, can be used to significantly constrain the photon parton distribution functions, such as those from the cTEQ, MRST, and NNPDF collaborations. We compare the data with predictions using these photon distributions, as well as the new LUXqed photon distribution. We study the impact of including these data on the NNPDF2.3QED, NNPDF3.0QED and cT14QEDinc fits. We find that these data place a useful and complementary cross-check on the photon distribution, which is consistent with the LUXqed prediction while suggesting that the NNPDF photon error band should be significantly reduced. Additionally, we propose a simple model for describing the two-photon production of W^＋W^- at the LHc. Using this model, we constrain the number of inelastic photons that remain after the experimental cuts are applied.

Developments in Humidity Standards and the Psychrometer Equation

Determination of the wet-bulb temperature at the surface of a material is the basis of one class ofhumidity measuring instruments, and is important in industrial applications such as dryer modelling and simula-tion. The psychrometer equation is a frequently used method of estimating wet-bulb temperature, and contains apsychrometer 'constant'. Analysis shows that this is in fact a variable coefficient affected by temperature, pressure,radiation and conduction effects, and the identity of the gas and vapour. Radiation and conduction affect the dif-ference between adiabatic saturation temperature and indicated wet-bulb temperature. Inconsistencies in currentlyrecommended values for the psychrometer coefficient, including published international standards, are identifiedand explained. Particular problems arise when the enhancement factor is applied to vapour pressure to accountfor non-ideality of gases. Special considerations are also needed for wet-bulb temperatures approaching the boilingpoint, where the psychrometer coefficient tends to zero. Self-consistent recommendations recently published in thenew British Standard BS1339 are given, which cover both the air-water system and a general vapour-gas system.

campylobacterjejuni in Musca domestica： An examination of survival and transmission potential in light of the innate immune responses of the house flies

The house fly, Musca domestica, has been implicated as a vector of campy- lobacter spp., a major cause of human disease. Little is known whether house flies serve as biological amplifying hosts or mechanical vectors for campylobacterjejuni. We in- vestigated the period after c. jejuni had been ingested by house flies in which viable c. jejuni colonies could be isolated from whole bodies, the vomitus and the excreta of adult M. domestica and evaluated the activation of innate immune responses of house flies to ingested c. jejuni over time. c. jejuni could be cultured from infected houseflies soon after ingestion but no countable c. jejuni colonies were observed 〉 24 h postingestion. We detected viable c. jejuni in house fly vomitus and excreta up to 4 h after ingestion, but no viable bacteria were detected 〉 8 h. Suppression subtractive hybridization identi- fied pathogen-induced gene expression in the intestinal tracts of adult house flies 4-24 h after ingesting c. jejuni. We measured the expression of immune regulatory （thor, JNK, and spheroide） and effector （cecropin, diptericin, attacin, defensing, and lysozyme） genes in c. jejuni-infected and -uninfected house flies using quantitative real time PcR. Some house fly factor, or combination of factors, eliminates c. jejuni within 24 h postingestion. Because c. jejuni is not amplified within the body of the housefly, this insect likely serves as a mechanical vector rather than as a true biological, amplifying vector for c. jejuni, and adds to our understanding of insect pathogen interactions.

Reduction of PDF uncertainty in the measurement of the weak mixing angle at the ATLAS experiment

We investigate the parton distribution function(PDF)uncertainty in the measurement of the effective weak mixing angle sin2θ^(l)_(eff)at the cERN Large Hadron collider(LHc).The PDF-induced uncertainty is large in proton-proton collisions at the LHc due to the dilution *** measurement of the Drell-Yan forward-backward asymmetry(ApB)at the LHc can be used to reduce the PDF uncertainty in the sin2θ^(l)_(eff)***,when including the full mass range of lepton pairs in the AFB data analysis,the correlation between the PDF updating procedure and the sin2θ^(l)_(eff) extraction leads to a sizable bias in the obtained sin2θ^(l)_(eff)*** our studies,we find that the bias can be significantly reduced by removing Drell-Yan events with invariant mass around the Z-pole region,while most of the sensitivity in reducing the PDF uncertainty ***,the lepton charge asymmetry in the^boson events as a function of the rapidity of the charged leptons,A±(ηl),is known to be another observable which can be used to reduce the PDF uncertainty in the sin2θ^(l)_(eff)*** constraint from A±(ηl)is complementary to that from AFB,and thus no bias affects the sin2θ^(l)_(eff)*** studies are performed using the error PDF Updating Method Package(ePump),which is based on Hessian updating *** this article,the cT14HERA2 PDF set is used as an example.

Identifying sediment discontinuities and solving dating puzzles using monitoring and palaeolimnological records

Palaeolimnological studies should ideally be based upon continuous, undisturbed sediment sequences with reliable chronologies. However for some lake cores, these conditions are not met and palaeolimnologists are often faced with dating puzzles caused by sediment disturbances in the past. This study chooses Esthwaite Water from England to illustrate how to identify sedimentation discontinuities in lake cores and how chronologies can be established for imperfect cores by correlation of key sediment signatures in parallel core records and with long-term monitoring data （1945-2003）. Replicated short cores （ESTH1, ESTH7, and ESTH8） were collected and subjected to loss-on-ignition, radiometric dating （210Pb, 137cs, and 14c）, particle size, trace metal, and fossil diatom analysis. Both a slumping and a hiatus event were detected in ESTH7 based on comparisons made between the cores and the long-term diatom data. Ordination analysis suggested that the slumped material in ESTH7 originated from sediment deposited around 1805-1880 AD. Further, it was inferred that the hiatus resulted in a loss of sediment deposited from 1870 to 1970 AD. Given the existence of three superior 14c dates in ESTH7, ESTH 1 and ESTH7 were temporally correlated by multiple palaeolimnological proxies for age-depth model development. High variability in sedimentation rates was evident, but good agreement across the various palaeo- limnological proxies indicated coherence in sediment processes within the coring area. Differences in sedimenta- tion rates most likely resulted from the natural morphology of the lake basin. Our study suggests that caution is required in selecting suitable coring sites for palaeolimno-logical studies of small, relatively deep lakes and that proximity to steep slopes should be avoided wherever possible.
