Understanding genome-wide diversity,inbreeding,and the burden of accumulated deleterious mutations in small and isolated populations is essential for predicting and enhancing population persistence and ***,these effects are rarely studied in limestone karst ***,we re-sequenced the nuclear genomes of 62 individuals of the Begonia masoniana complex(***,***,*** and ***)and investigated genomic divergence and genetic load for these four *** analyses revealed four distinct clusters corresponding to each species within the ***,there was only limited admixture between *** and *** occurring in overlapping geographic *** species experienced historical bottlenecks during the Pleistocene,which were likely caused by glacial climate *** detected an asymmetric historical gene flow between group pairs within this timeframe,highlighting a distinctive pattern of interspecific divergence attributable to karst geographic *** found that isolated populations of *** have limited gene flow,the smallest recent population size,the highest inbreeding coefficients,and the greatest accumulation of recessive deleterious *** findings underscore the urgency to prioritize conservation efforts for these isolated *** study is among the first to disentangle the genetic differentiation and specific demographic history of karst Begonia plants at the whole-genome level,shedding light on the potential risks associated with the accumulation of deleterious mutations over generations of ***,our findings may facilitate conservation planning by providing critical baseline genetic data and a better understanding of the historical events that have shaped current population structure of rare and endangered karst plants.

BACKGROUND Para-aortic lymph nodes(PALNs)are common sites for the regional spread of cervical squamous cell carcinoma(SCC).CASE SUMMARY We report the case of a 36-year-old woman who presented with cervical SCC with multiple bulky PALNs,largest measured 4.5 cm×5 cm×10 *** patient was treated with radical intent with definitive chemoradiation using sequential doseescalated adaptive radiotherapy,followed by maintenance *** patient achieved a complete response;she has been doing well since the completion of treatment with no evidence of the disease for 2 *** Regardless of the size of PALN metastases of cervical carcinoma origin,it is still treatable(with radical intent)via concurrent *** radiotherapy allows dose escalation with minimal toxicity.

Genome sequencing has revealed frequent mutations in Ras homolog family member A(RHOA) among various cancers with unique aberrant profiles and pathogenic effects, especially in peripheral T-cell lymphoma(PTCL). The discrete positional distribution and types of RHOA amino acid substitutions vary according to the tumor type, thereby leading to different functional and biological properties, which provide new insight into the molecular pathogenesis and potential targeted therapies for various tumors. However, the similarities and discrepancies in characteristics of RHOA mutations among various histologic subtypes of PTCL have not been fully elucidated. Herein we highlight the inconsistencies and complexities of the type and location of RHOA mutations and demonstrate the contribution of RHOA variants to the pathogenesis of PTCL by combining epigenetic abnormalities and activating multiple downstream pathways. The promising potential of targeting RHOA as a therapeutic modality is also outlined. This review provides new insight in the field of personalized medicine to improve the clinical outcomes for patients.

Inflammatory bowel disease(IBD)is a serious disorder,and exploration of active compounds to treat it is *** acidic polysaccharide named SUSP-4 was purified from Selaginella uncinata(Desv.)Spring,which contained galacturonic acid,galactose,xylose,arabinose,and rhamnose with the main chain structure of→4)-α-d-GalAp-(1→and→6)-β-d-Galp-(1→and the branched structure of→5)-α-l-Araf-(1→.Animal experiments showed that compared with Model group,SUSP-4 significantly improved body weight status,disease activity index(DAI),colonic shortening,and histopathological damage,and elevated occludin and zonula occludens protein 1(ZO-1)expression in mice induced by dextran sulfate sodium salt(DSS).16S ribosomal RNA(rRNA)sequencing indicated that SUSP-4 markedly downregulated the level of Akkermansia and *** results confirmed that SUSP-4 obviously elevated thiamine levels compared with Model mice by adjusting thiamine metabolism,which was further confirmed by a targeted metabolism *** transplantation experiments showed that SUSP-4 exerted an anti-IBD effect by altering the intestinal flora in mice.A mechanistic study showed that SUSP-4 markedly inhibited macrophage activation by decreasing the levels of phospho-nuclear factor kappa-B(p-NF-κB)and cyclooxygenase-2(COX-2)and elevating NF-E2-related factor 2(Nrf2)levels compared with Model *** conclusion,SUSP-4 affected thiamine metabolism by regulating Akkermania and inhibited macrophage activation to adjust NF-κB/Nrf2/COX-2-mediated inflammation and oxidative stress against *** is the first time that plant polysaccharides have been shown to affect thiamine metabolism against IBD,showing great potential for in-depth research and development applications.

The Response of carbonate System to Watershed Urbanization Process in a Semi-Arid River

Different from rivers in humid areas,the variability of riverine CO_(2) system in arid areas is heavily impacted by anthropogenic disturbance with the increasing urbanization and water *** this study,the water chemistry and the controls of carbonate system in an urbanized river(the Fenhe River)on the semi-arid Loess Plateau were *** water chemistry of the river water showed that the high dissolved inorganic carbon(DIC)concentration(about 37 mg L^(-1))in the upstream with a karst land type was mainly sourced from carbonate weathering involved by H_(2)CO_(3) and H_(2)SO_(4),resulting in an oversaturated partial pressure of CO_(2)(pCO_(2))(about 800μatm).In comparison,damming resulted in the widespread appearance of non-free flowing river segments,and aquatic photosynthesis dominated the DIC and pCO_(2) spatiality demonstrated by the enriched stable isotope of DIC(δ^(13)CDIC).Especially in the mid-downstream flowing through major cities in warm and low-runoff August,some river segments even acted as an atmospheric CO_(2) *** noteworthy is wastewater input leading to a sudden increase in DIC(>55 mg L^(-1))and pCO_(2)(>4500μatm)in the downstream of Taiyuan City,and in cold November the increased DIC even extended to the outlet of the *** results highlight the effects of aquatic production induced by damming and urban sewage input on riverine CO_(2) system in semi-arid areas,and reducing sewage discharge may mitigate CO_(2) emission from the rivers.

Effect of Fe_(2)O_(3)on the Structure,Physical Properties and Crystallization of CaO-Al_(2)O_(3)-SiO_(2)Glass

The calcium aluminosilicate-based glasses(CaO-Al_(2)O_(3)-SiO_(2),CAS)with different Fe_(2)O_(3)content(0.10wt%,0.50wt%,0.90wt%,and 1.30wt%)were prepared by traditional melt-quenching *** glass network structure,thermal and mechanical properties,and crystallization behavior changes were investigated by nuclear magnetic resonance spectrometer,Fourier-transform infrared spectro-photometer,X-ray diffractometer,differential scanning calorimetry and field emission scanning electron microscope *** change of Q^(n)in glass structures reveals the glass network connectivity decreases due to the increasing content of Fe_(2)O_(3)addition,resulting in the increasing of non-bridging number in glass *** glass densities slightly rise from 2.644 to 2.681 g/cm^(3),while Vickers’s hardness increases at first,from 6.469 to 6.901 GPa,then slightly drops to 6.745 GPa,with Fe_(2)O_(3)content *** is almost no thermal expansion coefficient change from different Fe_(2)O_(3)*** glass transmittance in visible range gradually decreases with higher Fe_(2)O_(3)content,resulting from the strong absorption of Fe^(2+)and Fe^(3+)*** calculated activation energy from thermal analysis results first decreases from 282.70 to 231.18 kJ/mol,and then increases to 244.02 kJ/mol,with the Fe_(2)O_(3)content increasing from 0.10wt%to 1.30wt%.Meanwhile,the maximum Avrami constant of 2.33 means the CAS glasses exhibit two-dimensional *** of the CAS glass-ceramics samples contain main crystal phase of anorthite,the microstructure appears lamellar and columnar crystals.

Unveiling the time-temperature dependence of metastability of supercooled liquid using nano-calorimetry

There is a lack of understanding of both the conversion of an unstable glass into a metastable supercooled liquid(MSL) upon heating and the metastability of MSLs. In this study, we investigated the time-and temperature-dependent metastability of an MSL using an advanced nano-calorimetric technique. The chosen Au-based metallic glass(Au MG) allowed adequate probing of its MSL in a temperature range between 10 and 70 K above the standard glass transition temperature. We found that the survival time of the MSL state is a quadratic function of temperature. Beyond this duration threshold, the sample undergoes fast crystallization even if it is below the crystallization temperature that is measured using differential scanning *** transmission electron microscopy, we observed the formation of clusters with a partially ordered lattice structure during relaxation in the Au MG sample fabricated using a nano-calorimeter. The atomic ordering within the clusters was enhanced by increasing time and temperature in the MSL region. Once the as-produced glass entered the MSL stage upon heating followed by a quenching stage at a given rate, the mechanical properties of the quenched glass remained the same regardless of its holding temperature and duration within the MSL region. This work provides insights into the glass-MSL-crystal transformation and offers guidance for designing standard metallic glasses for property characterizations.

Selection of Negative Charged Acidic Polar Additives to Regulate Electric Double Layer for Stable Zinc Ion Battery

Negative charged acidic polarity (NCAP) has been unveiled as the guideline for selecting additives to regulate electric double layer (EDL) for Zn-ion *** glutamate has been verified to regulate EDL structure with synergetic effects, including preferential adsorption on Zn anode and reconstruction of hydrated Zn-ion *** NCAP additives, Zn|Cu half-cell achieves a high Coulombic efficiency of 99.83% for 2000 cycles, and NH4V4O10|Zn full cell realizes a high-capacity retention of 82.1% for 3000 cycles. Zinc-ion batteries are promising for large-scale electrochemical energy storage systems, which still suffer from interfacial issues, e.g., hydrogen evolution side reaction (HER), self-corrosion, and uncontrollable dendritic Zn electrodeposition. Although the regulation of electric double layer (EDL) has been verified for interfacial issues, the principle to select the additive as the regulator is still misted. Here, several typical amino acids with different characteristics were examined to reveal the interfacial behaviors in regulated EDL on the Zn anode. Negative charged acidic polarity (NCAP) has been unveiled as the guideline for selecting additive to reconstruct EDL with an inner zincophilic H2O-poor layer and to replace H2O molecules of hydrated Zn2+ with NCAP glutamate. Taking the synergistic effects of EDL regulation, the uncontrollable interface is significantly stabilized from the suppressed HER and anti-self-corrosion with uniform electrodeposition. Consequently, by adding NCAP glutamate, a high average Coulombic efficiency of 99.83% of Zn metal is achieved in Zn|Cu asymmetrical cell for over 2000 cycles, and NH4V4O10|Zn full cell exhibits a high-capacity retention of 82.1% after 3000 cycles at 2 A g-1. Recapitulating, the NCAP principle posted here can quicken the design of trailblazing electrolyte additives for aqueous Zn-based electrochemical energy storage systems.
