Molecular characterization of two novel esterase genes from carmine spider mite, Tetranychus cinnabarinus （Acarina： Tetranychidae）

Two novel esterase complementary DNAs were identified and cloned from the insecticide-susceptible strain of Tetranychus cinnabarinus （Boisduval）（Acarina： Tetrany- chidae）, which were designated as TCE1 and TCE2, respectively. The cDNA of TCE1 gene contained an open reading frame （ORF） of 1701 bp encoding 567 amino acids, and a predicted molecular weight of 62.75 kDa, the cDNA of TCE2 contained an ORF of 1680 bp encoding 560 amino acids, and a predicted molecular weight of 63.14 kDa. TCE1 and TCE2 were submitted to GenBank, accession number EU 130461 and EU 130462. The well-conserved sequence motif, GXSXG, used as a signature pattern in the esterase fam- ily are present in both TCE1 and TCE2 （GQSAG in TCE1, whereas GESAG in TCE2）, indicating that these two genes are predicted to be esterases. Comparison of the deduced amino acid sequence with the published mite esterase sequence coming from Boophilus microplus showed that TCE1 shares 33.98% identity and TCE2 shares 33.46% identity. TCE1 and TCE2 share 46.4% identity. Quantitative real-time polymerase chain reaction revealed that expression level of the TCE2 gene was relatively higher than that of the TCE1 in all instars examined except the protonymph, and the expression level of these two esterase genes in adults of T. cinnabarinus was significantly higher than that in any other instars, respectively. T. cinnabarinus is an important agricultural mite pest and esterases are important in the metabolisms of insects and mites; the genomic information obtained in this study will contribute to esterase molecular biological study on mite pest species.

The stability issue has been acknowledged as the bottleneck in the practical application of perovskite photovoltaics,while the stabilized interface between the perovskites and charge transport layers dominates their stability performance under different ***,we developed a high-performance sulfurized zinc-titanium mixed oxide(ZTO-S)electron transport layer(ETL)to fabricate large-area efficient and long-term 85℃/85%RH stable perovskite solar *** scalably prepared ZTO-S using the facile spray coating method demonstrates excellent electron mobility close to that of Zn O,in addition to promoting the uniform crystallization of perovskite film across the entire module via the interaction between surface S and Pb^(2+).Furthermore,this novel coordination stabilized the interface and reduced the interfacial non-radiative recombination defects within the devices,yielding an efficient and stable performance for the *** efficiency of 21.73%and 17.50%was achieved for blade-coated 36 cm^(2)and 100 cm^(2)perovskite solar modules,*** addition,the encapsulated module(36 cm^(2))shows an attractive humidity and heat stability(85℃/85%RH)performance with a maintained 93.5%of the initial PCE over 1000 h.

Effects of Shexiangbaoxin pills on the expression of cardiac α1- and β-adrenergic receptor subtypes in rat hearts with heart failure induced by myocardial infarction

Background Chronic heart failure （CHF） had been characterized as an activated sympathetic system leading to the alteration of adrenergic receptor （AR） levels in the heart. Thus far, not much research has been done with regard to traditional Chinese medical treatment for CHF. We investigated the effect of Shexiangbaoxin pills （SXBXP） on the function of the heart and the expression of a1-AR and b-AR subtypes in the messenger RNA （mRNA） levels and protein levels of non-infarction left ventricular tissue from rats with CHF induced by myocardial infarction.Methods Models of CHF were established by left anterior descending coronary artery ligature. Fifty-four Wistar rats were randomly divided into five groups： normal control group （group A）, sham operation group （group B）, CHF model group （group C）, positive medicine control group （group D）, and small-dose SXBXP group （group E） and large-dose SXBXP group （group F）, deployed intragastrically. Cardiac function was examined by echocardiography before and after therapy; mRNA expressed levels were measured by semiquantitative reverse transcription polymerase chain reaction （RT-PCR） for b1-AR, b2-AR, b3-AR, a1A-AR, a1B-AR, and a1D-AR; protein levels were measured by Western blotting analysis for b1-AR, b2-AR, a1A-AR, a1B-AR, and a1D-AR in non-infarction left ventricular tissue.Results There was no significant difference in the left ventricular ejection fraction （LVEF） between groups A and B. Compared to group B, LVEF of groups C, D, E, and F were significantly decreased （P 〈0.01） before therapy. After therapy, compared to group C, LVEF of group F was significantly improved （P 〈0.05）. Compared to group B, b1-AR and a1B-AR expressed levels were markedly decreased （P 〈0.05）, a1A-AR and b3-AR were significantly increased （P 〈0.01） in group C, and in both mRNA and protein expressed levels b2-AR had no significant difference between groups B and C （P 〉0.05）. a1D-AR mRNA levels were unchanged in each group （P 〉0.05）, but a1D-AR

Maintaining open flowers is critical for successful pollination and depends on long-term water and carbon *** the relationship between how flower hydraulic traits are coordinated in different habitats is poorly ***,we hypothesize that the coordination and trade-offs between floral hydraulics and economics traits are independent of environmental *** test this hypothesis,we investigated a total of 27 flower economics and hydraulic traits in six aquatic and six terrestrial herbaceous species grown in a tropical botanical *** found that although there were a few significant differences,most flower hydraulics and economics traits did not differ significantly between aquatic and terrestrial herbaceous *** flower mass per area and floral longevity were significantly positively correlated with the time required for drying full-hydrated flowers to 70%relative water *** dry matter content was strongly and positively related to drought tolerance of the flowers as indicated by flower water potential at the turgor loss *** addition,there was a trade-off between hydraulic efficiency and the construction cost of a flower across *** results show that flowers of aquatic and terrestrial plants follow the same economics spectrum *** results suggest a convergent flower economics design across terrestrial and aquatic plants,providing new insights into the mechanisms by which floral organs adapt to aquatic and terrestrial habitats.

Identification of Sheep Endogenous Beta-Retroviruses with Uterus-Specific Expression in the Pregnant Mongolian Ewe

The sheep genome harbours approximately 20 copies of endogenous beta-retroviruses （enJSRVs）, and circumstantial evidence suggests that enJSRVs might play a role in mammalian reproduction, particularly placental morphogenesis. This study was aimed to assess the expression of mRNAs of an enJSRV and its receptor, HYAL2, in the uterus and conceptuses of Mongolian ewes throughout gestation, using real-time reverse transcription polymerase chain reaction and in situ hybridization analysis. The results showed that enJSRV and HYAL2 mRNAs were found to be expressed throughout gestation in the endometrium, chorion, placenta, and conceptus. The enJSRV mRNA was most abundant in the placenta on day 90 of pregnancy, in the endometrium on day 30 and 50, and in the chorion on day 70 and 110. However, HYAL2 mRNA was most abundant in the endometrium on day 30. These differences were all significantly different from each other （P〈0.01）. In situ hybridization showed that enJSRV and HYAL2 mRNAs were specifically expressed in endometrial luminal epithelium and glandular epithelium, trophoblastic giant binucleated cells （BNCs）, endometrial caruncles, placental cotyledons, stroma, trophectoderm, as well as multinucleated syncytia of the placenta and blood vessel endothelial cells. Collectively, little is known about the molecular mechanisms by which trophoblastic differentiation and multinucleated syncytia formation are regulated by enJSRVs. However, the temporal and spatial distributions of enJSRV expression in the uterus and conceptus indicate that differentiation of BNCs and the formation of a multinucleated syncytiotrophoblast involve enJSRV and possibly its cellular receptor, HYAL2. Therefore, enJSRV and HYAL2 appear to play important roles in the female reproductive physiology in this breed of sheep.

Analysis of QTLs for erucic acid and oil content in seeds on A8 chromosome and the linkage drag between the alleles for the two traits in Brassica napus

The history of canola breeding began with the discovery of germplasm with low erucic acid content in seeds of spring forage cultivar in the 1950＇s. FAEI mutations led to a dramatic decrease of the seed erucic acid content in Arabidopsis thaliana. The products of the two FAE1 loci, ***1 and ***1, showed additive effects to the level of erucic acid content in oilseed rape. Previous research be- lieved that the pleiotropy of FAE1 was responsible for the decrease in seed oil content along with the reduction of seed erucic acid content in the modem cultivars. TN DH population was developed from a canola cultivar Tapidor and a Chinese traditional cultivar Ningyou7. The population had been tested in 10 and 11 environments to map QTLs for the erucic acid content and oil content in seeds. As the map resolution increased, a novel QTL for seed erucic acid content was revealed, after Meta-analysis, 7 cM away from the most significant seed erucic acid content QTL where ***1 is located. Seven independent QTLs for seed oil content （qOC） were detected around the two seed erucic acid content QTLs （qEA） across 39.20 cM on linkage group AS. Two of the qOCs co-localized with the two qEAs, respectively, and were detected in a single environment. The other five qOCs were detected in 10 of 11 environments independent ofqEAS. Alleles from Tapi- dot in all the QTLs at the （~39.20 cM region contributed negative effects to either erueic acid content or oil content in seeds. Parallel, geno- typing showed that on 5 of the 7 QTLs regions, Tapidor alleles had the same genotypes with that in ＇Liho＇, the original low seed erucic acid content source. Through rounds of crossbreeding with oil-cropped eultivars and intensive selection for multi generations, Tapidor still had the inferior alleles for low seed oil content from ＇Liho＇, the forage rape. This showed a strong linkage drag of low seed oil content, which was controlled by the five qEA-independent qOCs, with low seed erucic acid

Effects of supercritical ammonia on bamboo pulping

We investigated the effects of supercritical ammonia on pulping of bamboo in a preliminary study,using testers such as differential scanning calorimetry（DSC）,scanning electron microscopy（SEM）,X-ray diffraction（XRD） and energy dispersive spectroscopy（EDS）.The results indicate that under supercritical conditions at 185-190°C,it is possible for the ammonia to realize bamboo *** with conventional kraft pulping,the products can meet the quality requirements of industrial paper.
