A controlled hydroponic experiment was undertaken to investigate Cd uptake in relation to the activity of Cd species in solution other than the free ion （Cd^2＋） by maintaining a constant Cd^2＋ activity under variable SO4^2- and Cl^- concentrations exposed to maize （Zea mays vat. cameron） plants. The objectives of these experiments were：（1） to distinguish and quantify the different uptake rates of free and inorganic-complexed Cd from nutrient solution, and （2） to model the uptake of Cd by maize with a Biotic ligand Model （BlM） in a system which facilitates the close examination of root characteristics. Results of the current experiments suggest that, in addition to the free ion, CdSO4^0 complexes are important factors in determining Cd uptake in nutrient solution by maize plants. Higher nominal SO4^2- concentrations in solution generally resulted in a greater Cd accumulation by maize plants than predicted by the Cd^2＋ activity. A better integration of the complete dataset for the 3 harvest times （6, 9 and 11 days after treatment） was achieved by including consideration of both the duration of Cd exposure and especially the root surface area to express Cd uptake. Similarly, the fit of the BlM was also improved when taking into account exposure time and expressing uptake in terms of root morphological parameters.

The timing of fruit maturity is an important trait in sweet cherry production and *** variation for phenology of fruit maturity in sweet cherry appears to be under strong genetic control,but that control might be complicated by phenotypic instability across *** such genotype-by-environment interaction(G×E)is a common phenomenon in crop plants,knowledge about it is lacking for fruit maturity timing and other sweet cherry *** this study,1673 genome-wide SNP markers were used to estimate genomic relationships among 597 weakly pedigree-connected individuals evaluated over two seasons at three locations in Europe and one location in the USA,thus sampling eight‘environments’.The combined dataset enabled a single meta-analysis to investigate the environmental stability of genomic *** disequilibrium among marker loci declined rapidly with physical distance,and ordination of the relationship matrix suggested no strong structure among *** most parsimonious G×E model allowed heterogeneous genetic variance and pairwise covariances among ***-sense genomic heritability was very high(0.60–0.83),as was accuracy of predicted breeding values(>0.62).Average correlation of additive effects among environments was high(0.96)and breeding values were highly correlated across *** indicated that genomic models can be used in cherry to accurately predict date of fruit maturity for untested individuals in new *** G×E for this trait indicated that phenotypes of individuals will be stable across similar *** analyses for other sweet cherry traits,for which multiple years of data are commonly available among breeders and cultivar testers,would be informative for predicting performance of elite selections and cultivars in new environments.

Implant associated infections are a critical health concern following orthopaedic surgery. Sustained local delivery of antibiotics has been suggested as a means of preventing these infections. Poly(D,l-lactide) (PDllA) is a biodegradable polymer that has been used to coat implants for the delivery of antibiotics and other bioactive molecules. While effective, these studies show that antibiotics are released in a burst profile. Here we evaluated a method for controlled release of gentamicin from implant surfaces using the palmitate alkyl salt to decrease its solubility in aqueous solution. Steel Kirschner wires (K-wires) were coated with Gentamicin-palmitate (GP)-PDllA, gentamicin sulphate (GS)-PDllA or vancomycin sulphate (VS)-PDllA, and elution of antibiotics from coated K-wires investigated using HPlC/MS/ MS. In contrast to burst antibiotic release from the GS-PDllA and VS-PDllA groups, GP was released in a slower sustained manner. Colonisation and initial attachment of Staphylococcus aureus Xen29 to gentamicin-coated K-wires was reduced by 90% when compared to the non-coated control group. However there was no statistical difference in recovery of bacteria from GS or GP groups. Bacteria recovered from VS-PDllA coated K-wires decreased by 36%. Bioluminescence emitted by S. aureus Xen29 was also reduced over seven days in the antibiotic control groups, demonstrating that growth and biofilm development over the longer term was impaired by antibiotic-PDllA coating. These results indicate that using alkyl salts of antibiotics may be an effective strategy for controlling the release of antibiotics from implants.

Ir（Ⅲ） complexes with high quantum yields for triplet excited-state formation, long triplet lifetimes, and bright luminescence are potential theranostic photodynamic therapy（PDT） reagents. More importantly, the triplet excited-state characteristics of these complexes are readily tuned via alternation of the ligands However, most of the currently studied Ir（Ⅲ） complex photosensitizers mainly absorb in the UV and blue spectral region, which is not suitable for potential clinical applications. To red-shift the ground-state absorption and meanwhile to retain the desired triplet excited-state characteristics, we synthesized severa series of cationic or neutral Ir（Ⅲ） complexes that have strong absorption in the visible to the red spectra region and are emissive in the red/NIR region. Their photophysical properties and in vitro/in vivo theranostic PDT effects were systematically evaluated. Alternation of the different types of ligands dramatically impacted the natures and energies of the lowest singlet and triplet excited states of these complexes. Many of the complexes can efficiently generate singlet oxygen in solutions and other reactive oxygen species in vitro, and exhibited strong PDT effects upon visible or red light activation. One of the complexes showed the largest phototherapeutic index（＞1600） for the reported Ir（Ⅲ） complexes toward the SKMEl28 cells upon white light activation. These complexes also produce strong intracellular luminescence which highlights their potential as theranostic PDT agents.

草本菝葜组(Smilax ***)是一个东亚-北美间断分布的类群。其中东亚的2种——牛尾菜(***)和白背牛尾菜(***)在形态特征和分子树上均界限清晰,而北美的草本菝葜在分类上则混杂不清——不同的研究者将其处理成l至1 5个种。最近基于叶绿体和核DNA的研究虽显示出北美草本菝葜是一个单系类群,却没能解决它们之间的系统发育关系。本研究运用AFlP数据对取自北美的33个草本菝葜群体,145个个体进行了系统发育分析,结果显示:1)加州北部狭域分布的***群体和北美东部分布的其他草本种分别构成一个单系;2)阿帕拉契亚山脉南部的***群体位于北美东部支系的基部;3)北美东部的其它群体的分布显示出明显的地理结构,而与形态和分类特征无明显关系;4)建议将北美草本菝葜处理成2个种——***和***。为了进一步验证以上结果,我们对这33个群体的292个体的3个叶绿体基因组片段(rpoC1内含子,trnG-psbK和trnC-ycf6)进行了序列分析。基于cpDNA和AFlP数据分析,提出如下假说:草本菝葜在晚上新世经白令陆桥从东亚迁往北美,其后受更新世冰期影响南迁,受北美西部山脉(科迪勒拉山系)隔离而形成加州北部和北美东部的两个支系;北美东部的草本菝葜可能在阿帕拉契亚山脉的两侧均存在冰期避难所,其中,阿帕拉契亚山脉西侧的中西部群体随着第四纪晚期的气候震荡发生了反复的群体扩张和收缩;阿帕拉契亚山脉作为基因流的屏障促进了其东西两侧的种群分化。东亚草本菝葜的亲缘地理学研究则选取牛尾菜33个群体(235个体)和白背牛尾菜12群体(106个体),对叶绿体基因组的2个片段(atpI-atpH和rbcl-atpB)作了序列分析。结果显示,鄂西巫山山脉至湘西北武陵山脉和皖苏浙交界处的天目山脉具有最丰富的单倍型,是可能的冰期避难所;其中,白背牛尾菜所有的6种单倍型聚为一支,与鄂西的牛尾菜群体关系最为接近,表明冰期生境的片段化可能导致了东亚草本菝葜在鄂西的隔离和物种形成;另外,日本、韩国以及台湾地区的单倍型较为单一,且常衍生自鄂西及皖苏浙交界地区的群体,据此推测经历了从大陆→台湾岛、日本列岛和朝鲜半岛的迁移事件。

Using directed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily receptor IlT2(synonyms:lIR1,MIR7,CD85j),we have selected a range of mutants with binding affinities enhanced by up to 168,000-fold towards the conserved region of major histocompatibility complex(MHC)class I *** in a dimeric form,either by chemical cross-linking with bivalent polyethylene glycol(PEG)derivatives or as a genetic fusion with human IgG Fc-fragment,the mutants exhibited a further increase in ligand-binding strength due to the avidity effect,with resident half-times(t1/2)on the surface of MHC I-positive cells of many *** novel compounds antagonized the interaction of CD8 co-receptor with MHC I in vitro without affecting the peptide-specific binding of T-cell receptors(TCRs).In both cytokine-release assays and cell-killing experiments the engineered receptors inhibited the activation of CD8^(+)cytotoxic T lymphocytes(CTls)in the presence of their target cells,with subnanomolar potency and in a dose-dependent *** a selective inhibitor of CD8^(+)CTl responses,the engineered high affinity IlT2 receptor presents a new tool for studying the activation mechanism of different subsets of CTls and could have potential for the development of novel autoimmunity therapies.

目的研究严重急性呼吸综合征(SARS)患者的临床和影像学特征并评价世界卫生组织(WHO)关于SARS诊断指南的准确性。试验设计前瞻性观察研究。研究地点香港新界一所教学医院急诊科新建立的SARS鉴别门诊。研究对象有SARS患者接触史的556名医院工作人员、患者及曾访问过该门诊的工作人员和患者的亲属。主要观察指标 SARS患者的确诊人数。结果 556名研究对象中,收入院141人,其中97人被确诊为SARS。97例确诊患者中,发热、寒战、乏力、肌痛、畏寒、纳差、呕吐、腹泻、颈痛等非呼吸道症状的发生率均显著高于其他患者。WHO关于诊断可疑SARS患者指南的准确性为83％,其阴性预计值为86％(95％可信区间为83％～89％),敏感性为26％(17％～36％),特异性为96％(93％～97％)。结论现行WHO关于诊断可疑SARS患者的指南,用于诊断入院前的患者尚不够敏感,而每天随诊、评价非呼吸道和全身的症状以及胸部X线检查可能是更好的鉴别工具。
