A new variant of concern for SARS-CoV-2,Omicron(B.1.1.529),was designated by the World Health Organization on November 26,2021.This study analyzed the viral genome sequencing data of 108 samples collected from patients infected with Omicron.First,we found that the enrichment efficiency of viral nucleic acids was reduced due to mutations in the region where the primers anneal to.Second,the Omicron variant possesses an excessive number of mutations compared to other variants circulating at the same time(median:62 vs.45),especially in the Spike gene.Mutations in the Spike gene confer alterations in 32 amino acid residues,more than those observed in other SARS-CoV-2 variants.Moreover,a large number of nonsynonymous mutations occur in the codons for the amino acid residues located on the surface of the Spike protein,which could potentially affect the replication,infectivity,and antigenicity of SARS-CoV-2.Third,there are 53 mutations between the Omicron variant and its closest sequences available in public databases.Many of these mutations were rarely observed in public databases and had a low mutation rate.In addition,the linkage disequilibrium between these mutations was low,with a limited number of mutations concurrently observed in the same genome,suggesting that the Omicron variant would be in a different evolutionary branch from the currently prevalent variants.To improve our ability to detect and track the source of new variants rapidly,it is imperative to further strengthen genomic surveillance and data sharing globally in a timely manner.

制备了一种环境友好型马来酸酐改性β-环糊精(MAH-β-CD),温和条件下将其与聚醚胺(JD230)通过静电作用合成了具有CO_(2)响应性的表面活性剂(MJD230)。MJD230能与稠油形成稳定的水包油乳液,进而有效降低溶液的表面张力。在CO_(2)调控下,MJD230可重复用于稠油的乳化降黏和破乳。利用FTIR对MJD230结构进行了表征。通过观察降黏率和乳液粒径优化MJD230的合成条件,同时对降黏效果和乳液的稳定性进行了考察。将在反应温度为60℃,反应时间为60min,MAH-β-CD与JD230物质的量比为2∶1条件下制备的MJD230配成质量分数为0.5%的MJD230水溶液,其与稠油按照体积比为3∶7乳化后,稠油降黏率可达99.19%。反应体系pH和电导率的可逆变化证明了MJD230溶液对CO_(2)的响应性,这为表面活性剂驱油和CO_(2)捕集相结合提高稠油采收率提供了可行的途径。

Methyl tert-butyl ether (MTBE),as a widely used gasoline additive,is suspected of being environmentally toxic.MTBE accumulates mainly in adipose tissue,but its effect on obesity or obesity-related metabolic disorders has not been well understood yet.Therefore,we examined the effect of MTBE on the adipose function and the related metabolic processes with both 3T3-L1 cell line and C57BL/6J mice model.We found that exposure to MTBE at the environmental relevant concentration (100 μmol/L) could significantly induce differentiation of preadipocyte and disturb insulin-stimulated glucose uptake of mature adipocyte.The in vivo observation in male mice showed a positive correlation of visceral white adipose tissue (vWAT) expansion and cell size increase with MTBE treatment in 14 weeks.Glucose tolerance and insulin sensitivity tests demonstrated that MTBE at 1000 μg/(kg·day) disturbed the systemic glucose metabolism in a gender-specific manner,which might be partly attributed to the alterations of gut microbiota community at genus level with respect to Akkermansia,Clostridium XIVb,and Megamonas.In summary,our study characterized the effect of MTBE on adipose tissue function and glucose homeostasis in vitro and in vivo,and revealed that systemic disorders of the glucose metabolism might be modulated by the related gut microbiota.

Rat calvarial osteoblasts were treated with lanthanum chloride（LaCl3） to explore its effect on the mineral crystalline phase during the process of osteoblast calcification in uitro.The results confirmed that La was readily deposited in the mineral component of the matrix.Employing high-resolution transmission electron microscopy and Fourier transform infrared microspectroscopy techniques,we demonstrated that features comparable to dicalcium phosphate dihydrate（DCPD） and octacalcium phosphate,and hydroxyapatite（HAP） were detected in the mineral phases in uitro.Particularly,LaCl3 treatment retarded conversion from DCPD-like phase into HAP during mineralization.In addition,La was introduced in DCPD powder during wet chemical synthesis.When compared with that of La-free DCPD,the dissolution rate of La-incorporated DCPD was lower,thereby leading to a delayed DCPD-to-HAP phase transformation.Thus,it can be concluded that LaCl3 treatment influences the kinetics of inorganic phase transition by decreasing the dissolution rate of DCPD.

目的探讨急性呼吸窘迫综合征(ARDS)患者机械通气治疗时采用肺牵张指数(SI)滴定呼气末正压(PEEP)的临床实用性。方法采用平行随机对照试验,选取2022年8月至2023年2月长江航运总医院重症医学科接诊的需要机械通气的中、重度ARDS患者,并随机分为SI指导PEEP滴定组(SI组)和压力-容积曲线(P-V曲线)吸气支低位拐点(LIP)指导PEEP滴定组(LIP组)。所有患者入科后均采用仰卧位通气,并将床头抬高30°,积极治疗原发病,俯卧位通气12 h/d,采用控制性肺膨胀法进行肺复张等肺保护性通气策略,在此基础上,SI组以SI滴定机械通气参数;LIP组以P-V曲线吸气支LIP+2 cmH2O(1 cmH2O≈0.098 kPa)滴定机械通气参数。于复张前及治疗1、3、5 d监测氧合指数(PaO_(2)/FiO_(2))以及肺动态顺应性(Cdyn)、气道峰压(Pip)等呼吸力学指标。对比两组患者治疗效果。结果最终SI组41例,LIP组40例,两组患者性别、年龄、疾病类型等一般资料比较差异均无统计学意义。SI组机械通气时间(d:9.47±3.36比14.68±5.52)、重症监护病房(ICU)住院时间(d:22.27±4.68比27.57±9.52)均较LIP组明显缩短(均P0.05〕。两组在复张前PaO_(2)/FiO_(2)及呼吸力学指标差异均无统计学意义。在治疗5 d时,SI组PaO_(2)/FiO_(2)较LIP组明显改善〔mmHg(1 mmHg≈0.133 kPa):225.57±47.85比198.32±31.59,P<0.05〕,Cdyn较LIP组明显升高(mL/cmH2O:47.39±6.71比35.88±5.35,P<0.01),Pip较LIP组明显降低(mmHg:35.85±5.77比43.87±6.68,P<0.05)。Kaplan-Meier生存曲线显示,两组28 d累积生存率差异无统计学意义(Log-Rank:χ^(2)=2.348,P=0.125)。结论应用SI滴定PEEP治疗ARDS患者可能改善预后。

Characteristics of the water-soluble components of aerosol particles in Hefei,China

Size-classified daily aerosol mass concentrations and concentrations of water-soluble inorganic ions were measured in Hefei,China,in four representative months between September 2012 and August 2013.An annual average mass concentration of 169.09μg/m^3 for total suspended particulate（TSP）was measured using an Andersen Mark-II cascade impactor.The seasonal average mass concentration was highest in winter（234.73μg/m^3）and lowest in summer（91.71μg/m^3）.Water-soluble ions accounted for 59.49%,32.90%,48.62%and 37.08%of the aerosol mass concentration in winter,spring,summer,and fall,respectively,which indicated that ionic species were the primary constituents of the atmospheric aerosols.The four most abundant ions were NO3^-,SO4^2-,Ca^2＋ and NH4^＋.With the exception of Ca^2＋,the mass concentrations of water-soluble ions were in an intermediate range compared with the levels for other Chinese cities.Sulfate,nitrate,and ammonium were the dominant fine-particle species,which were bimodally distributed in spring,summer and fall;however,the size distribution became unimodal in winter,with a peak at 1.1–2.1μm.The Ca^2＋ peak occurred at approximately 4.7–5.8μm in all seasons.The cation to anion ratio was close to 1.4,which suggested that the aerosol particles were alkalescent in Hefei.The average NO3^-/SO4^2-mass ratio was 1.10 in Hefei,which indicated that mobile source emissions were predominant.Significant positive correlation coefficients between the concentrations of NH4^＋ and SO4^2-,NH4^＋ and NO3^-,SO4^2-and NO3^-,and Mg^2＋ and Ca^2＋ were also indicated,suggesting that aerosol particles may be present as（NH4）2SO4,NH4HSO4,and NH4NO3.

目的探讨钙黏蛋白相关蛋白(cadherin associated protein beta 1,CTNNB1)、肿瘤抑制因子P53(tumor suppressor factor P53,TP53)蛋白在儿童肝母细胞瘤(hepatoblastoma, HB)组织中的表达及与病理特征、预后的关系。方法选取2017-06/2020-06月作者医院收治的72例HB患儿为研究对象,手术留取癌组织标本及对应的癌旁组织。连续随访3年,记录患儿的预后生存情况,并计算总生存率(overall survival, OS)。采用免疫组织化学法检测CTNNB1、TP53蛋白在HB患儿癌组织及癌旁组织中的表达情况;采用χ~2检验分析CTNNB1、TP53蛋白表达与HB患儿临床病理特征的关系;采用多因素Cox回归分析探讨HB患儿预后的影响因素。结果 HB患儿癌组织中CTNNB1阳性表达率(76.39%)高于对照组(43.06%),TP53阴性表达率(70.83%)高于对照组(38.89%)(P均<0.05)。初诊甲胎蛋白浓度≥100μg/L、POST-TEXT分期Ⅲ~Ⅳ期、肿瘤直径≥3 cm、有肿瘤侵袭或转移HB患儿的CTNNB1阳性表达率、TP53阴性表达率高于初诊甲胎蛋白<100μg/L、POST-TEXT分期Ⅰ~Ⅱ期、肿瘤直径<3 cm、无肿瘤侵袭或转移HB患儿(P均<0.05)。初诊甲胎蛋白≥100μg/L、POST-TEXT分期Ⅲ~Ⅳ期、有肿瘤侵袭或转移、CTNNB1阳性表达、TP53阴性表达的HB患儿3年OS低于初诊甲胎蛋白<100μg/L、POST-TEXT分期Ⅰ~Ⅱ期、无肿瘤侵袭或转移、CTNNB1阴性表达、TP53阳性表达的HB患儿(P均<0.05)。多因素Cox回归分析结果显示,POST-TEXT分期Ⅲ~Ⅳ期(HR=2.077,95%CI:1.423~3.032)、有肿瘤侵袭或转移(HR=2.291,95%CI:1.536~3.417)、CTNNB1阳性表达(HR=2.757,95%CI:1.781~4.268)、TP53阴性表达(HR=2.477,95%CI:1.635~3.753)是HB患儿预后的独立危险因素(P<0.05)。结论 HB患儿癌组织中CTNNB1主要呈阳性表达,而TP53主要呈阴性表达,二者与初诊甲胎蛋白、POST-TEXT分期、肿瘤直径、有肿瘤侵袭或转移密切相关,有望作为评估HB患儿预后的生物学指标。

【背景】解淀粉欧文氏菌（Erwinia amylovora）是仁果类果树火疫病的病原体，对全球苹果和梨的生产构成严重威胁。随着抗生素耐药菌株的出现，火疫病的防治面临巨大的挑战。【目的】分离一种新的裂解解淀粉欧文氏菌的噬菌体，并分析该噬菌体裂解相关基因的功能，为火疫病的防治提供新的选择。【方法】以解淀粉欧文氏菌Ea102为宿主菌，采用双层平板法从流行火疫病的果园土壤中分离噬菌体。通过噬菌斑和透射电镜观察其形态。用PacBio测序技术测定噬菌体基因组，SPAdes组装序列，RAST注释基因组。通过大肠杆菌原核表达系统分析噬菌体Kuerle的裂解机制。【结果】分离纯化出一株裂解解淀粉欧文氏菌的噬菌体，命名为Kuerle。Kuerle由二十面体衣壳的头部和短尾组成，潜伏期约为50 min，裂解量约为240 pfu/cell。噬菌体基因组全长75 599 bp,GC含量48.0%，末端有393 bp的重复序列，未发现与溶原调控相关的基因。共预测到85个开放阅读框（open reading frame,ORF），其中33个已知功能蛋白中包含一个由3 550 aa组成的巨大病毒颗粒相关的RNA聚合酶（virion-associated RNA polymerase,vRNAP），该vRNAP是Schitoviridae科噬菌体的主要特征之一。病毒粒子和基因结构表明噬菌体Kuerle属于有尾噬菌体类的Schitoviridae科病毒。聚集在DNA晚期表达区的3个裂解相关基因holin、endolysin和spanin组成了“裂解盒”。在大肠杆菌中Kuerle-holin的表达会抑制细胞的生长，而Kuerle-endolysin的表达可裂解细胞。二者共表达时Kuerle-holin会加速Kuerle-endolysin引起的细胞裂解。用叠氮化钠抑制细菌的一般分泌系统（Sec）或Kuerle-endolysin N端信号序列的缺失均会导致endolysin裂解功能丧失。上述结果表明噬菌体Kuerle具有pinholin-SAR endolysin裂解系统。Kuerle-holin(pinholin)使细胞质膜去极化以激活分泌的Kuerle-endolysin(SAR endolysin)降解肽聚糖层。【结论】Kuerle是一株烈性噬菌体并且Kuerle-endolysin抑菌效果显著，为后续火疫病生防试剂的制备提供了理论依据和研究材料。
