due to the low working efficiency of apple harvesting robots,there is still a long way to go for *** machine performance and extended operating time are the two research aspects for improving efficiencies of harvesting robots,this study focused on the extended operating time and proposed a round-the-clock operation *** to the influences of light,temperature,humidity,etc.,the working environment at night is relatively complex,and thus restricts the operating efficiency of the apple harvesting *** different artificial light sources(incandescent lamp,fluorescent lamp,and LED lights)were selected for auxiliary light according to certain rules so that the apple night vision images could be *** addition,by color analysis,night and natural light images were compared to find out the color characteristics of the night vision images,and intuitive visual and difference image methods were used to analyze the noise *** results showed that the incandescent lamp is the best artificial auxiliary light for apple harvesting robots working at night,and the type of noise contained in apple night vision images is Gaussian noise mixed with some salt and pepper *** preprocessing method can provide a theoretical and technical reference for subsequent image processing.

MHC polymorphism and disease-resistance to Edwardsiella tarda in six turbot(Scophthalmus maximus) families

This study examined genetic variation in the major histocompatibility complex(MHC) Class II B gene in turbot(Scophthalmus maximus) by virulent bacterial pathogen *** hundred fry from each of six families were infected with Edwardsiella tarda by intraperitoneal *** mortality ranged from 28.0% to 83.3%.Complete exon 2 and intron 1 sequences of MHC Class II B genes were amplified from five survivor and five non-survivor individuals per family using the clone-sequence ***-seven sequences from 60 individuals revealed 37 different alleles,25 of which were unique to this *** 25 unique alleles belonged to 16 major allele *** alleles were used to examine the association between alleles and resistance/susceptibility to *** alleles were present in an individual,suggesting a minimum of three loci or copies of the turbot MHC Class II B *** rate of non-synonymous substitution(d N) was 2.30 and 1.58 times higher than synonymous substitution(d S) in the peptide-binding regions(PBR) and non-PBR in whole families,respectively,which suggested balancing selection on exon 2 of the MHC Class II B gene in *** allele,Scma-DBB1*02,was significantly more prevalent in survivor stock than in non-survivor stock(P=0.001).Therefore,this allele might be associated with resistance to bacteria.A second allele,Scma-DBB1*10,was significantly more prevalent in non-survivor stock(P=0.021),and is likely associated with susceptibility to bacteria.

Comparative analysis of husk microstructure, fruit quality and concentrations of bioactive compounds of different pomegranate cultivars during low temperature storage

The deterioration degree of pomegranate husk,aroma components and antioxidant contents of pomegranate juice have important effects on postharvest storage quality and vary among *** the present study,‘Tunisia soft seed’,‘Jingpitian’and‘Lishanhong’cultivars from Shaanxi,China was used as raw *** were stored at 4±0.5℃ and 85-90%relative humidity for 90 d which aimed to elucidate storability *** husk changes and storage quality were *** results showed that‘Tunisia soft seed’maintained a good microstructure in husk and possessed a high value of soluble solids/titratable acid ratio and high contents of antioxidants(total phenols,total flavonoids,total anthocyanins,vitamin C)but suffered from high moisture and volatile aroma compounds losses.‘Jingpitian’exhibited lower moisture and aroma compounds losses,but showed significant husk browning,micro-cracks widening,lenticels blocking and wax layer disintegrating.‘Lishanhong’showed similar fruit quality characteristics to‘jingpitian’.These findings will contribute to elucidating the appearance change,microstructure and fruit quality differences among cultivars in postharvest pomegranates during cold storage and providing help for enterprises to discriminate and select varieties suitable for fresh sales or storage.

After the sequencing of citrus genomes,gene function annotation is becoming a new *** coexpression analysis can be employed for function annotation using publicly available microarray data *** this study,230 sweet orange(Citrus sinensis)microarrays were used to construct seven coexpression networks,including one condition-independent and six condition-dependent(Citrus canker,Huanglongbing,leaves,flavedo,albedo,and flesh)*** total,these networks contain 37633 edges among 6256 nodes(genes),which accounts for 52.11%measurable genes of the citrus ***,these networks were partitioned into functional modules using the Markov Cluster *** enriched Gene Ontology biological process terms and KEGG pathway terms were detected for 343 and 60 modules,***,independent verification of these networks was performed using another expression data of 371 *** study provides new targets for further functional analyses in citrus.

Prognostic value of lateral lymph node metastasis for advanced low rectal cancer

AIM： To evaluate the risk factors for lateral lymph node metastasis in patients with advanced low rectal cancer, in order to make the effective selection of patients who could benefit from lateral lymph node dissection, as well as the relationship of lateral lymph node metastasis with local recurrence and survival of patients with advanced low rectal cancer. METHODS： A total of 96 consecutive patients who underwent curative surgery with lateral pelvic lymphadenectomy for advanced lower rectal cancer were retrospectively analyzed. The relation of lateral lymph node metastasis with clinicopathologic characteristics, local recurrence and survival of patients was identified. RESULTS： Lateral lymph node metastasis was observed in 14.6% （14/96） of patients with advanced low rectal cancer. Lateral lymph node metastasis was detected in 10 （25.0%） of 40 patients with tumor diameter ≥ 5 cm and in 4 （7.1%） of 56 patients with tumor diameter 〈 5 cm. The difference between the significant （X^2 = 5.973, P = two groups was statistically 0.015）. Lateral lymph node metastasis was more frequent in patients with 4/4 diameter of tumor infiltration （7 of 10 cases, 70.0%）, compared with patients with 3/4, 2/4 and 1/4 diameter of tumor infiltration （3 of 25 cases, 12.0%; 3 of 45 cases, 6.7%; 1 of 16 cases, 6.3%）（X^2 = 27.944, P = 0.0001）. The lateral lymph node metastasis rate was 30.0% （9 of 30 cases）, 9.1% （4 of 44 cases） and 4.5% （1 of 22cases） for poorly, moderately and well-differentiated carcinoma, respectively. The difference between the three groups was statistically significant （X^2 = 8.569, P = 0.014）. Local recurrence was 18.8% （18 of 96 cases）, 64.3% （9 of 14 cases）, and 11.0% （9 of 82 cases） in patients with advanced low rectal cancer, in those with and without lateral lymph node metastasis, respectively. Global Distribution of Thermosteric Contribution to Sea Level Rising Trend

The sea level derived from TOPEX/Poseidon （T/P） altimetry data shows prominent long term trend and inter-annual variability. The global mean sea level rising rate during 1993-2003 was 2.9mm a^-1. The T/P sea level trend maps the geographical variability. In the Northern Hemisphere （15°-64°N）, the sea level rise is very fast at the mid-latitude （20°-40°N） but much slower at the high-latitude, for example, only 0.5 mm a^-1 in the latitude band 40°-50°N. In the Southern Hemisphere, the sea level shows high rising rate both in mid-latitude and high-latitude areas, for example, 5.1 mm a^-1 in the band 40°- 50°S. The global thermosteric sea level （TSL） derived from Ishii temperature data was rising during 1993-2003 at a rate of 1.2 mm a^-1 and accounted for more than 40% of the global T/P sea level rise. The contributions of the TSL distribution are not spatially uniform; for instance, the percentage is 67% for the Northern Hemisphere and only 29% for the Southern Hemisphere （15°-64°S） and the maximum thermosteric contribution appears in the Pacific Ocean, which contributes more than 60% of the global TSL. The sea level change trend in tropical ocean is mainly caused by the thermosteric effect, which is different from the case of seasonal variability in this area. The TSL variability dominates the T/P sea level rise in the North Atlantic, but it is small in other areas, and shows negative trend at the high-latitude area （40°-60°N, and 50°-60°S）. The global TSL during 1945-2003 showed obvious rising trend with the rate of about 0.3 mm a-l and striking inter-annual and decadal variability with period of 20 years. In the past 60 years, the Atlantic TSL was rising continuously and remarkably, contributing 38% to the global TSL rising. The TSL in the Pacific and Indian Ocean rose with significant in- ter-annual and decadal variability. Streptomyces griseus trypsin(SGT)is a bacteria-sourced trypsin that could be potentially applied to industrial insulin ***,SGT produced by microbial hosts displayed low catalytic efficiency and undesired preference to lysine *** this study,by engineering theαsignal peptide in Pichia pastoris,we increased the SGT amidase activity to 67.91 U mL^(−1)in shake flask ***,we engineered SGT by evolution-guided mutagenesis and obtained three variants A45S,V177I and E180M with increased catalytic *** this basis,we performed iterative combinatorial mutagenesis and constructed a mutant A45S/V177I/E180M which the amidase activity reached 98 U mL^(−1)in shake flasks and 2506 U mL^(−1)in 3-L fed-batch ***,single mutation T190 to S190 increased the substrate catalytic preference of R to K and the R/K value was improved to 7.5,which was 2 times better than the animal-sourced trypsin.

Targeting cancer stem cells in drug discovery: Current state and future perspectives

In recent decades,cancer stem cells(CSCs)have been increasingly identified in many ***-related signaling pathways and their functions provide new strategies for treating *** aberrant activation of related signaling pathways(e.g.,Wnt,Notch,and Hedgehog pathways)has been linked to multiple types of malignant tumors,which makes these pathways attractive targets for cancer *** display many characteristic features,such as self-renewal,differentiation,high tumorigenicity,and drug ***,there is an urgent need to develop new therapeutic strategies to target these pathways to control stem cell replication,survival,and *** crosstalk occurs among different signaling pathways and potentially leads to compensatory ***,multitarget inhibitors will be one of the main methods to overcome the drug resistance of *** small molecule inhibitors of components of signaling pathways in CSCs have entered clinical trials,and some inhibitors,such as vismodegib,sonidegib,and glasdegib,have been *** cells are susceptible to sonidegib and vismodegib resistance due to mutations in the Smo *** signal transducers and activators of transcription 3(STAT3)inhibitor BBI608 is being evaluated in a phase III trial for a variety of *** derivatives of BBI608 are the main focus of STAT3 inhibitor development,which is another strategy for CSC *** addition to the potential pharmacological inhibitors targeting CSCrelated signaling pathways,other methods of targeting CSCs are available,such as nano-drug delivery systems,mitochondrion targeting,autophagy,hyperthermia,immunotherapy,and CSC microenvironment *** addition,we summarize the latest advances in the clinical development of agents targeting CSC-related signaling pathways and other methods of targeting CSCs.
