One of the most pressing issues surrounding transgenic oilseed rape cultivation is the potential impact of GM genes entering neighboring non-GM oilseed rape plants, related weeds or closely related wild relatives. Following the principle of a case-by-case for transgenic crop safety assessment, we designed experiments to study pollen-mediated gene flow from herbicide-resistant transgenic oilseed rape to Chinese commercial varieties. The pollen dispersal was detected as the progeny of recipient plants that were identified as glufosinate resistant. The results indicated that pollen dispersal occurred up to 2000 m and its rate sharply decreased as the distance from the pollen source increased. However, the rate of long-distance pollen dispersal from 33 to 2000 m was less than 0.015% and did not present a gradual decrease, indicating the randomization of dispersal and pollination. Most pollen dispersed within a 4.5 m area around the transgenic pollen source plot, with a maximum of 1.19% at the sampling site of 1.4 m. Wind direction significantly affected the direction and the distance of pollen dispersal, resulting in the more and farther dispersal in southwest direction. However, the number of potentially pollinating bees was not correlated with direction and distance from the pollen source plot, suggesting that bee density and distribution would not contribute to the differences in pollen dispersal among different directions. This paper is the first report on research in a large-scale experiment of pollen-mediated gene flow un-der the China’s environmental conditions and provided scientific evidence for the effective commer-cialization of transgenic oilseed rape and its safe management. Our results also provided important data on pollen spatial distribution pattern.

Oilseed rape(Brassica napus) is an allotetraploid with two subgenomes descended from a common ancestor. Accordingly, its genome contains syntenic regions with many duplicate genes, some of which may have retained their original functions, whereas others may have diverged. Here, we mapped quantitative trait loci(QTL) for stem rot resistance(SRR), a disease caused by the fungus Sclerotinia sclerotiorum, and flowering time(FT) in a recombinant inbred line population. The population was genotyped using *** 60 K single nucleotide polymorphism arrays and phenotyped in six(FT) and nine(SSR) experimental conditions or environments. In total, we detected 30 SRR QTL and 22 FT QTL and show that some of the major QTL associated with these two traits were co-localized,suggesting a genetic linkage between them. Two SRR QTL on chromosome A2 and two on chromosome C2 were shown to be syntenic, suggesting the functional conservation of these regions. We used the syntenic properties of the genomic regions to exclude genes for selection candidates responsible for QTL-associated traits. For example, 152 of the 185 genes could be excluded from a syntenic A2–C2 region. These findings will help to elucidate polyploid genomics in future studies, in addition to providing useful information for B. napus breeding programs.

Agrobacterium-mediated gene transformation in Brassica crops is a time-con-suming and labor-cost method (tissue culture and plant regeneration) in a genotype-depen-dent way. Here we described a new method 'in situ pistil delivery (ISPD)' method with no tissue culture and adaptive to three Brassica species. Briefly this method can be stated as placement of a liquid drop containing binary vector DNA onto the cut surface of emasculated and artificial pollinated Brassica stigma. Transformation rate (Basta-resistant seedlings per 100 pistils) of different species ranged from 0.4% to 16.3%. Factors affecting transforma-tion success included timing and buffer type. PCR and Southern blot confirmed transgenic events. Reciprocal cross showed reporter gene steadily inherited in nuclear transformed pattern. GUS histochemical assay of transgenic immature seeds showed stained embroys and endosperm in the same seed, indicating DNA delivered through style and integrated into Brassica seed/genome after double fertilization.

目前,对于超高层结构进行动力弹塑性分析时,大多采用刚性地基假定,并忽略土-结构相互作用的影响。为了研究土-结构相互作用对超高层结构的影响,建立精细化的土体-地下室-上部结构三维非线性有限元模型,并对一栋高度366 m超高层塔楼进行了罕遇地震动力弹塑性分析。首先根据工程场地条件进行了一维等效线性化分析,以获取土层的地震反应。然后,利用波动法解决了在三维土层地震波动输入的问题,并验证了所采用方法的准确性。计算结果表明:与刚性地基假定相比,考虑土-结构相互作用会增加塔楼的自振周期;在预估的罕遇地震作用下,考虑土-结构相互作用后,塔楼的整体地震反应减小;塔楼层间位移角在低区增大,在高区减小,高区最大层间位移角减小,呈现变形向底部转移的趋势;塔楼高区墙体损伤减轻,结构整体刚度退化程度减小,同时基底剪力和倾覆弯矩降低;塔楼巨柱个别位置的内力增大,忽略土-结构相互作用的影响可能会导致设计结果偏于不安全;所提出的方法能够全面考虑场地土的影响,较为真实地模拟超高层结构的地震响应。

目的评估连续光电容积脉搏波算法应用于可穿戴设备时,检出心房颤动(房颤)和评估房颤负荷的准确性。方法本研究是一项自身对照的前瞻性队列研究,连续纳入2022年9至11月于首都医科大学附属北京安贞医院心内科住院治疗的254例房颤患者。所有受试者同时佩戴2种设备:(1)心电手表,用于采集光电容积脉搏波(W-PPG)和心电图(W-ECG);(2)心电贴,用于采集心电图(P-ECG)。分别以30 s数据片段和受试者个体为单位,将心电手表得出的测量结果与心电图专家判读心电贴结果相对比,采用受试者工作特征曲线评估其敏感度和特异度。按照房颤负荷比例均分为4个分段,计算由房颤分析软件得出的房颤负荷与心电图专家判别心电贴结果得出的房颤负荷差值。结果254例受试者的年龄为(63.04±11.04)岁,99例(38.98%)为女性,持续性房颤患者97例(38.19%)。以心电图专家判读心电贴的结果为标准,以片段为单位时,P-ECG算法的敏感度为94.86%(95%CI:94.81%~94.91%),特异度为99.30%(95%CI:99.28%~99.31%),W-PPG算法的敏感度为96.07%(95%CI:95.97%~96.18%),特异度为98.62%(95%CI:98.59%~98.65%);以个体为单位时,P-ECG算法的敏感度为92.55%(95%CI:87.57%~95.71%),特异度为96.39%(95%CI:93.45%~98.09%),W-PPG算法的敏感度为93.71%(95%CI:88.75%~96.67%),特异度为89.62%(95%CI:85.61%~92.65%);以单次采集W-ECG记录为单位时,W-ECG算法的敏感度为92.04%(95%CI:88.14%~94.78%),特异度为96.19%(95%CI:94.35%~97.47%)。房颤负荷评估方面,各负荷分布区间内W-PPG分析结果与心电图专家判读心电贴结果相差均小于2%。结论连续PPG算法应用于可穿戴设备检测房颤是一种可行策略。以心电图专家判读心电贴的结果为标准,通过智能手表连续监测PPG对于房颤诊断具有较高的准确性,同时也可有效地完成房颤负荷评估。
