Ubiquitin-specific protease 21 promotes tumorigenicity and stemness of colorectal cancer by deubiquitinating and stabilizing ZEB1

BACKGROUND Colorectal cancer(CRC)is one very usual tumor together with higher death rate.Ubiquitin-specific protease 21(USP21)has been confirmed to take part into the regulation of CRC progression through serving as a facilitator.Interestingly,the promotive function of USP21 has also discovered in the progression of CRC.ZEB1 has illustrated to be modulated by USP7,USP22 and USP51 in cancers.However,the regulatory functions of USP21 on ZEB1 in CRC progression need more invest-igations.AIM To investigate the relationship between USP21 and ZEB1 in CRC progression.METHODS The mRNA and protein expressions were assessed through RT-qPCR,western blot and IHC assay.The interaction between USP21 and ZEB1 was evaluated through Co-IP and GST pull down assays.The cell proliferation was detected through colony formation assay.The cell migration and invasion abilities were determined through Transwell assay.The stemness was tested through sphere formation assay.The tumor growth was evaluated through in vivo mice assay.RESULTS In this work,USP21 and ZEB1 exhibited higher expression in CRC,and resulted into poor prognosis.Moreover,the interaction between USP21 and ZEB1 was further investigated.It was demonstrated that USP21 contributed to the stability of ZEB1 through modulating ubiquitination level.In addition,USP21 streng-thened cell proliferation,migration and stemness through regulating ZEB1.At last,through in vivo assays,it was illustrated that USP21/ZEB1 axis aggravated tumor growth.CONCLUSION For the first time,these above findings manifested that USP21 promoted tumorigenicity and stemness of CRC by deubiquitinating and stabilizing ZEB1.This discovery suggested that USP21/ZEB1 axis may provide novel sights for the treatment of CRC.

Graphitic carbon nitride nanosheets mitigate cadmium toxicity in Glycine max L.by promoting cadmium retention in root and improving photosynthetic performance

Cadmium(Cd)pollution poses a serious threat to plant growth and yield.Nanomaterials have shown great application potential for alleviation of Cd toxicity to plants.In this study,we applied graphitic carbon nitride nanosheets(g-C_(3)N_(4)NSs)for alleviation of Cd-toxicity to soybean(Glycine max L.).The g-C_(3)N_(4)NSs supplementation significantly improved plant growth and reduced oxidative damage in the Cd-toxicated soybean seedlings through hydroponic culture.Particularly,the g-C_(3)N_(4)NSs dynamically regulated the root cell wall(RCW)components by increasing pectin content and modifying its demethylation via enhancing pectin methylesterase(PME)activity,therefore greatly enhanced stronger RCW-Cd retention(up to 82.8%)and reduced Cd migration to the shoot.Additionally,the g-C_(3)N_(4)NSs reversed the Cd-induced chlorosis,increased photosynthetic efficiency because of enhancement in F_v/F_mration,Y(Ⅱ)and sugars content.These results provide new insights into the alleviation of Cd toxicity to plants by g-C_(3)N_(4)NSs,and shed light on the application of low-cost and environmental-friendly carbon-based NMs for alleviating heavy metal toxicity to plants.

A DNA tetrahedron-based ferroptosis-suppressing nanoparticle: superior delivery of curcumin and alleviation of diabetic osteoporosis

Diabetic osteoporosis(DOP)is a significant complication that poses continuous threat to the bone health of patients with diabetes;however,currently,there are no effective treatment strategies.In patients with diabetes,the increased levels of ferroptosis affect the osteogenic commitment and differentiation of bone mesenchymal stem cells(BMSCs),leading to significant skeletal changes.To address this issue,we aimed to target ferroptosis and propose a novel therapeutic approach for the treatment of DOP.We synthesized ferroptosis-suppressing nanoparticles,which could deliver curcumin,a natural compound,to the bone marrow using tetrahedral framework nucleic acid(tFNA).This delivery system demonstrated excellent curcumin bioavailability and stability,as well as synergistic properties with tFNA.Both in vitro and in vivo experiments revealed that nanoparticles could enhance mitochondrial function by activating the nuclear factor E2-related factor 2(NRF2)/glutathione peroxidase 4(GPX4)pathway,inhibiting ferroptosis,promoting the osteogenic differentiation of BMSCs in the diabetic microenvironment,reducing trabecular loss,and increasing bone formation.These findings suggest that curcumin-containing DNA tetrahedron-based ferroptosissuppressing nanoparticles have a promising potential for the treatment of DOP and other ferroptosis-related diseases.

Solving Multi-Objective Vehicle Routing Problems with Time Windows: A Decomposition-Based Multiform Optimization Approach

In solving multi-objective vehicle routing problems with time windows (MOVRPTW),most existing algorithms focus on the optimization of a single problem formulation. However,little effort has been devoted to exploiting valuable knowledge from the alternate formulations of MOVRPTW for better optimization performance. Aiming at this insufficiency,this study proposes a decomposition-based multi-objective multiform evolutionary algorithm (MMFEA/D),which performs the evolutionary search on multiple alternate formulations of MOVRPTW simultaneously to complement each other. In particular,the main characteristics of MMFEA/D are three folds. First,a multiform construction (MFC) strategy is adopted to construct multiple alternate formulations,each of which is formulated by grouping several adjacent subproblems based on the decomposition of MOVRPTW. Second,a transfer reproduction (TFR) mechanism is designed to generate offspring for each formulation via transferring promising solutions from other formulations,making that the useful traits captured from different formulations can be shared and leveraged to guide the evolutionary search. Third,an adaptive local search (ALS) strategy is developed to invest search effort on different alternate formulations as per their usefulness for MOVRPTW,thus facilitating the efficient allocation of computational resources. Experimental studies have demonstrated the superior performance of MMFEA/D on the classical Solomon instances and the real-world instances.

Application of neoadjuvant chemoradiotherapy and neoadjuvant chemotherapy in curative surgery for esophageal cancer:A metaanalysis

BACKGROUND The effectiveness of neoadjuvant therapy in esophageal cancer(EC)treatment is still a subject of debate.AIM To compare the clinical efficacy and toxic side effects between neoadjuvant chemoradiotherapy(nCRT)and neoadjuvant chemotherapy(nCT)for locally advanced EC(LAEC).METHODS A comprehensive search was conducted using multiple databases,including PubMed,EMBASE,MEDlinE,Science Direct,The Cochrane Library,China National Knowledge Infrastructure,Wanfang Database,Chinese Science and Technology Journal Database,and Chinese Biomedical Literature Database Article.Studies up to December 2022 comparing nCRT and nCT in patients with EC were selected.RESULTS The analysis revealed significant differences between nCRT and nCT in terms of disease-free survival.The results indicated that nCRT provided better outcomes in terms of the 3-year overall survival rate(OSR)[odds ratio(OR)=0.95],complete response rate(OR=3.15),and R0 clearance rate(CR)(OR=2.25).However,nCT demonstrated a better 5-year OSR(OR=1.02)than nCRT.Moreover,when compared to nCRT,nCT showed reduced risks of cardiac complications(OR=1.15)and pulmonary complications(OR=1.30).CONCLUSION Overall,both nCRT and nCT were effective in terms of survival outcomes for LAEC.However,nCT exhibited better performance in terms of postoperative complications.

Assembly of highly efficient aqueous light-harvesting system from sequence-defined peptoids for cytosolic microRNA detection

Precisely controlled spatial distributions of artificial light-harvesting systems in aqueous media are of significant importance for mimicking natural light-harvesting systems;however,they are often restrained by the solubility and the aggregation-caused quenching effect of the hydrophobic chromophores.Herein,we report one highly efficient artificial light-harvesting system based on peptoid nanotubes that mimic the hierarchical cylindrical structure of natural systems.The high crystallinity of these nanotubes enabled the organization of arrays of donor chromophores with precisely controlled spatial distributions,favoring an efficient Förster resonance energy transfer(FRET)process in aqueous media.This FRET system exhibits an extremely high efficiency of 98.6%with a fluorescence quantum yield of 40%and an antenna effect of 29.9.We further demonstrated the use of this artificial light-harvesting system for quantifying miR-210 within cancer cells.The fluorescence intensity ratio of donor to acceptor is linearly related to the concentration of intercellular miR-210 in the range of 3.3–156 copies/cell.Such high sensitivity in intracellular detection of miR-210 using this artificial light-harvesting system offers a great opportunity and pathways for biological imaging and detection,and for the further creation of microRNA(miRNA)toolbox for quantitative epigenetics and personalized medicine.

Utilization of hydralazine as a reactive matrix for enhanced detection and on-MALDI-target derivatization of saccharides

Saccharides are a sort of ubiquitous and vital molecules within the whole life.However,the application of saccharides analysis with matrix-assisted laser desorption/ionization mass spectrometry(MALDI-MS)is restricted by their low ionization efficiency and the instability of the sialic acid fraction.Derivatization strategy based on nonreductive amination provides a good solution,however,this is often time consuming and may result in sample loss due to removal of excessive derivatization reagents.Herein,hydralazine(HZN)was utilized as a reactive matrix for labeling reducing saccharides directly on MALDI target which eliminated tedious sample preparation and avoided sample loss.After optimization,effective and reproducible on-MALDI-target derivatization of neutral and acidic saccharides was achieved in both positive and negative modes.Compared with 2,5-dihydroxybenzoic acid(DHB)and 9-aminoacridine(9-AA),HZN improved the detection sensitivity of reducing saccharides and provided more abundant fragment ions in MS/MS analysis.Moreover,26 kinds of neutral glycans and 5 kinds of sialic glycans were identified from ovalbumin(OVA)and bovine fetuin,respectively.Combined with the statistical models,this strategy could be used to distinguish and predict samples of 6 brands of beer,and discriminate 2 kinds of beer fermentation modes.In addition,HZN was applied for quantitative analysis of glucose in urine samples,and the obtained urine glucose concentrations of diabetic patients were consistent with the clinical test results,showing the potential of qualitative and quantitative analysis of reducing saccharides in complex samples.

Theoretical understanding of correlation between magnetic phase transition and the superconducting dome in high-Tc cuprates

Many issues concerning the origin of high-temperature superconductivity(HTS)are still under debate.For example,how the magnetic order varies with doping and its relationship with the superconducting temperature(Tc);and why Tcalways peaks near the quantum critical point.In this paper,taking hole-doped La_(2)CuO_(4)as a classical example,we employ the first-principles band structure and total energy calculations with Monte Carlo simulations to explore how the symmetry-breaking magnetic ground state evolves with hole doping and the origin of a dome-shaped superconductivity region in the phase diagram.We demonstrate that the local antiferromagnetic order and doping play key roles in determining the electron-phonon coupling,thus Tc.Initially,the La_(2)CuO_(4)possesses a checkerboard local antiferromagnetic ground state.As the hole doping increases,Tcincreases with the enhanced electron-phonon coupling strength.But as the doping increases further,the strength of the antiferromagnetic interaction weakens and spin fluctuation increases.At the critical doping level,a magnetic phase transition occurs that reduces the local antiferromagnetism-assisted electron-phonon coupling,thus diminishing the Tc.The superconductivity disappears in the heavily overdoped region when the ferromagnetic order dominates.These observations could account for why cuprates have a dome-shaped superconductivity region in the phase diagram.Our study,thus,contributes to a fundamental understanding of the correlation between doping,local magnetic order,and superconductivity of HTS.

Analysis on characteristics of extreme precipitation indices and atmospheric circulation in Northern Shanxi

This article utilizes daily precipitation data from 28 national meteorological stations in northern Shanxi Province spanning from 1972 to 2020, and the US NCEP/NCAR monthly average reanalysis and ERA5 monthly average reanalysis data. The study employs techniques such as empirical orthogonal function(EOF) decomposition, MannKendall mutation and other methods to investigate the spatiotemporal distribution of extreme precipitation index in northern Shanxi and their correlation with atmospheric circulation. The research results show that: the absolute index, relative index, intensity index and sustained dry period index(CDD) in the continuous index appear from southwest to northeast. The spatial distribution characteristics of the central region decrease, while the continuous wet period(CWD) decreases from the central to the east and west. The three indices Rx1day, Rx5day, and CWD mutated in 1978, 1975, and 1983 respectively, and other extreme precipitation indices all appeared in a sudden change from a low-value period to a high-value period occurred around 2010. In the high-value years of the summer extreme precipitation index, there is a significant negative anomaly in the height field in the mid-high latitude regions of Eurasia. Northern Shanxi is controlled by a broad low-pressure trough in the Lake Baikal area.Water vapor transported via the east, west, and south routes converges in the northern Shanxi region and encounters cold air from the north. There is a strong upward motion anomaly at 500 hPa in the troposphere, and the dynamic conditions of upper-level divergence and lower-level convergence lead to more summer extreme precipitation in the northern Shanxi region. Conversely, in the low-value years of the summer extreme precipitation index, northern Shanxi is affected by a strong high-pressure ridge north of Lake Baikal. There is a downward motion anomaly at 500 hPa, and the northern Shanxi region lacks water vapor. The cold and warm air cannot converge, and both the water vapor conditions and dynamic cond
