Breast cancer cells may disseminate early, before tumor diagnosis. disseminated tumor cells, or dTCs, reside in the bone marrow, and may persist for years or even decades. Some of these cells may be re-activated to resume aggressive growth, and eventually become overt bone metastases. Recent studies have begun to shed light on this complicated process and revealed multiple steps and intermediate states of colonizing dTCs. However, how cancer-host interactions evolve during this process needs to be further understood. Most of our current knowledge of the bone microenvironment is obtained through studies looking for the hematopoietic stem cell(HSC) niche. Although this long-standing question has not yet been resolved, our search for the HSC niche has resulted in a detailed map of various cell types in the bone marrow. Furthermore, various techniques used to find the HSC niche may also be adapted for finding the cancer cell niche. In this article, we will review the recent progress in both the dTC and HSC areas with a focus on their potential microenvironment niches. We will also discuss how to apply what we have learned from HSC studies to map dTCs in the bone context. We hope to stimulate thoughts and ideas to further elucidate the bone colonization process, and develop potential therapeutic interventions.

Provenance of Cored Sediments from Active Margin off Southwestern Taiwan deduced from Geochemical Constraints

The cored sediments sampled by R/V Marion dufrense in the active margin off southwestern Taiwan on the east side of the Manila Trench are mostly muds consisting mainly of quartz, feldspar, illite, chlorite ＋ kaolinite with a trace amount of montmorillonite. The cored sediments have higher Fe, Ti, Cr, Cs, Li, Ni, and V but lower ca, Na, Mn, Hf, Nb, Sr, Ta, U and Zr contents when compared with the upper continental crust（UCC, Taylor and McLennan, 1985）. The Eu/Eu＊ ratios of the sediments averaging 0.69 indicate that the recycling effect on the Eu anomaly may be minor and the contribution of mafic source rock was unimportant, which can also be depicted in the La-Th-Sc plot（Cullers, 1994） showing a ＂mixed origin＂ close to the felsic end member. The chemistry of the cored sediment can be explained by a mixing model designed by Ho and Chen（1996） with four end members i.e., greywacke, shale, quartzite and limestone, which are widely distributed in the Cenozoic formations of southern Taiwan with greywacke as the dominant contributor followed by shale.

To the Editor： Hepatocellular carcinoma （HCC） remains one of the commonest cancers worldwide especially in hepatitis B endemic regions. Its aggressive behavior is characterised by the natural history of increasing size, a tendency for vascular invasion into the hepatic veins and portal veins. Further growth into the inferior vena cava （IVC） and right atrium （ra） is an infrequent finding but signifies a pre-terminal event with a dismal prognosis.

Focal adhesions play an important role in cell spreading,migration,and overall mechanical *** relationship of cell structural and mechanical properties was investigated in the context of focal adhesion *** atomic force microscopy(AFM) and laser scanning confocal microscopy(LSCM) was utilized to measure single cell mechanics,in correlation with cellular morphology and membrane structures at a nanometer *** stages of focal adhesion were verified via confocal fluorescent studies,which confirmed three representative F-actin assemblies,actin dot,filaments network,and long and aligned fibrous bundles at ***-deformation profiles of living cells were measured at the single cell level,and displayed as a function of height deformation,relative height deformation and relative volume *** focal adhesion progresses,single cell compression profiles indicate that both membrane and cytoskeleton stiffen,while spreading increases especially from focal complex to focal ***,AFM imaging reveals morphological geometries of spherical cap,spreading with polygon boundaries,and elongated or polarized *** features are dominated by protrusions of 41-207 nm tall,short rods with 1-6 μm in length and 10.2-80.0 nm in height,and long fibrous features of 31-246 nm tall,*** protrusion is attributed to local membrane folding,and the rod and fibrous features are consistent with bilayer decorating over the F-actin *** collectively,the reassembly of F-actin during focal adhesion formation is most likely responsible for the changes in cellular mechanics,spreading morphology,and membrane structural features.

As a convenient passenger transit facility between floors with different heights, escalators have been extensively used in shopping malls, metro stations, airport terminals, etc. Compared with other vertical transit facilities including stairs and elevators, escalators usually have large transit capacity. It is expected to reduce pedestrian traveling time and thus improve the quality of pedestrian’s experiences especially in jamming conditions. However, it is noticed that pedestrians may present different movement patterns, e.g., queuing on each step of the escalator, walking on the left-side and meanwhile standing on the right-side of the escalator. These different patterns affect the actual escalator traffic volume and finally the passenger spatiotemporal distribution in different built environments. Thus, in the present study, a microscopic cellular automaton(ca) simulation model considering pedestrian movement behavior on escalators is built. Simulations are performed considering different pedestrian movement speeds, queuing modes, and segregation on escalators with different escalator *** actual escalator capacities under different pedestrian movement patterns are investigated. It is found that walking on escalators will not always benefit escalator transit volume improvement, especially in jamming conditions.

The family Alaudidae,larks,comprises 93-100 species(depending on taxonomy)that are widely distributed across Africa and Eurasia,with single species extending their ranges to North and northernmost South America and Australia.A decade-old molecular phylogeny,comprising~80%of the species,revealed multiple cases of parallel evolution and large variation in rates of morphological evolution,which had misled taxonomists into creating many non-monophyletic ***,we reconstruct the phylogeny of the larks,using a dataset covering one mitochondrial and 16 nuclear loci and comprising all except one of the currently recognised species as well as several recently proposed new species(in total 133 taxa;not all loci available for all species).We provide additional support using genome-wide markers to infer a genus-level phylogeny based on near-complete generic sampling(in total 51 samples of 44 taxa across 40 species).our results confirm the previous findings of rampant morphological convergence and divergence,and reveal new cases of paraphyletic *** propose a new subfamily classification,and also that the genus Mira.ra.is divided into four genera to produce a more balanced generic classification of the *** study supports recently proposed species splits as well as some recent lumps,while also questioning some of the *** comprehensive phylogeny will form an important basis for future studies,such as comparative studies of lark natural history,ecology,evolution and conservation.

Prognostic and predictive biomarkers in metastatic colorectal cancer anti-EGFR therapy

AIM: To reviewing genetic and epigenetic make-up of metastatic colorectal cancers (mCRCs) addicted to epidermal growth factor receptor (EGFR) ***: The present study summarizes the potential value of prognostic and predictive biomarkers in selecting mCRC patients treated with anti-EGFR therapy. A meta-analysis was performed using a systematic search of PubMed, Medline and Web of Science to identify eligible papers until March 21st, 2016 using these following terms: ‘‘colorectal cancer’’, “predictive biomarkers’’, “anti-EGFR therapy”, “KraS”, “NraS’’, “PIK3ca”, “TP53”, “PTEN”, ‘‘EGFR”, “MET”, “HER2”, “epiregulin”, “amphiregulin”, “prognostic biomarkers”, “BraF”, “miRNA” and “antibody-dependent cell-mediated cytotoxicity (AdCC) activity”. Two investigators independently evaluated and extracted data from each identified studies based on selected criteria of inclusion and ***: The introduction of agents targeting EGFR such as cetuximab and panitumumab increased overall survival of mCRCs. Nevertheless, it has firstly became evident that response rates to cetuximab regimens in unselected patient populations were typically lower than 30%. Clinical data confirmed the predictive value of raS mutations for resistance to cetuximab and panitumumab leading to the license of these monoclonal antibodies exclusively for the management of patients with raS-wild type colorectal cancers. So far the identification of predictive biomarkers have generated interesting, though preliminary and, at times, conflicting data on the importance of tumour mRNA levels of EGFR ligands, of activating mutations in other genes such as NraS and PIK3ca.
