Thrust Propagation in the Aqqikkol Lake Area, the East Kunlun Mountains, Northwestern China

The western segment of the East Kunlun Mountains is one of the poorly studied regions in northwestern China. Through a structural analysis of the typical sections, we have the following views: (1) There is a very well developed fault system in the western segment of the East Kunlun Mountains and thrust propagation, normal slip and decoupling are the chief deformation events in this area. (2) Although the thrusting started in the Late Carboniferous and Late Triassic-Early Jurassic, strong activity took place in the Miocene-Quaternary when the Kumkol basin was strongly downwarped. (3) The tectonic pattern of coexistence of N-directed thrust propagation and S-directed normal slip in this area is consistent with the general tectonic pattern of the northern Qinghai-Tibet plateau and also very similar to that of the Himalayan region on the southern margin of the Qinghai-Tibet plateau, but their directions between the thrust propagation are opposite and all the strong thrust propagations occurred from the Miocene-Pliocene to Quaternary, a period featuring strong collision between the Indian plate and the Eurasian plate and abrupt uplift of the Qinghai-Tibet plateau. This oppositely directed thrust propagation and normal slip reveal such kinematic characteristics as symmetric propagations of deep-seated materials towards the north and south beneath the Qinghai-Tibet plateau and gravitational sliding of superficial materials towards the interior of the plateau. Therefore, the establishment of the fault system in the study area may provide an approach to the study of deep processes of the northern Qinghai-Tibet plateau and the construction of a unified geodynamic model for the uplift of the Qinghai-Tibet plateau.

Zhigancao decoction is a traditional prescription for treating irregular pulse and palpitations in *** the monarch drug of Zhigancao decoction,the bioactive molecules of licorice against heart diseases remain *** established the HRESIMS-guided method leading to the isolation of three novel bicyclic peptides,glycnsisitins A-C(1-3),with distinctive C-C and C-O-C side-chain-toside-chain linkages from the roots of Glycyrrhiza uralensis(licorice).Glycnsisitin A demonstrated stronger cardioprotective activity than glycnsisitins B and C in an in vitro model of doxorubicin(DOX)-induced cardiomyocyte *** A treatment not only reduced the mortality of heart failure(HF)mice in a dose-dependent manner but also significantly attenuated DOX-induced cardiac dysfunction and myocardial *** set enrichment analysis(GSEA)of the differentially expressed genes indicated that the cardioprotective effect of glycnsisitin A was mainly attributed to its ability to maintain iron homeostasis in the ***,glycnsisitin A interacted with transferrin and facilitated its binding to the transferrin receptor(TFRC),which caused increased uptake of iron in *** findings highlight the key role of bicyclic peptides as bioactive molecules of Zhigancao decoction for the treatment of HF,and glycnsisitin A constitutes a promising therapeutic agent for the treatment of HF.

目的:探索与流感病毒核蛋白(NP)相互作用的宿主因子,研究其对流感病毒复制的影响,以及栀子环烯醚萜苷(IGE)抑制流感病毒的作用机制。方法:利用酵母双杂交系统筛选与流感病毒NP相互作用的宿主因子;通过免疫共沉淀实验对异质核糖核蛋白(HNRNPD)、氨基葡萄糖6磷酸脱氨酶1(GNPDA1)、Poly/rC结合因子1(PCBP1)、激活信号转导和转录激活因子(STAT)蛋白1抑制因子(PIAS1)进行验证;通过双荧光素酶实验比较PIAS1和HNRNPD对流感病毒复制的影响,并检测在转染核糖核蛋白体复合物(RNP)及PIAS1敲低的情况下,给予栀子环烯醚萜苷对流感病毒复制的影响。将ICR小鼠随机分为正常组、模型组、达菲组和IGE高、中、低剂量组,每组10只。除正常组外各组滴鼻感染甲型流感病毒FM1株建立病毒性肺炎模型。IGE高、中、低剂量组分别给予50、25、12.5 mg∙kg^(-1)IGE灌胃,达菲组给予27.5 mg∙kg^(-1)达菲灌胃,正常组和模型组灌服等体积蒸馏水,连续4 d。实验结束后,蛋白免疫印迹法检测肺组织中PIAS1、NP、磷酸化(p)-STAT3、STAT3、p-STAT1、STAT1的蛋白表达。结果:在酵母双杂交实验中,发现了16个流感病毒NP的潜在互作宿主靶点;免疫共沉淀实验发现HNRNPD和PIAS1可以与流感病毒NP相互作用;双荧光素酶报告实验中PIAS1敲低和过表达都可明显影响IAV RNP活性(P<0.05,P<0.01),HNRNPD对IAV RNP的影响差异无统计学意义;IGE高、低剂量组均可使流感病毒复制减少(P<0.05),并可逆转PIAS1敲低导致的流感病毒复制增加(P<0.05,P<0.01)。IGE各剂量组不同程度地降低感染小鼠肺组织PIAS1、NP、p-STAT3、p-STAT1、STAT1的表达量(P<0.05,P<0.01)。结论:PIAS1和流感病毒NP相互作用,并能够抑制流感病毒的复制,栀子环烯醚萜苷可能通过PIAS1/STAT1通路抑制IAV RNP的活性,从而发挥抗病毒作用。

DNA sequence comparative analysis of the 3pter-p26 region of human genome

Most proterminal regions of human chromosomes are GC-rich and gene-rich. Chromosome 3p is an exception. Its proterminal region is GC-poor, and likely to lose heterozygosity, thus causing a number of fatal diseases. Except one gap left in the telomeric position, the proterminal region of human chromosome 3p has been completely sequenced. The detailed sequence analysis showed: (i) the GC content of this region was 38.5%, being the lowest among all the human proterminal regions; (ii) this region contained 20 known genes and 22 predicted genes, with an average gene size of 97.5 kb. The previously mapped gene Cntn3 was not found in this region, but instead located in the 74 Mb position of human chromosome 3p; (iii) the interspersed repeats of this region were more active than the average level of the whole human genome, especially (TA)n, the content of which was twice the genome average; (iv) this region had a conserved synteny extending from 104.1 Mb to 112.4 Mb on the mouse chromosome 6, which was 8% larger in size, not in accordance with the whole genome comparison, probably because the 3pter-p26 region was more likely to lose neocleitides and its mouse synteny had more active interspersed repeats.

目的了解长春市儿童铅暴露现状及影响因素,为本地区儿童铅中毒防治工作提供科学依据。方法以长春市某儿童医院儿童保健科在2017年5月—2018年9月收治的804例就诊儿童作为研究对象,进行血铅检测和问卷调查,分析儿童铅暴露的特点。结果长春市儿童血铅水平为32.30(25.40,41.00)μg/L,男童和女童的血铅水平分别为33.00μg/L、32.00μg/L。其中53例血铅水平≥100μg/L,高血铅检出率为6.59%(高铅血症34例、轻度铅中毒7例、中度铅中毒11例、无重度铅中毒例数)。血铅正常组与异常组儿童在性别、父亲文化水平、啃指甲方面比较,差异无统计学意义(P>0.05);但在年龄、母亲文化水平、居住楼层、饭前洗手、吮吸手指、异食癖方面比较,差异有统计学意义(P<0.05)。儿童血铅异常的多因素分析显示:居住楼房较高、饭前洗手等是儿童血铅异常的保护因素(P<0.05);儿童年龄低、吮吸手指、异食癖是儿童血铅异常的危险因素(P<0.05)。结论长春市儿童的血铅水平及高铅血症不容忽视,尤其是3岁及以下婴幼儿、不良行为习惯和异食癖与血铅水平有密切关系。

钾盐是中国紧缺的战略性矿产资源,对保障国家粮食安全、社会稳定和经济可持续发展等具有重要意义和深远影响。中国钾盐资源勘查开发近70年,通过一代又一代钾盐人的共同努力,在钾盐勘查开发利用领域取得了显著成效。但由于受到中国地质构造与古气候等因素、钾盐成矿及保存等条件的影响,中国钾盐找矿工作难度大。中国钾盐产量难以满足需求,长期依赖进口,对外依存度大于50%,供需形势紧张。近年来,国内钾盐勘查总投入整体呈现收紧趋势,探矿权、采矿权数量也逐年下降。本文主要立足于钾盐资源分布与生产格局等,分析了中国钾盐资源勘查开发所面临的国际竞争等,提出应充分发挥中国现有钾盐产业优势和市场优势,加强顶层设计和统筹谋划,用好国内国外两种资源,进一步推进“一带一路”沿线国家钾盐勘查开发的合作进程,保障中国钾盐资源安全;发挥创新驱动作用,培育钾盐勘查开发的新质生产力,克服国内资源短板,引领国际钾盐行业可持续高质量发展。

Metallogenetic epoch and mechanism of the tungsten ore in Yushan, Beishan orogenic belt, Gansu

Using SHRIMP zircon U-Pb dating methods and cathodoluminescence (CL) technique, we have pre- cisely dated ore samples from tungsten ore deposit, ore-bearing volcanic-sedimentary strata and granite in the Yushan region within the east Tianshan-Beishan W-bearing zone. This study reveals Yu- shan tungsten ore deposit zone formed in 250 ± 9 Ma, and it is closely associated with the Permian granites and felsic volcanic rocks. This new type of tungsten ore deposit in the east Tianshan-Beishang multiple-metallogenetic province may probably be associated with the regionally right-literal shearing after the main Carboniferous collisional orogeny, and the formation of pull-apart basin and the felsic magmatism.

Pulmonary fibrosis(PF)is the pathological structure of incurable fibroproliferative lung diseases that are attributed to the repeated lung injury-caused failure of lung alveolar regeneration(LAR).Here,we report that repetitive lung damage results in a progressive accumulation of the transcriptional repressor SLUG in alveolar epithelial type II cells(AEC2s).The abnormal increased SLUG inhibits AEC2s from self-renewal and differentiation into alveolar epithelial type I cells(AEC1s).We found that the elevated SLUG represses the expression of the phosphate transporter SLC34A2 in AEC2s,which reduces intracellular phosphate and represses the phosphorylation of JNK and P38 MAPK,two critical kinases supporting LAR,leading to LAR ***3,a stress sensor,interacts with the E3 ligase MDM2 to suppress SLUG degradation in AEC2s by impeding MDM2-catalyzed SLUG *** SLUG degradation by disturbing the TRIB3/MDM2 interaction using a new synthetic staple peptide restores LAR capacity and exhibits potent therapeutic efficacy against experimental *** study reveals a mechanism of the TRIB3—MDM2—SLUG—SLC34A2 axis causing the LAR failure in PF,which confers a potential strategy for treating patients with fibroproliferative lung diseases.

Identification of interaction between HIV-1 glycoprotein 41 and integrase

Human immunodeficiency virus-1 （HIV-1）encodes 15 viral proteins. Protein-protein interactions play a large role in the function of these proteins. In this study, we attempted to identify novel interactions between the HIV-1 proteins to better understand the role played by viral protein-protein interactions in the life cycle of HIV-I. Genes encoding the 15 viral proteins from the HIV-1 strain AD8 were inserted into the plasmids of a yeast two-hybrid system. By screening 120 pairs of proteins, interactions between seven pairs were found. This led to the discovery of an interaction between the HIV-1 proteins integrase （IN） and glycoprotein 41 （gp41）, which was confirmed by both co-immunoprecipitation （Co-IP） assays and fluorescence resonance energy transfer （FRET） imaging in live cells. In addition, it was found that the amino acids at positions 76-100 of gp41 are required for it to bind to IN. Deletion of this region from gp41 prevented its interaction with IN and reduced the production of HIV-1 in 293T cells. This study provides new information on HIV-1 protein-protein interactions which improves the understanding of the biological functions of gp41 and IN during the virus life cycle.
