Influence of 6-shogaol potentiated on 5-fluorouracil treatment of liver cancer by promoting apoptosis and cell cycle arrest by regulating AKT/mTOR/MRP1 signalling

Currently, chemoresistance seriously attenuates the curative outcome of liver cancer. The purpose of our work was to investigate the influence of 6-shogaol on the inhibition of 5-fluorouracil (5-FU) in liver cancer. The cell viability of cancer cells was determined by MTT assay. Liver cancer cell apoptosis and the cell cycle were examined utilizing flow cytometry. Moreover, qRT–PCR and western blotting was used to analyse the mRNA and protein expression levels, respectively. Immunohistochemistry assays were used to examine multidrug resistance protein 1 (MRP1) expression in tumour tissues. In liver cancer cells, we found that 6-shogaol-5-FU combination treatment inhibited cell viability, facilitated G0/G1 cell cycle arrest, and accelerated apoptosis compared with 6-shogaol or 5-FU treatment alone. In cancer cells cotreated with 6-shogaol and 5-FU, AKT/mTOR pathway- and cell cycle-related protein expression levels were inhibited, and MRP1 expression was downregulated. AKT activation or MRP1 increase reversed the influence of combination treatment on liver cancer cell viability, apoptosis and cell cycle arrest. The inhibition of AKT activation to the anticancer effect of 6-shogaol-5-FU could be reversed by MRP1 silencing. Moreover, our results showed that 6-shogaol-5-FU combination treatment notably inhibited tumour growth in vivo. In summary, our data demonstrated that 6-shogaol contributed to the curative outcome of 5-FU in liver cancer by inhibiting the AKT/mTOR/MRP1 signalling pathway.

Sangguayin preparation(SGY-P) is refined from the traditional Chinese medicinal compound Sangguayin, which"clears heat and promotes fluid" and "tonifies kidney and spleen" for "Xiaoke", commonly known as ‘Diabetes mellitus’ in clinics.Previous studies have shown that SGY-P could reduce insulin resistance and repair damaged pancreas in db/db mice, but the underlying mechanisms were unclear. Here, we investigated whether treatment with SGY-P could protect pancreatic β-cells from apoptosis and uncovered the underlying mechanisms. db/db mice were used to observe the hypoglycemic and islet protective effect in vivo. Apoptosis was induced in mouse insulinoma 6(MIN6) cells by palmitate, following which the cells were treated with SGY-P for elucidating the anti-apoptotic mechanism in vitro. Cell viability and nuclear morphology were detected by CCK-8 assay and Hoechst 33258 staining. The expression levels of apoptosis-, endoplasmic reticulum(ER) stress-, and autophagy-related proteins were measured by western blot. The results showed that SGY-P reduced fasting blood glucose, pancreatic pathological changes, and islet β-cell apoptosis in db/db mice. Palmitate-induced apoptosis in MIN6 cells was decreased by SGY-P treatment. Hence, SGY-P therapy exhibited a protective effect on pancreatic β-cells by decreasing the expression of cleaved caspase-3, cleaved PARP and Bax, and increasing Bcl-2 by suppressing ER stress(Bip/XBP1/IRE1α/CHOP/Caspase-12) and autophagy(LC3/p62/Atg5) pathways.2/Atg5 pathways.

目的研究虎杖苷对D-半乳糖致衰老模型小鼠的调节作用。方法将56只ICR小鼠(雌雄各半)分为正常组、模型组、阳性组、虎杖苷低、中、高治疗组,通过每日颈后部皮下注射D-半乳糖(500 mg·kg^(-1))建立衰老模型。造模期间,阳性组灌胃盐酸多奈哌齐片(0.75 mg·kg^(-1)),治疗组灌胃虎杖苷(40、70、100 mg·kg^(-1)),正常组给予等量生理盐水。通过筑巢实验、新型物体识别实验与Morris水迷宫实验评估小鼠学习认知能力;取小鼠心、肝、脾、肾、胸腺计算脏器指数;苏木精-伊红(HE)染色观察小鼠全脑组织病理变化;ELISA检测小鼠血清与全脑组织中T-SOD、MDA、GSH-Px、AchE水平;Western blot检测小鼠海马组织Keap1、Nrf2、HO-1蛋白表达水平。结果较模型组相比,阳性组与虎杖苷低、中、高剂量组小鼠的筑巢能力、对新物体的识别能力与水下寻找站台的能力有所上升;脏器指数增加;大脑皮层与海马组织的神经元损伤明显改善;血清与脑组织中T-SOD、GSH-Px活性升高,MDA、AchE活性下降;海马组织中Nrf2、HO-1蛋白表达水平上升,Keap1蛋白表达水平降低。结论虎杖苷对D-半乳糖致衰老模型小鼠的学习认知障碍具有改善作用,其机制可能与Keap1/Nrf2/HO-1通路有关。

A Possible X-ray Quasi-periodic Oscillation in the Narrow-line Seyfert 1 Galaxy Mrk 142

A possible quasi-periodic oscillation（QPO） at frequency 7.045 × 10-5Hz is found in the narrow-line Seyfert 1galaxy Mrk 142 in the data of XMM-Newton collected on 2020 April 11. We find that the QPO signal is statistically significantly larger than the 5σ level and highly coherent with quality factor Q > 5 at the 0.3–10 keV band by using the method of the Lomb–Scargle Periodogram, the Weighted Wavelet Z-transform and the REDFIT.We analyze the data in 0.3–0.6 keV, 0.6–1 keV, 1–3 keV and 3–10 keV energy bands, and find obvious QPO signals at 0.3–0.6 keV and 1–3 keV bands. We then analyze the time-average spectra and time variability at the QPO frequency of 7.045 × 10-5Hz, and use a model to fit them. We find that the QPO signal mainly comes from the X-ray hot corona.

目的建立特异性检测三七粉掺伪比例的荧光定量聚合酶链式反应(quantitative polymerase chain reaction, qPCR)方法。方法三七粉经适宜的方法进行核酸提取后,以经过鉴定的正品三七药材粉末核酸提取物作为定量基准,采用特异性引物运用qPCR技术SYBR Green染料法进行扩增,以ΔRn为纵坐标,以百分含量的对数值作为横坐标,绘制标准曲线,建立三七粉掺伪比例相对定量方法。结果在相对含量1%～20%范围内相关系数(r)为0.998,线性关系良好,用人工掺伪25%、50%、75%的样本验证定量方法准确性,准确度在103.2%～122.6%之间,相对标准偏差(relative standard deviation, RSD)均小于15%。结论该方法的线性范围、准确度、精密度均良好,可以用于三七粉掺伪比例的定量测定;另由于伪品没有特异性扩展曲线,该方法还可用于三七粉的真伪鉴别。

Neurophysiological predictors of aphasia recovery in patients with large left-hemispheric infarction: a mismatch negativity study

Background:Although the rehabilitation of aphasia has been extensively studied,the prediction of language outcome still has not received sufficient attention.The aim of this study was to predict the language outcome using mismatch negativity(MMN)in patients with large left-hemispheric infarction.Methods:MMN was elicited by an oddball paradigm in which a standard tone(1000 Hz)and deviant tone(1500 Hz)were presented at 90%and 10%of the number of tones,respectively.The mean amplitudes and laterality indexes(LIs)of MMN were measured over the prefrontal,frontal,central,parietal,temporal,and perisylvian electrodes and both hemispheres during the first 7 days(session 1)and 10 to 20 days(session 2)post-onset.Mixed three-way analysis of variance(ANOVA)was used to investigate differences in these factors between two aphasia groups(the good recovery group and poor recovery group).The predictive value of the most significant LI was also compared with the score of National Institutes of Health Stroke Scale score and low-density volume on computed tomography.Results:A total of 18 patients were enrolled in this study.Mixed three-way ANOVA showed no interaction effect of session×region of interest(ROI)×group(F[3.59,57.38]=1.301,P=0.282)and no interaction effect of ROI×group(F[1.81,29.01]=0.71,P=0.487)and session×group(F[1.00,16.00]=0.084,P=0.776)for MMN amplitude.No interaction effect of session×ROI×group(F[1.79,28.58]=0.62,P=0.530),but an interaction effect of session×group(F[1.00,16.00]=5.21,P=0.036)was found for LIs.In the poor recovery group,the LIs of MMN over all the ROIs,except the parietal area,became more negative at session 2 than those at session 1(P-0.36 over the perisylvian area sugge

Mixing states and secondary formation processes of organic nitrogen-containing single particles in Guangzhou,China

Organic nitrogen(ON)compounds play a significant role in the light absorption of brown carbon and the formation of organic aerosols,however,the mixing state,secondary formation processes,and influencing factors of ON compounds are still unclear.This paper reports on the mixing state of ON-containing particles based on measurements obtained using a highperformance single particle aerosol mass spectrometer in January 2020 in Guangzhou.The ON-containing particles accounted for 21% of the total detected single particles,and the particle count and number fraction of the ON-containing particles were two times higher at night than during the day.The prominent increase in the content of ON-containing particles with the enhancement of NO_xmainly occurred at night,and accompanied by high relative humidity and nitrate,which were associated with heterogeneous reactions between organics and gaseous NO_(x)and/or NO_(3)radical.The synchronous decreases in ON-containing particles and the mass absorption coefficient of water-soluble extracts at 365 nm in the afternoon may be associated with photo-bleaching of the ON species in the particles.In addition,the positive matrix factorization analysis found five factors dominated the formation processes of ON particles,and the nitrate factor(33%)mainly contributed to the production of ON particles at night.The results of this study provide unique insights into the mixing states and secondary formation processes of the ON-containing particles.

Carbonyl compounds have attracted considerable attention due to their extensive applications in drug discovery.Furthermore,they are important synthetic intermediates for the construction of carbon-carbon and carbon-heteroatom bonds.Transition-metal-catalyzed carbonylation via the insertion of co is one of the most efficient and straightforward strategies to access carbonyl compounds.However,most of the transition-metal-catalyzed carbonylative reactions require expensive and toxic noble-metal catalysts.Therefore,there is a growing demand for the exploration of nickel-catalyzed carbonylative reactions via the insertion of CO due to the earth abundance and low cost of nickel.Compared with the well-established palladium-catalyzed carbonylative reactions,nickel-catalyzed analogous transformations have been relatively underdeveloped.This is primarily because CO strongly binds to nickel,often resulting in catalyst poisoning.In recent years,some research groups have focused on using CO surrogates or NN_(2) pincer nickel catalyst to circumvent the formation of Ni(CO)_(4).Nickel-catalyzed carbonylation has been applied in the construction of carbonyl-containing compounds,such as ketones,carboxylic acids,thioesters,acyl chloride and carboxamides.

Frequency and associated factors of accommodation and non-strabismic binocular vision dysfunction among medical university students

AIM:To investigate the frequency and associated factors of accommodation and non-strabismic binocular vision dysfunction among medical university students.METHODS:Totally 158 student volunteers underwent routine vision examination in the optometry clinic of Guangxi Medical University.Their data were used to identify the different types of accommodation and nonstrabismic binocular vision dysfunction and to determine their frequency.Correlation analysis and logistic regression were used to examine the factors associated with these abnormalities.RESULTS:The results showed that 36.71%of the subjects had accommodation and non-strabismic binocular vision issues,with 8.86%being attributed to accommodation dysfunction and 27.85%to binocular abnormalities.Convergence insufficiency(CI)was the most common abnormality,accounting for 13.29%.Those with these abnormalities experienced higher levels of eyestrain(χ2=69.518,P<0.001).The linear correlations were observed between the difference of binocular spherical equivalent(SE)and the index of horizontal esotropia at a distance(r=0.231,P=0.004)and the asthenopia survey scale(ASS)score(r=0.346,P<0.001).Furthermore,the right eye's SE was inversely correlated with the convergence of positive and negative fusion images at close range(r=-0.321,P<0.001),the convergence of negative fusion images at close range(r=-0.294,P<0.001),the vergence facility(VF;r=-0.234,P=0.003),and the set of negative fusion images at far range(r=-0.237,P=0.003).Logistic regression analysis indicated that gender,age,and the difference in right and binocular SE did not influence the emergence of these abnormalities.CONCLUSION:Binocular vision abnormalities are more prevalent than accommodation dysfunction,with CI being the most frequent type.Greater binocular refractive disparity leads to more severe eyestrain symptoms.

Background The aim of this study was to review current delivery room(DR)resuscitation intensity in Chinese tertiary neonatal intensive care units and to investigate the association between DR resuscitation intensity and short-term outcomes in preterm infants born at 24+0_31+6 weeks gestation age(GA).Methods This was a retrospective cross-sectional study.The source population was infants born at 24+0_31+6 weeks'GA who were enrolled in the Chinese Neonatal Network 2019 cohort.Eligible infants were categorized into five groups:(1)regular care;(2)oxygen supplementation and/or continuous positive airway pressure(O2/CPAP);(3)mask ventilation;(4)endotracheal intubation;and(5)cardiopulmonary resuscitation(CPR).The association between DR resuscitation and shortterm outcomes was evaluated by inverse propensity score-weighted logistic regression.Results Of 7939 infants included in this cohort,2419(30.5%)received regular care,1994(25.1%)received O,/CPAP,1436(18.1%)received mask ventilation,1769(22.3%)received endotracheal intubation,and 321(4.0%)received CPR in the DR.Advanced maternal age and maternal hypertension correlated with a higher need for resuscitation,and antenatal steroid use tended to be associated with a lower need for resuscitation(P<0.001).Severe brain impairment increased significantly with increasing amounts of resuscitation in DR after adjusting for perinatal factors.Resuscitation strategies vary widely between centers,with over 50%of preterm infants in eight centers requiring higher intensity resuscitation.Conclusions Increased intensity of DR interventions was associated with increased mortality and morbidities in very preterm infants in China.There is wide variation in resuscitative approaches across delivery centers,and ongoing quality improvement to standardize resuscitation practices is needed.
