Evaluation of microRNA expression profiles in human sperm frozen using permeable cryoprotectant-free droplet vitrification and conventional methods

For sperm cryopreservation,the conventional method,which requires glycerol,has been used for a long *** addition,the permeable cryoprotectant-free vitrification method has been continuously *** the differences of cryopreservation effects between the two methods have being studied,differences in microRNA(miRNA)profiles between them remain *** this study,we investigated the differences in miRNA expression profiles among conventional freezing sperm,droplet vitrification freezing sperm and fresh human *** also analyzed the differences between these methods in terms of differentially expressed miRNAs(DEmiRs)related to early embryonic development and paternal *** results showed no significant differences between the cryopreservation methods in terms of sperm motility ratio,plasma membrane integrity,DNA integrity,mitochondrial membrane potential,acrosome integrity,and ultrastructural ***,sperm miRNA-sequencing showed differences between the two methods in terms of the numbers of DEmiRs(2. and 19 with vitrification using a nonpermeable cryoprotectant and the conventional method,respectively)in postthaw and fresh sperm *** related to early embryonic development and paternal epigenetics mainly included common DEmiRs between the *** results showed that the differences between conventional freezing and droplet vitrification were minimal in terms of miRNA expression related to embryonic development and *** in sperm miRNA expression due to freezing are not always detrimental to embryonic *** study compared differences in miRNA expression profiles before and after cryopreservation between cryopreservation by conventional and vitrification *** offers a new perspective to evaluate various methods of sperm cryopreservation.

BACKGROUND Splenic injury following endoscopic retrograde cholangiopancreatography(ERCP)is a rare *** literature contains around 30 articles reporting various degrees of splenic injuries resulting from ERCP since the first report of splenic rupture after ERCP in *** SUMMARY This report describes a case of splenic hematoma and stent displacement in a 69-year-old male patient who developed these conditions 7 days after undergoing ERCP and *** patient had bile duct stenosis caused by a malignant tumor that was obstructing the bile *** diagnosis was confirmed by epigastric computed tomography and magnetic resonance *** patient was successfully treated with percutaneous transhepatic cholangial drainage,endoscopic pyloric stent placement,and conservative *** causes of splenic injury following ERCP are *** ERCP has the potential to cause splenic *** a patient experiences symptoms such as abdominal pain,decreased blood pressure,and altered hematology after the procedure,it's important to be thoroughly investigated for postoperative bleeding and splenic injury.

It is well known that when the random errors are iid. with finite variance, the week and the strong consiStency of LS estimate of multiple regression coefficients are equivalent. This note, by constructing a counter-example, shows that this equivalence no longer holds true in case that the random errors possess only the r-th moment with 1≤5 T ＜ 2.

The electron-phonon(el-ph)and phonon-phonon interactions play a key role in determining electronic and thermal transport properties,respectively,in promising two-dimensional(2.)semiconductor *** this study,we investigated el-ph interactions using Wannier-Fourier interpolation method and renormalized phonon scattering considering finite-temperature effects in Bi_(2.TeSe_(2.*** re-sults show that the optical phonon modes dominate the carrier scattering,where level repulsion induced by crystal field splitting and spin-orbit coupling(SOC)effect effectively suppresses intervalley scattering,leading to high hole ***,the strong anharmonicity in Bi_(2.TeSe_(2.monolayer results in the temperature-dependent softening of its optical phonons,along with a corresponding variation in interatomic force constants(IFCs).As a result,the lattice thermal conductivity is remarkably low and exhibits weak temperature ***,the predicted dimensionless thermoelectric figure of merit exceeds unity in the range of 2.0-800 K,indicating the potential of Bi_(2.TeSe_(2.monolayer for thermoelectric *** work provides new insights into the elimination of intervalley scat-tering by crystal field splitting and SOC effects,and paves the way for the evaluation of thermoelectric properties of materials with complex scattering mechanisms and strong anharmonicity.

Antitumor therapies based on adoptively transferred T cells or oncolytic viruses have made significant progress in recent years,but the limited efficiency of their infiltration into solid tumors makes it difficult to achieve desired antitumor effects when used *** this study,an oncolytic virus(rVSV-LCMVG)that is not prone to induce virus-neutralizing antibodies was designed and combined with adoptively transferred T *** transforming the immunosuppressive tumor microenvironment into an immunosensitive one,in B16 tumor-bearing mice,combination therapy showed superior antitumor effects than *** occurred whether the OV was administered intratumorally or *** therapy significantly increased cytokine and chemokine levels within tumors and recruited CD8^(+)T cells to the TME to trigger antitumor immune *** with adoptively transferred T cells and subsequent oncolytic virotherapy sensitizes refractory tumors by boosting T-cell recruitment,downregulating the expression of PD-1,and restoring effector T-cell *** offer a combination therapy with greater translational value,mRNA vaccines were introduced to induce tumor-specific T cells instead of adoptively transferred T *** combination of OVs and mRNA vaccine also displays a significant reduction in tumor burden and prolonged *** study proposed a rational combination therapy of OVs with adoptive T-cell transfer or mRNA vaccines encoding tumor-associated antigens,in terms of synergistic efficacy and mechanism.

Spinal cord injuries impose a notably economic burden on society,mainly because of the severe after-effects they *** the ongoing development of various therapies for spinal cord injuries,their effectiveness remains ***,a deeper understanding of metabolism has opened up a new therapeutic opportunity in the form of metabolic *** this review,we explore the metabolic changes that occur during spinal cord injuries,their consequences,and the therapeutic tools available for metabolic *** spinal cord metabolism is characterized by independent cellular metabolism and intercellular metabolic ***,spinal cord injury results in metabolic disorders that include disturbances in glucose metabolism,lipid metabolism,and mitochondrial *** metabolic disturbances lead to corresponding pathological changes,including the failure of axonal regeneration,the accumulation of scarring,and the activation of *** rescue spinal cord injury at the metabolic level,potential metabolic reprogramming approaches have emerged,including replenishing metabolic substrates,reconstituting metabolic couplings,and targeting mitochondrial therapies to alter cell *** available evidence suggests that metabolic reprogramming holds great promise as a next-generation approach for the treatment of spinal cord *** further advance the metabolic treatment of the spinal cord injury,future efforts should focus on a deeper understanding of neurometabolism,the development of more advanced metabolomics technologies,and the design of highly effective metabolic interventions.

Dynamic Spacer Installation of Multifunctionalities into Metal-Organic Frameworks for Spontaneous One-Step Ethylene Purification from a Ternary C_(2.-Hydrocarbons Mixture

Simultaneous ethane and acetylene removal from a C_(2.-gases mixture(C_(2.H_(6),C_(2.H_(4),and C_(2.H_(2.)through a one-step separation process for ethylene purification is of great importance yet challenging in petrochemical industry,owing to their similar molecule sizes and physical ***,a series of multifunctionalized metal–organic frameworks(MOFs),LIFM-XYY-1∼8(LIFM stands for Lehn Institute of Functional Materials,and XYY are the initials of the first author),are constructed via a dynamic spacer installation(DSI)approach to optimize the pore-nanospaces for efficient C_(2.H_(4) isolation from the ternary C_(2.-gases *** of variable organic-spacers into the prototypical MOFs,LIFM-2. or PCN-700,results in dramatically improved pore volume/surface area,contracted pore size,and functionalized pore surface,which in turn bring out high C_(2.-gases uptake capacities,enhanced C_(2.H_(6) and C_(2.H_(2. adsorption selectivities over C_(2.H_(4),and fast adsorption kinetics,providing an effective strategy to achieve delicate trade-off among these indexes for adequate separation ***,optimized LIFM-XYY-7 presents four-times C_(2.H_(6) and C_(2.H_(2. adsorption capacities than *** breakthrough experiments reveal that poly-grade C_(2.H_(4)(>99.9%)can be obtained from binary or ternary C_(2.-hydrocarbon mixtures through a single separation *** with themolecular simulations,this work demonstrates a promising protocol of porenanospace engineering via multi-functional optimization by the DSI approach to screen out MOFs for a formidable task.

Dear Editor,Major depressive disorder(MDD)is one of the most common psychiatric illnesses that significantly increase the risk of suicide.1 Stress-triggered dysfunctions of microglia have been identified as a commonly occurred pathological feature of MDD.1–3 Microglial dysfunction contributes to the pathogenesis of MDD via immunoresponses/neuroinflammation-mediated neural damage and pathological synapse loss-mediated neural circuit disruption.1 Although the involvement of microglia in MDD has been widely investigated,the molecular mechanisms underlying microglial dysfunction remain largely ***,we identified glutaminase 1(Gls1)as one key protein that participates in microglial dysfunction.4–6 Gls1 catalyzes the hydrolysis of glutamine to produce glutamate in the brain.4 Besides its well-known role in excitatory neurotoxicity,we found Gls1 up-regulation in microglia in animal models of Alzheimer’s disease and ischemic stroke.4,7 Gls1 activates microglia to overproduce cytokines and release inflammatory extracellular vesicles,therefore leading to neuroinflammation in animal models of Alzheimer’s disease and ischemic stroke.4–6 More importantly,Gls1 has been found to be up-regulated in the brains of MDD patients,and microglial Gls1 deficiency mitigated LPS-induced depression-like behaviors.8 However,LPS exposure is not an appropriate model to mimic MDD phenotypes,leaving the involvement of Gls1 in MDD an undetermined question.

Enhancing solid-state lithium metal battery performance via indium-based modification of electrolytes and lithium metal surfaces: mechanistic insights and optimization

Argyrodite-based solid-state lithium metal batteries exhibit significant potential as next-generation energy storage ***,their practical applications are constrained by the intrinsic poor stability of argyrodite towards Li metal and exposure to air/***,an indium-involved modification strategy is employed to address these *** optimized doping yields a high Li-ion conductivity of 7.5 mS cm-1for Li5.54In0.02.S4.47O0.03Cl1.5electrolyte,accompanied by enhanced endurance against air/moisture and bare Li *** retains 92.0%of its original conductivity after exposure to air at a low dew point of-60°C in dry ***,a composite layer comprising Li–In alloy and LiF phases is generated on the surface of lithium metal anode via the reaction between In F3and molten *** layer effectively mitigates Li dendrite growth by creating a physical barrier from the robust LiF phase,while the Li–In alloy induces uniform Li-ion deposition and accelerates Li transport dynamics across the interphase between the solid electrolyte/Li ***,the In-doped electrolyte facilitates the in-situ generation of Li–In alloy within its voids,reducing local current density and further inhibiting lithium dendrite ***,the combination of the Li5.54In0.02.S4.47O0.03Cl1.5electrolyte and the InF3@Li anode provides exceptional electrochemical performances in both symmetric cells and solid-state lithium metal batteries across different operating ***,the LiNbO3@LiNi0.7Co0.2.n0.1O2.Li5.54In0.02.S4.47O0.03Cl1.5/InF3@Li cell delivers a high discharge capacity of 167.8 mAh g-1at 0.5 C under 2.°C and retains 80.0%of its initial value after 400 *** work offers a viable strategy for designing functional interfaces with enhanced stability for sulfide-based solid-state lithium batteries.
