A target lipidomics approach to investigate the acute inflammatory irritation induced by indolealkylamines from Chansu water fraction in rats

Chansu has demonstrated adverse reactions in clinical settings,which is associated with its toxicity and limits its clinical *** there are methodological limitations for drug safety *** the current study,ultra-high performance liquid chromatography,lipidomic profiling,and molecular docking were used to systemically assess Chansu-induced acute inflammatory irritation and further identify the underlying drug *** with the EtOAc extract,Chansu water fraction containing indolealkylamines caused acute inflammatory irritation in rats,including acute pain(spontaneous raising foot reaction),and inflammation(paw edema).At the molecular level,lipids analysis revealed significantly higher levels of pro-inflammatory mediators of the COX and LOX ***,anti-inflammatory mediators from the CYP 450,ALA,and DHA pathways markedly decreased after exposure to Chansu water ***,four indolealkylamines from Chansu showed a high theoretical affinity to a known irritation target,5-HT_(2A)*** results suggest that Chansu-induced inflammatory irritation is related to the distinct dysregulation of inflammatory lipids,and peripheral 5-HT_(2A)R is a potential target for irritation *** strategy used in this study can be a crucial approach in the safety evaluation of natural medicinal substances.

Environmental Embrittlement in A_3B-type Intermetallic Alloys

Environmental embrittlement in A3B-type intermetallics based on Ni3Al and Fe3Al has been studied in this paper. For the Ni3Al doped with 120 wt ppm B and Ni,(Al,***) doped with 80 wt ppm B,their elongation and ultimate tensile strength decreased in the sequence:of vacuum > air >hydrogen. while for Ni,(Al,Mn) doped with 400 wt ppm B no envifonmental degradation was ob served, although a -Ni3(Al,Mn) alloy without B showed a decrease in ductility when tested in air in stead of oxygen. It is supposed that boron and hydrogen compete for the occupation of interstitial sites near grain boundaries. If boron content is sufficiently low, hydrogen embrittlement occurs ;however, if its content is sufficiently high. boron addition is capable of eliminating envjronmental ef fect in Ni3Al-based alloysi As to the micromechanism of hydrogen embrittlement in Ni3Al+B. S EM in situ observations showed that both grain boundary decohesion and a high stress concentration con tributed to hydrogen-assisted jntergranu lar cracking in this alloy. For the Fe3Al and Fe3 (***) *** mechanical properties depended strongly on grain size / grain shape and testing environment. A strain rate effect on ductiIity and fracture strength was also observed in the Fe3Al and Fe,(Al,Cr)+B aIloys. Preoxidation increased the ductility of the Fe,(Al,Cr)+B alloy. All these results can be rationalized from a hypothesis that surface reaction is the controlling process in embrittling Fe3Al-based alloys.

Climate changes drive phytoplankton community through complementarity and selection effects in a large mesotrophic reservoir

Understanding the response of the phytoplankton community to climate change is essential for reservoir *** analyzed a long-term data series(2009–2020)on the phytoplankton community in a large mesotrophic reservoir in the wet season to investigate the impacts of temperature and precipitation increases caused by climate change on the functioning and trait composition of the phytoplankton *** the last twelve years,the 3-month accumulative precipitation increased from 291.03 mm to 590.91 mm,and the surface water temperature increased from 25.06℃to 26.49℃in wet season,*** changes caused a higher water level,stronger thermal stratification and lower nitrogen concentration in Daxi *** dynamic equilibrium model indicated that the increased precipitation and water temperature-related environmental changes would result in a more diverse and productive phytoplankton *** effects of increasing water temperature and precipitation on the niche complementarity and selection effects within the phytoplankton community were analyzed using structural equation model by means of the functional divergence index and functional evenness index,respectively,elucidating the reasons for the increase in cyanobacteria in the absence of a significant increase in nutrient *** on these results,it is advisable that more stringent phosphorus control standards might be conducted to reduce the risks of cyanobacteria proliferation in the context of global warming.

“One stone, two birds”: Salt template enabling porosity engineering and single metal atom coordinating toward high-performance zinc-ion capacitors

Zinc-ion hybrid capacitors (ZIHCs) have received increasing attention as energy storage devices owing to their low cost,high safety,and environmental ***,their progress has been hampered by low energy and power density,as well as unsatisfactory long-cycle stability,mainly due to the lack of suitable electrode *** this context,we have developed manganese single atoms implanted in nitrogen-doped porous carbon nanosheets (MnSAs/NCNs) using a metal salt template method as cathodes for *** metal salt serves a dual purpose in the synthesis process:It facilitates the uniform dispersion of Mn atoms within the carbon matrix and acts as an activating agent to create the porous *** applied in ZIHCs,the MnSAs/NCNs electrode demonstrates exceptional performance,including a high capacity of 203 m Ah g^(-1),an energy density of 138 Wh kg^(-1)at 68 W kg^(-1),and excellent cycle stability with 91%retention over 10,000 *** calculations indicate that the introduced Mn atoms modulate the local charge distribution of carbon materials,thereby improving the electrochemical *** work demonstrates the significant potential of carbon materials with metal atoms in zinc-ion hybrid capacitors,not only in enhancing electrochemical performance but also in providing new insights and methods for developing high-performance energy storage devices.

Functions of S100B in cardiovascular disease

S100B is an EF chiral protein, which is acidic and mainly located in Schwann cells and glial cells of the nervous system. It has the functions of regulating calcium homeostasis and protein phosphorylation, activating glial cells and regulating their migration and proliferation. At the same time, S100B is also expressed in myocardial tissue, which can reduce the concentration of calcium ions in myocardial cells, thus reducing myocardial contractility. It can also negatively regulate cardiac hypertrophy by inhibiting the activity of P38 kinase and reducing MHC expression, and induce apoptosis by extracellular mechanism namely interacting with receptor of advanced glycosylation end product (RAGE) and activating the extracellular regulatory kinase (ERK1/2) and p53 signal. The intracellular and extracellular effects of S100B are promising therapeutic targets for cardiovascular diseases.

针对延迟容忍移动传感器网络提出一种基于广播的副本消除数据传输机制(CRD).CRD机制中,传感器节点利用基站在频率f1上的大功率广播信息完成自身转发概率的计算和冗余消息副本的清理,基于转发概率在频率f2上完成节点间的消息转发.同时,CRD采用消息生存时间和消息转发域值M来完成消息队列的管理.仿真结果表明,与其他数据传输机制相比,CRD能达到传输成功率、传输延迟和通信开销的有效平衡.

Assembly and Immunogenicity of Human Papillomavirus Type 16 Major Capsid Protein(HPV16 L1) in Pichia pastoris

In this study, a recombinant Pichia pastoris expression system was developed to express HPV16 L1 protein that was driven by a strong AOX1 promoter. HPV16L1 gene was cloned into vector pPICZ,αB. HPV16 L1 protein expression induced by methanol was screened by using sodium dedecyl sulfate-polyacrylamide gel electrophoresis （SDSPAGE） and Western blotting. The results indicate that the HPVl6 L1 protein is secreted by the recombinant P. pastoris, and the purified HPV16 L1 protein can self-assemble into vires-like particles（ VLPs）, which show a good immunogenicity and induces high-titer antibody in mice.

Efficient photocatalytic reduction of CO_(2)to formate by a molecular noble metal-free system

Reducing CO_(2)to carbon-neutral solar fuels can not only alleviate the energy demand but also mitigate the greenhouse *** present,developing an efficient non-noble metal photo/electrocatalytic system for converting CO_(2)to a specific product beyond CO is still a big ***,we report a new cobalt complex bearing 2,2′-bi-1,10-phenanthroline ligand that can photocatalytically reduce CO_(2)to formate(HCOO^(-)).Turnover number(TON)for HCOO^(-)up to 677 with 99%selectivity can be achieved using organic dyes purpurin as photosensitizer and 1,3-dimethyl-2-phenyl-2,3-dihydro-1H-benzo[d]imidazole(BIH)as sacrificial reductant in a CO_(2)-saturated NMP/TEA(v/v,3:1)solution under visible light *** potential electrolysis at-1.41 V(***/Ag Cl)in the presence of 12 mM Et_(3)NHBF_(4)also produced HCOO^(-)with Faradaic efficiency of 39%.Spectroscopic and electrochemical studies suggest that a[CoI-H]species is the key intermediate for CO_(2)reduction.

The gas explosion in residential building has always been a highly concerned *** in homogeneous mixtures have been extensively ***,mixtures are often inhomogeneous in the practical scenarios due to the differences in the densities of methane and *** order to investigate the effects of gas explosions in inhomogeneous mixtures,experimental studies involving gas leakage and explosion are conducted in a full-scale residential building to reproduce the process of gas *** fitting the dimensionless buoyancy as a function of dimensionless height and dimensionless time,a distribution model of gas in large-scale spaces is established,and the mechanism of inhomogeneous distribution of methane is also be ***,the stratified reconstruction method(SRM)is introduced for efficiently setting up inhomogeneous concentration fields in *** simulation results highlight that for the internal overpressure,the distribution of methane has no effect on the first overpressure peak(ΔP1),while it significantly influences the subsequent overpressure peak(ΔP2),and the maximum difference between the overpressure of homogeneous and inhomogeneous distribution is174.3%.Moreover,the initial concentration distribution also has a certain impact on the external overpressure.

Systematic experimental investigation on pressure build-up characteristics of water-jet injection into a molten LBE pool

In the scenario of a steam generator tube rupture accident in a lead-cooled fast reactor,secondary circuit subcooled water under high pressure is injected into an ordinary-pressure primary vessel,where a molten lead-based alloy(typically pure lead or lead-bismuth eutectic(LBE))is used as the *** clarify the pressure build-up characteristics under water-jet injection,this study conducted several experiments by injecting pressurized water into a molten LBE pool at Sun Yat-sen *** obtain a further understanding,several new experimental parameters were adopted,including the melt temperature,water subcooling,injection pressure,injection duration,and nozzle *** detailed analyses,it was found that the pressure and temperature during the water-melt interaction exhibited a consistent variation trend with our previous water-droplet injection mode LBE ***,the existence of a steam explosion was confirmed,which typically results in a much stronger pressure *** the non-explosion cases,increasing the injection pressure,melt-pool temperature,nozzle diameter,and water subcooling promoted pressure build-up in the melt ***,a limited enhancement effect was observed when increasing the injection duration,which may be owing to the continually rising pressure in the interaction vessel or the isolation effect of the generated steam *** of whether a steam explosion occurred,the calculated mechanical and kinetic energy conversion efficiencies of the melt were relatively small(not exceeding 4.1%and 0.7%,respectively).Moreover,the range of the conversion efficiency was similar to that of previous water-droplet experiments,although the upper limit of the jet mode was slightly lower.
