Increased susceptibility of algal symbionts to photo-inhibition resulting from the perturbation of coral gastrodermal membrane trafficking

The stability of cnidarian-dinoflagellate endosymbioses is dependent upon communication between the host gastrodermal cell and the symbionts housed within it. Although the molecular mechanisms remain to be elucidated, existing evidence suggests that the establishment of these endosymbioses may involve the sorting of membrane proteins. The present study examined the role of host gastrodermal membranes in regulating symbiont （genus Symbiodinium） photosynthesis in the stony coral Euphyllia glabrescens. In comparison with the photosynthetic behavior of Symbiodinium in culture, the Symbiodinium populations within isolated symbiotic gastrodermal cells （SGCs） exhibited a significant degree of photo-inhibition, as determined by a decrease in the photochemical efficiency of photosystem II （Fv/Fm）. This photo-inhibition coincided with increases in plasma membrane perturbation and oxidative activity in the SGCs. Membrane trafficking in SGCs was examined using the metabolism of a fluo- rescent lipid analog, N-[5-（5,7-dimethyl boron dipyrromethene difluoride）-l-pentanoyl]-D-erythro-Sphingosylpbosphoryl- choline （BODIPY-Sphingomyelin or BODIPY-SM）. Light irradiation altered both membrane distribution and trafficking of BODIPY-SM, resulting in metabolic changes. Cholesterol depletion of the SGC plasma membranes by methyl-13-cyclodextrin retarded BODIPY-SM degradation and further augmented Symbiodinium photo-inhibition. These results indicate that Symbio- dinium photo-inhibition may be related to perturbation of the host gastrodermal membrane, providing evidence for the pivotal role of host membrane trafficking in the regulation of this environmentally important coral-dinoflagellate endosymbiosis.

Evaluating the Compatibility Mechanism of Shengxian Decoction Based on an Excretion Study of 18 Bioactive Constituents in Rat Biosamples

Introduction:The Shengxian decoction(SXT),a Chinese herbal medication used to treat heart failure,is composed of Astragali Radix,Anemarrhenae Rhizoma,Bupleuri Radix,Cimicifuage Rhizoma,and Platycodonis *** of the excretion of the active compounds in this herbal medication is vital for investigating the underlying mechanisms behind its ***,this remains ***:Liquid chromatography coupled with mass spectrometry was performed in both positive and negative modes to assay 18 constituents of SXT from rat urinary,fecal,and biliary ***:The methodology was validated and showed good linearity(r≥0.9),precision,stability,repeatability,and *** relative standard deviations and relative errors for the precision and accuracy did not exceed 15%.The recoveries of all the compounds ranged from 77.37%to 114.82%,and the matrix effect was between 82.53%and 105.71%.The results suggested that,when combined with Platycodonis Radix,the levels of 14 constituents from the four herbs were reduced in the urine,while the levels of six constituents were reduced in the ***:The comparative excretion results indicated that Platycodonis Radix reduced the excretion of compounds in SXT,demonstrating the compatibility mechanism and holistic properties of these compounds,favoring the pharmacological effects of SXT.

A froth velocity measurement method based on improved U-Net++semantic segmentation in flotation process

Realgar-indigo naturalis formula for the treatment of patients with relapsed and arsenic trioxide-resistant acute promyelocytic leukemia:A case series

Introduction There is currently no standard treatment for relapsed and arsenic trioxide(ATO)-resistant acute promyelocytic leukemia(APL).Here,we report a case series of realgar-indigo naturalis formula(RIF)for the successful treatment of patients with relapsed and ATO-resistant *** presentation Two patients in the first relapse and one in the second relapse failed to achieve hematologic complete remission(HCR)when reinduced by ATO;the other five patients progressed to relapse during ATO-based regimens for post-remission *** eight patients received RIF in three doses per day totaling 130 mg/kg(≤30 pills)as induction therapy and achieved HCR at a median time of 46.5 *** received 5 years of post-remission therapy,which consisted of combined chemotherapy followed by *** this period,the patients did not experience renal dysfunction or QT interval *** the last follow-up,three patients survived without relapse,two patients survived with a second or third relapse and third or fourth remission,and the other three patients relapsed for a third or fourth time and *** 5-year overall survival and event-free survival rates were 75.0%(95%confidence interval[CI]:31.5–93.1)and 37.5%(95%CI:5.6–71.7),*** RIF for induction therapy and RIF combined with chemotherapy for post-remission therapy may represent an effective and safe protocol for the treatment of patients with relapsed and ATO-resistant APL.

Tailoring microenvironment for efficient CO_(2)electroreduction through nanoconfinement strategy

The electrocatalytic conversion of CO_(2)to produce fuels and chemicals holds great promise,not only to provide an alternative to fossil feedstocks,but also to use renewable electricity to convert and recycle the greenhouse gas CO_(2)to mitigate climate ***,the selectivity and reaction rates for the conversion of CO_(2)into desirable carbon-based products,especially multicarbon products with high added value,are still insufficient for commercial applications,which is attributed to insufficiently favourable microenvironmental conditions in the vicinity of the *** construction of catalysts/electrodes with confined structures can effectively improve the reaction microenvironment in the vicinity of the electrodes and thus effectively direct the reaction towards the desired *** this review,we firstly introduce the effects of the microenvironment at the electrode-electrolyte interface including local pH,local intermediate concentration,and local cation concentration on CO_(2)reduction reaction(CO_(2)RR)as well as the mechanism of action,and then shed light on the microenvironmental modulation within the confined space,and finally and most importantly,introduce the design strategy of CO_(2)RR catalyst/electrode based on the confinement effect.

Spin-orbit interaction induced Casimir-Lifshitz torque between two-dimensional electron gases

We investigate theoretically the Casimir interaction between two parallel two-dimensional electron gases(2DEGs)with Rashba and Dresselhaus spin-orbit interactions(SOIs),based on the quantum field *** derive an analytical expression for the polarization tensor and obtain the solution to the motion equation of the electromagnetic(EM)*** calculate the CasimirLifshitz torque(CLT)between two parallel 2DEGs with Rashba and Dresselhaus SOIs,which can be tuned by changing the relative strength between two *** anisotropic Casimir energy between 2DEGs caused by the interplay between the SOIs of 2DEGs offers a new origin of CLT and a new type of optoelectronic device.

N6-methyladenosine(m^(6)A)has been established as the most prevalent chemical modification in message RNA(mRNA),playing an essential role in determining the fate of RNA *** of m^(6)A has been revealed to lead to abnormal physiological conditions and cause various types of human *** studies have delineated the genetic regulatory maps for m^(6)A methylation by mapping the quantitative trait loci of m^(6)A(m^(6)A-QTLs),thereby building up the regulatory circuits linking genetic variants,m^(6)A,and human complex ***,we review the recent discoveries concerning the genetic regulatory maps of m^(6)A,describing the methodological and technical details of m^(6)A-QTL identification,and introducing the key findings of the cis-and trans-acting drivers of m^(6)*** further delve into the tissue-and ethnicity-specificity of m^(6)A-QTL,the association with other molecular phenotypes in light of genetic regulation,the regulators underlying m^(6)A genetics,and importantly,the functional roles of m^(6)A in mediating human complex ***,we discuss potential research avenues that can accelerate the translation of m^(6)A genetics studies toward the development of therapies for human genetic diseases.

SsdchA is a novel secretory cellobiohydrolase driving pathogenicity in Sclerotinia sclerotiorum

The necrotrophic fungus, Sclerotinia sclerotiorum, employs an array of cell wall-degrading enzymes(CWDEs), including cellulase, to dismantle host cell walls. However, the molecular mechanisms through which S. sclerotiorum degrades cellulose remain elusive. Here, we unveil a novel secretory cellobiohydrolase, SsdchA, characterized by a signal peptide and a Glyco_hydro_7(GH7) domain. SsdchA exhibits a robust expression of during early infection stages. Interestingly, colony morphology and growth rates remain unaffected across the wild-type, SsdchA deletion strains and SsdchA overexpression strains on potato dextrose agar(PDA) medium. Nevertheless, the pathogenicity and cellobiohydrolase activity decreased in the SsdchA deletion strains, but enhanced in the SsdchA overexpression strains. Moreover,the heterologous expression of SsdchA in Arabidopsis thaliana leads to reduced cellulose content and heightened susceptibility to S. sclerotiorum. Collectively, our data underscore the pivotal role of the novel cellobiohydrolase SsdchA in the pathogenicity of S. sclerotiorum.

The breaking of spin symmetry in the single-particle resonances in deformed nuclei

The exploration of spin symmetry (SS) in nuclear physics has been instrumental in identifying atomic nucleus *** this study,we solve the Dirac equation from the relativistic mean field (RMF) in complex momentum *** investigated SS and its breaking in single-particle resonant states within deformed nuclei,with a focus on the illustrative *** was the initial discovery of a resonant spin doublet in a deformed nucleus,with the expectation of the SS approaching the continuum *** increasing single-particle energy,the splitting of the resonant spin doublets widened *** escalating splitting implies diminishing adherence to the SS,indicating a departure from the expected behavior as the energy levels *** also analyzed the width of the resonant states,showing that lower orbital angular momentum resonances possess shorter decay times and that SS is preserved within broad resonant doublets,as opposed to narrow resonant *** the radial density of the upper components for the bound-state and resonant-state doublets,it becomes evident that while SS is well-preserved in the bound states,it deteriorates in the resonant *** impact of nuclear deformation (β_(2)) on SS was examined,demonstrating that an increase in β_(2) resulted in higher energy and width splitting in the resonant spin doublets,which is attributed to increased component ***,the sensitivity of spin doublets to various potential parameters such as surface diffuseness (a),radius (R),and depth (Σ0) is discussed,emphasizing the role of these parameters in *** study provides valuable insights into the behavior of spin doublets in deformed nuclei and their interplay with the nuclear structure,thereby advancing our understanding of SS in the resonance state.

Lithium–oxygen battery with ultrahigh theoretical energy density is considered a highly competitive next-generation energy storage device,but its practical application is severely hindered by issues such as difficult decomposition of discharge products at ***,we have developed N-doped carbon anchored atomically dispersed Ru sites cathode catalyst with open hollow structure(h-RuNC)for Lithium–oxygen *** one hand,the abundance of atomically dispersed Ru sites can effectively catalyze the formation and decomposition of discharge products,thereby greatly enhancing the redox *** the other hand,the open hollow structure not only enhances the mass activity of atomically dispersed Ru sites but also improves the diffusion efficiency of catalytic ***,the excellent activity from atomically dispersed Ru sites and the enhanced diffusion from open hollow structure respectively improve the redox kinetics and cycling stability,ultimately achieving a high-performance lithium–oxygen battery.
