Effects of hypoxia,hyperoxia on the regulation of expression and activity of matrix metalloproteinase-2 in hepatic stellate cells

AIM: To study the effects of hypoxia, hyperoxia on the regulation of expression and activity of matrix metalloproteinase-2 (MMP-2) in hepatic stellate cells (HSC). METHODS: The expressions of MMP-2, tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) and membrane type matrix metalloproteinase-1 (MT1-MMP) in cultured rat HSC were detected by immunocytochemistry (ICC) and in situ hybridization (ISH). The contents of MMP-2 and TIMP-2 in culture supernatant were detected with ELISA and the activity of MMP-2 in supernatant was revealed by zymography. RESULTS: In the situation of hypoxia for 12h, the expression of MMP-2 protein was enhanced (hypoxia group positive indexes: 5.7 +/- 2.0, n=10; control: 3.2 +/- 1.0, n = 7; P<0.05), while TIMP-2 protein was decreased in HSC (hypoxia group positive indexes: 2.5 +/- 0.7, n = 10; control: 3.6 +/- 1.0, n = 7; P < 0.05), and the activity (total A) of MMP-2 in supernatant declined obviously (hypoxia group: 7.334 +/- 1.922, n = 9; control: 17.277 +/- 7.424, n = 11; P < 0.01). Compared the varied duration of hypoxia, the changes of expressions including mRNA and protein level as well as activity of MMP-2 were most notable in 6h group. The highest value(A(hypoxia)-A(control)) of the protein and the most intense signal of mRNA were in the period of hypoxia for 6h, along with the lowest activity of MMP-2. In the situation of hyperoxia for 12h, the contents (A(450)) of MMP-2 and TIMP-2 in supernatant were both higher than those in the control, especially the TIMP-2 (hyperoxia group: 0.0499 +/- 0.0144, n = 16; control: 0.0219 +/- 0.0098, n = 14; P < 0.01), and so was the activity of MMP-2 (hyperoxia group: 5.252 +/- 0.771, n = 14; control: 4.304 +/- 1.083, n = 12; P < 0.05), and the expression of MT1-MMP was increased. CONCLUSION: HSC is sensitive to the oxygen, hypoxia enhances the expression of MMP-2 and the effect is more marked at the early stage; hyperoxia mainly raises the activity of MMP-2.

Building of hFⅨ transgenic mice by spermatogenic cells

Human FⅨ expression vector pCMVⅨ was packaged by EffecteneTM reagent and injected into mice seminiferous tubules with glass pipettes. The expressional frame of pCMVⅨ was examined by PCR and Southern blotting among 41 progenies. There were 2 (4%) mice being integrated with hFⅨ gene into chromosomes. 4.6 ng/mL of hFⅨ protein was expressed in plasma of one mouse, which was tested by ELISA. We demonstrated that building of transgenic animals by spermatogonial stem cells is an efficient method. Meanwhile, it has also been proved to be an alternative choice for mammary gland bioreactor.

Phylogenetic relationships of host insects of Cordyceps sinensis inferred from mitochondrial Cytochrome b sequences

This study used the sequence of the mitochondrial Cytochrome b(Cytb)to estimate phylogenetic relationships among host Hepialidae insects of Cordyceps *** DNA of host insect was extracted from the dead larva head part of 18 cordyceps populations and 2 species of Hepialus,and the Cytb fragment of host insect was amplified with PCR *** nucleotide sequence alignments and their homologous sequences of 24 species host Hepialidae insects of Cordyceps sinensis were obtained from GenBank and were used to construct phylogenetic trees based on neighbor-joining *** results showed that genus Bipectilus diverged earlier than genus Hepialus and *** host insects of Cordyceps sinensis have multitudinous species with different morphological characteristics and geographical *** interspecific genetic differentiations are obvious in ***,the genus Hepialus might be considered as polyphyletic *** sequences have abundant variations among the host insects of Cordyceps sinensis on spe- cific and generic *** divergence rate of Cytb sequences among the species in Hepialus ranged from 0.23% to 9.24%,except that Hepialus pratensis and Hepialus jinshaensis have the same *** sequence can be used for species identification of host insects of Cordyceps sinensis,but further confirmation in more host insect species is *** obtain the Cytb sequence of host insect by ampli- fying DNA extracted from the head part of dead larva in cordyceps turns out to be an effective and accurate approach,which will be useful for studies on phylogeny and genetic structure of host insects of cordyceps populations,especially for analyzing relationships between C. sinensis and its host insects.

The author studies M-D Riemann problems for a quasilinear nonstrictly hyperbolic system. The initial data are taken as three different constants in three sections divided by three rays starting from the origin. From each direction of these rays two waves coming from infinity are allowed. All possible local singularity structures are carefully studied and classified. Then based on such analysis,existence and global singularity structure of the solution are obtained under some assumptions.

Achieving high maize yields and efficient phosphorus(P)use with limited environmental impacts is one of the greatest challenges in sustainable maize *** plant density is considered an effective approach for achieving high maize ***,the low mobility of P in soils and the scarcity of natural P resources have hindered the development of methods that can simultaneously optimize P use and mitigate the P-related environmental footprint at high plant *** this study,meta-analysis and substance flow analysis were conducted to evaluate the effects of different types of mineral P fertilizer on maize yield at varying plant densities and assess the flow of P from rock phosphate mining to P fertilizer use for maize production in china.A significantly higher yield was obtained at higher plant densities than at lower plant *** application of single superphosphate,triple super-phosphate,and calcium magnesium phosphate at high plant densities resulted in higher yields and a smaller environmental footprint than the application of diammonium phosphate and monoammonium *** scenario analyses suggest that combining the optimal P type and application rate with a high plant density could increase maize yield by 22%.Further,the P resource use efficiency throughout the P supply chain increased by 39%,whereas the P-related environmental footprint decreased by 33%.Thus,simultaneously optimizing the P type and application rate at high plant densities achieved multiple objectives during maize production,indicating that combining P management with cropping techniques is a practical approach to sustainable maize *** findings offer strategic,synergistic options for achieving sustainable agricultural development.
