Rbm8a regulates neurogenesis and reduces Alzheimer's disease-associated pathology in the dentate gyrus of 5×FAD mice

Alzheimer’s disease is a prevalent and debilitating neurodegenerative condition that profoundly affects a patient’s daily functioning with progressive cognitive decline,which can be partly attributed to impaired hippocampal neurogenesis.Neurogenesis in the hippocampal dentate gyrus is likely to persist throughout life but declines with aging,especially in Alzheimer’s disease.Recent evidence indicated that RNA-binding protein 8A(Rbm8a)promotes the proliferation of neural progenitor cells,with lower expression levels observed in Alzheimer’s disease patients compared with healthy people.This study investigated the hypothesis that Rbm8a overexpression may enhance neurogenesis by promoting the proliferation of neural progenitor cells to improve memory impairment in Alzheimer’s disease.Therefore,Rbm8a overexpression was induced in the dentate gyrus of 5×FAD mice to validate this hypothesis.Elevated Rbm8a levels in the dentate gyrus triggered neurogenesis and abated pathological phenotypes(such as plaque formation,gliosis reaction,and dystrophic neurites),leading to ameliorated memory performance in 5×FAD mice.RNA sequencing data further substantiated these findings,showing the enrichment of differentially expressed genes involved in biological processes including neurogenesis,cell proliferation,and amyloid protein formation.In conclusion,overexpressing Rbm8a in the dentate gyrus of 5×FAD mouse brains improved cognitive function by ameliorating amyloid-beta-associated pathological phenotypes and enhancing neurogenesis.

Imbalance of Circulating Follicular Regulatory and Follicular Helper T Cell Subpopulations Is Associated with Disease Progression and Serum CYFRA 21-1 Levels in Patients with Non-small Cell Lung Cancer

Objective This study aimed to investigate the changes of follicular helper T(TFH)and follicular regulatory T(TFR)cell subpopulations in patients with non-small cell lung cancer(NSCLC)and their significance.Methods Peripheral blood was collected from 58 NSCLC patients at different stages and 38 healthy controls.Flow cytometry was used to detect TFH cell subpopulation based on programmed death 1(PD-1)and inducible co-stimulator(ICOS),and TFR cell subpopulation based on cluster determinant 45RA(CD45RA)and forkhead box protein P3(FoxP3).The levels of interleukin-10(IL-10),interleukin-17a(IL-17a),interleukin-21(IL-21),and transforming growth factor-β(TGF-β)in the plasma were measured,and changes in circulating B cell subsets and plasma IgG levels were also analyzed.The correlation between serum cytokeratin fragment antigen 21-1(CYFRA 21-1)levels and TFH,TFR,or B cell subpopulations was further explored.Results The TFR/TFH ratio increased significantly in NSCLC patients.The CD45RA^(+)FoxP3^(int) TFR subsets were increased,with their proportions increasing in stages Ⅱ to Ⅲ and decreasing in stage IV.PD-1^(+)ICOS+TFH cells showed a downward trend with increasing stages.Plasma IL-21 and TGF-β concentrations were increased in NSCLC patients compared with healthy controls.Plasmablasts,plasma IgG levels,and CD45RA^(+)FoxP3^(int) TFR cells showed similar trends.TFH numbers and plasmablasts were positively correlated with CYFRA 21-1 in stages Ⅰ-Ⅲ and negatively correlated with CYFRA 21-1 in stage IV.Conclusion Circulating TFH and TFR cell subpopulations and plasmablasts dynamically change in different stages of NSCLC,which is associated with serum CYFRA 21-1 levels and reflects disease progression.

Surface reconstruction of heterostructures in alkaline medium towards enhanced electrocatalytic hydrogen evolution

Constructing heterostructures has proved to be a successful strategy to fabricate electrocatalysts with high efficiency for water splitting.However,the structure evolution in alkaline hydrogen evolution reaction lacks investigation and the specific active center remains disputable.Herein,we take the well-designed Ni3S2@VO2heterostructures as a model to investigate the electrocatalytic activity and the surface reconstruction process of heterostructure catalysts in alkaline electrolyte.The Ni3S2@VO2heterostructures,with Ni3S2nanorods as the core and VO2nanoflakes as the shell,coupled with the high conductive Ni3S2,the hydrophilic VO2and modulated electronic structures at the interfaces,exhibited prominent activity and superior stability at various current densities.Further,the ex-situ characterizations confirmed that the surface reconstruction from Ni3S2@VO2into Ni3S2@amorphous-Ni（OH）2in alkaline media could optimize the water dissociation barrier and exposed large active area,thereby contributing to improved electrocatalytic performance.Our study not only introduces novel high-performance electrocatalysts for hydrogen evolution reaction（HER）,but also provides a new avenue for re-examining hetero-structural catalysts in alkaline solutions.

To assess whether a development strategy will be profitable enough,production forecasting is a crucial and difficult step in the process.The development history of other reservoirs in the same class tends to be studied to make predictions accurate.However,the permeability field,well patterns,and development regime must all be similar for two reservoirs to be considered in the same class.This results in very few available experiences from other reservoirs even though there is a lot of historical information on numerous reservoirs because it is difficult to find such similar reservoirs.This paper proposes a learn-to-learn method,which can better utilize a vast amount of historical data from various reservoirs.Intuitively,the proposed method first learns how to learn samples before directly learning rules in samples.Technically,by utilizing gradients from networks with independent parameters and copied structure in each class of reservoirs,the proposed network obtains the optimal shared initial parameters which are regarded as transferable information across different classes.Based on that,the network is able to predict future production indices for the target reservoir by only training with very limited samples collected from reservoirs in the same class.Two cases further demonstrate its superiority in accuracy to other widely-used network methods.

Tailored architecture of composite electrolyte for all-solid-state sodium batteries with superior rate performance and cycle life

Seeking for composite electrolytes reinforced all-solid-state sodium ion batteries with superior long lifespan and rate performance remains a great challenge.Here,a unique strategy to tailor the architecture of composite electrolyte via inserting polymer chains into a small quantity of sulfate sodium grafted C_(48)0H_(28)O_(32)Zr_(6)(UIOSNa)is proposed.The intimate contact between polymer segments and UIOSNa with limited pore size facilitates the anion immobilization of sodium salts and reduction of polymer crystallinity,thereby providing rapid ion conduction and reducing the adverse effect caused by the immigration of anions.The tNa+grafting of-SO_(3)Na groups on fillers allows the free movement of more sodium ions to further improve and ionic conductivity.Consequently,even with the low content of UIOSNa fillers,a high ionic conductivity of 6.62×10^(-4) S·cm^(-1) at 60℃ and a transference number of 0.67 for the special designed composite electrolyte are achieved.The assembled all-solid-state sodium cell exhibits a remarkable rate performance for 500 cycles with 95.96%capacity retention at a high current rate of 4 C.The corresponding pouch cell can stably work for 1000 cycles with 97.03%capacity retention at 1 C,which is superior to most of the reported composite electrolytes in the literature.

Construction of LiCl/LiF/LiZn hybrid SEI interface achieving high-performance sulfide-based all-solid-state lithium metal batteries

Sulfide-based all-solid-state lithium metal batteries(ASSLMBs)have received extensive attention due to their high energy density and high safety,while the poor interface stability between sulfide electrolyte and lithium metal anode limits their development.Hence,a hybrid SEI(LICl/Li F/Li Zn)was constructed at the interface between Li_(5.5)PS_(4.5)Cl_(1.5)sulfide electrolyte and lithium metal.The Li Cl and Li F interface phases with high interface energy effectively induce the uniform deposition of Li^(+)and reduce the overpotential of Li^(+)deposition,while the Li Zn alloy interface phase accelerates the diffusion of lithium ions.The synergistic effect of the above functional interface phases inhibits the growth of lithium dendrites and stabilizes the interface between the sulfide electrolyte and lithium metal.The hybrid SEI strategy exhibits excellent electrochemical performance on symmetric batteries and all-solid-state batteries.The symmetrical cell exhibits stable cycling performance over long duration over 500 h at 1.0 mA cm^(-2).Moreover,the LiNbO_(3)@NCM712/Li_(5.5)PS_(4.5)Cl_(1.5)/Li-10%Zn F_(2)battery exhibits excellent cycle stability at a high rate of 0.5 C,with a capacity retention rate of 76.4%after 350 cycles.

The High Altitude Detection of Astronomical Radiation(HADAR)experiment,which was constructed in Tibet,China,combines the wide-angle advantages of traditional EAS array detectors with the high-sensitivity advantages of focused Cherenkov detectors.Its objective is to observe transient sources such as gamma-ray bursts and the counterparts of gravitational waves.This study aims to utilize the latest AI technology to enhance the sensitivity of HADAR experiments.Training datasets and models with distinctive creativity were constructed by incorporating the relevant physical theories for various applications.These models can determine the type,energy,and direction of the incident particles after careful design.We obtained a background identification accuracy of 98.6%,a relative energy reconstruction error of 10.0%,and an angular resolution of 0.22°in a test dataset at 10 TeV.These findings demonstrate the significant potential for enhancing the precision and dependability of detector data analysis in astrophysical research.By using deep learning techniques,the HADAR experiment’s observational sensitivity to the Crab Nebula has surpassed that of MAGIC and H.E.S.S.at energies below 0.5 TeV and remains competitive with conventional narrow-field Cherenkov telescopes at higher energies.In addition,our experiment offers a new approach for dealing with strongly connected,scattered data.

Crystallinity engineering of carbon nitride protective coating for ultra-stable Zn metal anodes

Ineffective control of dendrite growth and side reactions on Zn anodes significantly retards commercialization of aqueous Zn-ion batteries.Unlike conventional interfacial modification strategies that are primarily focused on component optimization or microstructural tuning,herein,we propose a crystallinity engineering strategy by developing highly crystalline carbon nitride protective layers for Zn anodes through molten salt treatment.Interestingly,the highly ordered structure along with sufficient functional polar groups and pre-intercalated Kþendows the coating with high ionic conductivity,strong hydrophilicity,and accelerated ion diffusion kinetics.Theoretical calculations also confirm its enhanced Zn adsorption capability compared to commonly reported carbon nitride with amorphous or semi-crystalline structure and bare Zn.Benefiting from the aforementioned features,the as-synthesized protective layer enables a calendar lifespan of symmetric cells for 1100 h and outstanding stability of full cells with capacity retention of 91.5%after 1500 cycles.This work proposes a new conceptual strategy for Zn anode protection.

Development of a prognostic scoring system for hepatic vena cava Budd-Chiari syndrome with hepatocellular carcinoma

Background:Hepatocellular carcinoma(HCC)is a serious complication of hepatic vena cava Budd-Chiari syndrome(HVC-BCS)that significantly reduces the survival time of patients.Our study aimed to analyze the prognostic factors influencing the survival of HVC-BCS patients with HCC and to develop a prognostic scoring system.Methods:The clinical and follow-up data of 64 HVC-BCS patients with HCC who received invasive treatment at the First Affiliated Hospital of Zhengzhou University between January 2015 and December 2019 were retrospectively analyzed.Kaplan-Meier curves and log-rank tests were used to analyze the survival curve of patients and the difference in prognoses between the groups.Univariate and multivariate Cox regression analyses were performed to analyze the influence of biochemical,tumor,and etiological characteristics on the total survival time of patients,and a new prognostic scoring system was developed according to the regression coefficients of the independent predictors in the statistical model.The prediction efficiency was evaluated using the time-dependent receiver operating characteristics curve and concordance index.Results:Multivariate analysis showed that serum albumin level7 cm(HR=8.623,95%CI:3.771-19.715,P<0.001),and inferior vena cava stenosis(HR=3.612,95%CI:1.646-7.928,P=0.001)were independent predictors of survival.A prognostic scoring system was developed according to the above-mentioned independent predictors,and patients were classified into grades A,B,C and D.Significant differences in survival were found among the four groups.Conclusions:This study successfully developed a prognostic scoring system for HVC-BCS patients with HCC,which is helpful for clinical evaluation of patient prognosis.
