Cycling and Tai Chi Chuan exercises exert greater immunomodulatory effect on surface antigen expression of human hepatitis B virus

Background Both athletes with intensive exercise and aged people may have weakened immunity against virus infection. This study aimed to evaluate whether people undergoing aerobic exercises including competitive cyclists with moderate training （CMT） and middle-aged people practicing Tai Chi Chuan （TCC） exercise have higher immunity against hepatitis B virus than age-matched sedentary controls including college students （CSC） and middle-aged people （MSC）. Methods Human peripheral blood mononuclear cells from competitive cyclists and sedentary controls were stimulated by phytohemagglutinin （PHA） to prepare conditioned medium （MNC-CM） for the assessment of inhibitory effects on hepatitis B surface antigen （HBsAg） expression in human hepatoma Hep3B cells. Results The inhibitory effects on the relative HBsAg expression of CMT＇s and TCC＇s MNC-CM were greater than those of the controls. The CMT＇s MNC-CM prepared from 5 μg/ml PHA decreased HBsAg expression to 61.5%, whereas that of CSC remained at 83.8%. Similarly, this expression by treatment of TCC group＇ MNC-CM was 68.4% whereas that of MSC group was 84.3%. The levels of cytokines such as interferon-γ（IFN-γ）, tumor necrosis factor-α （TNF-α）, IFN-α and interleukin-1β （IL-1β） in the MNC-CM from the CMT and TCC groups were greater than those in the controls. Antibody neutralization of CMT＇s MNC-CM and addition of recombinant cytokines into CSC＇s MNC-CM indicated that IFN-γ, TNF-α and IFN-α had synergistic effects against HBsAg expression. Similar blocking effect was noted in TCC versus MSC groups. Conclusion These results suggest that the immunomodulatory response to suppress HBsAg expression in CMT and TCC with moderate aerobic exercise is greater than that in age-matched sedentary controls.

Background The change of anaerobic exercise abilities during and after a high-altitude expedition or hypoxic exposure is not well studied. To evaluate the effects of an extreme-altitude expedition on anaerobic performance, the lO-second supramaximal test and endocrine hormones were evaluated before and after an expedition to Peak Lenin. Methods Four subjects （3 male and 1 female, age （30.5＋16.5） years） were recruited into the study. Three sets of tests were performed, including a basic test at sea level and 20 days before first arrival at the base camp （3600 m）, a middle test done at day after returning from the summit to the base camp and the post test at the lOth day after return to the sea level. Both the supramaximal test, performed by a cycle ergometer, and body composition, performed by bioelectrical impedance analysis, were completed before the basic test and post test. The endocrine hormones including cortisol, growth hormone, testosterone, noradrenaline, adrenaline, dopamine, glucagon and β-endorphin were measured at all tests. Results Comparing the conditions before and after the expedition, the body measurement parameters were decreased alter the expedition, i.e., body weight （-4.22%, P〈O.05）, fat-free mass （-2.09%, P〈O.01） and body fat （-8.95%, P=0.172）. The peak power relative/body weight ratio （PP/BW） was similar （（9.70±1.97） vs （9.11±1.80） W/kg, P=-0.093）, while mean power/body weight ratio （MP/BW） was reduced significantly after the expedition （（9.14±1.77） vs （8.33±1.74） W/kg, P 〈0.05）. Peak power/fat-free mass （PP/FFM）, mean power/fat-free mass （MP/FFM） and fatigue index （FI） were significantly lower after the expedition （PP/FFM：（11.95±1.71） vs （10.99±1.59） W/kg, P 〈0.05; MP/FFM：（11.26±1.50） vs （10.04±1.55） W/kg, P〈O.O05; FI （85.55±4.17）% vs （77.25±4.40）%, P〈O.05）. Hormone assays showed a significant increase of noradrenaline （basic vs middle, P〈O.05） as well as decrease of adrenaline （P〈O.05）. Nicotiana tabacum and Nicotiana benthamiana are widely used models in plant biology research.However,genomic studies of these species have lagged.Here we report the chromosome-level reference genome assemblies for N.benthamiana and N.tabacum with an estimated 99.5%and 99.8%completeness,respec-tively.Sensitive transcription start and termination site sequencing methods were developed and used for accurate gene annotation in N.tabacum.Comparative analyses revealed evidence for the parental origins and chromosome structural changes,leading to hybrid genome formation of each species.Interestingly,theantiviral silencinggenesRDR1,RDR6,DCL2,DCL3,andAGO2were lost from one or both subgenomes in N.benthamiana,while both homeologs were kept in N.tabacum.Furthermore,the N.benthamiana genome encodes fewer immune receptors and signaling components than that of N.tabacum.These find-ings uncover possible reasons underlying the hypersusceptible nature of N.benthamiana.We developed the user-friendly Nicomics(http:/***/Nicomics/)web server to facilitate better use of Nicotiana genomic resources as well as gene structure and expression analyses.

Unveiling the secrets of diatom-mediated calcification: Implications for the biological pump

Siliceous diatoms are one of the most prominent actors in the oceans, and they account for approximately 40%of the primary production and particulate organic carbon export flux. It is believed that changes in carbon flux caused by variations in diatom distribution can lead to significant climate shifts. Although the fundamental pathways of diatom-driven carbon sequestration have long been established, there are no reports of CaCO3precipitation induced by marine diatom species. This manuscript introduces novel details regarding the enhancement of aragonite precipitation during photosynthesis in Skeletonema costatum in both artificial and natural seawater. Through direct measurements of cell surfaces via a pH microelectrode and zeta potential analyzer, it was determined that the diatom-mediated promotion of CaCO3precipitation is achieved through the creation of specific microenvironments with concentrated [CO32–] and [Ca2+] and/or the dehydrating effect of adsorbed Ca2+.Based on this mechanism, it is highly plausible that diatom-mediated calcification could occur in the oceans, an assertion that was supported by the significant deviation of total alkalinity (TA) from the conservative TA-salinity mixing line during a Skeletonema costatum bloom in the East China Sea and other similar occurrences. The newly discovered calcification pathway establishes a link between particulate inorganic and organic carbon flux and thus helps in the reassessment of marine carbon export fluxes and CO2sequestration efficiency. This discovery may have important ramifications for assessing marine carbon cycling and predicting the potential effects of future ocean acidification.

Dangling Ligand Enables an Iron Hexa-Coordinated Molecular Catalyst for Water Oxidation by Photocatalysis

Photocatalytic water oxidation reaction by transition metal complexes remains a challenge because of their poor stability under irradiation,especially for earth-abundant metal catalysts.In this regard,ironbased water oxidation catalysts are prone to hydrolysis and/or dissociate the ligands to form nanoparticles under a real catalytic condition.Herein,we describe a unique hexa-coordinated catalyst 1[Fe^(II)(Py_(3)tacn)Cl_(2)]and its reference 2[Fe^(II)(PhPy_(2)tacn)Cl_(2)]with a dangling pyridyl ligand and a phenyl group,respectively.We anticipated that the dynamically open and close coordination behaviors of the pyridyl ligand enabled balance of the reactivity and stability of catalyst 1.To our delight,the“open form”of catalyst 1 provided a free coordination site,and the“close form”guaranteed its molecular integrity,resulting in a water oxidation reaction with high efficiency and robustness.The turnover number and turnover frequency values of 2332 and 60 s^(−1)are the highest known to date among iron-based homogeneous water oxidation systems under visible light irradiation.

Lithium–oxygen battery with ultrahigh theoretical energy density is considered a highly competitive next-generation energy storage device,but its practical application is severely hindered by issues such as difficult decomposition of discharge products at present.Here,we have developed N-doped carbon anchored atomically dispersed Ru sites cathode catalyst with open hollow structure(h-RuNC)for Lithium–oxygen battery.On one hand,the abundance of atomically dispersed Ru sites can effectively catalyze the formation and decomposition of discharge products,thereby greatly enhancing the redox kinetics.On the other hand,the open hollow structure not only enhances the mass activity of atomically dispersed Ru sites but also improves the diffusion efficiency of catalytic molecules.Therefore,the excellent activity from atomically dispersed Ru sites and the enhanced diffusion from open hollow structure respectively improve the redox kinetics and cycling stability,ultimately achieving a high-performance lithium–oxygen battery.

Tendon injuries often lead to joint dysfunction due to the limited self-regeneration capacity of tendons.Repairing tendons is a major challenge for surgeons and imposes a signif-icant financial burden on society.Therefore,there is an urgent need to develop effective stra-tegies for repairing injured tendons.Tendon tissue engineering using hydrogels has emerged as a promising approach that has attracted considerable interest.Hydrogels possess excellent biocompatibility and biodegradability,enabling them to create an extracellular matrix-like growth environment for cells.They can also serve as a carrier for cells or other substances to accelerate tendon repair.In the past decade,numerous studies have made significant prog-ress in the preparation of hydrogel scaffolds for tendon healing.This review aims to provide an overview of recent research on the materials of hydrogel-based scaffolds used for tendon tis-sue engineering and discusses the delivery systems based on them.

Photodegradation of xylene isomers:Kinetics,mechanism,secondary pollutant formation potential and health risk evaluation

Photodegradation technology has been widely applied in the purification of industrial aromatic hydrocarbons.However,whether this technology efficiently removes the pollutants to prevent secondary pollution and health risk is still unclear.Here,the photodegradation processes of three xylenes were compared under designed reaction atmospheres and light sources.Xe lamp showed poor photodegradation ability toward xylenes,no matter in N_(2) or N_(2)+O_(2)system,while much higher photodegradation performance of xylenes were obtained under ultraviolet(UV)and vacuum ultraviolet(VUV)irradiation,especially in N_(2)+O_(2)+VUV system,where 97.9%of m-xylene,99.0%of o-xylene or 87.5%of p-xylene with the initial concentration of 860 mg/m^(3) was removed within 240 min.The xylenes underwent three processes of photo-isomerization,photodecomposition and photo-oxidation to produce intermediates of aromatics,alkanes and carbonyls.Among them,the photo-isomerization products showed the highest concentration percentage(e.g.,≥50%in o-xylene system),confirming that photo-isomerization reaction was the dominated photodegradation process of xylenes.Moreover,these isomerized products not only contributed about 97%and91%to the formation potential of O3(OFP)and secondary organic aerosols(SOAFP),but also displayed obvious non-carcinogenic risk,although one of photodecomposition product—benzene showed the highest occupational exposure risk.Therefore,the secondary pollution and health risks of photodegradation products of xylenes were non-ignorable,although the OFP,SOAFP and health risks of the generated products reduced at least 4.5 times in comparison with that of the degraded xylenes.The findings are helpful for the appropriate application of this technology in the purification of industrial organic waste gas.

Isotopic characteristics and source analysis of atmospheric ammonia during agricultural periods in the Xichuan area of the Danjiangkou Reservoir

Nitrogen deposition is an important means of exogenous nitrogen input in reservoir water.Agricultural activities around the reservoir lead to a sharp increase in the concentration of ammonia in the atmosphere,which poses a threat to the reservoir water body.Clarifying the contribution of agricultural ammonia release to atmospheric NH_(x)(gaseous NH_(3)and particulate NH_(4)^(+)),in the reservoir area can provide a theoretical foundation for local reactive nitrogen control.We collected atmospheric NH_(3)and NH_(4)^(+)samples during the agricultural periods and analyzed the isotopic characteristics of atmospheric NH_(x)and the contribution rates of different ammonia sources in the Xichuan area of the Danjiangkou Reservoir.The results showed that the initialδ~(15)N values of NH_(3)(-30.0‰to–7.2‰)and particulate NH_(4)^(+)(–33‰to+4.9‰for finer and coarser particles,respectively)are different,and their contribution ratios from dissimilar ammonia sources are also different,among which NH_(4)^(+)is more susceptible to meteorological factors.However,since the atmospheric NH_(x)in the Xichuan area is mainly gaseous NH_(3),the final sources of atmospheric ammonia nitrogen source depend on gaseous NH_(3).Agricultural sources(59%-74%)were the main NH_(3)sources in this area.Among them,the fertilizer use emission was dominant;it had the highest contribution rate in summer during the agricultural period and a more prominent impact in areas with less human interference.Reasonable regulation of the application of high-ammonia releasing fertilizer,especially during the agricultural period in summer,is an effective way to reduce the threat of atmospheric ammonia to water health.
