Prostate cancer(PCa)is one of the most common malignancies in males worldwide,and its development and progression involve the regulation of multiple metabolic pathways.Alterations in lipid metabolism affect the proliferation and metastatic capabilities of PCa cells.Cancer cells increase lipid synthesis and regulate fatty acid oxidation to meet their growth and energy demands.Similarly,changes occur in amino acid metabolism in PCa.Cancer cells exhibit an increased demand for specific amino acids,and they regulate amino acid transport and metabolic pathways to fulfill their proliferation and survival requirements.These changes are closely associated with disease progression and treatment response in PCa cells.Therefore,a comprehensive investigation of the metabolic characteristics of PCa is expected to offer novel insights and approaches for the early diagnosis and treatment of this disease.

A digital data-acquisition system based on XIA LLC products was used in a complex nuclear reaction experiment using radioactive ion beams.A flexible trigger system based on a field-programmable gate array(FPGA)parametrization was developed to adapt to different experimental sizes.A user-friendly interface was implemented,which allows converting script language expressions into FPGA internal control parameters.The proposed digital system can be combined with a conventional analog data acquisition system to provide more flexibility.The performance of the combined system was veri-fied using experimental data.

Creep constitutive model considering nonlinear creep degradation of fractured rock

Stability analysis of underground constructions requires a model study of rock masses’ long-term performance. Creep tests under different stress conditions was conducted on intact granite and granite samples fractured at 30° and 45° angles. The experimental results indicate that the steady creep strain rates of intact and fractured rock present an exponential increase trend with the increase of stress level. A nonlinear creep model is developed based on the experimental results, in which the initial damage caused by fracture together with the damage caused by constant load have been taken into consideration. The fitting analysis results indicated that the model proposed is more accurate at identifying the full creep regions in fractured granite, especially the accelerated stage of creep deformation. The least-square fit error of the proposed creep model is significantly lower than that of Nishihara model by almost an order of magnitude. An analysis of the effects of elastic modulus, viscosity coefficient, and damage factors on fractured rock strain rate and creep strain is conducted. If no consideration is given to the effects of the damage, the proposed nonlinear creep model can degenerate into to the classical Nishihara model.

Small extracellular vesicles from hypoxia-preconditioned bone marrow mesenchymal stem cells attenuate spinal cord injury via miR-146a-5p-mediated regulation of macrophage polarization

Spinal cord injury is a disabling condition with limited treatment options.Multiple studies have provided evidence suggesting that small extracellular vesicles(SEVs)secreted by bone marrow mesenchymal stem cells(MSCs)help mediate the beneficial effects conferred by MSC transplantation following spinal cord injury.Strikingly,hypoxia-preconditioned bone marrow mesenchymal stem cell-derived SEVs(HSEVs)exhibit increased therapeutic potency.We thus explored the role of HSEVs in macrophage immune regulation after spinal cord injury in rats and their significance in spinal cord repair.SEVs or HSEVs were isolated from bone marrow MSC supernatants by density gradient ultracentrifugation.HSEV administration to rats via tail vein injection after spinal cord injury reduced the lesion area and attenuated spinal cord inflammation.HSEVs regulate macrophage polarization towards the M2 phenotype in vivo and in vitro.Micro RNA sequencing and bioinformatics analyses of SEVs and HSEVs revealed that mi R-146a-5p is a potent mediator of macrophage polarization that targets interleukin-1 receptor-associated kinase 1.Reducing mi R-146a-5p expression in HSEVs partially attenuated macrophage polarization.Our data suggest that HSEVs attenuate spinal cord inflammation and injury in rats by transporting mi R-146a-5p,which alters macrophage polarization.This study provides new insights into the application of HSEVs as a therapeutic tool for spinal cord injury.

The repair and regeneration of bone defects are highly challenging orthopedic problems.Recently,Mg-based implants have gained popularity due to their unique biodegradation and elastic modulus similar to that of human bone.The aim of our study is to develop a magnesium alloy with a controllable degradation that can closely match bone tissue to help injuries heal in vivo and avoid cytotoxicity caused by a sudden increase in ion concentration.In this study,we prepared and modified Mg-3Zn,Mg-3Zn-1Y,and Mg-2Zn-1Mn by hot extrusion,and used Mg-2.5Y-2.5Nd was as a control.We then investigated the effect of additions of Y and Mn on alloys'properties.Our results show that Mn and Y can improve not only compression strength but also corrosion resistance.The alloy Mg-2Zn-1Mn demonstrated good cytocompatibility in vitro,and for this reason we selected it for implantation in vivo.The degraded Mg-2Zn-1Mn implanted a bone defect area did not cause obvious rejection and inflammatory reaction,and the degradation products left no signs of damage to the heart,liver,kidney,or brain.Furthermore,we find that Mg-2Zn-1Mn can promote an osteoinductive response in vivo and the formation of bone regeneration.

Tendon injuries often lead to joint dysfunction due to the limited self-regeneration capacity of tendons.Repairing tendons is a major challenge for surgeons and imposes a signif-icant financial burden on society.Therefore,there is an urgent need to develop effective stra-tegies for repairing injured tendons.Tendon tissue engineering using hydrogels has emerged as a promising approach that has attracted considerable interest.Hydrogels possess excellent biocompatibility and biodegradability,enabling them to create an extracellular matrix-like growth environment for cells.They can also serve as a carrier for cells or other substances to accelerate tendon repair.In the past decade,numerous studies have made significant prog-ress in the preparation of hydrogel scaffolds for tendon healing.This review aims to provide an overview of recent research on the materials of hydrogel-based scaffolds used for tendon tis-sue engineering and discusses the delivery systems based on them.

Major volcanic eruptions(MVEs)have attracted increasing attention from the scientific community.Previous studies have explored the climatic impact of MVEs over the past two millennia.However,proxy-based reconstructions and climate model simulations indicate divergent responses of global and China’s regional climates to MVEs.Here,we used multiple data from observations,reconstructions,simulations,and assimilations to summarize the historical facts of MVEs,the characteristics and mechanisms of their climatic impact,and directions for future research.We reviewed volcanic datasets and determined intensive MVE periods;these periods corresponded to the years 530–700,1200‒1460,and 1600‒1840 CE.After tropical MVEs,a substantial cooling effect is observed throughout the globe and China on the interannual-interdecadal time scales but an inconsistent cooling magnitude is detected between reconstructions and simulations.In the first summer after tropical MVEs,a decrease in global and monsoonal precipitation is observed.In reconstructions and simulations,an increased precipitation is seen for the Yangtze River Basin,while large uncertainties in precipitation changes are present for other regions of China.Decadal drought can be induced by frequent eruptions and volcanism superimposed on low solar irradiation and internal variability.MVEs affect climate directly through the radiative effect and indirectly by modulating internal variability,such as the El Niño‒Southern Oscillation(ENSO)and Atlantic Multidecadal Oscillation(AMO).However,changes in the phase,amplitude,and periodicity of ENSO and AMO after MVEs and the associated mechanisms remain controversial,which could account for model-reconstruction disagreements.Moreover,other internal variability,uncertainties in reconstruction methods and aerosol‒climate models,and climate background may also induce model-reconstruction disagreements.Knowledge gaps and directions for future research are also discussed.

目的探讨踝关节镜辅助下经跗骨窦入路内固定治疗SandersⅡ、Ⅲ型跟骨骨折的疗效。方法采用踝关节镜辅助下经跗骨窦入路内固定治疗23例跟骨骨折患者。记录骨折愈合情况、Bohler角、Gissane角、跟骨高度及宽度、踝关节活动度、术后并发症发生情况,采用AOFAS踝-后足评分评价踝关节功能。结果患者均获得随访,时间10~16个月。切口均一期愈合,未发生切口感染或坏死、骨折不愈合等并发症。术后10~12周骨折线均模糊,骨痂形成,骨折愈合。Bohler角、Gissane角、跟骨高度及宽度术后6个月均较术前改善(P<0.05)。末次随访时,患者均能完全负重行走,步态正常;踝关节背伸15°~25°,跖屈30°~45°;采用AOFAS踝-后足评分评价功能:优17例,良5例,可1例,优良率22/23。结论踝关节镜辅助下经跗骨窦入路内固定治疗SandersⅡ、Ⅲ型跟骨骨折,骨折复位精准,术中透视次数少、创伤小,术后足部功能恢复满意、并发症发生率低。

Background:Cancer-related fatigue(CRF)is a common and debilitating symptom experienced by patients with advanced-stage cancer,especially those undergoing antitumor therapy.This study aimed to evaluate the efficacy and safety of Renshenguben(RSGB)oral solution,a ginseng-based traditional Chinese medicine,in alleviating CRF in patients with advanced hepatocellular carcinoma(HCC)receiving antitumor treatment.Methods:In this prospective,open-label,controlled,multicenter study,patients with advanced HCC at BCLC stage C and a brief fatigue inventory(BFI)score of≥4 were enrolled.Participants were assigned to the RSGB group(RSGB,10 mL twice daily)or the control group(with supportive care).Primary and secondary endpoints were the change in multidimensional fatigue inventory(MFI)score,and BFI and functional assessment of cancer therapy-hepatobiliary(FACT-Hep)scores at weeks 4 and 8 after enrollment.Adverse events(AEs)and toxicities were assessed.Results:A total of 409 participants were enrolled,with 206 assigned to the RSGB group.At week 4,there was a trend towards improvement,but the differences were not statistically significant.At week 8,the RSGB group exhibited a significantly lower MFI score(P<0.05)compared to the control group,indicating improved fatigue levels.Additionally,the RSGB group showed significantly greater decrease in BFI and FACT-Hep scores at week 8(P<0.05).Subgroup analyses among patients receiving various antitumor treatments showed similar results.Multivariate linear regression analyses revealed that the RSGB group experienced a significantly substantial decrease in MFI,BFI,and FACT-Hep scores at week 8.No serious drug-related AEs or toxicities were observed.Conclusions:RSGB oral solution effectively reduced CRF in patients with advanced HCC undergoing antitumor therapy over an eight-week period,with no discernible toxicities.These findings support the potential of RSGB oral solution as an adjunctive treatment for managing CRF in this patient population.
