Utilizing engineered extracellular vesicles as delivery vectors in the management of ischemic stroke:a special outlook on mitochondrial delivery

Ischemic stroke is a secondary cause of mortality worldwide,imposing considerable medical and economic burdens on *** vesicles,serving as natural nanocarriers for drug delivery,exhibit excellent biocompatibility in vivo and have significant advantages in the management of ischemic ***,the uncertain distribution and rapid clearance of extracellular vesicles impede their delivery *** utilizing membrane decoration or by encapsulating therapeutic cargo within extracellular vesicles,their delivery efficacy may be greatly ***,previous studies have indicated that microvesicles,a subset of large-sized extracellular vesicles,can transport mitochondria to neighboring cells,thereby aiding in the restoration of mitochondrial function post-ischemic *** extracellular vesicles have also demonstrated the capability to transfer mitochondrial components,such as proteins or deoxyribonucleic acid,or their sub-components,for extracellular vesicle-based ischemic stroke *** this review,we undertake a comparative analysis of the isolation techniques employed for extracellular vesicles and present an overview of the current dominant extracellular vesicle modification *** the complex facets of treating ischemic stroke,we also delineate various extracellular vesicle modification approaches which are suited to different facets of the treatment ***,given the burgeoning interest in mitochondrial delivery,we delved into the feasibility and existing research findings on the transportation of mitochondrial fractions or intact mitochondria through small extracellular vesicles and microvesicles to offer a fresh perspective on ischemic stroke therapy.

Explaining the dynamics of the sub-relativistic electron third belt in the Earth's radiation belts by using medium Earth orbit satellite observations

The Earth's electron radiation belts typically exhibit a two-belt ***,observations from the Van Allen Probes revealed the existence of a three-belt *** structure consists of an inner belt,a slot region,a remnant belt,a“second slot,”and a new outer belt(or the“third belt”).The formation of the structure involves both the partial loss of the original outer belts and the formation of the third *** processes are likely associated with radial diffusion induced by ultra-low-frequency(ULF)*** this study,we mainly analyzed electron flux data from medium Earth orbit(MEO)navigation satellites M17–M19 to supplement the observational evidence for the sub-relativistic electron(~100–500 keV)three-belt *** of substorm injections and ULF waves during the three-belt event was identified,suggesting they played significant roles in the formation and evolution of the third *** injections may introduce new electron populations to the third belt,whereas ULF waves may influence the evolution of the third belt through radial *** the end of the three-belt event,the compression of the magnetosphere by shocks may lead to the dropout of the third belt because of the magnetopause shadowing effect and outward radial diffusion,ultimately disrupting the three-belt *** study provides more evidence for the presence of a sub-relativistic electron three-belt structure and offers an analysis of the evolutionary mechanisms of the third belt,which may contribute to a comprehensive understanding of the electron three-belt structure in the radiation belts.

High-performance porous carbon foams via catalytic pyrolysis of modified isocyanate-based polyimide foams for electromagnetic shielding

Porous carbon skeletons(PCSs)derived from isocyanate-based aromatic polyimide foams(PIFs)by high-temperature pyrolysis are very promising in the fabrication of high-performance polymer composite foams for electromagnetic interference(EMI)shielding due to their efficient conductive networks and facile preparation ***,severe volumetric shrinkage and low graphitization degree is not conducive to enhancing the shielding efficiency of the ***,ferric acetylacetonate and carbon-nanotube coating have been introduced in isocyanate-based PIFs to greatly suppress the serious shrinkage during pyrolysis and improve the graphitization degree of the final carbon foams through the Fe-catalytic graphitization process,thereby endowing them with better EMI-shielding performance even at lower pyrolysis temperature compared to the control ***,compressible polydimethylsiloxane(PDMS)composite foams with the as-prepared carbon foams as prefabricated PCSs have also been fabricated,which could provide not only stable shielding effectiveness(SE)performance even after a thousand compressions,but also multiple functions of Joule heating,thermal insulation and infrared *** study offers a feasible route to prepare high-performance PCSs in a more energy-efficient manner via PIF pyrolysis,which is very promising in the manufacture of multifunctional conductive polymer composite foams.

Insight into synergistic antioxidation mechanisms of butyl hydroxyanisole with common synthetic antioxidants

Butyl hydroxyanisole(BHA) is usually blended with other synthetic antioxidants to improve the antioxidative property due to synergistic antioxidation. However, the synergistic antioxidation mechanisms of BHA with synergists have not been revealed yet. Thus, the antioxidation of BHA with butylated hydroxytoluene(BHT), tert-butylhydroquinone(TBHQ), or propyl gallate(PG) was investigated in the 2,2-azobis(2-amidino-propane) dihydrochloride oxidizing system. The contents of BHA, BHT, TBHQ,PG, and transformation products were measured by high-performance liquid chromatography. Transformation products were identified with liquid chromatography-mass spectrometry and gas chromatography–mass spectrometry. Results showed that synergistic antioxidation occurred between BHA and BHT, TBHQ, or PG. The synergistic antioxidation effect of BHA and BHT was attributed to the regeneration of BHA by BHT. Transformation products of BHA and BHT(compounds 6 and 7, dimers of BHA and BHT) had little contribution due to the relatively low content(<0.6%). The synergistic antioxidation effect of BHA and TBHQ or BHA and PG was attributed to the protective mechanism of TBHQ or PG on BHA. No transformation products were detected of BHA and TBHQ. Transformation products of BHA and PG(compounds 9 and 10, dimers of BHA and PG) had limited contribution due to the relatively low percentage(<7%). Therefore, BHA and BHT performed competitive antioxidation,while BHA and TBHQ or PG performed protective antioxidation.

Validation of neuropathic pain assessment tools among Chinese patients with painful diabetic peripheral neuropathy

Objective:This study aims to evaluate the reliability and validity of neuropathic pain assessment tools among Chinese patients with painful diabetic peripheral neuropathy(PDPN).Methods:One hundred patients with PDPN and 70 patients with non-neuropathic pain were recruited from five grade III general hospitals in *** was assessed using the Leeds Assessment of Neuropathic Symptoms and Signs(LANSS),Douleur Neuropathique 4 questionnaire(DN4),and Brief Pain Inventory for Painful Diabetic Peripheral Neuropathy(BPI-DPN).Reliability was evaluated by internal consistency of the Cronbach's a coefficient and Guttman *** validity was analyzed by factor analysis and Spearman correlation *** and specificity were also ***:The Cronbach's a coefficients of the LANSS,DN4,and BPI-DPN were 0.735,0.750,and 0.898,*** Guttman split-half coefficients of the LANSS,DN4,and BPIDPN were 0.660,0.726,and 0.849,*** cumulative contributions of the LANSS,DN4,and BPI-DPN to the total variance were 61.945%,57.010%,and 66.056%,*** items of the LANSS,DN4,and BPI-DPN presented high factorial loads,ranging from 0.387 to 0.841,0.137 to 0.948,and 0.487 to 0.953,*** LANSS and DN4 exhibited sensitivities of 58.0%and 82.7%,respectively,and specificity of 97.1%.Conclusions:The LANSS or DN4 can be used to detect neuropathic pain in Chinese patients with *** BPI-DPN can be employed to monitor the effectiveness of pain intervention.

Donkey whey proteins ameliorate dextran sulfate sodium-induced ulcerative colitis in mice by downregulating the S100A8-TRAF6-NF-κB axis-mediated inflammatory response

Donkey milk has a variety of physiological functions, including antibacterial and anti-inflammatory. Donkey whey proteins(DWPs), as the main functional component in donkey milk, its inhibitory effect on colitis is still unclear. In this study, the inhibitory effect and potential mechanism of DWPs on dextran sulfate sodium(DSS)-induced colitis were investigated. Firstly, the DWPs and bovine milk whey proteins(BWPs)were characterized using proteomics. Then, we administered DWPs and BWPs to mice with colitis via oral gavage. The results of immunohistochemistry and flow cytometry indicated that DWPs increased T regulatory cell accumulation and increased the abundance of the cluster of differentiation 205+(CD205+)macrophages compared to those with BWPs and in model groups. In addition, DWPs exhibited a more remarkable ability to inhibit pro-inflammatory proteins(S100A8, TRAF6, and NF-κB)expression and inflammatory secretion than BWPs. In addition, DWPs significantly decreased NF-κB and CD86 levels more than BWPs or the negative control in both LPS-stimulated human peripheral blood mononuclear cells or cell lines. These findings indicate that DWPs comprise a promising anti-colitis functional food, and this work has established a foundation for future research on these compounds.

The earliest paper currencies in existence in China were handcrafted during the Yuan *** currencies were scientifically excavated from different ruins or tombs,whereas scientific analyses of the papers are *** study used optical and scanning electron microscopy to examine the fibers collected in Yuan dynasty paper currencies in conjunction with the Herzberg staining *** differences in circulation period,paper fibers in both Zhi Yuan Tong Xing Bao Chao(two Guan)and Zhong Tong Yuan Bao Jiao Chao(one Guan and 500 Wen,issued in Zhi Zheng period)were identified as similar papermaking materials,bast fibers of mulberry *** results indicate that mulberry bark,a durable papermaking material used since ancient times,was mainly utilized as a raw material in these Yuan dynasty paper *** fiber identification work solved the critical problem of papermaking material in the Yuan dynasty paper currency and provided important information for conserving these precious cultural relics.

In-situ oxidized tungsten disulfide nanosheets achieve ultrafast photocatalytic extraction of uranium through hydroxyl-mediated binding and reduction

Photoreduction of hexavalent uranium(U(VI))by semiconductor provides a novel and effective avenue for uranium ***,the traditional metal oxide and sulfide semiconductors suffer from the lack of confinement sites to U(VI),which resulted in the long period(~1 h)to achieve a high U(VI)extraction efficiency of>90%.Herein,we successfully constructed WS2 nanosheets and created in-situ oxidized domains on the surfaces(O-WS_(2))to promote the uranium extraction and the corresponding removal *** this system,the O_(7.7-)WS_(2) nanosheets exhibited a considerable U(VI)extraction efficiency of>90%within 20 min in 8 mg·L^(–1) U(VI)-containing solution,which represented the highly efficient U(VI)removal *** 200 mg·L^(–1) U(VI)-containing solution,the O7.7-WS2 nanosheets exhibited an extraction capacity of 652.4 mg·g^(-1).The mechanism study revealed that the oxidized surface tended to trap hydrogen atom and in-situ form hydroxyl groups in defect *** by a series of experiment,such as kinetic isotope effect,1H nuclear magnetic resonance(NMR)spectra,and X-ray absorption near-edge structure(XANES)spectra,the in-situ formed hydroxyl groups participated in the uranium reduction,which dramatically enhanced uranium extraction kinetics and efficiency.

Biomass-based aligned carbon networks with double-layer construction for tunable electromagnetic shielding with ultra-low reflectivity

Nowadays,carbon frameworks derived from natural biomaterials have attracted extensive attention for electromagnetic interference(EMI)shielding due to their renewability and ***,it is critical and challenging to achieve effective regulation of shielding effectiveness(SE)as well as weaken the strong EM reflection of highly conductive biomass-based carbon ***,commercial cotton pads with oriented structure were selected as carbonaceous precursor to fabricate aligned carbon networks by pyrolysis,and the EMI SE of the samples with increased temperature of 800-1000℃ can be accurately controlled in the effective range of~21.7-29.1,~27.7-37.1 and~32.7-43.3 d B with high reflection coefficient of>0.8 by changing the cross-angle between the electric-field direction of incident EM waves and the fiber-orientation direction due to the occurrence of opposite internal electric ***,the further construction of Salisbury absorber-liked double-layer structure could result in an ultralow reflection coefficient of only~0.06 but enhanced SE variation range up to~38.7-49.3 d B during the adjustment of cross-angle,possibly due to the destructive interference of EM waves in the double-layer carbon *** work would provide a simple and effective way for constructing high-performance biomass carbon materials with adjustable EMI shielding and ultra-low reflectivity.

Treatment of hemophilic arthropathy by immunomodulatory extracellular vesicle delivered by liposome hybrid nanoparticles

In individuals afflicted with hemophilia,characterized by a deficiency of coagulation factor VIII(FVIII),the occurrence of spontaneous recurrent intra-articular hemorrhage precipitates the emergence of hemophilic arthropathy(HA).Although clotting factor replacement therapy reduces joint bleeding clinically,clotting factors need to be injected frequently due to the rapid diffusion of the ***,a novel drug delivery approach may be developed to improve the drug ***-derived extracellular vesicles(PEVs)are known to possess anti-inflammatory and hemostatic properties and could be used as a potential HA *** this study,we constructed a PEV-LS@FVIII nanotherapeutic system by combining thioketal(TK),liposomes(LS),and FVIII to form the LS@FVIII complexes,and then hybridizing PEV with LS@*** results demonstrated that PEVLS@FVIII could efficiently facilitate FVIII delivery and specifically target the injured knee *** in vitro and in vivo studies showed a reduction in the M1 phenotype of macrophages and an enhancement of the M2 phenotype,compared to FVIII free ***,PEV-LS@FVIII appeared to alleviate HA-induced cartilage *** conclusion,our findings demonstrate that PEV-LS@FVIII could delay the progression of HA by targeting bleeding joints,modulating macrophage polarization to suppress inflammation,and mitigating cartilage damage.
