Transdermal treatment for malignant melanoma by aptamer-modified tetrahedral framework nucleic acid delivery of vemurafenib

Melanoma is one of the most malignant skin tumors, whose high invasion is generally associated with BRAF gene mutation. Although new chemotherapeutic drugs, such as vemurafenib, have been developed to inhibit the growth of melanoma, these drugs are usually administered intravenously or orally, resulting in toxic side effects on major tissues and organs. Tetrahedral framework nucleic acids(tFNAs) are a novel type of DNA nanostructures with excellent biocompatibility and versatility which have been proven to penetrate through skin barrier with ease. In this study, we prepared t FNAs with vemurafenib and connected DNA aptamer AS1411 at the apex of t FNAs(AS1411-tFNAs/vemurafenib). On one hand, AS1411-tFNAs/vemurafenib could kill melanoma cells by blocking the mutated BRAF gene in vitro. Compared with free vemurafenib, AS1411-tFNAs/vemurafenib had no obvious toxicity to normal cells. On the other hand,AS1411-tFNAs could transfer vemurafenib to cross through the skin barrier and permeate into tumor tissues. In vivo, transdermal delivery of AS1411-t FNAs/vemurafenib could inhibit the growth of human A375melanoma, whose inhibiting effect was stronger than intravenous administration of vemurafenib. These results demonstrated the application prospects of tFNAs combined with chemotherapeutic drugs in skin tumors.

The vaccine-induced innate immune response is essential for the generation of an antibody *** date,how Ad5-vectored vaccines are influenced by preexisting anti-Ad5 antibodies during activation of the early immune response remains ***,we investigated the specific alterations in GP1,2-specific IgG-related elements of the early immune response at the genetic,molecular,and cellular levels on days 0,1,3,and 7 after Ad5-EBOV *** a causal multiomics analysis,distinct early immune responses associated with GP1,2-specific IgG were observed in Ad5-EBOV recipients with a low level of preexisting anti-Ad5 *** study revealed the correlates of the Ad5-EBOV-induced IgG response and provided mechanistic evidence for overcoming preexisting Ad5 immunity during the administration of Ad5-vectored vaccines.

Tetrabromobisphenol A and hexabromocyclododecane in sediments from the Zhujiang(Pearl) River Estuary and South China Sea

Marine sediments collected from the Zhujiang(Pearl) River Estuary(ZRE) and South China Sea(SCS) were utilized to study the occurrence and spatial distribution of tetrabromobisphenol A(TBBPA) and hexabromocyclododecane(HBCDD).The levels of TBBPA and HBCDD in sediments ranged from not detected(nd) to 6.14 ng/g dry weight(dw) and nd to 0.42 ng/g *** concentrations in marine sediments were substantially higher than *** concentrations of TBBPA and HBCDD in the ZRE sediments were significantly greater than those in the SCS.α-HBCDD(48.7%) and γ-HBCDD(46.2%) were the two main diastereoisomers of HBCDD in sediments from the ZRE,with minor contribution of β-HBCDD(5.1%).HBCDD were only found in one sample from the northern *** enantiomeric fraction of α-HBCDD in sediments from the ZRE was obviously greater than 0.5,indicating an accumulation of(+)-α-*** enantiomers of HBCDD were not measured in sediments from the *** work highlighted the environmental behaviors of TBBPA and HBCDD in marine sediments.

The Naoli River Basin(NRB),a pivotal agricultural production area in China,is poised to undergo substantial impacts on water resources due to projected climate and land use/cover(LULC)*** its significance in the context of China’s expanding farmland construction in the NRB,there exists limited research on the potential repercussions of future shifts in runoff,soil water content(SWC),and evapotranspiration(ET)on crop productivity and water availability(both in terms of quantity and timing).This study employs future LULC maps and an ensemble of ten CMIP6 Global Climate Models(GCMs)across three scenarios to drive the well-calibrated distributed hydrological model,*** objective of present study is aimed on projecting hydrological consequences under climate and land use/land cover changes in near-term(2026–2050),middle-term(2051–2075),and far-term(2076–2100)future in comparison to the baseline period of 1990–*** consistently indicate an increase trend in annual average ET,runoff,and SWC in the NRB across all three future periods under the three SSP *** changes emerge as the primary driver influencing regional hydrological processes in the near ***,under high-emission scenarios,monthly runoff and SWC are projected to significantly increase in March but decrease in April during the middle and far future periods compared to the *** shift is attributed to the anticipated warming of winter and spring,leading to a transition in peak snowmelt from April to ***,the expansion of cropland intensifies crop evapotranspiration demand,potentially exacerbating water stress during the early stages of crop growth in *** findings underscore the importance of addressing the substantial impacts of climate change and land use planning on regional water cycling *** planning to mitigate water shortages during the initial stage of future crop growth is crucial for ensuring food security and managing wat

Preparation and characteristic study of Schottky diodes based on Ga_(2)O_(3)thin films

This study uses atomic layer deposition(ALD)to grow Ga_(2)O_(3)films on SiO_(2)substrates and investigates the influence of film thickness and annealing temperature on film *** diode devices are fabricated based on the grown Ga_(2)O_(3)films,and the effects of annealing temperature,electrode size,and electrode spacing on the electrical characteristics of the devices are *** results show that as the film thickness increases,the breakdown voltage of the fabricated devices also increases.A Schottky diode with a thickness of 240 nm can achieve a reverse breakdown voltage of 300 *** film quality significantly improves as the annealing temperature of the film *** a voltage of 5 V,the current of the film annealed at 900℃is 64 times that of the film annealed at 700℃.The optimum annealing temperature for Ohmic contact electrodes is 450℃.At 550℃,the Ohmic contact metal tends to burn,and the performance of the device is *** the electrode spacing increases the forward current of the device but decreases the reverse breakdown *** the Schottky contact electrode size increases the forward current,but the change is not significant,and there is no significant change in the reverse breakdown *** device also performs well at high temperatures,with a reverse breakdown voltage of 220 V at 125℃.

Andrias davidianus(Chinese giant salamander,CGS)is the largest and oldest extant amphibian species in the world and is a source of prospective functional food in ***,the progress of functional peptides mining was slow due to lack of reference genome and protein sequence *** this study,we illustrated full-length transcriptome sequencing to interpret the proteome of CGS meat and obtain 10703 coding DNA *** functional annotation and amino acid composition analysis,we have discovered various genes related to signal transduction,and 16 genes related to *** have also found vast variety of functional peptides through protein coding sequence(CDS)analysis by comparing the data obtained with the functional peptide ***-Pro-Ile predicted by the CDS analysis was released from the CGS meat through enzymatic hydrolysis,suggesting that our approach is *** study suggested that transcriptomic analysis can be used as a reference to guide polypeptide mining in CGS meat,thereby providing a powerful mining strategy for the bioresources with unknown genomic and proteomic sequences.

MOF-derived 1D CGO Cathode for Efficient Solid Oxide Electrolysis Cells

Solid oxide electrolysis cells(SOECs)provide a promising way for converting renewable energy into chemical ***,NiO/CGO(nickel-gadolinium doped ceria)cermet has shown its excellent properties in ionic and electronic conductivity under reducing ***,we developed a novel 1D NiO/CGO cathode through a cerium metal-organic framework(MOF)derived *** cathode’s 1D nanostructure integrated with a microchannel scaffold facilitates enhanced mass transport,providing vertically aligned pathways for CO_(2)and H_(2)O ***,the 1D framework increases the number of interfacial sites and reduces ion diffusion distances,thereby simplifying electron/ion ***,this advanced cathode achieved a significant breakthrough in SOEC performance,maintaining efficient CO_(2)and H_(2)O electrolysis at an extraordinary current density of 1.41 A/cm^(2)at 1.5 V and excellent stability over 24 h at 850℃.The enhanced performance of this newly developed cathode not only achieves a remarkable 100%improvement compared to those of NiO/CGO cathodes with varying geometrical configurations but also surpasses those of commercial NiO/CGO catalysts by an outstanding 40%when tested under identical *** development of the 1D NiO/CGO enhances the efficiency and durability of ceramic cathodes for CO_(2)and H_(2)O co-electrolysis in SOECs and improves the scalability and effectiveness of SOECs in renewable energy applications.

Lysine-specific demethylase 4 A(KDM4A,also named JMJD2A,KIA0677,or JHDM3A)is a demethylase that can remove methyl groups from histones H3K9me2/3,H3K36me2/3,and H1.4K26me2/*** evidence suggests that KDM4A is not only involved in body homeostasis(such as cell proliferation,migration and differentiation,and tissue development)but also associated with multiple human diseases,especially ***,an increasing number of studies have shown that pharmacological inhibition of KDM4A significantly attenuates tumor progression in vitro and in vivo in a range of solid tumors and acute myeloid *** there are several reviews on the roles of the KDM4 subfamily in cancer development and therapy,all of them only briefly introduce the roles of KDM4A in cancer without systematically summarizing the specific mechanisms of KDM4A in various physiological and pathological processes,especially in tumorigenesis,which greatly limits advances in the understanding of the roles of KDM4A in a variety of cancers,discovering targeted selective KDM4A inhibitors,and exploring the adaptive profiles of KDM4A ***,we present the structure and functions of KDM4A,simply outline the functions of KDM4A in homeostasis and non-cancer diseases,summarize the role of KDM4A and its distinct target genes in the development of a variety of cancers,systematically classify KDM4A inhibitors,summarize the difficulties encountered in the research of KDM4A and the discovery of related drugs,and provide the corresponding solutions,which would contribute to understanding the recent research trends on KDM4A and advancing the progression of KDM4A as a drug target in cancer therapy.
