miR-34c inhibits proliferation and enhances apoptosis in immature porcine Sertoli cells by targeting the SMAD7 gene

MicroRNAs(miRNAs) are implicated in swine spermatogenesis via their regulations of cell proliferation, apoptosis, and differentiation. Recent studies indicated that miR-34 c is indispensable in the late steps of spermatogenesis. However, whether miR-34 c plays similar important roles in immature porcine Sertoli cells remain unknown. In the present study, we conducted two experiments using a completely randomised design to study the function roles of miR-34 c. The results from experiment I demonstrated that the relative expression level of miR-34 c in swine testicular tissues increased(P=0.0017) quadratically with increasing age, while the relative expression level of SMAD family member 7(SMAD7) decreased(P=0.0009) with curve. Furthermore, miR-34 c expression levels showed a significant negative correlation(P=0.013) with SMAD7 gene expression levels. The results from experiment II indicated that miR-34 c directly targets the SMAD7 gene using a luciferase reporter assay, and suppresses(P<0.05) SMAD7 mRNA and protein expressions in immature porcine Sertoli cells. Overexpression of miR-34 c inhibited(P<0.05) proliferation and enhanced(P<0.05) apoptosis in the immature porcine Sertoli cells, which was supported by the results from the Cell Counting Kit-8(CCK-8) assay, the 5-Ethynyl-2′-deoxyuridine(EdU) assay, and the Annexin V-FITC/PI staining assay. Furthermore, knockdown of SMAD7 via small interfering RNA(siR NA) gave a similar result. It is concluded that miR-34 c inhibits proliferation and enhances apoptosis in immature porcine Sertoli cells by targeting the SMAD7 gene.

Electroreduction of CO_(2)into chemicals is of great importance in the global carbon *** noble-metal based catalysts and single-atom catalysts(SACs)are known to be active for CO_(2)electroreduction reaction(CO_(2)RR),the high cost of noble-metal and the lack of effective synthesis approaches to prepare SACs have tremendously hindered the ***-metal doped carbon materials have attracted great interest because of their reasonable cost,chemical stability and excellent electrical ***,the design and fabrication of highly efficient non-metal doped carbon electrocatalysts for CO_(2)RR to meet industry demands still remains a big ***,triphenylphosphine@covalent triazine frameworks(CTFs)composites were employed as precursors to fabricate N,P dual-doped porous carbon catalysts PCTF-X-Y(X represents the carbonization temperature,and Y represents the mass ratio of CTF to triphenylphosphine)for CO_(2)*** to the high specific surface areas and synergistic effect between N and P,the obtained PCTF-1000-5 exhibited high selectivity for CO production up to 84.3%at–0.7 V versus the reversible hydrogen electrode(***)and long-term durability over 16 h,which are better than the reported N,P dual-doped carbon catalysts in aqueous *** work provides a new way to design and fabricate non-metal catalysts for electrocatalysis.

Calcium fluoride(CaF_(2))is an ideal adsorbent for the dehydration of gaseous hydrogen fluoride(HF)containing water *** this work,a novel CaF_(2)absorbent,spherical CaF_(2)nanoaggregates(NAs)with a closely packed structure,was proposed and efficiently fabricated by spray drying *** the building blocks of CaF_(2)NAs,the CaF_(2)nanoparticles(NPs)were prepared by the addition of excess calcium ions(Ca^(2+))or fluorine ions(F−)in the *** results indicated that the CaF_(2)NPs synthesized by excess Ca^(2+)and the corresponding NAs exhibited much better water vapor adsorption properties than their counterparts by excess F−,owing to higher zeta *** importantly,whether excess Ca^(2+)or F^(−),CaF_(2)NAs had further enhanced water vapor adsorption capacity compared to primary CaF_(2)NPs,possibly owing to their unique nano-micro secondary structures and higher surface *** work has great potential in the development of high-performance absorbents for separating moisture from corrosive gas HF.

目的分析我国医疗卫生资源配置的空间差异和动态演进情况。方法利用2012—2021年我国31个省份(直辖市、自治区)的医疗卫生资源数据,评价指标选取医疗卫生财力、物力、人力和服务资源的4项指标:人均卫生费用、每千人医疗卫生机构数、每千人卫生技术人员数和医师每日担负诊疗人次数,使用全局熵值法计算各省历年配置水平指数,应用Dagum基尼系数和核密度估计方法分析我国医疗卫生资源配置水平的区域差异和动态演进特征。结果Dagum基尼系数结果显示,2012—2021年间我国医疗卫生资源配置水平基尼系数由0.1805上升至0.1972,后波动下降至0.1187。核密度估计结果显示,核密度曲线中心位置随年份的增加从0.2左右逐渐右移,稳定在0.3左右。结论我国医疗资源配置水平逐年提高,资源配置空间总体差距整体上逐年缩小。各地区仍存在不平衡的发展现状和差异化的资源配置水平,影响总体区域差距的主要因素是区域间差距。

Studies on silicon-containing fragrance raw materials——I. Syntheses and structure-odor relationship of acetals of 4-trimethylsilyl-3-cyclohexenone and 4-trimethylsilylcyclohexanone and their carbon counterparts

In order to investigate the structure-odor relationship of odoriferous compounds and search for new aroma chemicals, seven acetals of 4-trimethylsilyl-3-cyclohexenone and their carbon counterparts were synthesized by Birch reduction of 4-substituted anisoles and then acetalization. Seven acetals of 4-trimethylsilylcyclohexanone and their carbon counterparts were synthesized similarly. Structures of all new compounds were determined by MS, IR and ~1H NMR, and their characteristic odors were evaluated as well. The characteristic odors of the acetals formed by 4-substituted-3-cyclohexenone and 1, 2-diols are fruity and woody. The acetals formed from 1, 3-diols are woody, and formed with 1, 4-diols are very faint in odor. Odors of acetals of 4-substituted cyclohexanone are all very weak. As a whole, odors of organosilicon compounds are weaker, but somewhat more delicate than their carbon counter- parts.

Highly efficient electroreduction of CO_(2) by defect single-atomic Ni-N_(3) sites anchored on ordered micro-macroporous carbons

Microporous supports typically fail to fully expose active sites for electrolytes and CO_(2) molecules,and this usually results in low current density for the electrocatalytic CO_(2) reduction reaction(CO_(2)RR).To overcome the biggest obstacle and facilitate commercial applications,defective single-atomic Ni-N_(3) sites anchored to ordered micro-macroporous N-doped carbon(Ni-N/OMC)have been prepared by the pyrolysis of the Ni-ZIF-8@PS(ZIF=zeolitic imidazolate framework)and are intended to provide enhanced CO_(2)RR with a current density at an industrial *** Ni-ZIF-8@PS is constructed of nickel-based ZIF-8 embedded in the three-dimensional(3D)highly ordered polystyrene spheres(PS).The 3D ordered micro-macroporous architecture of Ni-N/OMC could facilitate the mass transfer of substrates to the accessible defective single-atomic Ni-N_(3) sites through micropores(0.6 nm)and macropores(~200 nm)interconnected by 50 nm *** a flow cell,Ni-N/OMC exhibits almost 100.0%CO Faraday efficiency(FECO)between−0.2 and−1.1 V *** and an industrial level CO partial current density of 208 mA cm^(−2).It has a turnover frequency of 1.5×10^(5) h^(−1) at−1.1 V *** in 1 M KOH electrolyte,which exceeds that of most reported nickel-based *** excellent CO_(2)RR performance for Ni-N/OMC makes it a state-of-the-art electrocatalyst for CO_(2)*** calculations show that the defective Ni-N_(3) site can lower the energy of*COOH formation compared with that of the Ni-N4 site,thereby accelerating CO_(2)***-N/OMC can also be utilized as a cathodic catalyst in Zn-CO_(2) battery,exhibiting high CO selectivity in the discharge process and excellent *** work paves a pathway to rational design of highly efficient electrocatalysts with 3D hierarchically ordered micro-macroporous architecture for CO_(2)RR towards industrial production and commercial applications.

Application of Brain-Computer-Interface in Awareness Detection Using Machine Learning Methods

The awareness detection in patients with disorders of consciousness currently relies on behavioral observations and CRS-R tests,however,the misdiagnosis rates have been relatively *** this study,we applied brain-computer interface(BCI)to awareness detection with a passive auditory stimulation paradigm.12 subjects with normal hearing were invited to collect electroencephalogram(EEG)based on a BCI communication system,in which EEG signals are transmitted *** necessary preprocessing,RBF-SVM and EEGNet were used for algorithm realization and *** a single subject,RBF-SVM can distinguish his(her)name stimuli awareness with classification accuracies ranging from 60-95%.EEGNet was used to learn all subjects'data and improved accuracy to 78.04%for characteristics finding and model ***,we completed the supplementary analysis work from the time domain and time-frequency *** study applied BCI communication to human awareness detection,proposed a passive auditory paradigm,and proved the effectiveness,which could be an inspiration for brain,mental or physical diseases diagnosis and detection.

甘草次酸调控铁死亡缓解舒尼替尼肝损伤的机制研究

目的探讨甘草次酸(GA)对舒尼替尼诱导的肝损伤的保护作用及机制。方法首先提取美国FDA药物不良反应数据库(FAERS)中使用舒尼替尼与其他治疗转移性肾细胞癌和胃肠道间质瘤的药物的肝脏不良事件(AEs)数据,采用贝叶斯法计算药物与不良反应的信号关联强度,对比舒尼替尼与其他抗肿瘤药物的肝毒性风险。其次在动物水平上,将小鼠随机分为正常对照组、舒尼替尼组(120 mg·kg^(-1))、舒尼替尼+GA低剂量组(10 mg·kg^(-1))、舒尼替尼+GA高剂量组(20 mg·kg^(-1)),每组5只,各组小鼠每日灌胃给药,连续2周。给药结束后检测小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)水平;苏木素伊红(HE)染色观察肝组织病理变化;试剂盒测定肝组织脂质过氧化物(LPO)、丙二醛(MDA)及铁(Fe)含量;Western blot分别检测核因子E2相关因子2(Nrf2)、溶质载体家族7成员11(SLC7A11)、血红素氧合酶(HO-1)、谷胱甘肽过氧化物酶4(GPX4)的蛋白表达情况。结果FAERS数据库信号挖掘分析发现舒尼替尼相比于其他治疗转移性肾细胞癌和胃肠道间质瘤的药物,与肝脏AEs存在的信号关联性更为显著,尤其与黄疸、高胆红血素症、AST升高、肝衰竭紧密相关。在动物实验结果中,与正常对照组相比,舒尼替尼组血清ALT、AST水平显著升高;HE染色结果显示明显的肝细胞肝索排列紊乱、空泡样变及炎性细胞浸润;LPO、MDA和Fe含量明显升高;Nrf2、SLC7A11、HO-1、GPX4蛋白均表达降低。与GA联合后,ALT、AST水平明显降低;肝组织病理损伤程度得到一定的恢复;LPO、MDA和Fe水平降低;Nrf2、SLC7A11、HO-1及GPX4蛋白表达增加。结论GA可有效缓解舒尼替尼所致的肝损伤,其作用机制可能与抑制铁死亡有关。

Global Convergence of a Modified Spectral CD Conjugate Gradient Method

In this paper,we present a new nonlinear modified spectral CD conjugate gradient method for solving large scale unconstrained optimization *** direction generated by the method is a descent direction for the objective function,and this property depends neither on the line search rule,nor on the convexity of the objective ***,the modified method reduces to the standard CD method if line search is *** some mild conditions,we prove that the modified method with line search is globally convergent even if the objective function is *** numerical results show that the proposed method is very promising.
