Immunogenicity and immunizing protection effect of GAMA gene DNA vaccine on Plasmodium berghei

Objective: To explore the effect of immunogenicity and immunizing protection of GAMA gene DNA vaccine, which was related with merozoite, ookinete and sporozoite invasion. Methods:Gene fragments were obtained using PCR technique and eukaryotic expression vector(containing immunostimulatory sequence) was built. BALB/c mice were divided into PBS control group, empty vector control group and study group and were immunized at week 0, 3 and 6 respectively. Blood was collected 2 weeks after each immunization and serum was separated to detect the Ig G, Ig G1 and Ig G2 a levels. Spleen of mice was obtained for preparation of splenic mononuclear cell and the cytokine IL-4 and IFN-αlevels were detected. Indirect immunofluorescence and western blot were employed to verify the specificity of antiserum. Sporozoite and merozoite invasion were used respectively to detect the immune protective effect 2 weeks after the third immunization. Ookinete conversion rate in vitro and oocyst numbers of mosquito stomach were observed to evaluate the transmission-blocking levels. Results: In GAMA DNA vaccine group: antiserum could be combined with recombinant protein specifically and green fluorescence signals of merozoite, ookinete and sporozoite were observable, while specific fragments and fluorescence signals were not observable in empty vector group. Compared with control group, specific Ig G in DNA vaccine immunity group significantly increased(P<0.01), and Ig G1 and Ig G2 a all increased(P<0.01). IL-4, IFN-αcontent in study group significantly increased, compared with control group(P<0.01). GAMA DNA vaccine immunity could not obviously block the erythrocyte-stage infection(caused by sporozoite invasion); compared with control group, liver worm load was slightly reduced(P<0.05), and antiserum ookinete numbers(cultured in vitro) had no significant difference with oocyst numbers of mosquito stomach in DNA vaccine group. Optimal design of hot rolling process for C-Mn steel by combining industrial data-driven model and multi-objective optimization algorithm

A successful mechanical property data-driven prediction model is the core of the optimal design of hot rolling process for hot-rolled strips. However, the original industrial data, usually unbalanced, are inevitably mixed with fluctuant and abnormal values. Models established on the basis of the data without data processing can cause misleading results, which cannot be used for the optimal design of hot rolling process. Thus, a method of industrial data processing of C-Mn steel was proposed based on the data analysis. The Bayesian neural network was employed to establish the reliable mechanical property prediction models for the optimal design of hot rolling process. By using the multi-objective optimization algorithm and considering the individual requirements of costumers and the constraints of the equipment, the optimal design of hot rolling process was successfully applied to the rolling process design for Q345B steel with 0.017% Nb and 0.046% Ti content removed. The optimal process design results were in good agreement with the industrial trials results, which verify the effectiveness of the optimal design of hot rolling process.

Objective Traditional epidemiological studies have shown that C-reactive protein(CRP)is associated with the risk of cardiovascular diseases(CVDs).However,whether this association is causal remains ***,Mendelian randomization(MR)was used to explore the causal relationship of CRP with cardiovascular outcomes including ischemic stroke,atrial fibrillation,arrhythmia and congestive heart *** We performed two-sample MR by using summary-level data obtained from Japanese Encyclopedia of Genetic association by Riken(JENGER),and we selected four single-nucleotide polymorphisms associated with CRP level as instrumental *** estimates were calculated with the inverse-variance weighted(IVW),penalized weighted median and weighted ***-Egger regression was used to explore *** No significant causal association of genetically determined CRP level with ischemic stroke,atrial fibrillation or arrhythmia was found with all four MR methods(all Ps>0.05).The IVW method indicated suggestive evidence of a causal association between CRP and congestive heart failure(OR:1.337,95%CI:1.005–1.780,P=0.046),whereas the other three methods did *** clear pleiotropy or heterogeneity were *** Suggestive evidence was found only in analysis of congestive heart failure;therefore,further studies are ***,no causal association was found between CRP and the other three cardiovascular outcomes.

Characterization of Organic Aerosol at a Rural Site in the North China Plain Region:Sources,Volatility and Organonitrates

The North China Plain(NCP)is a region that experiences serious aerosol pollution.A number of studies have focused on aerosol pollution in urban areas in the NCP region;however,research on characterizing aerosols in rural NCP areas is comparatively *** this study,we deployed a TD-HR-AMS(thermodenuder high-resolution aerosol mass spectrometer)system at a rural site in the NCP region in summer 2013 to characterize the chemical compositions and volatility of submicron aerosols(PM_(1)).The average PM_(1)mass concentration was 51.2±48.0μg m^(−3) and organic aerosol(OA)contributed most(35.4%)to PM_(1).Positive matrix factorization(PMF)analysis of OA measurements identified four OA factors,including hydrocarbon-like OA(HOA,accounting for 18.4%),biomass burning OA(BBOA,29.4%),lessoxidized oxygenated OA(LO-OOA,30.8%)and more-oxidized oxygenated OA(MO-OOA,21.4%).The volatility sequence of the OA factors was HOA>BBOA>LO-OOA>MO-OOA,consistent with their oxygen-to-carbon(O:C)***,the mean concentration of organonitrates(ON)was 1.48−3.39μg m−3,contributing 8.1%-19%of OA based on cross validation of two estimation methods with the high-resolution time-of-flight aerosol mass spectrometer(HRToF-AMS)*** analysis shows that ON were more correlated with BBOA and black carbon emitted from biomass burning but poorly correlated with ***,volatility analysis for ON further confirmed that particulate ON formation might be closely associated with primary emissions in rural NCP areas.

Underground pressure appearance laws analysis for fully mechanized top coal slice caving on high-dipping thick coal seams

Taking Adaohai Coal Mine as the example, underground pressure appearance laws of fully mechanized top coal slice caving on high-dipping and thick coal seams. Through site visit, theoretical analysis and discrete element calculation, the research shows that, as the mining deepens, underground stress of lower sublevels is more obvious and higher than that of upper sublevels and is higher in the air return roadway than that in the air intake roadway in the area that is near to the top coal. Because the top coal is thick and gangue is caved above the support, underground pressure to the working face is relatively gentle. Immediate roof will mainly fall down along the floor. Main roof and the rock bed above the main roof will move to the mined out area along the fault in the early stage and then fall down with the mined out area later. In addition, roof pressure mainly periodically appears in two directions along the trend and the dip.
