Circular RNAs(circRNAs)have been recognized as pivotal regulators in tumorigenesis,yet the biological functions as well as molecular mechanisms of the majority of circRNAs in hepatocellular carcinoma(HCC)remain *** sought to unveil the expression profile and biological role of circMYBL2 in *** microarray analyses were conducted to probe the expression profile of circMYBL2 in HCC cells,and qRT‒PCR analysis was then performed in HCC cell lines and tissues,revealing significant upregulation of *** experiments were conducted to evaluate the biological function of circMYBL2 in HCC ***,bioinformatics analysis,qRT‒PCR analysis,luciferase reporter assays,and western blot analysis were employed to investigate the interplay among circMYBL2,miR-1205,and ***2 was found to exhibit marked upregulation in tumor tissues as well as HCC cell *** expression of circMYBL2 increased the proliferation and migration of HCC cells,whereas circMYBL2 knockdown elicited contrasting ***,our results indicated that circMYBL2 promoted E2F1 expression and facilitated HCC progression by sponging *** findings revealed that circMYBL2 contributed to HCC progression through the circMYBL2/miR-1205/E2F1 axis,suggesting the potential of circMYBL2 as a novel target for HCC treatment or a prognostic biomarker for HCC.

The spreading of cancer cells from the primary tumor site to other parts of the body, known as metastasis, is the leading cause of cancer recurrence and mortality in patients with triple-negative breast cancer(TNBC). Overexpression of epidermal growth factor receptor(EGFR)is observed in approximately 70% of TNBC patients. EGFR is crucial for promoting tumor metastasis and associated with poor ***, it is vital to identify effective therapeutic strategies targeting EGFR inhibition. Ononin, an isoflavonoid found in various plants,such as clover and soybeans, has been shown to have anticancer properties in several cancers. In the present study, we aimed to investigate the effects of ononin on TNBC lung metastasis and the associated molecular pathways. We used various assays, including cell viability,colony formation, Transwell, wound healing, ELISA, Western blotting, and staining techniques, to achieve this objective. The results demonstrated that ononin effectively suppressed cellular proliferation and induced apoptosis, as evidenced by the cell viability assay, colony formation assay, and expression of apoptosis markers, and reduced the metastatic capabilities of TNBC cells. These effects were achieved through the direct suppression of cell adhesion, invasiveness and motility. Furthermore, in TNBC xenograft lung metastatic models, ononin treatment significantly reduced tumor growth and lung metastasis. Additionally, ononin reversed the epithelial-mesenchymal transition(EMT) by downregulating the expression of EMT markers and matrix metalloproteinases, as confirmed by Western blot analysis. Furthermore, ononin treatment reduced EGFR phosphorylation and suppressed the PI3K, Akt, and m TOR signaling pathways, which was further confirmed using EGFR agonists or inhibitors. Importantly, ononin treatment did not exert any toxic effects on liver or kidney function. In conclusion, our findings suggest that ononin is a safe and potentially therapeutic treatment

目的探索circ0000799经miR-1287-5p/GPX4轴对铁死亡促进三阴乳腺癌脑转移的影响机制。方法检测circ0000799在乳腺癌细胞系及乳腺癌组织中的表达。沉默亲代三阴乳腺癌细胞MDA-MB-231和子代脑转移细胞MDA-MB-231-BM中circ0000799的表达。比较各组细胞生长、侵袭能力、脑转移结节数目。使用双荧光素酶报告基因实验验证circ0000799、miR-1287-5p、GPX4调控关系。检测circ0000799对铁死亡相关蛋白谷胱甘肽过氧化酶4(GPX4)表达和总谷胱甘肽/氧化型谷胱甘肽比的影响。结果与人正常乳腺上皮细胞(MCF-10A)或癌旁组织比较,circ0000799在三阴乳腺癌细胞系及乳腺癌组织中上调,且在子代三阴乳腺癌细胞系及脑转移灶中上调最为显著(P<0.05)。与sh-circCTR组比较,sh-circ0000799组细胞生长、侵袭能力降低(P<0.05)。sh-circCTR组、sh-circ0000799组、sh-circ0000799+RSL3组脑转移结节数量依次降低(P<0.05)。双荧光素酶报告基因实验显示,circ0000799可与miR-1287-5p相互作用,miR-1287-5p可与GPX4相互作用。与sh-circCTR组比较,sh-circ0000799组miR-1287-5表达增加,总谷胱甘肽/氧化型谷胱甘肽比和GPX4表达降低(P<0.05)。与miR-CTR组比较,miR-1287-5p组GPX4 mRNA表达降低(P<0.05)。结论circ0000799在三阴乳腺癌中高表达且可能通过miR-1287-5p/GPX4轴促进三阴乳腺癌脑转移。

Is surgical axillary staging necessary in women with T1 breast cancer who are treated with breast-conserving therapy?

Background:In the post-Z0011 trial era,the need to perform surgical axillary staging for early-stage breast cancer patients,who are treated with breast-conserving therapy(BCT),is being *** conducted a retrospective cohort study using the Surveillance,Epidemiology,and End Results(SEER)database to evaluate the safety of waiving surgical axillary staging in patients with T1 breast cancer treated with ***:A total of 166,615 eligible patients diagnosed between 2000 and 2012 were divided into staging(sentinel lymph node biopsy or axillary lymph node dissection)and non-staging(no lymph node examined or only needle aspiration biopsy of lymph nodes)*** score matching(PSM)was performed to balance disparities between the two *** analysis with the Cox proportional hazards model was used to assess factors related to breast cancer-specific survival(BCSS).Results:Although the tumor size at time of presentation was decreasing over years,the rate of surgical axillary stag-ing increased from 93.3%to 96.9%.The 5-year BCSS rates of the whole cohort(before PSM)and matched cohort(after PSM)were 98.0%and 97.5%.Within the matched cohort,the BCSS was significantly longer in the staging group than in the non-staging group(P0.05).Race,marital status,hormone receptors,and chemotherapy were not associated with the favorable impact of surgical axillary staging on BCSS(P>0.05).Conclusion:Although surgical axillary staging remains important for T1 breast cancer patients treated with BCT,it might be unnecessary for patients with old age,small tumor,grade I disease,or favorable histological types.

The Persistence and Zonal Scale of Atmospheric Dipolar Modes

This study investigates the relationship between the persistence and the zonal scale of atmospheric dipolar modes(DMs). Results from the daily data of ERA5 and the long-term output of an idealized atmospheric model show that the atmospheric DMs with a broader(narrower) zonal scale dipolar structure possess a longer(shorter) persistence. A detailed vorticity budget analysis indicates that the persistence of a hemispheric-scale DM(1/1 DM) and a regional or sectoral DM(1/8 DM) in the model both largely rely on the persistence of the nonlinear eddy forcing. Linear terms can indirectly reduce the persistence of the anomalous nonlinear eddy forcing in a 1/8 DM by modifying the baroclinicity via the arousal of anomalous vertical motions. Therefore, the atmospheric DMs with a broader(narrower) zonal scale possess a longer(shorter) persistence because the effects of the linear terms are less(more) pronounced when the atmospheric DMs have better(worse) zonal symmetry. Further analyses show that the positive eddy feedback effect is weak or even absent in a 1/8DM and the high-frequency eddy forcing acts more like a concomitant phenomenon rather than a leading driving factor for a 1/8 DM. Thus, the hemispheric-scale DM and the regional or sectoral DMs are different, not only in their persistence but also in their dynamics.

The burgeoning demand for modern electronic devices and electric vehicles has driven the development of efficient,reliable,and environmentally friendly batteries[1,2].Lithium–sulfur(Li–S)batteries with high theoretical capacity(1672 mA h g1)and energy density(2600 W h kg1),have garnered significant interest in both academic and industrial research[3,4].However,the widespread production and commercialization of Li–S batteries are impeded by the inherent characteristics of sulfur and lithium,along with their complex electrochemical ***,challenges such as the polysulfide shuttle effect and the slow kinetics of the sulfur nucleation/decomposition reaction hinder capacity utilization and cycling *** overcome these challenges,innovative electrocatalyst strategies aimed at enhancing activity have been explored.

The learning curve for transurethral enucleation with bipolar energy for benign prostate hyperplasia:a single-surgeon experience of 494 patients

This study was performed to investigate the learning curve of transurethral enucleation with bipolar energy(TUEB)for benignprostatic *** study involved 494 consecutive patients who underwent TUEB for benign prostatic hyperplasia fromAugust 2018 to March 2022 by one surgeon(SJJ,Seoul National University Bundang Hospital,Seongnam,Korea).The patientswere followed up at 1 week,1 month,3 months,and 6 months *** evaluate the learning curve of TUEB,perioperativeparameters including the enucleation ratio(enucleated tissue weight/transitional zone volume),TUEB efficiency(enucleatedtissue weight/operation time),and enucleation efficiency(enucleated tissue weight/enucleation time)were *** and postoperative complications were also assessed,including the International Prostate Symptom Score(IPSS),IPSSquality-of-life(QoL)score,and uroflowmetry *** patients’median age was 72(interquartile range[IQR]:66–78)years,and the estimated prostate volume and transitional zone volume were 63.0(IQR:46.0–90.6)ml and 37.1(IQR:24.0–60.0)ml,*** enucleation ratio,TUEB efficiency,and enucleation efficiency were 0.60(IQR:0.46–0.54)g ml−1,0.33(IQR:0.22–0.46)g min−1,and 0.50(IQR:0.35–0.72)g min−1,respectively,plateauing after 70 *** functional outcomes,including total IPSS,IPSS QoL score,and uroflowmetry outcomes,significantly improved at 6 months after TUEB(all P<0.05),but without significant differences over the learning ***-five(13.2%)patients developed complications after TUEB,21.5%of whom experienced major complications(Clavien–Dindo grade≥3).The rate of major complications declined as the number ofTUEB cases increased(P=0.013).Our results suggest that the efficiency of TUEB stabilized within 70 procedures.
