MicroRNA-219 alleviates glutamate-induced neurotoxicity in cultured hippocampal neurons by targeting calmodulin-dependent protein kinase Ⅱ gamma

Septic encephalopathy is a frequent complication of sepsis,but there are few studies examining the role of micro RNAs（mi Rs） in its *** this study,a mi R-219 mimic was transfected into rat hippocampal neurons to model mi R-219 overexpression.A protective effect of mi R-219 was observed for glutamate-induced neurotoxicity of rat hippocampal neurons,and an underlying mechanism involving calmodulin-dependent protein kinase II γ（Ca MKIIγ） was *** R-219 and Ca MKIIγ m RNA expression induced by glutamate in hippocampal neurons was determined by quantitative real-time reverse transcription-polymerase chain reaction（q RT-PCR）.After neurons were transfected with mi R-219 mimic,effects on cell viability and apoptosis were measured by 3-（4,5-dimethylthiazolyl-2）-2,5-diphenyltetrazolium bromide（MTT） assay and flow *** addition,a luciferase reporter gene system was used to confirm Ca MKIIγ as a target gene of mi *** blot assay and rescue experiments were also utilized to detect Ca MKIIγ expression and further verify that mi R-219 in hippocampal neurons exerted its effect through regulation of Ca MKIIγ.MTT assay and q RT-PCR results revealed obvious decreases in cell viability and mi R-219 expression after glutamate stimulation,while Ca MKIIγ m RNA expression was ***,flow cytometry,and caspase-3 activity assays showed that mi R-219 overexpression could elevate glutamate-induced cell viability,and reduce cell apoptosis and caspase-3 ***,luciferase Ca MKIIγ-reporter activity was remarkably decreased by co-transfection with mi R-219 mimic,and the results of a rescue experiment showed that Ca MKIIγ overexpression could reverse the biological effects of mi ***,these findings verify that mi R-219 expression was decreased in glutamate-induced neurons,Ca MKIIγ was a target gene of mi R-219,and mi R-219 alleviated glutamate-induced neuronal excitotoxicity by negatively controlling Ca MKIIγ expression.

Use of two-stage dough mixing process in improving water distribution of dough and qualities of bread made from wheat–potato flour

The two-stage dough mixing process was innovated to improve the qualities of bread made from potato flour(PF) and wheat flour at a ratio of 1:1(w/w). The final dough was first prepared from wheat flour before being added with PF. The effects of the method on enhancing the dough qualities were verified, and the distribution of water in gluten-gelatinized starch matrix of the doughs was investigated. We observed that the bread qualities were improved, as reflected by the increase of specific volume from 2.26 to 2.96 m L g^–1 and the decrease of crumb hardness from 417.93 to 255.57 g. The results from rheofermentometric measurements showed that the dough mixed using the developed mixing method had higher maximum dough height value, time of dough porosity appearance, and gas retention coefficient, as well as enhanced gluten matrix formation compared to that mixed by the traditional mixing method. The results from low-field nuclear magnetic resonance confirmed that the competitive water absorption between gluten and gelatinized starch could restrict the formation of gluten network in the dough mixed using the traditional mixing process. Using the novel mixing method, gluten could be sufficiently hydrated in stage 1, which could then weaken the competitive water absorption caused by gelatinized starch in stage 2;this could also be indicated by the greater mobility of proton in PF and better development of gluten network during mixing.

Age-and Microbiota-Dependent Cell Stemness Plasticity Revealed by Cattle Cell Landscape

Newborn ruminants are considered functionally monogastric *** poor understanding of cellular differences between newborn and mature ruminants prevents the improvement of health and performance of domestic ***,we performed the single-cell RNA sequencing on the rumen,reticulum,omasum,abomasum,duodenum,jejunum,ileum,cecum,colon,rectum,liver,salivary gland,and mammary gland from newborn and adult cattle.A comprehensive single-cell transcriptomic atlas covering 235,941 high-quality single cells and 78 cell types was deciphered.A Cattle Cell Landscape database(http://***)was established to elaborately display the data and facilitate effective annotation of cattle cell types and subtypes for the broad research *** measuring stemness states of epithelial cells in each tissue type,we revealed that the epithelial cells from newborn forestomach(rumen,reticulum,and omasum)were more transcriptionally indistinct and stochastic compared with the adult stage,which was in contrast to those of abomasum and intestinal *** rapid forestomach development during the early life of calves was driven by epithelial progenitor-like cells with high DNA repair activities and ***,in the forestomach tissues of newborn calves,the Megasphaera genus was involved in regulating the transcriptional plasticity of the epithelial progenitor-like cells by DNA methylation regulation.A novel cell type,the STOML3+cell,was found to be *** apparently plays a crucial role in stemness maintenance of its own and cholangiocytes in the hepatic *** results reveal that the age-and microbiota-dependent cell stemness plasticity drives the postnatal functional maturity of ruminants.

Clinical application of free/total PSA ratio in the diagnosis of prostate cancer in men over 50 years of age with total PSA levels of 2.0-25.0 ng ml^(−1) in Western China

The goal of this study was to investigate the clinical application of free/total prostate-specific antigen(F/T PSA)ratio,considering the new broad serum total PSA(T-PSA)“gray zone”of 2.0–25.0 ng ml^(−1)in differential diagnosis of prostate cancer(PCa)and benign prostate diseases(BPD)in men over 50 years in Western China.A total of 1655 patients were included,528 with PCa and 1127 with *** T-PSA,free PSA(F-PSA),and F/T PSA ratio were *** operating characteristic curves were used to assess the efficiency of PSA and F/T PSA *** were 47.4%of cancer patients with T-PSA of 2.0–25.0 ng ml^(−1).When T-PSA was 2.0–4.0 ng ml^(−1),4.0–10.0 ng ml^(−1),and 10.0–25.0 ng ml^(−1),the area under the curve(AUC)of F/T PSA ratio was 0.749,0.769,and 0.761,*** best AUC of F/T PSA ratio was 0.811 when T-PSA was 2.0–25.0 ng ml^(−1),with a specificity of 0.732,a sensitivity of 0.788,and an optimal cutoff value of 15.5%.The AUC of F/T PSA ratio in different age groups(50–59 years,60–69 years,70–79 years,and≥80 years)was 0.767,0.806,0.815,and 0.833,respectively,and the best sensitivity(0.857)and specificity(0.802)were observed in patients over 80 *** T-PSA trend was in accordance with the Gleason score,tumor node metastasis(TNM)stage,and American Joint Committee on Cancer prognosis ***,the F/T PSA ratio can facilitate the differential diagnosis of PCa and BPD in the broad T-PSA“gray zone”.Serum T-PSA can be a Gleason score and prognostic indicator.

Facile Preparation of a Porous Biochar Derived from Waste Crab Shell with High Removal Performance for Diesel

In this study,a porous biochar material derived from waste crab shell was prepared by one-step hydrothermal carbonization and acetic acid activation method at 180°C,which was characterized by SEM,BET,XRD and *** results show that the as-prepared crab shell biochar(CSB)exhibits a fluffy irregular layered structure with abundant pores and oxygen-containing functional groups,which can facilitate the adsorption of diesel using *** addition,batch adsorption experiments had been performed,effects of initial diesel concentration,adsorption time,adsorbent dosage and pH on the diesel adsorption using CSB were analyzed,which could be observed that CSB has high removal efficiency for diesel,and the maximum removal rate is up to 80.1%.The adsorption isotherms and kinetic studies were also investigated to determine the adsorption mechanism of diesel using CSB,the results show that the Langmuir model and the pseudo-second-order model are more suitable for describing the adsorption of diesel using CSB,indicating that the adsorption of diesel oil by CSB is monolayer chemical *** study will provide a theoretical basis for the high-value utilization of waste crab shell,which has a great potential in the treatment of oil spill.
