Objective This literature review aims to summarize the methods of isolation, expansion, preservation of human umbilical cord mesenchymal stem cells （hUCMSCs）, for comprehensive practical use in preclinical research and clinical trials. differentiation and understanding and Data sources All the literature reviewed was published over the last 10 years and is listed in PubMed and Chinese National Knowledge Infrastructure （CNKI）. Studies were retrieved using the key word ＂human umbilical cord mesenchymal stem cells＂. Results Explants culture and enzymatic digestion are two methods to isolate hUCMSCs from WJ and there are modifications to improve these methods. Culture conditions may affect the expansion and differentiating orientations of hUCMSCs. In addition, hUCMSCs can maintain their multi-potential effects after being properly frozen and thawed. Conclusion Considering their multi-potential, convenient and non-invasive accessibility, low immunogenicity and the reported therapeutic effects in several different preclinical animal models, hUCMSCs have immense scope in regeneration medicine as a substitute for MSCs derived from bone marrow or umbilical cord blood.

Background Signal transducer and activator of transcription 3 （STAT3） is usually constitutively activated in a variety of malignancies. It directly contributes to tumorigenesis, invasion, and metastasis. The surgical treatment of breast cancer has made no breakthroughs in terms of treatment effect, in spite of its long history. Current biotherapies bring a note of optimism to breast cancer treatment. To explore the possibility of a siRNA targeted STAT3 blocking treatment for over-activated tumor cells, we evaluated the efficacy of a STAT3 siRNA on human breast cancer cells in vitro and in vivo. Methods Three MCF-7 human breast cancer cell lines were tested： control MCF-7 cells, non-specific siRNA transfected MCF-7 cells and STAT3 siRNA transfected MCF-7 cells. Expression of STAT3 in MCF-7 cells was inhibited by RNA interference （RNAi）. The STAT3 mRNA and protein levels were detected by semi-quantity RT-PCR and Western blotting. Cell proliferation and apoptosis were determined by MTT method and flow cytometry. The three groups of MCF-7 cells mentioned above were transplanted subcutanuously into nude mice and their tumorgenic ability observed. The STAT3 mRNA and protein levels of the samples from tumors in different groups were determined by semi-quantity RT-PCR and Western blotting and compared. Results In STAT3 siRNA transfected MCF-7 cells, the expressions （STAT3/13-actin） of STAT3 mRNA （0.327±0.020） and protein （0.153±0.006） were significantly lower than that in control MCF-7 cells （mRNA 1.093±0.018, protein 1.374±0.022） and non-specific siRNA transfected MCF-7 cells （mRNA 1.035_±0.050, protein 1.320±0.033）（P 〈0.05）. MTT showed that cell proliferation was significantly reduced and the cell growth inhibition ratio in the STAT3-siRNA group was （44.00±5.10）%, significantly higher than that in non-specific siRNA transfected MCF-7 cells （（16.10_±1.05）%, P 〈0.05）. Since requirement dependency extraction is a cognitively challenging and error-prone task,this paper proposes an automatic requirement dependency extraction method based on integrated active learning *** this paper,the coefficient of variation method was used to determine the corresponding weight of the impact factors from three different angles:uncertainty probability,text similarity difference degree and active learning variant prediction divergence *** combining the three factors with the proposed calculation formula to measure the information value of dependency pairs,the top K dependency pairs with the highest comprehensive evaluation value are selected as the optimal *** the optimal samples are continuously added into the initial training set,the performance of the active learning model using different dependency features for requirement dependency extraction is rapidly ***,compared with other active learning strategies,a higher evaluation measure of requirement dependency extraction can be achieved by using the same number of ***,the proposed method using the PV-DM dependency feature improves the weight-F1 by 2.71%,the weight-recall by 2.45%,and the weight-precision by 2.64%in comparison with other strategies,saving approximately 46%of the labelled data compared with the machine learning approach.

The emerging biomass-based epoxy vitrimers hold great potential to fulfill the requirements for sustainable development of *** the existence of dynamic chemical bonds in vitrimers often reduces both the thermal and mechanical properties of epoxy resins, it is challenging to produce recyclable epoxy vitrimers with both excellent mechanical properties and good thermal stability. Herein, a monomer 4-(((5-(hydroxymethyl)furan-2-yl)methylene)amino)phenol(FCN) containing furan ring with potential to form high density of hydrogen bonding among repeating units is designed and copolymerized with glycerol triglycidyl ether to yield epoxy resin(FCN-GTE), which intrinsically has dual hydrogen bond networks, dynamic imine structure and resultant high performance. Importantly, as compared to the BPA-GTE, the FCN-GTE exhibits significantly improved mechanical properties owing to the increased density of hydrogen bond network and physical crosslinking interaction. Furthermore, density functional theory(DFT) calculation and in situ FTIR analysis is conducted to decipher the formation mechanism of hydrogen bond network. In addition, the FCN-GTE possesses superior UV shielding, chemical degradation, and recyclability because of the existence of abundant imine bonds. Notably, the FCN-GTE-based carbon fiber composites could be completely recycled in an amine *** study provides a facile strategy for synthesizing recyclable biomass-based high-performance epoxy vitrimers and carbon fiber composites.
