Effects of amyloid precursor protein peptide APP96-110,alone or with human mesenchymal stromal cells,on recovery after spinal cord injury

Delivery of a peptide(APP96-110),derived from amyloid precursor protein(APP),has been shown to elicit neuroprotective effects following cerebral stroke and traumatic brain *** this study,the effect of APP96-110 or a mutant version of this peptide(mAPP96-110)was assessed following moderate(200 kdyn,(2 N))thoracic contusive spinal cord injury(ScI)in adult Nude *** received a single tail vein injection of APP96-110 or mAPP96-110 at 30 minutes post-ScI and were then assessed for functional improvements over the next 8 weeks.A cohort of animals also received transplants of either viable or non-viable human mesenchymal stromal cells(hMScs)into the Sc lesion site at one week post-injury to assess the effect of combining intravenous APP96-110 delivery with hMSc *** were perfused 8 weeks post-ScI and longitudinal sections of spinal cord analyzed for a number of factors including hMSc viability,cyst size,axonal regrowth,glial reactivity and macrophage *** of sensorimotor function revealed occasional significant differences between groups using Ladderwalk or Ratwalk tests,however there were no consistent improvements in functional outcome after any of the ***96-110 alone,and APP96-110 in combination with both viable and non-viable hMScs significantly reduced cyst size compared to ScI *** treatments with donor hMScs also significantly increased βIII tubulin^(+),glial fibrillary acidic protein(GFAP^(+))and laminin+expression,and decreased ED1^(+)expression in *** preliminary study demonstrates that intravenous delivery of APP96-110 peptide has selective,modest neuroprotective effects following ScI,which may be enhanced when combined with hMSc ***,the effects are less pronounced and less consistent compared to the protective morphological and cognitive impact that this same peptide has on neuronal survival and behaviour after stroke and traumatic brain *** while the efficacy of a

Awareness of the adverse impact of air pollution on attention-related performance such as learning and driving is rapidly ***,there is still little known about the underlying neurocognitive *** an adapted dot-probe task paradigm and event-related potential(ERP)technique,we investigated how visual stimuli of air pollution influence the attentional allocation *** were required to make responses to the onset of a target presented at the left or right visual *** probable location of the target was forewarned by a cue(pollution or clean air images),appearing at either the target location(attention-holding trials)or the opposite location(attention-shifting trials).Behavioral measures showed that when cued by pollution images,subjects had higher response accuracy in attention-shifting *** analysis results revealed that after the cue onset,pollution images evoked lower N300 amplitudes,indicating less attentioncapturing effects of dirty *** the target onset,pollution cues were correlated with the higher P300 amplitudes in attention-holding trials but lower amplitudes in attention-shifting *** indicates that after visual exposure to air pollution,people need more neurocognitive resources to maintain attention but less effort to shift attention *** findings provide the first neuroscientific evidence for the distracting effect of air *** conclude with several practical implications and suggest the ERP technique as a promising tool to understand human responses to environmental stressors.

Genetic susceptibility to metabolic associated fatty liver disease(MAFLD)is complex and poorly *** characterization of the genetic background of hepatic fat content would provide insights into disease etiology and causality of risk *** performed genome-wide association study(GWAS)on two noninvasive definitions of hepatic fat content:magnetic resonance imaging proton density fat fraction(MRI-PDFF)in 16,050 participants and fatty liver index(FLI)in 388,701 participants from the United Kingdom(UK)Biobank(UKBB).Heritability,genetic overlap,and similarity between hepatic fat content phenotypes were analyzed,and replicated in 10,398 participants from the University Medical center Groningen(UMcG)Genetics Lifelines Initiative(UGLI).Meta-analysis of GWASs of MRI-PDFF in UKBB revealed five statistically significant loci,including two novel genomic loci harboring cREB3L1(rs72910057-T,P=5.40E−09)and GcM1(rs1491489378-T,P=3.16E−09),respectively,as well as three previously reported loci:PNPLA3,TM6SF2,and *** of FLI in UKBB identified 196 genome-wide significant loci,of which 49 were replicated in UGLI,with top signals in ZPR1(P=3.35E−13)and FTO(P=2.11E−09).Statistically significant genetic correlation(rg)between MRI-PDFF(UKBB)and FLI(UGLI)GWAS results was found(rg=0.5276,P=1.45E−03).Novel MRI-PDFF genetic signals(cREB3L1 and GcM1)were replicated in the FLI *** identified two novel genes for MRI-PDFF and 49 replicable loci for *** a difference in hepatic fat content assessment between MRI-PDFF and FLI,a substantial similar genetic architecture was *** is identified as an easy and reliable approach to study hepatic fat content at the population level.

cholesterol and Sericin as First Aid for Damaged cells

cells are surrounded by a double-layered phospholipid cell membrane responsible for the isolation of intracellular contents, active regulation of uptake from the extracellular environment, and intercellular connection and communication. These cell membranes must be intact and functionally active for cell survival and biological functioning. compromised damage repair mechanisms usually result in impaired cellular homeostasis, leading to early or late problems. chronic myopathies, certain myocardial diseases, aging, and acute or chronic neurodegenerative diseases (like Parkinson and Alzheimer) are directly related to cell membrane damage. This study examined the effect of a cholesterol-loaded nanoparticle (methyl-beta cyclodextrin) or the silk protein sericin on cell membrane and DNA integrity and cell viability in an in vitro cell damage model (frozen-thawed rabbit sperm cells). The cells were stored in liquid nitrogen (-196°c), thawed in small batches, and treated with cholesterol-loaded cyclodextrin or sericin before incubation at 35°c for 4 hours. cell membrane integrity, DNA damage, and viability rates were assessed immediately after thawing and after the incubation period. The administration of sericin and cholesterol in a cell damage model increased cell survival and reduced DNA damage over a 4-hour post-thaw incubation period, suggesting their potential use as a “first aid” intervention at the cellular level.

Systematic study of cluster radioactivity within the generalized liquid drop model

cluster radioactivity is studied within the generalized liquid drop model(GLDM),in which the shell correction energy,pairing energy,and cluster preformation factor are *** calculations show significant improvements and can reproduce the experimental data within a factor of 8.04 after considering these physical *** addition,the systematic trend of the cluster preformation factors is discussed in terms of the N_(p)N_(n)scheme to study the influence of the valence proton-neutron interaction and shell effect on cluster *** is found that log10Pcis linearly related to N_(p)N_(n).This is in agreement with a recent study[*** et al.,***.c 108,014325(2023)],in which log10Pc,obtained using different theoretical models and treatment methods than those used in this study,also had a linear relationship with N_(p)N_(n).combined with the work by Qi et al.,this study suggests that the linear relationship between log10Pcand N_(p)N_(n)is model-independent and both the shell effect and valence proton-neutron interaction play essential roles in cluster *** analytical formula is proposed to calculate the cluster preformation factor based on the N_(p)N_(n)*** addition,the cluster preformation factors and the cluster radioactivity half-lives of some heavy nuclei are predicted,which can provide a reference for future experiments.

Purpose:The aim of this study was to review information about risk factors for lower extremity running injuries in both short-distance(mean running distance-20 km/week and-10 km/session)and long-distance runners(mean running distance>20 km/week and>10 km/session).Methods:Electronic databases were searched for articles published up to February *** cohort studies using multivariable analysis for the assessment of individual risk factors or risk models for the occurrence of lower extremity running injuries were *** reviewers independently selected studies for eligibility and assessed risk of bias with the Quality in Prognostic Studies *** GRADE approach was used to assess the quality of the ***:A total of 29 studies were included:17 studies focused on short-distance runners,11 studies focused on long-distance runners,and 1 study focused on both types of runners.A previous running-related injury was the strongest risk factor for an injury for long-distance runners,with moderate-quality *** injuries not attributed to running was the strongest risk factor for an injury for short-distance runners,with high-quality *** body mass index,higher age,sex(male),having no previous running experience,and lower running volume were strong risk factors,with moderate quality evidence,for short-distance ***-quality evidence was found for all risk models as predictors of runningrelated injuries among short-and long-distance ***:Several risk factors for lower extremity injuries have been identified among short-and long-distance runners,but the quality of evidence for these risk factors for running-related injuries is *** injuries seem to have a multifactorial origin both in short-and long-distance runners.
