Background Juvenile loc.lized scleroderma(JLS)is a rare pediatric disease characterized by inflammation and skin thick *** is associated with deep tissue and extracutaneous involvement that often results in functional impairment and growth *** article provides an overview of the disease with a focus on active features and *** sources We searched databases including PubMed,Elsevier and Medline and Wanfang,reviewing publications from 2013 to *** earlier publications were also *** linear scleroderma is the most common JLS *** lines of evidence suggest that JLS is an autoimmune *** involvement is common and can present before the onset of skin *** skin features are associated with disease activity,and activity can also manifest as arthritis,myositis,uveitis,seizures,and growth *** immunosuppressive treatment,commonly methotrexate with or without glucocorticoids,greatly improves outcome and is recommended for treating JLS patients with active disease and moderate or higher *** term monitoring is needed because of the disease's chronicity and the high frequency of relapses off of *** JLS is associated with a risk for disabling and disfiguring morbidity for the growing *** active disease is important for guiding treatment,but often difficult because of the paucity of markers and lack of a universal skin activity *** studies of JLS pathophysiology are needed to allow the identification of biomarkers and therapeutic *** effectiveness treatment studies are also needed to work towards optimizing care and outcome.

The immune system has a remarkable ability to respond to seemingly endless *** essence,a productive immune response takes place along a well defined but treacherous line,that is to effectively eradicate pathogens,and at the same time avoid causing damage to self *** type of response is fine-tuned,at least in part,by a complex array of pathways that either promote or inhibit the activation of innate and adaptive immune *** effort has been focused on pathways that can support immune *** this article,we review specifically pathways that can inhibit immune responses and maintain immune homeostasis,highlighting our recent understanding on the role of inhibitory receptors that selectively engage the self MHc class I molecules and the B7 superfamily members,we also discuss the inhibitory Fc receptors and inhibitory cytokines and how such pathways,either individually or collectively,regulate innate and adaptive immune ***,we summarize new emerging approaches on how such negative pathways can be therapeutically modulated in various disease settings.

Immune tolerance is a highly regulated state and involves diverse *** to the induction of tolerance is the targeted modulation of T-cell activities(both effector and regulatory),in which transcription factors play a significant *** nuclear factor kappa-B(NF-κB)family is a family of transcription factors that not only are critically involved in diverse T-cell responses but also are regulated by many mechanisms to maintain tolerance and T-cell ***-κB,as a transcription factor,has been extensively studied in recent decades,and the molecular mechanisms that regulate NF-κB activities have been well ***,recent studies have revealed exciting new roles for NF-κB;in addition to its transcriptional activity,NF-κB can also activate diverse epigenetic mechanisms that mediate extensive chromatin remodeling of target genes to regulate T-cell *** this review article,we highlight recent discoveries and emerging opportunities in targeting NF-κB family members as well as their associated chromatin modifiers in the induction of immune tolerance and in the clinical treatment of immune diseases.

This is a review paper on the performances of both conventional and energy-absorbing rockbolts manifested in laboratory tests. characteristic parameters such as ultimate load, displacement and energy absorption are reported, in addition to load-displacement graphs for every type of rockbolt. conventional rockbolts refer to mechanical rockbolts, fully-grouted rebars and frictional rockbolts. According to the test results, under static pull loading a mechanical rockbolt usually fails at the plate; a fully-grouted rebar bolt fails in the bolt shank at an ultimate load equal to the strength of the steel after a small amount of displacement; and a frictional rockbolt is subjected to large displacement at a low yield load. Under shear loading, all types of bolts fail in the shank. Energy-absorbing rockbolts are developed aiming to combat instability problems in burst-prone and squeezing rock conditions. They absorb deformation energy either through ploughing/slippage at predefined load levels or through stretching of the steel bolt. An energy-absorbing rockbolt can carry a high load and also accommodate significant rock displacement, and thus its energy-absorbing capacity is high. The test results show that the energy absorption of the energy-absorbing bolts is much larger than that of all conventional bolts. The dynamic load capacity is smaller than the static load capacity for the energy-absorbing bolts displacing based on ploughing/slippage while they are approximately the same for the D-Bolt that displaces based on steel stretching.

china is breaking through the petrodollar system,establishing RMB-dominating crude oil futures *** country is achieving a milestone in its transition to energy finance market *** study explores the price leadership of china's crude oil futures and identifies its price co-movement to uncover whether it truly shakes up the global oil spots ***,we find that for oil spots under different gravities,china's oil futures is only a net price information receiver from light-,medium-,and heavy-gravity oil spots,but it has a relatively stronger price co-movement with these three ***,for oil spots under different sulfur contents,china's oil futures still has weak price leadership in sweet,neutral,and sour oil spots,but it has strong co-movement with ***,for oil spots under different geographical origins,china's oil futures shows price leadership in East Asian and Australian oil spots at the medium-and longrun time scales and strong price co-movement with East Asian,Middle Eastern,Latin American and Australian oil ***'s oil futures may not have good price leadership in global spots market,but it features favorable price co-movement.

Impact drop tests are routinely used to examine the dynamic performance of *** impact tests have been carried out in the past decades on independently designed,constructed and operated testing *** laboratory has developed testing procedures;thus,the results are often reported in different ways by various *** inconsistency in testing procedures and reporting formats presents a challenge when comparing results from different laboratories.A series of impact tests of identical rockbolts was carried out using the direct impact method(*** mass free-fall method)on the rigs in four laboratories in different *** purpose of these tests was to investigate the level of consistency in the results from the four *** rig demonstrated a high level of repeatability,but differences existed between the various *** differences would suggest that there is noticeable equipment-dependent bias when test results obtained from different laboratories are *** was also observed that the energy dissipated for the plastic displacement of the bolt was smaller than the impact energy in the *** average impact load(AIL)and the ultimate plastic displacement(D)of the bolt describe the ultimate dynamic performance of the *** the case where the bolt does not rupture,the specific plastic energy(SPE)is an appropriate parameter in describing the impact performance of the *** other relevant parameters are the first peak load(FPL)and the initial stiffness(K)of the bolt *** information from this test series will guide the formulation of standardised testing procedures for dynamic impact tests of rockbolts.

Understanding the role of humic substances in soils is important for developing and utilizing organic fertilizers or soil amendments for sustainable *** objective of this study was to determine the effects of different fractions of humic acids derived from Leonardite on enzymatic activities and bioavailability of nutrients in a *** experiment was carried out by mixing different factions of humic acids with a soil and incubated for 70 *** treatments included five factions of humic acids(HS1(low molecular weight),HS2(medium molecular weight)),HS3(large molecular weight,SED(sediment of fractions),HS(mixture of HS1,HS2,and HS3)),raw Leonardite(IM)and a control(no addition of humic acid).Experimental results showed that application of humic acids significantly improved acid and alkaline phosphatase activities,especially with *** substances with high molecular weights significantly inhibit urease activity,and the optimal application rate was 600 kg/hm2 of humic substances with the high molecular *** of NH4+-N were decreased with increasing humus *** treatments(HSmix,HS1,HS2,HS3,IM,SED)did not affect the soil contents of ca,although soil concentrations of K,P,cu,Zn were increase significantly when small molecular weight humus(HS1)was applied.

T-cell-based immunotherapy is gaining momentum in cancer treatment;however,our comprehension of the transcriptional regulation governing T cell antitumor activity remains *** objective of this study was to explore the function of interferon regulatory factor 4(IRF4)in antitumor cD8^(+)T cells using the TRAMP-c1 prostate cancer and B16F10 melanoma *** achieve this,we generated an Irf4^(GFP-DTR) mouse strain and discovered that cD8^(+)tumor-infiltrating lymphocytes(TILs)expressing high levels of *** exhibited a more differentiated PD-1high cell *** administering diphtheria toxin to tumor-bearing Irf4^(GFP-DTR) mice,we partially depleted ***^(+)TILs and observed an accelerated tumor *** specifically explore the function of IRF4 in antitumor cD8^(+)T cells,we conducted 3 adoptive cell therapy(AcT)***,depleting ***^(+)cD8^(+)TILs derived from AcT significantly accelerated tumor growth,emphasizing their crucial role in controlling tumor ***,deleting the Irf4 gene in antitumor cD8^(+)T cells used for AcT led to a reduction in the frequency and effector differentiation of cD8^(+)TILs,completely abolishing the antitumor effects of ***,we performed a temporal deletion of the Irf4 gene in antitumor cD8^(+)T cells during AcT,starting from 20 days after tumor implantation,which significantly compromised tumor ***,sustained expression of IRF4 is essential for maintaining cD8^(+)T cell immunity in the melanoma model,and these findings carry noteworthy implications for the advancement of more potent immunotherapies for solid tumors.
