Oral treatment with plecanatide or dolcanatide attenuates visceral hypersensitivity via activation of guanylate cyclase-c in rat models

AIM To investigate the effects of plecanatide and dolcanatide on maintenance of paracellular permeability, integrity of tight junctions and on suppression of visceral hypersensitivity. METHODS Transport of fluorescein isothiocyanate(FITc)-dextran was measured to assess permeability across cell monolayers and rat colon tissues. Effects of plecanatide and dolcanatide on the integrity of tight junctions in caco-2 and T84 monolayers and on the expression and localization of occludin and zonula occludens-1(ZO-1) were examined by immunofluorescence microscopy. Anti-nociceptive activity of these agonists was evaluated in trinitrobenzene sulfonic acid(TNBS)-induced inflammatory as well as in non-inflammatory partial restraint stress(PRS) rat models. Statistical significance between the treatment groups in the permeability studies were evaluated using unpaired *** Treatment of T84 and caco-2 monolayers with lipopolysaccharide(LPS) rapidly increased permeability, which was effectively suppressed when monolayers were also treated with plecanatide or dolcanatide. Similarly, when T84 and caco-2 monolayers were treated with LPS, cell surface localization of tight junction proteins occludin and ZO-1 was severely disrupted. When cell monolayers were treated with LPS in the presence of plecanatide or dolcanatide, occludin and ZO-1 were localized at the cell surface of adjoining cells, similar to that observed for vehicle treated cells. Treatment of cell monolayers with plecanatide or dolcanatide without LPS did not alter permeability, integrity of tight junctions and cell surface localization of either of the tight junction proteins. In rat visceral hypersensitivity models, both agonists suppressed the TNBS-induced increase in abdominal contractions in response to colorectal distension without affecting the colonic wall elasticity, and both agonists also reduced colonic hypersensitivity in the PRS model. cONcLUSION Our results suggest that activation of Gc-c signaling mi

Genomic change in hepatitis B virus associated with development of hepatocellular carcinoma

AIM: To determine the genomic changes in hepatitis B virus(HBV) and evaluate their role in the development of hepatocellular carcinoma(Hcc) in patients chronically infected with genotype c ***: Two hundred and forty chronic hepatitis B(cHB) patients were subjected and followed for a median of 105 mo. Hcc was diagnosed in accordance with AASLD guidelines. The whole X, S, basal core promoter(BcP), and precore regions of HBV were sequenced using the direct sequencing ***: All of the subjects were infected with genotype c HBV. Out of 240 cHB patients, 25(10%) had c1653 T and 33(14%) had T1753 V mutation in X region; 157(65%) had A1762T/G1764 A mutations in BcP region, 50(21%) had G1896 A mutation in precore region and 67(28%) had pre-S deletions. Hcc occurred in 6 patients(3%). The prevalence of T1753 V mutation was significantly higher in patients who developed Hcc than in those without Hcc. The cumulative occurrence rates of Hcc were 5% and 19% at 10 and 15 years, respectively, in patients with T1753 V mutant, which were significantly higher than 1% and 1% in those with wild type HBV(P cONcLUSION: The presence of T1753 V mutation in HBV X-gene significantly increases the risk of Hcc development in patients chronically infected with genotype c HBV.

Mineral chemistry and Geochemistry as Proxy for Petrogenetic Evaluation of charnockites: Evidences from Marandahalli, Salem Block of Southern India

The charnockites of Marandahalli, Dharmapuri District, Tamil Nadu India, form a part of the southern granulitic terrain in the Salem Block (SB). The charnockites are medium to coarse grained, inequigranular, with porphyroblastic and granophyric textures. They are composed of plagioclase feldspars (49 - 66 vol.%), K-feldspar (1 - 5 vol.%) and quartz (21 - 34 vol.%). The mafic minerals include pyroxene (2 - 7 vol.%) and biotite (1 - 12 vol.%). The accessory minerals include magnetite and apatite. Orthopyroxene compositions (MgO 16 - 22 wt.%) fall within the hypersthene series (En55.03Fs43.57Wo1.4) and yield peak metamorphic temperatures of 800˚c - 1000˚c. Biotites show K2O, MgO, FeO, TiO2, and Al2O3 concentrations consistent with magmatic formation, and Ti-in-biotite thermometry suggests temperatures of ~740˚c - 800˚c. Plagioclase compositions (An26-33) plot in the oligoclase field, while K-feldspar (Or93-99) plots in the orthoclase/microcline field. A ternary feldspar geothermometer indicates a retrograde temperature of 2 (65 - 68 wt.%), Na2O > K2O, high caO (3 - 5 wt.%), MgO (2.5 - 4 wt.%) and Fe2O3 (2 - 6 wt.%). The concentration of V, Rb and Ni, and LREE-rich and HREE-depleted patterns of the rocks indicate low degree of partial melting.

Ultrasound curriculum taught by first-year medical students： A four-year experience in Tanzania

BAcKGROUND: Diagnostic imaging is an integral aspect of care that is often insufficient, if not altogether absent, in rural and remote regions of low to middle income countries(LMIcs) such as Tanzania. The introduction of ultrasound can significantly impact treatment in these countries due to its portability, low cost, safety, and usefulness in various medical assessments. This study reviews the implementation of a four-week ultrasound course administered annually from 2013–2016 in a healthcare professional school in Mwanza, Tanzania by first-year allopathic US medical ***: Participants(n=582, over 4 years) were recruited from the Tandabui Institute of Health Sciences and Technology to take the ultrasound course. Subjects were predominantly clinical officer students, but other participants included other healthcare professional students, practicing healthcare professionals, and school employees. Data collected includes pre-course examination scores, post-course examination scores, course quiz scores, demographic surveys, and postcourse feedback surveys. Data was analyzed using two-tailed t-tests and the single factor analysis of variance(ANOVA).RESULTS: For all participants who completed both the pre-and post-course examinations(n=229, 39.1% of the total recruited), there was a significant mean improvement in their ultrasound knowledge of 42.5%, Pcal students can effectively teach a point of care ultrasound course to healthcare professional students within four weeks in Tanzania. Future investigation into the level of long-term knowledge retention, impact of ultrasound training on knowledge of human anatomy and diagnostic capabilities, and how expansion of an ultrasound curriculum has impacted access to care in rural Tanzania is warranted.

本文论述了用离散予估器补偿纯滞后的必要性。离散补偿器技术以广义的一阶和二阶脉冲传递函数为基础,它的设计方法也能方便地推广到高阶模型。所谓补偿器的设计,就是用比例调节器和负载予估器使静态偏差为零。采用与 Steiglitz-McBride 迭代前置波器模拟相结合的最小二乘回归,就是根据开环对象的输入—输出数据,去获得模型的脉冲传递函数系数和模型的纯滞后。一阶和二阶离散予估器两者都在 DEcPDP 15/35计算机实现的,而且也在小型的水温度控制系统进行了模拟试验。离散史密特予估器和标准离散 PI 调节器可以作为与它们比较的基础。

Bioactive proteins from pipefishes

Objective:To screen antimicrobial potence of some pipefish species collected from Tuticorin coastal ***:Antimicrobial activity of pipefishes in methanol extract was investigated against 10 bacterial and 10 fungal human pathogenic ***:Among the tested strains,in centriscus scutatus,pipefish showed maximum zone of inhibition against Vibrio cholerae(8 mm)and minimum in the sample of Hippichthys cyanospilos against Klebseilla pneumoniae(2 mm).In positive control,maximum zone of inhibition was recorded in Vibrio cholerae(9 mm)and minimum in Klebseilla pneumoniae,and Salmonella paratyphi(5 mm).chemical investigation indicated the presence of peptides as evidenced by ninhydrin positive spots on thin layer chromatography and presence of *** SDS PAGE,in centriscus scutatus,four bands were detected in the gel that represented the presence of proteins in the range nearly 25.8-75 *** Hippichthys cyanospilos,five bands were detected in the gel that represented the presence of proteins in the range nearly 20.5-78 *** result of FT-IR spectrum revealed that the pipe fishes extracts compriseed to have peptide derivatives as their predominant chemical ***:It can be conclude that this present investigation suggests the tested pipe fishes will be a potential source of natural bioactive compounds.

The crystal structure of spiro[2.2”]acenaphthene-1”-onespiro[3.3’]-5’-(2,3-dichlorophenylmethylidene)-1’-methylpipe-ridin-4’-one-4-(2,3-dichorophenyl) octahydroindolizine was elucidated by single crystal X ray diffraction. The title compound c37H30cl4N2O2, crystallizes in the orthorhombic system, space group P212121 with a = 8.4610(4) , b = 16.0926(6) , c= 23.8997(11) and Z = 4. The central piperidine ring adopts twisted conformation, the piperidine of octahydroindolizine ring is in chair conformation and the pyrrole ring is in slightly twisted envelope conformation. Details of the synthesis, NMR, crystal structure determination and intra- and inter- molecular interactions of the compound are given.

Absorption and emission modulation in a MoS2–GaN（0001） heterostructure by interface phonon–exciton coupling

Semiconductor heterostructures based on layered two-dimensional transition metal dichalcogenides(TMDs)interfaced to gallium nitride(Ga N)are excellent material systems to realize broadband light absorbers and emitters due to their close proximity in the lattice *** surface properties of a polar semiconductor such as Ga N are dominated by interface phonons,and thus the optical properties of the vertical heterostructure are influenced by the coupling of these carriers with *** activation of different Raman modes in the heterostructure caused by the coupling between interfacial phonons and optically generated carriers in a monolayer MoS_2–Ga N(0001)heterostructure is *** excitonic states in MoS_2 are close to the interband energy state of intraband defect state of Ga *** functional theory(DFT)calculations are performed to determine the band alignment of the interface and revealed a type-I *** close proximity of the energy levels and the excitonic states in the semiconductors and the coupling of the electronic states with phonons result in the modification of carrier relaxation *** of the excitonic absorption states in MoS_2 is measured by transient optical pump-probe spectroscopy and the change in emission properties of both semiconductors is measured by steady-state photoluminescence(PL)emission *** is significant red-shift of the c excitonic band and faster dephasing of carriers in MoS_***,optical excitation at energy higher than the bandgap of both semiconductors slows down the dephasing of carriers and energy exchange at the *** and blue-shifted PL emission is observed in MoS_*** N band-edge emission is reduced in intensity at room temperature due to increased phonon-induced scattering of carriers in the Ga N *** results demonstrate the relevance of interface coupling between the semiconductors for the development of optical and electronic applications.

The combination of kidney paired donation(KPD) with desensitization represents a promising method of increasing the rate of living donor kidney transplantation(LDKT) in immunologically challenging patients. Patients who are difficult to match and desensitize due to strong donor specific antibody are may be transplanted by a combination of desensitization and KPD protocol with more immunologically favorable donor. We present our experience of combination of desensitization protocol with three-way KPD which contributed to successful LDKT in highly sensitized end stage renal disease patient. All recipients were discharged with normal and stable allograft function at 24 mo follow up. We believe that this is first report from India where three-way KPD exchange was performed with the combination of KPD and desensitization. The combination of desensitization protocol with KPD improves access and outcomes of LDKT.
