Glioblastoma （GBM） is the most common and most aggressive primary brain tumor in adults. The existence of a small population of stem-like tumor cells that efficiently propagate tumors and resist cytotoxic therapy is one proposed mechanism leading to the resilient behavior of tumor cells and poor prognosis. In this study, we performed an in-depth analysis of the DNA methylation landscape in GBM- derived cancer stem ceils （GScs）. Parallel comparisons of primary tumors and GSc lines derived from these tumors with normal controls （a neural stem cell （NSc） line and normal brain tissue） identified groups of hyper- and hypomethylated genes that display a trend of either increasing or decreasing methylation levels in the order of controls, primary GBMs, and their counterpart GSc lines, respectively. Interestingly, concurrent promoter hypermethylation and gene body hypomethylation were observed in a subset of genes including MGMT, AJAP1 and PTPRN2. These unique DNA methylation signatures were also found in primary GBM-derived xenograft tumors indicating that they are not tissue culture-related epigenetic changes. Integration of GSc-specific epigenetic signatures with gene expression analysis further identified candidate tumor suppressor genes that are frequently down-regulated in GBMs such as SPINT2, NEFM and PENK. Forced re-expression of SPINT2 reduced glioma cell proliferative capacity, anchorage independent growth, cell motility, and tumor sphere formation in vitro. The results from this study demonstrate that GScs possess unique epigenetic signatures that may play important roles in the pathogenesis of GBM.

The present study aimed to purify and characterize one polygalacturonase from L. gongylophorus (PGaseLg), the symbiotic fungus of Atta sexdens. The enzyme was isolated by salting out of crude extract followed by two chromatographic steps. PGaseLG was identified with MS analysis and molecular exclusion chromatography revealed the monomeric nature of a protein with an estimated molecular weight of about 39 kDa. PGaseLg has an optimum temperature of 60°c and optimum ph activity at 5.0. Using polygalacturonate as a substrate, the calculations of KM, Vmax and kcat were 0.65 mg·mL-1, 1800 μmol·min-1·mg-1 and 35.97 s-1, respectively. The enzyme was stable for more than 3 h at 50°c at ph 5.0;otherwise, at lower or higher ph values, the PGaseLg was less stable. The influence of several metals, EDTA and β-mercaptoethanol on enzyme activity was also determined. Thin layer chromatography (TLc) analyses indicated that PGaseLg is an exopolygalacturonase.

Methods and Results:Response surface methodology(RSM) was applied to evaluate the effects of variables,namely the concentrations of wheat bran,soybean meal,KhPO,MnSO·hO and cuSO·5hO on alpha-galactosidase production in the solid substrate.A fractional factorial design(FFD) was firstly used to isolate the main factors that affected the production of alpha-galactosidase and the central composite experimental design(ccD) was then adopted to derive a statistical model for optimizing the composition of the fermentation *** experimental results showed that the optimum fermentation medium for alpha-galactosidase production by Aspergillus foetidus ZU-G1 was composed of 8-2137 g wheat bran,1·7843 g soybean meal,0·001 g MnSO·hO and 0·001 g cuSO·5hO in 10 g dry matter fermentation medium. conclusions:After incubating 96 h in the optimum fermentation medium, alpha-galactosidase activity was predicted to be 2210·76 U g dry matter in 250 ml shake *** the present study,alpha-galactosidase activity reached 2207·19 U g dry matter. Significance and Impact of the Study:Optimization of the solid substrate was a very important measure to increase enzyme activity and realize industrial production of *** process of alpha-galactosidase production in laboratory scale may have the potential to scale-up.

Based on the monitoring of the main water quality parameters in the inverted A2 /O process employed in The No.4 Wastewater Treatment Plant of Xi’an city,this paper mainly analyzed contaminant removals during over half a year *** results demonstrated that the total removal efficiencies of cODcr,TN,Nh3-N,TP and SS could reach to 90.5%,65.2%,82.0%,54.8% and 95.8%,*** addition,the treatment process showed a strong resistance for loading impact,resulting in relatively stable effluent ***,a lower removal efficiency of TP in the bioreactor and sludge floating in the secondary settling tank were *** deep analyzing,some countermeasures were ***,it is expected that the conclusions derived from this study may be valuable for design and operation management of the similar Wastewater Treatment Plants in the future.

目的:评估食管闭锁合并(或不合并)气管食管瘘(EA/TEF)患儿的长期(不局限于儿童时期)并发症和临床结局。本研究旨在通过临床症状、诊断试验和治疗干预评估一家三甲医院EA/TEF患儿吞咽和呼吸系统并发症。方法:纳入2011-2014年间先天性EA/TEF患儿,收集这些患儿的人口统计特征、疾病分类和分型、临床症状、影像学、内镜下及支气管镜下发现及其治疗情况。结果:共计43例患儿纳入研究,其中以白人居多,中位年龄8岁(四分位间距320岁)。20例(62.5%,20/32)为c型异常(EA合并远端TEF)。21例(48.8%)有烧心症状,19例(44.1%)有反酸症状,31例(72.1%)吞咽固体食物困难。26例患儿接受钡餐检查,其中17例(65.4%)狭窄,20例(76.9%)动力不足,4例(15.4%)出现复发瘘。30例患儿接受上消化道内镜检查,其中21例(70.0%)发现狭窄,6例(20.0%)出现需外科干预的复发瘘。8例患儿行胃底折叠术。呼吸功能评估显示31例(72.1%)患儿有咳嗽和气梗,32例有呼吸困难和哮喘,19例(44.2%)患儿有反复的呼吸系统感染。其他发现包括气管软化(86.7%)和限制性肺病(54.5%)。结论:EA/TEF患儿残留的食管和气管具有较高的病理风险,需要长期随访来监测临床症状和治疗反应。
