FtsH proteases are membrane-embedded proteolytic complexes important for protein quality control and regulation of various physiological processes in bacteria,mitochondria,and *** most cyanobacteria,the model species Synechocystis *** 6803 contains four FtsH homologs,FtsH1–***1–FtsH3 form two hetero-oligomeric complexes,FtsH1/3 and FtsH2/3,which play a pivotal role in acclimation to nutrient deficiency and photosystem Ⅱ quality control,***4 differs from the other three homologs by the formation of a homo-oligomeric complex,and together with Arabidopsis thaliana AtFtsH7/9 orthologs,it has been assigned to another phylogenetic group of unknown *** results exclude the possibility that Synechocystis FtsH4 structurally or functionally substitutes for the missing or non-functional FtsH2 subunit in the FtsH2/3 ***,we demonstrate that FtsH4 is involved in the biogenesis of photosystem Ⅱ by dual regulation of high light-inducible proteins(Hlips).FtsH4 positively regulates expression of Hlips shortly after high light exposure but is also responsible for Hlip removal under conditions when their elevated levels are no longer *** provide experimental support for Hlips as proteolytic substrates of *** labeling of FtsH4 enabled us to assess its localization using advanced microscopic *** show that FtsH4 complexes are concentrated in well-defined membrane regions at the inner and outer periphery of the thylakoid *** on the identification of proteins that co-purified with the tagged FtsH4,we speculate that FtsH4 concentrates in special compartments in which the biogenesis of photosynthetic complexes takes place.

There is a strong drive towards hyperresolution earth system models in order to resolve finer scales of motion in the *** problem of obtaining more realistic representation of terrestrial fluxes of heat and water,however,is not just a problem of moving to hyperresolution grid *** is much more a question of a lack of knowledge about the parameterisation of processes at whatever grid scale is being used for a wider modelling *** grid scales cannot alone solve the problem of this hyperresolution *** paper discusses these issues in more detail with specific reference to land surface parameterisations and flood inundation *** importance of making local hyperresolution model predictions available for evaluation by local stakeholders is *** is expected that this will be a major driving force for improving model performance in the future.

SFRP4 expression correlates with epithelial mesenchymal transitionlinked genes and poor overall survival in colon cancer patients

BAcKGROUND colon cancer is among the most commonly diagnosed cancers in the United States with an estimated 97220 new cases expected by the end of *** affects 1.2 million people around the world and is responsible for about 0.6 million deaths every *** decline in overall incidence and mortality over the past 30 years,there continues to be an alarming rise in early-onset colon cancer cases(creted frizzled-related protein 4(SFRP4),is over-expressed in early-onset colon ***4 is speculated to play an essential role in cancer by inhibiting the epithelial mesenchymal transition(EMT).AIM To investigate the correlation between SFRP4 expression and EMT-linked genes in colon cancer and how it affects patient *** SFRP4 expression relative to that of EMT-linked genes and survival analysis were performed using the University of california Santa cruz cancer Browser *** SFRP4 was found to be co-expressed with the EMT-linked markers cDH2,FN1,VIM,TWIST1,TWIST2,SNAI1,SNAI2,ZEB1,ZEB2,POSTN,MMP2,MMP7,MMP9,and ***4 expression negatively correlated with the EMTlinked suppressors cLDN4,cLDN7,TJP3,MUc1,and *** expression of SFRP4 and the EMT-linked markers was higher in mesenchymal-like samples compared to epithelial-like samples which potentially implicates SFRP4-EMT mechanism in colon ***,patients overexpressing SFRP4 presented with poor overall survival(P=0.0293).cONcLUSION considering the implication of SFRP4 in early-onset colon cancer,particularly in the context of EMT,tumor metastasis,and invasion,and the effect of increased expression on colon cancer patient survival,SFRP4 might be a potential biomarker for early-onset colon cancer that could be targeted for diagnosis and/or disease therapy.

Erectile dysfunction （ED） commonly results from endothelial dysfunction of the systemic vasculature. Although phosphodiesterase type 5 （PDE-5） inhibitors are effective at treating most cases of ED, they must be taken routinely and are ineffectual for a meaningful number of men. In recent years gene and stem cell-based therapies targeted at the penile endothelium have been gaining momentum in preclinical studies. These early studies reveal that gene and stem cell-based therapies may be both enduring and efficacious, and may eventually lead to a cure for ED. The following review will highlight our current understanding of endothelial-specific gene and stem cell-based therapies performed to date in a number of experimental animal models.

Rural implementation of the perioperative surgical home:A case-control study

BAcKGROUND Perioperative surgical home(PSH)is a novel patient-centric surgical system developed by American Society of Anesthesiologist to improve outcomes and patient *** has proven success in large urban health centers by reducing surgery cancellation,operating room time,length of stay(LOS),and readmission ***,only limited studies have assessed the impact of PSH on surgical outcomes in rural *** To evaluate the newly implemented PSH system at a community hospital by comparing the surgical outcomes using a longitudinal case-control *** The research study was conducted at an 83-bed,licensed level-III trauma rural community hospital.A total of 3096 TJR procedures were collected retrospectively between January 2016 and December 2021 and were categorized as PSH and non-PSH cohorts(n=2305).To evaluate the importance of PSH in the rural surgical system,a case-control study was performed to compare TJR surgical outcomes(LOS,discharge disposition,and 90-d readmission)of the PSH cohort against two control cohorts[control-1 PSH(c1-PSH)(n=1413)and control-2 PSH(c2-PSH)(n=892)].Statistical tests including chi-square test or Fischer’s exact test were performed for categorical variables and Mann-Whitney test or Student’s t-test were performed for continuous *** general linear models(Poisson regression and binomial logistic regression)were performed to fit adjusted *** The LOS was significantly shorter in PSH cohort compared to two control cohorts(median PSH=34 h,c1-PSH=53 h,c2-PSH=35 h)(P valuecohort had lower percentages of discharges to other facilities(PSH=3.5%,c1-PSH=15.5%,c2-PSH=6.7%)(P valuecal difference observed in 90-d readmission between control and PSH ***,the PSH implementation reduced the 90-d readmission percentage(PSH=4.7%,c1-PSH=6.1%,c2-PSH=3.6%)lower than the national average 30-d readmission percentage which is 5.5%.The PSH system was effecti

The onset of alcoholic liver disease (ALD) is initiated by different cell types in the liver and a number of different factors including: products derived from ethanol-induced inflammation, ethanol metabolites, and the indirect reactions from those metabolites. Ethanol oxidation results in the production of metabolites that have been shown to bind and form protein adducts, and to increase inflammatory, fibrotic and cirrhotic responses. Lipopolysaccharide (LPS) has many deleterious effects and plays a significant role in a number of disease processes by increasing inflammatory cytokine release. In ALD, LPS is thought to be derived from a breakdown in the intestinal wall enabling LPS from resident gut bacterial cell walls to leak into the blood stream. The ability of adducts and LPS to independently stimulate the various cells of the liver provides for a two-hit mechanism by which various biological responses are induced and result in liver injury. Therefore, the purpose of this article is to evaluate the effects of a two-hit combination of ethanol metabolites and LPS on the cells of the liver to increase inflamma-tion and fi brosis, and play a role in the development and/or progression of ALD.
