Despite improvements in adjuvant therapies for gastric cancer in recent years, the disease is characterized by high recurrence rates and a dismal prognosis. The major improvement in the treatment of recurrent or metastatic gastric cancer in recent years has been the incorporation of trastuzumab, a monoclonal antibody that inhibits human epidermal growth factor receptor 2(HER2) heterodimerization, after the demonstrated predictive value of the overexpression and/or amplification of this receptor. Beyond HER2, other genetic abnormalities have been identified, and these mutations may be targetable by tyrosine kinase inhibitors or monoclonal antibodies. The demonstration of four distinct molecular subtypes of gastric cancer by the cancer Genome Atlas study highlight the enormous heterogeneity of the disease and its complex interplay between genetic and epigenetic alterations and provide a roadmap to implement genome-guided personalized therapy in gastric cancer. In the present review, we aim to discuss, from a clinical point of view, the genomic landscape of gastric cancer described in recent studies, the therapeutic insights derived from these findings, and the clinical trials that have been conducted and those in progress that take into account tailored therapies for gastric cancer.

Tailoring the partitioning of proteins using ionic liquids as adjuvants in polymer-polymer aqueous biphasic systems

Aqueous biphasic systems(ABS)are promising for proteins purification;however,when dealing with samples comprising several proteins,the selectivity towards a target protein is difficult to achieve.In this work,the addition of ionic liquids(ILs)as adjuvants(5 wt%and 10 wt%)in ABS composed of polyethylene glycol(PEG)and dextran was investigated to tailor proteins partitioning between the coexisting phases.The liquid-liquid phase diagrams were determined at 298 K,followed by partition studies of three proteins(bovine serum albumin(BSA),immunoglobulin G(IgG),and cytochrome c(cyt c)).Partition coefficients of IgG and BSA indicate the preference of both proteins to the dextran-rich phase,whereas cyt c has no preferential partitioning between the phases.The addition of chloride-based ILs as adjuvants allows to increase or decrease the partition coefficients,thus tailoring the proteins partitioning between the phases.BSA partitioning essentially depends on the IL content in each phase,whereas cyt c and IgG partitioning is ruled by the ILs chemical structure and established interactions.molecular docking was carried out to address the ILs effect on the proteins partitioning,supporting experimental observations,while identifying the specific interactions occurring.The partition of each protein in polymer-salt ABS with ILs as adjuvants was determined,demonstrating the higher tailoring ability of polymer-polymer ABS when adding ILs.Finally,the partition of each protein in presence of the remaining ones was determined,as well as the selectivity of the studied systems to separate each pair of proteins,paving the way for their use in liquidliquid chromatography.

currently,in the era of big data and 5G communication technology,electromigration has become a serious reliability issue for the miniaturized solder joints used in microelectronic devices.Since the effective charge number(Z*)is considered as the driving force for electromigration,the lack of accurate experimental values for Z* poses severe challenges for the simulation-aided design of electronic materials.In this work,a data-driven framework is developed to predict the Z* values of cu and Sn species at the anode based LIQUID,cu_(6)Sn_(5) intermetallic compound(Imc)and Fcc phases for the binary cu-Sn system undergoing electromigration at 523.15 K.The growth rate constants(kem)of the anode Imc at several magnitudes of applied low current density(j=1×10^6 to 10×10^6A/m^2)are extracted from simulations based on a 1D multi-phase field model.A neural network employing Z* and j as input features,whereas utilizing these computed kemdata as the expected output is trained.The results of the neural network analysis are optimized with experimental growth rate constants to estimate the effective charge numbers.For a negligible increase in temperature at low j values,effective charge numbers of all phases are found to increase with current density and the increase is much more pronounced for the Imc phase.The predicted values of effective charge numbers Z* are then utilized in a 2D simulation to observe the anode Imc grain growth and electrical resistance changes in the multi-phase system.As the work consists of the aspects of experiments,theory,computation,and machine learning,it can be called the four paradigms approach for the study of electromigration in Pb-free solder.Such a combination of multiple paradigms of materials design can be problem-solving for any future research scenario that is marked by uncertainties regarding the determination of material properties.

multicomponent alloys show intricate microstructure evolution,providing materials engineers with a nearly inexhaustible variety of solutions to enhance material properties.multicomponent microstructure evolution simulations are indispensable to exploit these opportunities.These simulations,however,require the handling of high-dimensional and prohibitively large data sets of thermodynamic quantities,of which the size grows exponentially with the number of elements in the alloy,making it virtually impossible to handle the effects of four or more elements.In this paper,we introduce the use of tensor completion for highdimensional data sets in materials science as a general and elegant solution to this problem.We show that we can obtain an accurate representation of the composition dependence of high-dimensional thermodynamic quantities,and that the decomposed tensor representation can be evaluated very efficiently in microstructure simulations.This realization enables true multicomponent thermodynamic and microstructure modeling for alloy design.

BAcKGROUND While endoscopic ultrasound(EUS)-guided fine needle aspiration(FNA)is considered a preferred technique for tissue sampling for solid lesions,fine needle biopsy(FNB)has recently been developed.AIm To compare the accuracy of FNB vs FNA in determining the diagnosis of solid lesions.mETHODS A retrospective,multi-center study of EUS-guided tissue sampling using FNA vs FNB needles.measured outcomes included diagnostic test characteristics(i.e.,sensitivity,specificity,accuracy),use of rapid on-site evaluation(ROSE),and adverse events.Subgroup analyses were performed by type of lesion and diagnostic yield with or without ROSE.A multivariable logistic regression was also performed.RESULTS A total of 1168 patients with solid lesions(n=468 FNA;n=700 FNB)underwent EUS-guided sampling.mean age was 65.02±12.13 years.Overall,sensitivity,specificity and accuracy were superior for FNB vs FNA(84.70%vs 74.53%;99.29%vs 96.62%;and 87.62%vs 81.55%,respectively;Pcificity,and accuracy of FNB alone were similar to FNA+ROSE[(81.66%vs 86.45%;P=0.142),(100%vs 100%;P=1.00)and(88.40%vs 85.43%;P=0.320].There were no difference in diagnostic yield of FNB alone vs FNB+ROSE(P>0.05).multivariate analysis showed no significant predictor for better accuracy.On subgroup analyses,FNB was superior to FNA for non-pancreatic lesions;however,there was no difference between the techniques among pancreatic lesions.One adverse event was reported in each group.cONcLUSION FNB is superior to FNA with equivalent diagnostic test characteristics compared to FNA+ROSE in the diagnosis of non-pancreatic solid lesions.Our results suggest that EUS-FNB may eliminate the need of ROSE and should be employed as a first-line method in the diagnosis of solid lesions.

We consider nested or multiscale models to study the effect of the temporal evolution of the disease within the host in the population dynamics of the disease,for one and two infectious agents.We assumed a coupling between the within-host infection rate and the between-host transmission rate.The age of infection within each individual in a population affects the probability of transmission of the disease to a susceptible host and this will affect the temporal evolution of the disease in the host population.To analyze the infection within the host,we consider bacterial-like and viral-like infections.In the model for two infectious agents,we found that,when strain 2 has a basic reproduction number R02 greater than the basic reproduction number R01 of strain 1,strain 2 replaces strain 1 in the population.However,if R02>R01 but the values are closer,the replacement does not occur immediately and both strains can coexist for a long time.We applied the model to a scenario in which patients infected with the hepatitis c virus(HcV)are cleared of HcV when super-infected with the hepatitis A virus(HAV).We compared the time for the replacement of HcV by HAV in the population considering instantaneous and noninstantaneous replacement within the individuals.The model developed can be generalized for more than two infectious agents.

BAcKGROUND The literature indicates that the enteric nervous system is affected in inflammatory bowel diseases(IBDs)and that the P2X7 receptor triggers neuronal death.However,the mechanism by which enteric neurons are lost in IBDs is unknown.AIm To study the role of the caspase-3 and nuclear factor kappa B(NF-κB)pathways in myenteric neurons in a P2X7 receptor knockout(KO)mouse model of IBDs.mETHODS Forty male wild-type(WT)c57BL/6 and P2X7 receptor KO mice were euthanized 24 h or 4 d after colitis induction by 2,4,6-trinitrobenzene sulfonic acid(colitis group).mice in the sham groups were injected with vehicle.The mice were divided into eight groups(n=5):The WT sham 24 h and 4 d groups,the WT colitis 24 h and 4 d groups,the KO sham 24 h and 4 d groups,and the KO colitis 24 h and 4 d groups.The disease activity index(DAI)was analyzed,the distal colon was collected for immunohistochemistry analyses,and immunofluorescence was performed to identify neurons immunoreactive(ir)for calretinin,P2X7 receptor,cleaved caspase-3,total caspase-3,phospho-NF-κB,and total NF-κB.We analyzed the number of calretinin-ir and P2X7 receptor-ir neurons per ganglion,the neuronal profile area(μm^(2)),and corrected total cell fluorescence(cTcF).RESULTS cells double labeled for calretinin and P2X7 receptor,cleaved caspase-3,total caspase-3,phospho-NF-κB,or total NF-κB were observed in the WT colitis 24 h and 4 d groups.The number of calretinin-ir neurons per ganglion was decreased in the WT colitis 24 h and 4 d groups compared to the WT sham 24 h and 4 d groups,respectively(2.10±0.13 vs 3.33±0.17,Pcantly different between the KO groups.The calretinin-ir neuronal profile area was increased in the WT colitis 24 h group compared to the WT sham 24 h group(312.60±7.85 vs 278.41±6.65,Pclear profile area was decreased in the WT colitis 4 d group compared to the WT sham 4 d group(104.63±2.49 vs 117.41±1.14,Pmber of P2X7 receptor-ir neurons per ganglion was decrea

Objective: To evaluate the trypanocidal, leishmanicidal and cytotoxic activity of Eugenia jambolana(E. jambolana) and Eugenia uniflora(E. uniflora) extracts and fractions.methods: The products were characterized by Lc–mS. Antiparasitic assays were performed and cytotoxicity was evaluated in fibroblastos. In vitro assays were performed using spectrophotometric evaluation. All assays were performed in thrice.Results: The results showed that the extracts and the tannic fraction from E. jambolana inhibited 100% of the epimastigote lines. The ethanolic extract was the most efficient in all concentrations tested against the three parasite strains. In the cytotoxicity assay the flavonoid fraction showed low toxicity. All E. uniflora samples showed cytotoxicity at the highest concentration tested, but the extract showed no toxic effect on the fibroblasts at the lowest concentration. The flavonoid and tannic fractions were more efficient against Leishmania braziliensis promastigotes compared to the extract. However, the extracts and the tannic fraction were more effective against Leishmania infantum strains. The effect on epimastigote cells was observed at all concentrations tested, with all E. uniflora samples. However, the samples were more effective at the highest concentration, where there was inhibition in 100% of the Trypanosoma cruzi strains.conclusions: The species E. jambolana and E. uniflora presented antiparasitic activity against all tested parasite strains, indicating that these species can serve as an alternative therapy as they were efficient in the tests performed. The E. uniflora extract and the E. jambolana flavonoid fraction presented a low cytotoxicity, opening the floor for new biological studies.
