A whole-systems approach for assessing measures to improve arable ecosystem sustainability

There is increasing pressure on the agricultural industry to maintain or increase production of high-quality food while maintaining long-term environmental *** and EU policies and practices have been developed and implemented in an attempt to improve the sustainability and efficiency of arable farming and satisfy these potentially conflicting ***,to the authors’knowledge,there has been no attempt to quantify whether new interventions have the desired effect on improving sustainability at a whole-systems *** studies focus on one,or a few,elements of a specific system and therefore fail to account for trade-offs and conflicts between the many different interacting ***,we propose a whole-systems approach based on a suite of indicators for a complete and holistic assessment of the efficacy of policies to improve economic,environmental,and ecological sustainability.

The endoplasmic reticulum: A dynamic and well-connected organelle

The endoplasmic reticulum forms the first compart- ment in a series of organelles which comprise the secretory pathway, it takes the form of an extremely dynamic and pleomorphic membrane-bounded network of tubules and cisternae which have numerous different cellular functions. In this review, we discuss the nature of endoplasmic reticulum structure and dynamics, its relationship with closely associated organelles, and its possible function as a highway for the distribution and delivery of a diverse range of structures from metabolic complexes to viral particles.

Association between calcium sensing receptor gene polymorphisms and chronic pancreatitis in a US population:Role of serine protease inhibitor Kazal 1type and alcohol

AIM： To test the hypothesis that calcium sensing receptor （cASR） polymorphisms are associated with chronic pancreatitis （cP）, and to determine whether serine protease inhibitor Kazal 1type （SPINK1） N34S or alcohol are necessary co-factors in its etiology. METHODS： Initially, 115 subjects with pancreatitis and 66 controls were evaluated, of whom 57 patients and 21 controls were predetermined to carry the high-risk SPINK1 N34S polymorphism. We sequenced cASR gene exons 2, 3, 4, 5 and 7, areas containing the majority of reported polymorphisms and novel mutations. Based on the initial results, we added 223 patients and 239 controls to analyze three common nonsynonymous single nucleotide polymorphisms （SNPs） in exon 7 （A986S, R990G, and Q1011E）. RESULTS： The cASR exon 7 R990G polyrnorphism was significantly associated with cP （OR, 2.01; 95% cI, 1.12-3.59; P = 0.015）. The association between cASR R990G and cP was stronger in subjects who reported moderate or heavy alcohol consumption （OR, 3.12; 95% cI, 1.14-9.13; P = 0.018）. There was no association between the various cASR genotypes and SPINK1 N34S in pancreatitis. None of the novel cASR polymorphisms reported from Germany and India was detected. cONcLUSION： Our United States-based study confirmed an association of cASR and cP and for the first time demonstrated that cASR R990G is a significant risk factor for cP. We also conclude that the risk of cP with cASR R990G is increased in subjects with moderate to heavy alcohol consumption.

Involvement of Lipid Rafts and cellular Actin in AcMNPV GP64 Distribution and Virus Budding

GP64 is the major envelope glycoprotein associated with the budded virus (BV) of Autographa californica nucleopolyhedrovirus (AcMNPV) and is essential for attachment and budding of BV particles. confocal microscopy and flotation assays established the presence of lipid raft domains within the plasma membranes of AcMNPV-infected Sf9 cells and suggested the association of GP64 with lipid rafts during infection. GP64 and filamentous actin (F-actin) were found to co-localise at the cell cortex at 24 and 48 hpi and an additional restructuring of F-actin during infection was visualised, resulting in a strongly polarised distribution of both F-actin and GP64 at the cell cortex. Depletion of F-actin, achieved by treatment of Sf9 cells with latrunculin B (LB), resulted in the redistribution of GP64 with significant cytoplasmic aggregation and reduced presence at the plasma membrane. Treatment with LB also resulted in reduced production of BV in Sf9 cells. Analysis of virus gene transcription confirmed this reduction was not due to decreased trafficking of nucleocapsids to the nucleus or to decreased production of infectious progeny nucleocapsids. Reduced BV production due to a lack of GP64 at the plasma membrane of AcMNPV-infected Sf9 cells treated with LB, suggests a key role for F-actin in the egress of BV.

The multiplexed,point-of-care measurement of specific antibodies could improve the speed with which diseases are diagnosed and their treatment *** this end,we are developing E-DNA scaffold sensors,which consist of a rigid,nucleic acid“scaffold”attached on one end to an electrode and presenting both a redox reporter and an epitope on the *** the absence of antibody,the reporter efficiently transfers electrons when interrogated ***-induced steric hindrance limits movement,reducing electron transfer in a manner that is both easily measured and quantitatively related to target *** we have used monoclonal antibodies to explore the analytical performance of E-DNA sensors,showing that they support the rapid,single-step,quantitative detection of multiple antibodies in small volume ***,in contrast,we employ authentic human samples to better explore the platform’s clinical ***,we developed E-DNA sensors targeting three HIV-specific antibodies and then compared the analytical and clinical performance of these against those of gold standard serological *** so we find that,although the multistep amplification of an ELISA leads to a lower detection limits,the clinical sensitivity of ELISAs,E-DNA sensors and lateral-flow dipsticks are indistinguishable across our test *** thus appears that,by merging the quantitation and multiplexing of ELISAs with the convenience and speed of dipsticks,E-DNA scaffold sensors could significantly improve on current serological practice.
